Mutations in the global transcriptional activator EP300/KAT3B are being reported in aggressive malignancies. However, the mechanistic contri … e-stranded DNA gaps. Importantly, we find that the loss of EP300 results in decreased expression of BRCA2 protein leading to sensitivity to treatments that are cytotoxic to BRCA-deficient cancers. Overall, we demonstrate that EP300-mutated cells recapitulate features of BRCA-deficient cancers.

BACKGROUND/AIM: Several clinical trials have investigated homologous recombination deficiency and BRCA1/2 status to select ovarian cancer patients for treatment with poly(ADP-ribose) polymerase-inhibitors (PARPi), but less attention has been given to other DNA-damage response (DDR) pathways. Therefore, we investigated somatic single/multiple nucleotide variants and small insertions/deletions in exonic and splice-site regions of 356 DDR genes to examine whether genes other than BRCA1/2 are altered.
MATERIALS AND METHODS: Whole-exome sequencing data from eight high-grade serous adenocarcinoma (

The breast cancer susceptibility gene (BRCA) is an important tumor suppressor gene, including BRCA1 and BRCA2, a biomarker that assesses the risk of breast cancer and influences a patient's individualized treatment options. BRCA1/2 mutation (BRCAm) increases the risk of breast cancer. However, breast-conserving surgery is still an option for BRCAm, and prophylactic mastectomy and nipple-sparing mastectomy may also reduce the risk of breast cancer. BRCAm is sensitive to Poly (ADP-ribose) polymerase inhibitor (PARPi) therapy due to specific types of DNA repair defects, and its combination with o

Chromosome instability (CIN) is a cancer hallmark that drives tumour heterogeneity, phenotypic adaptation, drug resistance and poor prognosis. High-grade serous ovarian cancer (HGSOC), one of the most chromosomally unstable tumour types, has a 5-year survival rate of only ~30% - largely due to late diagnosis and rapid development of drug resistance, e.g., via CIN-driven ABCB1 translocations. However, CIN is also a cell cycle vulnerability that can be exploited to specifically target tumour cells, illustrated by the success of PARP inhibitors to target homologous recombination deficiency (HRD).

Thymomas comprise a group of rare epithelial neoplasms of the anterior mediastinum. Whereas localized disease carries a favorable prognosis, the majority of patients with metastatic thymomas experience progression or recurrence over a 10-year period. Although targeted therapies have become standard of care in many malignancies, no clinically actionable mutations have consistently been identified in metastatic thymomas. Here, we describe a patient with an aggressive thymoma complicated by extensive pleural metastases. Over a 16-year period, she progressed on multiple treatment regimens. To iden

Purpose Over the past three decades, the incidence rate of breast cancer (BC) among Arab women has continually increased. However, data on the prevalence of BRCA1/2 mutations are scarce. Although the population in Saudi Arabia is at large homogeneous and consanguinity is common, especially in the central, eastern, and southern regions of the country, the prevalence of BRCA1 and BRCA2 mutations and the characteristics of BC are not well studied in the country. Methods This prospective observational study intended to determine the prevalence of BRCA1 and BRCA2 mutations and sought to examine the

Preventive surgical procedures for inherited risk of breast cancer Forwomen with a demonstrated BRCA1 or BRCA2 mutation, the cumulative risk of developing invasive breast cancer before the age of 70 years is about 50-85% and the risk of developing invasive epithelial ovarian cancer is 20-60%. Regular surveillance including physical examination and imaging is offered to mutation carriers and the options for risk-reducing surgery are discussed. Although bilateral prophylactic mastectomy is a drastic intervention, it significantly reduces the incidence of breast cancer. For mutation carriers with

Women in general have a 10% risk of developing breast cancer and a 2-3% chance of ovarian cancer in their life-times. Mutations in BRCA-1 and BRCA-2 are present in only a small portion (5-10%) of all breast cancers. Carriers of mutations in these genes have a greater risk of cancer, especially before menopause in the case of BRCA-1 carriers. In addition, their risk of contralateral breast cancer is significantly higher than for the general population (4.2-53% vs. 2%). The grade of contralateral tumours in these patients is more aggressive. BRCA-2 hereditary breast cancer seems more heterogeneo

BACKGROUND: In the kin-cohort design, a volunteer with or without disease (the proband) agrees to be genotyped, and one obtains information on the history of a disease in first-degree relatives of the proband. From these data, one can estimate the penetrance of an autosomal dominant gene, and this technique has been used to estimate the probability that Ashkenazi Jewish women with specific mutations of BRCA1 or BRCA2 will develop breast cancer.
METHODS: We review the advantages and disadvantages of the kin-cohort design and focus on dichotomous outcomes, although a few results on time-to-dise

