A wireless magnetoelectric-driven strategy to boost nose-to-brain drug delivery with сore-shell nanotransducers.

Targeted therapeutic delivery to specific regions of the central nervous system (CNS) is a promising approach for treating localized pathologies such as neuropathic pain or viral infections. The systemic administration of drugs is often inefficient, as it distributes medication throughout the body, including non-targeted CNS areas, rather than concentrating it in the affected neural tissues. Leveraging axonal transport for targeted drug delivery could enable precise therapeutic interventions, such as antiviral, antineuropathic, or regenerative treatments, selectively directed to specific gangl…

Age-related accumulation of phosphorylated mitofusin 2 protein in retinal ganglion cells correlates with glaucoma progression.

Dysregulation of axonal bioenergetics is likely a key mechanism in the initiation and progression of age-related neurodegenerative diseases. Glaucoma is a quintessential neurodegenerative disorder characterized by progressive deterioration of the optic nerve (ON) and eventual death of retinal ganglion cells (RGCs). Age and elevation of intraocular pressure are key risk factors in glaucoma, but the common early hallmarks of decreased axonal transport and increased bioenergetic vulnerability likely underlie disease initiation. We examined the correlation between bioenergetics and axonal transpor…

Time-course of nociceptive disorders induced by chronic loose ligatures of the rat sciatic nerve and changes of the acetylcholinesterase transport along the ligated nerve.

Changes in the axonal transport of acetylcholinesterase (AChE) were studied in the painful mononeuropathy induced by setting 4 loose ligatures around the right sciatic nerve of the rat. Since changes in the axonal transport of AChE can be used to assess axonal degeneration/regeneration, we used this marker to investigate whether the time course of pain-related behavioral disorders observed following chronic constriction injury (CCI) to the sciatic nerve are related to the time course of the regeneration of the injured axons. In addition, a comparison was made between changes in AChE observed i…

Selective damage to large cells in the cat retinogeniculate pathway by 2,5-hexanedione.

The neurotoxic hexacarbon 2,5-hexanedione (2,5-HD), which produces transport abnormalities and swellings in the large diameter fibers of the … tion caused (1) axonal swellings which were not selective for fiber size; (2) a selective defect in axonal transport with later neuronal degeneration and shrinkage that were limited to large cells; and (3) a loss of flicker resolution that may reflect dysfunction of large ganglion cells.

Polarity of axoplasmic microtubules in the olfactory nerve of the frog.

Pieces of olfactory nerve of the bullfrog were extracted in a tubulin assembly buffer medium containing detergents. With incubation at 37 degrees C in such medium containing soluble tubulin, ribbons of protofilaments are formed on the surfaces of microtubules, with the ribbons curving in a clockwise or counterclockwise direction. The direction of hooking reflects the polarity of the microtubule. In nerve pieces oriented such that cross sections could be viewed toward the perikarya of the axons, over 90% of the ribbons on microtubules showed a clockwise orientation. When observers were looking

Pathogenic KIF5C mutation disrupts dendritic spine maturation and mitochondrial trafficking in neurodevelopmental disorders.

KIF5C, a kinesin-1 motor protein critical for neuronal cargo transport, has been clinically associated with developmental delay and intellectual disability (DD/ID), although its pathogenic mechanisms are yet to be elucidated. Building on our prior identification of a de novo heterozygous KIF5C variant in a patient with DD/ID, a conditional knock-in mouse model was constructed to determine disease pathogenesis. The mutant mice exhibited core clinical phenotypes, including growth retardation, microcephaly, and deficits in social and spatial memory. Electrophysiological recordings revealed a decr

Neurochemical architecture of the filum terminale in the rat.

Contrary to the widespread assumption, the filum terminale in the rat possesses a precise glial and neuronal organization. The processes of glial fibrillary acidic protein-stained astrocytes form a rich, three dimensional array. The crescent shaped white matter could be outlined with antibody detecting oligodendrocytes. The neurons in the filum terminale, labeled with neuron-specific nuclear protein, are distributed in a small midline group (dorsal nucleus) dorsal to and in two symmetrical clusters at both sides of the central canal (lateral nuclei). Influence of temperature on slow flow in populations of regenerating axons with different elongation velocities.

The slow intra-axonal transport mechanisms may be subdivided into 4 elementary processes: (a) protein synthesis; (b) release of the slow material from the cell body; (c) transport of the molecules along the axon; and (d) deposition of the material from the moving phase into the immobile axonal constituents. These 4 components of the slow flow mechanisms are differently affected by temperature and react differently to axonal regeneration. They have been studied at temperatures ranging from 10 to 35 degrees C, in 3 different populations of regenerating C-fibers of the garfish olfactory nerve. Th…

Interaction of tau protein with the dynactin complex.

