The Synergistic Effects of rhArg with Bcl-2 Inhibitors or Metformin Co-Treatment in Multiple Cancer Cell Models.

BACKGROUND: Recombinant human arginase (rhArg) has been proven to exhibit an anticancer effect via arginine starvation. To further improve the efficacy of rhArg, we examined the feasibility of a combination strategy with Bcl-2 inhibitors (ABT263 and ABT199) or an antidiabetic drug (metformin) and investigated the mechanistic basis for these strategies. METHODS: The combination effects were evaluated in a panel of human cancer cell lines modeling pancreatic ductal carcinoma (PDAC), triple-negative breast cancer (TNBC), colorectal cancer (CRC) and glioblastoma (GBM). Western blot analysis was u

Genotoxic stressors mimicking synovial microenvironment modulate B cell fate in RA-FLS co-culture.

Rheumatoid arthritis (RA) is characterized by persistent synovial inflammation in which fibroblast-like synoviocytes (RA-FLS) and B cells interact within a potentially genotoxic microenvironment. This study investigates how naive B cells respond to cytotoxic stress when supported by RA-FLS in a co-culture model, using viability assays, time resolved γ-H2AX MFI, RT-qPCR of DNA damage response and differentiation genes, and flow cytometric analysis of subsets and surface markers. Naive B cells exhibited greater loss of viability than RA-FLS, and time-resolved γ-H2AX MFI showed an earlier signal

An engineered ferritin nanocage co-delivery system for targeted hair cells protection and hearing loss attenuation.

RATIONALE: Hearing loss is a common sensory disorder with few treatment options, mainly due to the blood-labyrinth barrier and lack of effective targeted delivery systems. Small molecules such as LLY283 and RG108 protect hair cells (HCs), but clinical use is limited by poor cellular uptake, off-target toxicity, and inefficient cochlear delivery. METHODS: We developed a ferritin (Fn)-based nanocage (LR@Fn) to co-deliver RG108 and LLY283 via local injection through the round window membrane. Fn offers natural HC targeting, a hollow cavity, and good biocompatibility. Drugs were loaded using a pH

Fish-shaped erythrocytes and pincer cells co-occur in distinct hematological disorders and are associated with anemia severity.

OBJECTIVES: Red blood cell (RBC) morphology in peripheral blood smears (PBS) remains essential for the differential diagnosis of anemia and other hematologic disorders. As yet unclassified poikilocytes, fish-shaped RBCs and pincer (mushroom-shaped) cells are rarely observed but have been described as "bizarre erythrocytes" and linked to hereditary spherocytosis. This study aimed to investigate their relevance across a spectrum of hematologic diseases and provide a basis for future morphological standardization. METHODS: Peripheral blood smears from 251 specimens were systematically reviewed,

Not just a viral co-receptor: the role of CCR5 in HIV cure after allogeneic stem cell transplantation.

PURPOSE OF REVIEW: Allogeneic hematopoietic stem cell transplantation (allo-HSCT) remains the only approach with confirmed cases of HIV cure. This review highlights the multifaceted role of CCR5 in this context, emphasizing that its impact extends far beyond functioning as a viral entry co-receptor, and outlines how donor and host CCR5 genotypes may influence reservoir depletion, transplant-related immune processes, and sustained remission. RECENT FINDINGS: Transplantation with CCR5Δ32/Δ32 donor cells is the most well documented approach to achieving HIV cure. However, growing evidence indica

Immune Dysregulation in HIV-TB Co-Infection: Role of Cytokines and T Cell Biomarkers-A Narrative Review.

Immune dysregulation is a hallmark of human immunodeficiency virus (HIV) infection, characterized by persistent immune activation and systemic inflammation that drive T cell exhaustion and senescence, contributing to disease progression and non-AIDS comorbidities, most notably tuberculosis (TB). With rising HIV prevalence, the incidence of HIV-TB co-infection continues to rise, highlighting the need to understand their immunopathological interplay. This narrative review aims to examine the association between immune dysregulation in HIV-TB co-infection, with a focus on cytokine profiles and im

Mucin hydrogel fibers enable oxygen-limited co-culture of Bifidobacterium bifidum and intestinal epithelial cells.

