Pseudomonas aeruginosa is the gram-negative bacterial rod which is often isolated from chronic aural discharge. This microorganism may also cause necrotizing infection of the external auditory canal in certain patients with impaired host-defense mechanisms. Involvement of the inner ear by this microbe is extremely rare. In this communication, we report a case of pseudomonas labyrinthitis which resulted from traumatic middle ear injury. Infection produced massive granulations and extensive bone destruction of the otic capsule. This case shows that while P.aeruginosa is usually an avirulent oppo

There is increased interest in characterizing and describing the effects of space stressors on human microflora. This study describes virulence-related phenotypes of two human yeast commensals, Rhodotorula mucilaginosa and Candida parapsilosis, isolated from the International Space Station (ISS). The strains were compared with ATCC control strains to provide insights into adaptation and phenotypic switching of fungal species in spacecraft environments. Strains were grown in media that induce filamentation and capsule production. Antimycotic susceptibility was determined after exposure of liqui

Streptococcus suis serotype 2 (SS2) is an emerging zoonotic agent responsible for a variety of diseases. The septicemia caused by SS2 suggests that it can evade the bactericidal effects of innate immune cells. However, the mechanisms by which SS2 evades innate immunity remain largely unknown. Neutrophils are critical components of innate immunity for antimicrobial defense. In this study, we identified a cell surface protein (CSP) insertion mutant in an SS2 transposon mutant library that significantly induces neutrophil extracellular traps (NETs) and shows poor survival compared to the parent S

BACKGROUND: Klebsiella aerogenes can cause ventilator-associated pneumonia by forming biofilms, and it is frequently associated with multidrug resistance. Phages are good antibiotic alternatives with unique advantages. There has been a lack of phage therapeutic explorations, kinetic studies, and interaction mechanism research targeting K. aerogenes.
METHODS: Plaque assay, transmission electron microscopy and whole-genome sequencing were used to determine the biology, morphology, and genomic characteristics of the phage. A mouse pneumonia model was constructed by intratracheal/endobronchial de

Bacterial vaginosis (BV) is the most common cause of vaginal discomfort in women. It is characterised by abnormal vaginal microbiota with a depletion of lactobacilli and predominance of anaerobic microorganisms, mainly Gardnerella vaginalis and Atopobium vaginae. Although antibiotics represent an effective therapeutic option in the short-term, recurrent infections still remain a serious problem. Nowadays, evidence exists about the efficacy of probiotics for the management of BV. The aim of the current double blind, randomised clinical trial was to assess the efficacy of a probiotic mixture, in

BACKGROUND: Pneumococcal meningitis (PM) is a life-threatening disease with a high case-fatality rate and elevated risk for serious neurological sequelae. In this study, we investigated the contribution of three major virulence factors of Streptococcus pneumoniae, the capsule, pneumococcal surface protein A (PspA) and C (PspC), to the pathogenesis of experimental PM.
METHODS: Mice were challenged by the intracranial route with the serotype 4 TIGR4 strain (wt) and three isogenic mutants devoid of PspA, PspC, and the capsule. Survival, bacterial counts, and brain histology were carried out. To

The global burden of invasive pneumococcal diseases, including pneumonia and sepsis, caused by Streptococcus pneumoniae, a Gram-positive bacterial pathogen, remains a major global health risk. The success of pneumococcus as a pathogen can be attributed to its ability to regulate the synthesis of capsular polysaccharide (CPS) during invasive disease. We previously reported that deletion of a putative lysine decarboxylase (LDC; ΔSP_0916) in pneumococcal serotype 4 (TIGR4) results in reduced CPS. SP_0916 locus is annotated as either an arginine or a LDC in pneumococcal genomes. In this study, by

While significant protection from pneumococcal disease has been achieved by the use of polysaccharide and polysaccharide-protein conjugate vaccines, capsule-independent protection has been limited by serotype replacement along with disease caused by nonencapsulatedStreptococcus pneumoniae(NESp). NESp strains compose approximately 3% to 19% of asymptomatic carriage isolates and harbor multiple antibiotic resistance genes. Surface proteins unique to NESp enhance colonization and virulence despite the lack of a capsule even though the capsule has been thought to be required for pneumococcal patho