Tau is an axonal microtubule-associated protein involved in microtubule assembly and stabilization. Mutations in Tau cause frontotemporal dementia and parkinsonism linked to chromosome 17 (FTDP-17), and tau aggregates are present in Alzheimer's disease and other tauopathies. The mechanisms leading from tau dysfunction to neurodegeneration are still debated. The dynein-activator complex dynactin has an essential role in axonal transport and mutations in its gene are associated with lower motor neuron disease. We show here for the first time that the N-terminal projection domain of tau binds to …

Membrane turnover and receptor trafficking in regenerating axons.

Peripheral axonal regeneration requires surface-expanding membrane addition. The continuous incorporation of new membranes into the axolemma allows the pushing force of elongating microtubules to drive axonal growth cones forwards. Hence, a constant supply of membranes and cytoskeletal building blocks is required, often for many weeks. In human peripheral nerves, axonal tips may be more than 1 m away from the neuronal cell body. Therefore, in the initial phase of regeneration, membranes are derived from pre-existing vesicles or synthesised locally. Only later stages of axonal regeneration are

Regulation of Axonal Microtubule Polarity Orientation in Different Kinds of Neurons.

Microtubules (MTs) in the axon are nearly all oriented with plus-end-out, and this pattern of organization is important for regulating the morphology and cytoplasmic composition of the axon, and for defining the cargoes transported in the anterograde and retrograde directions in the axon. Over the past several years, studies on vertebrate neurons (and also insect neurons) have been conducted by many investigators to understand the mechanisms that establish this pattern and maintain it over the life of the neuron and in the face of potential corruption. Studies on cultured rat sympathetic neuro

Nerve injury-on-a-chip uncovers extracellular matrix and mitochondrial roles in axonal regeneration.

Peripheral nerve regeneration depends on interactions among extracellular matrix cues, neuronal bioenergetics, and therapeutic signal presentation, yet progress is limited by the lack of physiologically relevant quantitative in vitro models to evaluate biomaterials and mitochondria-targeted therapies. Here, we present a nerve injury-on-a-chip (NI-Chip) that integrates dorsal root ganglion explants, aligned nanofibers, compartmentalized microfluidics, and precisely controlled axotomy to enable real-time, quantitative analysis of axonal regeneration dynamics. Extracellular matrix composition sig

LAMP1 and LAMP2A localise to axonal organelles with distinct motility dynamics and partially overlapping molecular signatures in human neurons.

LAMP1 and LAMP2A (an isoform of LAMP2) are abundant proteins of late endosomal/lysosomal compartments that are often used interchangeably to label what is assumed to be the same organelle population, potentially obscuring distinct physiological roles. Here, we characterised the axonal transport dynamics of LAMP1- and LAMP2A-positive compartments in human induced pluripotent stem cell (hiPSC)-derived cortical neurons. We found that LAMP1-positive organelles move slower in the retrograde direction, pause more frequently, and display a broader anterograde velocity distribution than LAMP2A-positiv…

Investigating how changes in the levels of kinesins impact neuronal health in Drosophila and human iPSC-derived neurons AD model.

Alzheimer's disease (AD) is the leading cause of dementia and the most common neurodegenerative disorder. Understanding the molecular pathology of AD may help identify new ways to reduce neuronal damage. In the past decades, Drosophila has become a powerful tool in modelling mechanisms underlying human diseases. Here, we investigate how the expression of the human 42-residue β-amyloid (Aβ) carrying the E22G pathogenic 'Arctic' mutation (Aβ42Arc) affects axonal health and behaviour in Drosophila. We find that Aβ42Arc flies present aberrant neurons, with altered axonal transport of mitochondria …

Target-Derived Neurotrophic Factor Deprivation Puts Retinal Ganglion Cells on Death Row: Cold Hard Evidence and Caveats.

Glaucoma and other optic neuropathies are characterized by axonal transport deficits. Axonal cargo travels back and forth between the soma and the axon terminus, a mechanism ensuring homeostasis and the viability of a neuron. An example of vital molecules in the axonal cargo are neurotrophic factors (NTFs). Hindered retrograde transport can cause a scarcity of those factors in the retina, which in turn can tilt the fate of retinal ganglion cells (RGCs) towards apoptosis. This postulation is one of the most widely recognized theories to explain RGC death in the disease progression of glaucoma a…

Tubulin transport in neurons.

A question of broad importance in cellular neurobiology has been, how is microtubule cytoskeleton of the axon organized? It is of particular … to visualize the footprints of the tubulin wave as it polymerized and depolymerized during its slow axonal transport. In contrast, depolymerization resistant fluorescent microtubules did not move when injected in neurons. We show that tubulin subunits, not microtubules, are the primary form of tubulin transport in neurons.
