Lactic acid bacteria (LAB) play a pivotal role in food fermentation by shaping microbial ecosystems and enhancing product safety through the production of antimicrobial metabolites. This study investigated the selective utilization of indigestible polysaccharides extracted from Cordyceps militaris (IPC) and their application in combination with a bacteriocin-like inhibitory substance (BLIS)-producing Leuconostoc mesenteroides AHL9a starter culture during mustard green fermentation. IPC selectively promoted the growth of Le. mesenteroides AHL9a, reaching 8.91 log CFU/mL after 24 h, and signific

Listeria monocytogenes is a Gram-positive facultative intracellular bacterium that is ubiquitous in nature and the causative agent of listeriosis. The outbreak-derived serotype 4b strain L. monocytogenes strain WS1, sequence type (ST) 558, sublineage (SL) 558, was previously found to have unusual pathogenicity, with ability to cause fetal damage in the first trimester of pregnancy. Search of the WS1 genome for novel and unique genomic features identified a putative lantibiotic island on the chromosome of WS1 and all tested strains of SL558 and two other putative emerging serotype 4b clones, cl

Pediococcus pentosaceus 732, Lactococcus lactis subsp. lactis 431, and Lactococcus lactis 808, bacteriocinogenic strains previously isolated from kimchi and banana, were investigated for their safety, beneficial properties and in vitro inhibition of pathogens such as Listeria monocytogenes ATCC 15313 and Staphylococcus simulans KACC 13241 and Staphylococcus auricularis KACC 13252. The results of performed physiological, biochemical, and biomolecular tests suggest that these strains can be deemed safe, as no virulence genes were detected in their DNA. Notably, only the gad gene associated with

Melioidosis, caused by Burkholderia pseudomallei, is an enigmatic infectious disease that afflicts individuals in many tropical and developing regions. Treatment is hampered by the organism's innate antibiotic resistance and the disease's non-pathognomic presentation. Recently, added attention has been given to this organism due to its classification as a potential biowarfare agent. Therefore, methods of preventing acquisition of infection are needed. We investigated antagonism between Burkholderia spp and B. pseudomallei derived from the same ecological niche in a melioidosis endemic region i

The anti-listerial effect of pediocin PA-1 and its producing strain, Pediococcus acidilactici UL5, was investigated in vivo using an ICR mouse model. The effect of intra-gastric administration of a single dose of P. acidilactici UL5 (4 x 10(10) CFU/animal) on the propagation of Listeria monocytogenes LSD348 in intestine, liver and spleen was negligible. P. acidilactici UL5 did not appear competitive with the mouse intestinal flora and was not detectable in fecal samples collected two days after administration. However, double-agar-layer activity assay showed the ability of P. acidilactici UL5

The rapid escalation of antimicrobial resistance (AMR) has rendered many conventional antibiotics ineffective, emphasizing the need for alternative therapeutic approaches. Probiotics have emerged as promising biotherapeutic agents capable of inhibiting multidrug-resistant (MDR) pathogens through diverse mechanisms, including secretion of antimicrobial metabolites (bacteriocins, organic acids, short-chain fatty acids, and hydrogen peroxide), competitive exclusion, quorum-sensing interference, and immune modulation. However, their clinical application is limited by poor stability under environme…

Lactobacillus helveticus is a probiotic bacterium widely used in the food industry. In this study, we evaluate the safety and probiotic prop … associated with these traits. It contains gene clusters potentially involved in the biosynthesis of bacteriocins Helveticin J and Enterolysin A, as well as encoding multiple genes of carbohydrate-active enzymes. Moreover, KM7 demonstrated excellent exopolysaccharide-producing capability, and glucose supplementation in the medium enhanced yield. These findings …

Streptomyces is a Gram-positive bacterium, belonging to the family Streptomycetaceae and order Streptomycetales. Several strains from different species of Streptomyces can be used to promote the health and growth of artificially cultured fish and shellfish by producing secondary metabolites including antibiotics, anticancer agents, antiparasitic agents, antifungal agents, and enzymes (protease and amylase). Some Streptomyces strains also exhibit antagonistic and antimicrobial activity against aquaculture-based pathogens by producing inhibitory compounds such as bacteriocins, siderophores, hydr…

The discovery that Streptococcus pneumoniae uses a competence-stimulating peptide (CSP) to induce competence for natural transformation, and that other species of the mitis and the anginosus streptococcal groups use a similar system, has expanded the tools to explore gene function and regulatory pathways in streptococci. Two other classes of pheromones have been discovered since then, comprising the bacteriocin-inducing peptide class found in Streptococcus mutans (also named CSP, although different from the former) and the SigX-inducing peptides (XIP), in the mutans, salivarius, bovis, and pyo

The <<Taq Man>> assay (PE Applied Biosystems) combines PCR with concomitant release of fluorogenic nucleotides for immediate product detection by fluorometry. Yersinia pestis, the etiological agent of bubonic plague, expresses species-specific genes known to be located on two unique plasmids (9.6-kb pPCP and 100.9-kb pMT). Pesticin (pst) is a unique gene located on pPCP which encodes for a bacteriocin. Using fluorogenic probe coupled PCR as few as three copies of pst targets were detected from total Y. pestis genomic DNA. The pst probe used in this report was positive only for pesticinogenic i
