Viral cross-class transmission results in disease of a phytopathogenic fungus.

Interspecies transmission of viruses is a well-known phenomenon in animals and plants whether via contacts or vectors. In fungi, interspecies transmission between distantly related fungi is often suspected but rarely experimentally documented and may have practical implications. A newly described double-strand RNA (dsRNA) virus found asymptomatic in the phytopathogenic fungus Leptosphaeria biglobosa of cruciferous crops was successfully transmitted to an evolutionarily distant, broad-host range pathogen Botrytis cinerea. Leptosphaeria biglobosa botybirnavirus 1 (LbBV1) was characterized in L.

Pathogenicity and Growth Conditions Modulate Fonsecaea Extracellular Vesicles' Ability to Interact With Macrophages.

Chromoblastomycosis (CBM) is a chronic cutaneous and subcutaneous mycosis caused by black, dimorphic, and filamentous fungi of the Herpothrichiellaceae family, such as species of the genus Fonsecaea. These fungi can switch between the saprophytic forms (conidia and hyphae) and the pathogenic form, the muriform cells (MCs), which is considered an essential mechanism for fungal virulence. Nearly all types of cells can produce membranous structures formed by a lipid bilayer that communicate extracellularly with other cells, known as "extracellular vesicles" (EVs), which may act as virulence facto

First Isolation of Neoscytalidium dimidiatum from Human Dermatomycosis in Japan.

Neoscytalidium dimidiatum is a common fungus that causes non-dermatophyte dermatomycosis in tropical regions, but there have been no reports of infection with N. dimidiatum in Japan. Here, we report the first isolation of N. dimidiatum from human dermatomycosis in Japan. A 62-year-old healthy Japanese male had been treated with oral terbinafine for tinea pedis diagnosed from a microscopic examination in 2003 with a lesion that was intractable. In 2020, re-identification by sequencing the internal transcribed spacer regions and the D1/D2 domain of the large-subunit (LSU) ribosomal RNA gene reve

An amyotrophic lateral sclerosis hot spot in the French Alps associated with genotoxic fungi.

Between 1990 and 2018, 14 cases of amyotrophic lateral sclerosis (ALS) were diagnosed in residents of, and in visitors with second homes to, a mountainous hamlet in the French Alps. Systematic investigation revealed a socio-professional network that connected ALS cases. Genetic risk factors for ALS were excluded. Several known environmental factors were scrutinized and eliminated, notably lead and other chemical contaminants in soil, water or home-grown vegetation used for food, radon and electromagnetic fields. Some lifestyle-related behavioral risk factors were identified: Prior to clinical

Tracing the Origin and Evolutionary History of Pyricularia oryzae Infecting Maize and Barnyard Grass.

Blast disease is a notorious fungal disease leading to dramatic yield losses on major food crops such as rice and wheat. The causal agent, Pyricularia oryzae, encompasses different lineages, each having a different host range. Host shifts are suspected to have occurred in this species from Setaria spp. to rice and from Lolium spp. to wheat. The emergence of blast disease on maize in Iran was observed for the first time in the north of the country in 2012. We later identified blast disease in two additional regions of Iran: Gilan in 2013 and Golestan in 2016. Epidemics on the weed barnyard gras

Improvement of antifungal activity of a culture filtrate of endophytic Bacillus amyloliquefaciens isolated from kiwifruit and its effect on postharvest quality of kiwifruit.

This study reports the optimization of culture conditions and medium components of M9, an endophytic strain of Bacillus amyloliquefaciens, which we had previously isolated from kiwifruit, and evaluation of its effect on kiwifruit postharvest protection against soft rot. The method of one-factor-at-a-time was used to determine the optimum culture conditions, and response surface methodology was applied to optimize the medium constituents. After optimization, a high rate of antifungal activity (73.12% decrease in decay rate) by M9 culture filtrate against the soft rot fungal pathogen, Botryospha

Clinical and histopathological changes in rainbow trout Oncorhynchus mykiss experimentally infected with fungus Bradymyces oncorhynchi.

Bradymyces oncorhynchi is a poorly known melanised fungal species that has been isolated only from a hyperaemic focus near the enlarged spleen of a rainbow trout. Although the pathogenicity of this species to fish is suspected, it has not been fully confirmed. Four laboratory experiments were conducted to test the effect of the fungus on the health of rainbow trout fingerlings. Mycelia were cultivated under different conditions to increase inoculum variability and to test the impact of the conditions on the pathogenicity. The inoculum was subsequently administered by the intraperitoneal route.

Whole Genome Sequence Resource of the Asian Pear Scab Pathogen Venturia nashicola.

Venturia nashicola, the cause of scab disease of Asian pears, is a host-specific, biotrophic fungus. It is restricted to Asia and is regarded as a quarantine threat outside this region. European pear displays nonhost resistance (NHR) to V. nashicola and Asian pears are nonhosts of V. pyrina (the cause of European pear scab disease). The host specificity of these two fungi is likely governed by differences in their effector arsenals, with a subset hypothesized to activate NHR. The Pyrus-Venturia pathosystem provides an opportunity to dissect the underlying genetics of nonhost interactions in th

Diversity and ecology of cultivable fungi isolated from the thermal soil gradients in Deception Island, Antarctica.