The intestinal mucus layer forms the interface between the gut microbiota and the epithelium, and in vitro platforms are needed to study host-microbe interactions at this interface. However, such platforms must reconcile conflicting conditions: obligately anaerobic bacteria require a low-oxygen environment, whereas epithelial cells require conditions compatible with barrier maintenance. In addition, simple co-culture systems lack a glycan-presenting mucus matrix. Here, we established a co-culture platform in a conventional Transwell format without complex microfluidic devices by integrating a

Chitosan-PEG Nanoparticles for Co-Delivery of Paclitaxel and KRAS G12D-Directed siRNA to Pancreatic Cancer Cells.

Pancreatic ductal adenocarcinoma (PDAC) remains one of the most lethal malignancies due to limited responsiveness to chemotherapy and the high prevalence of oncogenic Kirsten rat sarcoma viral oncogene homolog (KRAS) mutations. Co-delivery of cytotoxic agents and small interfering RNA (siRNA) is a potential combination strategy, but the two cargos have distinct physicochemical and intracellular-delivery requirements. Here, we developed a chitosan-polyethylene glycol (CP)-based polymeric nanoparticle platform for the cotreatment of paclitaxel (PTX) and small interfering RNA (siRNA) targeting KR

Cellular determinants influence the red blood cell adsorption efficiency of poly(amine-co-ester) nanoparticles.

Many poly(amine-co-ester) (PACE) nanoparticles, drug delivery vehicles for nucleic acid and small molecule cargoes, accumulate in the liver and spleen following intravenous administration, limiting delivery to nonhepatosplenic tissues. Red blood cell (RBC) hitchhiking, a strategy in which nanoparticles are nonspecifically adsorbed to RBCs prior to administration, has been used to modulate nanoparticle biodistribution, enabling enrichment in organs immediately downstream from the site of vascular infusion. We find that scarcely investigated cellular determinants-namely, storage duration, membra

Single-cell RNA-sequencing co-expression analysis with CFTR and CF modifier genes in lung tissue.

While cystic fibrosis is caused by loss-of-function variants in the Cystic Fibrosis Transmembrane Conductance Regulator (CFTR), other modifier genes have been shown to associate with disease severity. Co-expression of modifiers with CFTR in normal tissue indicates a cooperative relationship and suggests the potential for compensation in the presence of CFTR dysfunction. We examined the co-expression relationships with CFTR in the lung using single cell RNA sequencing to pinpoint cell types and their modifiers involved in the cystic fibrosis lung phenotype and support target prioritization for

2B4 co-stimulation and dasatinib modulation enhance anti-CD19 CAR-NK-92 cell cytotoxicity.

INTRODUCTION: Chimeric Antigen Receptor (CAR)-based therapies have transformed cancer treatment, especially in hematological malignancies. While the impact of co-stimulatory domains on CAR-T cell efficacy is well established, the optimal signaling modules for CAR-natural killer (CAR-NK) cells remain less defined. Identifying NK-tailored co-stimulatory domains is essential for maximizing CAR-NK cytotoxicity and clinical potential. METHODS: Using the NK-92 cell line as a controlled proof-of-concept platform, we engineered CAR19 constructs incorporating NK-specific co-stimulatory domains, includ

Unravelling T cell exhaustion through co-inhibitory receptors and its transformative role in cancer immunotherapy.

Persistent stimulation from cancer antigens leads to T lymphocytes (T cells) exhaustion, with up-regulated expression of co-inhibitory receptors, including programmed death-1 (PD-1), cytotoxic T lymphocyte-associated antigen-4 (CTLA-4), lymphocyte-activation gene 3 (LAG-3), T cell immunoglobulin and mucin domain 3 (TIM-3) and T cell immunoreceptor with Ig and ITIM domains (TIGIT). These receptors collectively impair T cell function via distinct molecular pathways, contributing to immune evasion and cancer progression. This review highlights the therapeutic promise of immune checkpoint inhibito

[Anticipation of pharmacotherapeutic problems prior to allo stem cell transplantation in patients with co-morbidities (SFGM-TC)].