INTRODUCTION: Multilocus inherited neoplasia alleles syndrome (MINAS) can pair a germline pathogenic BRCA1/2 variant with a pathogenic mismatch-repair (MMR) variant, but current guidelines manage the two inherited pathways separately.
AREAS COVERED: We searched PubMed, Embase, the American Society of Clinical Oncology Library, European Society for Medical Oncology OncologyPro, and Society of Gynecologic Oncology library from 2011 to 17 May 2026. It distinguishes a dual germline carrier from single-track, dual-defective, and biomarker-discordant tumors. For mismatch-repair immunohistochemistry

BACKGROUND: Pathogenic variants in BRCA1 and BRCA2 are well known drivers of homologous recombination deficiency in ovarian carcinoma. Somatic BRCA mutations are observed in a minority of ovarian cancers. The co-occurrence of both BRCA1 and BRCA2 pathogenic somatic mutations in a single tumor is rare, and to our knowledge this has not been described in endometrioid ovarian carcinoma. We describe a case of endometrioid ovarian carcinoma harboring double somatic BRCA1 and BRCA2 pathogenic variants, with negative germline testing. CASE PRESENTATION: A 50-year-old woman underwent right oophorectom

The period for reclassifying the variants' pathogenicity is too long for patients with advanced or recurrent cancers with variants of uncertain significance that can be objectively re-evaluated to be (likely) pathogenic according to accumulated evidence. At present, we have three breast cancer patients with the germline variant BRCA2 c.7847C>T, which was genetically evaluated to be likely pathogenic by the paper in Cancer Science.

PURPOSE: To effectively reduce cancer burden, genetic testing programs should identify high-risk individuals prior to cancer development, when risk-reduction strategies can be implemented. We evaluated trends in BRCA1/BRCA2 testing use after implementation of a publicly funded testing program.
METHODS: We conducted a retrospective, near population-based study of women who underwent BRCA1/BRCA2 testing in Ontario, Canada, (2007-2016) (n = 15,986). Temporal trends were evaluated using linear and Poisson regression.
RESULTS: Although annual utilization of testing increased over time (p < 0.001)

Breast and ovarian cancers are the second and fifth leading causes of cancer-related death among women in the United States. Compared to non-carriers, BRCA mutation carriers are subject to a higher risk of developing breast and ovarian cancers. Prophylactic surgeries including prophylactic mastectomy and prophylactic bilateral salpingo-oophorectomy can significantly reduce the incidence risks of breast and ovarian cancers for BRCA mutation carriers. However, both prophylactic surgeries are permanent one-time interventions. Determining the optimal age for BRCA carriers to undergo such surgeries

Patients with prostate cancer and homologous recombination repair alterations face worse outcomes. Early use of PARP inhibitors, especially in BRCA-mutated cancers, is promising. However, optimal timing, sequencing, and benefits for non-BRCA alterations remain unclear, underscoring the need for well-designed clinical trials and personalized treatment discussions. OBJECTIVE: The BRCA1/2 mutation status testing is the global standard of care for breast cancer patients with a family history of cancer. BRCA1/2 mutations are known to be ethno-specific. For some ethnic groups of the Northern Asia (Buryats, Yakuts, Altaians, Tuvans, Khakasses, etc.) the founder mutations in the BRCA1/2 genes have not been revealed. This systematic review was conducted to assess the prevalence of BRCA1/2 mutation in breast cancer patients inhabiting Eastern Europe and Northern Asia (or Siberia).
METHODS: A total of 23,561 studies published between 2014 and 2024 were analyzed,

OBJECTIVE: Hereditary cancer syndromes constitute 5-10% of all cancers. The development of next-generation sequencing technologies has made it possible to examine many hereditary cancer syndrome-causing genes in a single panel. This study's goal was to describe the prevalence and the variant spectrum using NGS in individuals who were thought to have a hereditary predisposition for cancer.
MATERIAL AND METHOD: Analysis was performed for 1254 who were thought to have a familial predisposition for cancer. We excluded 46 patients who were carrying BRCA1/2 variants in this study, for focusing on t

The cumulative risk for breast and ovarian cancer is high in BRCA1 or BRCA2 mutation carriers. The lifetime risk of breast cancer by the age of 80 years is respectively up to 72% and 69% in BRCA1 and BRCA2 mutation carriers. The risk of ovarian cancer is higher (44%) in BRCA1 than in BRCA2 (17%) mutation carriers. Breast and ovarian cancers tend to arise earlier in BRCA1 mutation carriers. Breast cancers in BRCA1 mutation carriers are more frequently (up to 70%) triple negative while the majority (up to 80%) of breast cancers in BRCA2 mutation carriers are hormone sensitive. Many issues remain

BACKGROUND: Homologous recombination deficiency (HRD) phenotype will sensitize tumors to poly (ADP-ribose) polymerases inhibitors and platinum. However, previous studies did not focus on the prevalence of HRD among Chinese breast cancer (BC) patients.
METHODS: One hundred and forty-seven BC patients were included in this study. Their HRD status was assessed by Genomic Scar Score (GSS), which was determined according to the length, site, and type of copy number. HRD was defined as positive when a harmful BRCA1/2 mutation was detected or GSS ≥50.
RESULTS: Our data revealed that 9.5% of the 147