We surveyed the diversity of cultivable fungi isolated from cold and hot volcanic soils of Deception Island, Antarctica. Seventy-four fungal isolates were identified; these belonged to 17 taxa in the genera Aspergillus, Penicillium, Pseudogymnoascus, Purpureocillium, and Mortierella. The fungal assemblages showed low diversity, richness, and dominance indices. The Aspergillus taxa were dominant in the soils at 0 °C, 50 °C, and 100 °C. Aspergillus lacticoffeatus, Aspergillus cf. ruber, Penicillium citrinun, and Purpureocillium sodanum were present only in soils having a temperature of 100 °C. A

Strain identification and metabolites isolation of Aspergillus capensis CanS-34A from Brassica napus.

An isolate (CanS-34A) of Aspergillus from a healthy plant of oilseed rape (Brassica napus) was identified based on morphological characterization and multi-locus phylogeny using the sequences of internal transcribed spacer (ITS)-5.8S rDNA region, BenA (for β-tubulin), CaM (for calmodulin) and RPB2 (for RNA polymerase II). The results showed that CanS-34A belongs to Aspergillus capensis Hirooka et al. The antifungal metabolites produced by CanS-34A in potato dextrose broth (PDB) were extracted with chloroform. Three antifungal metabolites were isolated and purified from the chloroform extract o

Evaluation of bioactivity and control efficacy of tetramycin against Corynespora cassiicola.

Tetramycin is a novel polyene antibiotic that has exhibited excellent inhibitory activity against many plant pathogens. In this study, the sensitivity of Corynespora cassiicola to tetramycin was assessed in vitro using a series of 91 isolates, and its effects on hyphae and conidia were evaluated. Preventive and curative efficacies of tetramycin against Corynespora leaf spot were evaluated using detached cucumber leaves and potted cucumber plants. The control efficacy of tetramycin against the pathogen was also determined under field conditions. Measurement of baseline sensitivity suggested tha

Ethylnaphthoquinone derivatives as inhibitors of indoleamine-2, 3-dioxygenase from the mangrove endophytic fungus Neofusicoccum austral SYSU-SKS024.

Bioassay-guided fractionation of the dichloromethane extract of the fungus Neofusicoccum austral SYSU-SKS024 led to the isolation of three new ethylnaphthoquinone derivatives, neofusnaphthoquinone A (1), 6-(1-methoxylethy1)-2,7-dimethoxyjuglone (2), (3R,4R)-3-methoxyl-botryosphaerone D (6), together with six known analogs (3-5 and 7-9). Their structures were elucidated by spectroscopic analysis and single crystal X-ray diffraction analysis. Neofusnaphthoquinone A (1) is the third example of the unsymmetrical naphthoquinone dimer, which is rarely found in natural source. All of the isolates wer

Narrow windrow burning canola (Brassica napus L.) residue for Sclerotinia sclerotiorum (Lib.) de Bary sclerotia destruction.

BACKGROUND: Since the introduction of herbicide-tolerant varieties of canola (Brassica napus L.) in 1993, global plantings have increased resulting in an increased incidence of Sclerotinia sclerotiorum (Lib.) de Bary infections. Developments in narrow windrow burning techniques to destroy the seed of multiple herbicide-resistant weeds provide an opportunity to also intercept and heat-treat the S. sclerotiorum inoculum source, termed sclerotia, before it re-enters the soil to infect susceptible crop species in successive years. RESULTS: Preliminary kiln studies determined that a temperature of

The Epichloë festucae antifungal protein has activity against the plant pathogen Sclerotinia homoeocarpa, the causal agent of dollar spot disease.

Epichloë spp. are naturally occurring fungal endophytic symbionts of many cool-season grasses. Infection by the fungal endophytes often confers biotic and abiotic stress tolerance to their hosts. Endophyte-mediated disease resistance is well-established in the fine fescue grass Festuca rubra subsp. rubra (strong creeping red fescue) infected with E. festucae. Resistance to fungal pathogens is not an established effect of endophyte infection of other grass species, and may therefore be unique to the fine fescues. The underlying mechanism of the disease resistance is unknown. E. festucae produce

Phylogenomic analyses reveal the diversity of laccase-coding genes in Fonsecaea genomes.

The genus Fonsecaea comprises black yeast-like fungi of clinical relevance, including etiologic agents of chromoblastomycosis and cerebral phaeohyphomycosis. Presence of melanin and assimilation of monoaromatic hydrocarbons and alkylbenzenes have been proposed as virulence factors. Multicopper oxidase (MCO) is a family of enzymes including laccases, ferroxidases and ascorbate oxidases which are able to catalyze the oxidation of various aromatic organic compounds with the reduction of molecular oxygen to water. Additionally, laccases are required for the production of fungal melanins, a cell-wa

PREVALENCE AND DISTRIBUTION OF PSEUDOGYMNOASCUS DESTRUCTANS IN MICHIGAN BATS SUBMITTED FOR RABIES SURVEILLANCE.

Since 2006, bat populations in North America have suffered devastating mortality from an emerging disease known as white-nose syndrome (WNS). The causal agent of WNS is the fungus Pseudogymnoascus destructans. In April 2014, WNS was discovered in little brown bats ( Myotis lucifugus ) in Michigan, US, and has since been documented in 12 counties. Because current surveillance for WNS focuses primarily on mine-hibernating species in winter, it is subject to geographic, species, and seasonal bias. To investigate species affected and potential associations of gender, seasonal life cycle, and regio
