Identification of a novel synonymous mutation in the human β -Ureidopropionase Gene UPB1 affecting pre-mRNA splicing.

β-Ureidopropionase is the third enzyme of the pyrimidine degradation pathway and it catalyzes the conversion of N-carbamyl-β-alanine and N-carbamyl-β-aminoisobutyric acid to β-alanine and β-aminoisobutyric acid, respectively, and ammonia and CO2. To date, only 16 genetically confirmed patients with a complete ß-ureidopropionase deficiency have been reported. Here, we report the clinical, biochemical, and molecular analysis of a newly identified patient with β-ureidopropionase deficiency. Mutation analysis of the UPB1 gene showed that the patient was compound heterozygous for a novel synonymous

Multiplex analysis of sphingolipids using amine-reactive tags (iTRAQ).

Ceramides play a crucial role in divergent signaling events, including differentiation, senescence, proliferation, and apoptosis. Ceramides are a minor lipid component in terms of content; thus, highly sensitive detection is required for accurate quantification. The recently developed isobaric tags for relative and absolute quantitation (iTRAQ) method enables a precise comparison of both protein and aminophospholipids. However, iTRAQ tagging had not been applied to the determination of sphingolipids. Here we report a method for the simultaneous measurement of multiple ceramide and monohexosylc

Brucella abortus choloylglycine hydrolase affects cell envelope composition and host cell internalization.

Choloylglycine hydrolase (CGH, E.C. 3.5.1.24) is a conjugated bile salt hydrolase that catalyses the hydrolysis of the amide bond in conjugated bile acids. Bile salt hydrolases are expressed by gastrointestinal bacteria, and they presumably decrease the toxicity of host's conjugated bile salts. Brucella species are the causative agents of brucellosis, a disease affecting livestock and humans. CGH confers Brucella the ability to deconjugate and resist the antimicrobial action of bile salts, contributing to the establishment of a successful infection through the oral route in mice. Additionally,

Sequential treatment with SN-38 followed by 5-fluorouracil shows synergistic cytotoxic activity in small cell lung cancer cells.

Despite the high response rates of small cell lung cancer (SCLC) to first-line cisplatin-based chemotherapies, most patients with SCLC will eventually experience disease progression. Accordingly, novel chemotherapeutic regimens are desired. This in vitro study was carried out in order to develop novel chemotherapeutic regimens containing 5-fluorouracil (5-FU) or oral fluoropyrimidine for SCLC. 5-FU was combined with other standard drugs for SCLC (cisplatin, etoposide, an active metabolite of irinotecan and amrubicin) in different schedules. The combination effects were analyzed by a 3-(4,5-dim

Severe deficiency of the fatty acid amide hydrolase (FAAH) activity segregates with the Huntington's disease mutation in peripheral lymphocytes.

The search for peripheral markers of neurodegenerative diseases aims at identifying molecules that could help in monitoring the effects of future therapeutics in easily accessible cells. Here we focused on the involvement of the endocannabinoid system in Huntington's disease (HD). We assayed peripheral lymphocytes from HD patients and healthy controls, and found that the activity of the fatty acid amide hydrolase (FAAH), the enzyme that degrades the endocannabinoid anandamide (AEA), was dramatically decreased (down to less than 10%) in HD compared to healthy subjects. Concomitantly, the endoge

Inhibition of chlamydial infection in the genital tract of female mice by topical application of a peptide deformylase inhibitor.

Chlamydia trachomatis is an obligate intracellular bacterium responsible for a number of health problems, including sexually transmitted infection in humans. We recently discovered that C. trachomatis infection in cell culture is highly susceptible to inhibitors of peptide deformylase, an enzyme that removes the N-formyl group from newly synthesized polypeptides. In this study, one of the deformylase inhibitors, GM6001, was tested for potential antichlamydial activity using a murine genital C. muridarum infection model. Topical application of GM6001 significantly reduced C. muridarum loading i

Effects of liquid storage on amidase activity, DNA fragmentation and motility of turkey spermatozoa.

Short-term liquid storage of turkey semen is of great interest in the management of turkey reproduction due to the extensive use of artificial insemination. This study examined changes in DNA fragmentation (using a comet assay), sperm motility characteristics (using computer-aided sperm analysis), and amidase activity (using a colorimetric assay) of turkey sperm stored for 24 and 48 h. In addition we found that turkey spermatozoa contain besides acrosin, additional two serine proteinases of molecular weight of 34 and 42 kDa. We found that, after 48 h of liquid storage, decreases in sperm motil

A biotinidase Km variant causing late onset bilateral optic neuropathy.

A patient with a newly recognised variant of biotinidase deficiency presented with acute loss of vision at the age of 10 years. Progressive bilateral optic neuropathy, spastic paraparesis, and a predominantly motor type neuropathy developed over the next five years. Metabolic investigations revealed biotin depletion causing multiple carboxylase deficiency. The basic defect was a biotin recycling disorder due to a mutant biotinidase with residual activity of 4.4% assayed routinely. Biocytin excretion in urine was only slightly increased. Further investigations on plasma biotinidase revealed bip

Biotinidase in the porcine cerebrum.

Biotinidase activities found in porcine brains (n = 3) were as follows: cerebrum, 4.4 +/- 0.2 pmol/min per milligram of protein; cerebellum, 7.6 +/- 0.3 pmol/min per milligram of protein; medulla, 2.9 +/- 0.3 pmol/min per milligram of protein. These values are relatively high compared with the activities in rat or guinea pig brains. Subcellular distribution of biotinidase was found mainly in the soluble cytoplasmic fraction (S3), i.e., in the supernatant of 0.32 M sucrose S2 solution after ultracentrifugation at 105,000g for 90 min. This is in contrast to the guinea pig livers, in which the su

Acetaminophen nephrotoxicity in the rat: quantitation of renal metabolic activation in vivo.