The biofilm process in Streptococcus pneumoniae (pneumococcus) is described. Virtually all wild-type pneumococci are capable of the biofilm formation. The pneumococcal capsule may reduce the biofilm production, and the propensity to form biofilms has a reverse correlation with the amount of the capsule material. Invasive pneumococcal isolates and noninvasive strains that persist in the nasopharynx have different biofilm potential. A number of issues related to effector and regulatory factors in the pneumococcal biofilms are discussed in this review. In the summary, a biofilm may be essential o

UNLABELLED: The capsular polysaccharide synthesis ( cps ) locus of Neisseria meningitidis is implicated in invasive meningococcal disease. The synthesis ( synABCD ) and transport ( ctrABCD ) operons are transcribed in opposite directions from a common intergenic region and expression is negatively regulated by the bacterial two-component system misR/misS and thermosensitive RNA folding. However, these mechanisms do not fully explain the stationary phase responses and the cis-acting elements remain to be fully characterized. Using GFP reporter gene and site-directed mutagenesis, cis-regulatory

INTRODUCTION: Legitimate concerns about the quality of environmental waters are on the rise. As a consequence, efforts are being made by both the scientific community and policymakers to develop proficient methods for the monitoring and detection of pollutants of emerging concerns (PECs). In line with the European Union's "Zero Pollution" action plan, there is a growing need to develop real-time, multiplexed, on-site monitoring systems. The study explores the encapsulation of engineered Pseudomonas putida cells designed to produce a luminescent response upon exposure to specific analytes. The

INTRODUCTION: Subclinical intestinal inflammation and gut dysbiosis have been reported in patients with spondyloarthritis (SpA). In common practice, rheumatologists are increasingly confronted with patients with inflammatory rheumatism who are on gluten-free diets (GFDs), despite the lack of reliable data from controlled studies. This study aims to determine the impact of a GFD on the quality of life of patients with axial SpA.
METHODS AND ANALYSIS: The GlutenSpA study is a 24-week, randomised, double-blinded, placebo-controlled, multicentre trial. Patients with axial SpA (n=200) will follow

After a short introduction (chapter 1) methods of measuring gastrointestinal pH are described in chapter 2. The methods are divided into intubation techniques and tubeless methods, and the advantages and disadvantages are discussed. Measurements with pH-sensitive, radiotransmitting capsules are highlighted, and methodological problems with these capsules are described. Chapter 3 concerns the gastrointestinal pH profile of healthy subjects. The intraluminal pH is rapidly changed from highly acid in the stomach to about pH 6 in the duodenum. The pH gradually increases in the small intestine from

Hypervirulent Klebsiella pneumoniae (hvKP) poses a significant public health threat due to its ability to cause severe invasive infections with higher mortality than classical strains. Understanding its pathogenesis is essential for devising effective treatment strategies. We analyze shared virulence-associated genes from three hvKP transposon mutagenesis libraries and identify wzi as a key virulence determinant of hvKP. We demonstrate that Wzi deficiency reduces CPS production in K. pneumoniae, differing from its function as a CPS anchor in Escherichia coli. Importantly, Wzi exerts a pivotal

Invasive meningococcal disease (IMD) is always caused by Neisseria meningitidis, presents a significant clinical challenge with high mortality rates (up to 20%) and not only neurological sequelae, but also physical and psychological sequelae in survivors. While vaccination has effectively reduced the incidence of traditional serogroups like A and C in China, the emergence of serogroup replacement due to capsular switching-horizontal gene transfer of capsular genes-threatens ongoing public health efforts. This study reports firstly the identification of a ST-7 NmX strain (20220811) in Jiangsu P

Burkholderia pseudomallei is the causative agent of melioidosis and is a major mediator of sepsis in its endemic areas. Because of the low LD(50) via aerosols and resistance to multiple antibiotics, it is considered a Tier 1 select agent by the CDC and APHIS. B. pseudomallei is an encapsulated bacterium that can infect, multiply, and persist within a variety of host cell types. In vivo studies suggest that macrophages and neutrophils are important for controlling B. pseudomallei infections, however few details are known regarding how neutrophils respond to these bacteria. Our goal is to descri

Burkholderia pseudomallei has been shown to produce more than one capsular polysaccharide (CPS). Analysis of the B. pseudomallei genome has revealed that the organism contains four CPS operons (I-IV). One of these operons (CPS III) was selected for further study. Comparative sequencing analysis revealed that the genes encoding CPS III are present in B. pseudomallei and Burkholderia thailandensis but not in Burkholderia mallei. In this study, CPS III was not found to contribute to the virulence of B. pseudomallei. Strains containing mutations in CPS III had the same LD(50) value as the wild-typ