Renal cortical necrosis induced by acetaminophen (APAP) may be related to generation of reactive intermediates by two mechanisms of metabolic activation, direct P-450 dependent metabolic activation (P-450) or metabolic activation subsequent to deacetylation of APAP to p-aminophenol (PAP). Generation of arylating intermediates by both pathways of metabolic activation was quantified in cyclohexamide (HEX)-pretreated or naive rats in vivo with specifically labeled [14C]APAP. The association of each type of metabolic activation with APAP-induced nephrotoxicity was determined in Fischer 344 (F344)

Lysosomal involvement in cellular turnover of plasma membrane sphingomyelin.

At least two isoenzymes of sphingomyelinase (sphingomyelin cholinephosphohydrolase, EC 3.1.4.12), including lysosomal acid sphingomyelinase and nonlysosomal magnesium-dependent neutral sphingomyelinase, catalyse the degradation of sphingomyelin in cultured human skin fibroblasts. A genetically determined disorder of sphingomyelin metabolism, type A Niemann-Pick disease, is characterized by a deficiency of lysosomal acid sphingomyelinase. To investigate the involvement of lysosomes in the degradation of cellular membrane sphingomyelin, we have undertaken studies to compare the turnover of plasm

Corilagin alleviates Staphylococcus aureus pathogenicity by interacting with amidase and α-hemolysin.

Staphylococcus aureus amidase cleaves the peptidoglycan that connect parent and offspring to promote bacterial proliferation, and it also affects the formation of bacterial biofilm. The critical roles of α-hemolysin (Hla) in the pathogenicity of S. aureus are indispensable. Compounds that inhibit the functions of these two proteins can provide a new strategy and a promising agent for the development of S. aureus infection inhibitors. In this study, it was found that corilagin bound to the active pocket of amidase and interacted with His370, Glu324, His382 and Ala288, the binding inhibited the

Anticonvulsant Effects of Synthetic N-(3-Methoxybenzyl)oleamide and N-(3-Methoxybenzyl)linoleamide Macamides: An In Silico and In Vivo Study.

Epilepsy is a chronic neurological disorder that affects nearly 50 million people worldwide. Experimental evidence suggests that epileptic neurons are linked to the endocannabinoid system and that inhibition of the FAAH enzyme could have neuroprotective effects by increasing the levels of endogenous endocannabinoid anandamide. In this context, the use of macamides as therapeutic agents in neurological diseases has increased in recent years. With a similar structure to anandamide, several theories point to the FAAH-macamide interaction as a possible cause of FAAH enzymatic inhibition. In this w

Identification of novel zinc-binding inhibitors against key microbial metallohydrolase DapE in Klebsiella pneumoniae: an integrated ligand-based virtual screening, molecular docking, molecular dynamics, and MM/PBSA approach.

Klebsiella pneumoniae (K. pneumoniae) has emerged as a prominent multidrug-resistant pathogen in healthcare settings and is ranked among the top three critical priority pathogens by the World Health Organization. Owing to the surge in antibiotic resistance and resulting treatment failures, there is an urgent need for alternative therapeutic approaches. N-succinyl-L, L-diaminopimelic acid desuccinylase (DapE), a crucial metalloenzyme in the lysine biosynthesis pathway in K. pneumoniae, is essential for protein synthesis and the cross-linking of the bacterial peptidoglycan cell wall. Comprehensive evaluation of the MeltPro MTB/PZA assay for prediction of pyrazinamide resistance in multidrug-resistant tuberculosis.

UNLABELLED: Resistance to pyrazinamide (PZA) poses significant challenges to tuberculosis (TB) management, and prediction of susceptibility to PZA has been challenging. This study examined PZA resistance-associated gene mutations in 125 multidrug-resistant clinical isolates of Mycobacterium tuberculosis (MTB) from Wenzhou, China. Phenotypic drug-susceptibility testing (pDST) for PZA was performed on clinical isolates using the MGIT 960 system to accurately determine resistance. Subsequently, whole-genome sequencing (WGS) was conducted on the 125 isolates to identify genetic mutations linked to

Vanin-2 is expressed in peripheral blood T cells and upregulated in patients with systemic lupus erythematosus.

Members of the vanin gene family include VNN1, VNN2, and VNN3 in humans. Although the functions of vanins have been widely examined in myeloid cells, their expression and functions have not been clarified in T lymphocytes. This study aimed to elucidate the significance of Vanin-2 (VNN2) on human peripheral blood T lymphocytes and study its expression in systemic lupus erythematosus (SLE). The differential expression of Vanins was analyzed by bioinformatics. VNN2 expressions in peripheral blood T-cell subsets were analyzed by single-cell RNA sequencing data and flow cytometry. Impact of Stress on the Endocannabinoid System: A [11C]-CURB Positron Emission Tomography Study in Early Psychosis: Les effets du stress sur le système endocannabinoïde : étude par tomographie par émission de positons avec l'indicateur radioactif [11C-CURB] dans la psychose précoce.

BACKGROUND: Stress and traumatic experiences are well-established risk factors for psychiatric disorders. Stressful events can induce symptoms of anxiety and depression and may lead to overt psychosis, especially when there is an innate biological vulnerability. This study explores the role of the stress-regulating endocannabinoid system, specifically the activity of the enzyme fatty acid amid hydrolase (FAAH), a key regulatory enzyme for endocannabinoids, in association with stress by analysing data from healthy individuals and patients with psychosis. METHODS: We performed a post-hoc explor