Allogeneic hematopoietic stem cell transplantation (allo-HSCT) frequently involves patients with multiple co-morbidities. The drugs prescribed for these conditions may lead to complications during the transplant procedure, either through their mechanism of action, their adverse reaction profile or the risk of pharmacological interactions with the drugs prescribed for the allo-HSCT procedure and its complications. Therefore, each patient's drug-related risks must be assessed prior to allo-HSCT. Various therapeutic decisions may be considered to limit the occurrence of complications. The aim of

Poly[(N-acryloyl glycine)-co-(acrylamide)]-induced cell growth inhibition in heparanase-driven malignancies.

In the present work, glycine, the monomer N-acryloylglycine (NAG), and polymeric units of poly[(N-acryloylglycine)-co-(acrylamide)] p(NAG-co-Ac) are examined using density functional theory (DFT), and experimental evidence is provided for their use in the therapy of cancer with a poor prognosis. Glycine plays a pivotal role in cell survival, and most anti-cancer agents alter glycine metabolomics and suppress cancer cell proliferation. Herein, we have utilized Frontier Molecular Orbital theory (FMO), and the results revealed that the introduction of acrylamide/divinyl benzene into the glycine-b

Library size-stabilized metacells construction enhances co-expression network analysis in single-cell data.

Single-cell RNA sequencing (scRNA-seq) deciphers cell type-specific co-expression networks to resolve biological functions but remains constrained by data sparsity and compositional biases. Conventional metacells construction strategies mitigate sparsity by aggregating transcriptionally similar cells but often neglect systematic biases introduced by compositional data. This problem leads to spurious co-expression correlations and obscuring biologically meaningful interactions. Through mathematical modeling and simulations, we demonstrate that uncontrolled library size variance in traditional m

Co-delivery of ripasudil and dexamethasone in trabecular meshwork cells for potential prevention of GC-induced ocular hypertension.

The purpose of this in vitro study is to investigate whether the co-delivery of Ripasudil (Rip) can suppress the expression of genes related to glaucoma pathogenesis induced by prolonged dexamethasone (Dex) use, which can lead to ocular hypertension and potentially glaucoma formation. The effects of Rip (10 μM) on Dex (100 nM)-treated human trabecular meshwork (TM) cells were tested through co-delivery and sequential treatments using RNA-seq. Genome-wide analysis was performed using gene ontology (GO) enrichment and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analysis with Enrichr a

Stem-like CD8+ T cells preserve HBV-specific responses in HBV/HIV co-infection.

OBJECTIVE: Chronic HBV infection disproportionately affects people living with HIV, who are often excluded from functional cure studies. This study investigates CD8+ T cell profiles in HBV mono-infection versus HBV/HIV co-infection, examining the impact of long-term therapy on virus-specific responses with the goal of informing therapeutic strategies for immune restoration. DESIGN: We analysed CD8+ T cell responses in 61 participants (HBV n=20, HBV/HIV n=20, HIV n=21), on suppressive antiviral therapy. We assessed transcriptomic and proteomic profiles, focusing on exhaustion markers alongside

Neural Crest-Derived Co-localization: A Case of Granular Cell Tumor Arising Beneath a Nevus Spilus.

Granular cell tumors (GCTs) are rare neoplasms believed to originate from Schwann cells, exhibiting immunohistochemical and ultrastructural features consistent with neural crest derivation. Similarly, a nevus spilus (NS) is a pigmented skin lesion composed of lentiginous background macules with superimposed darker macules or papules, also thought to arise from neural crest-derived melanocytes. We report a unique case of a 27-year-old female who presented with a painful subcutaneous nodule on the left medial midback, overlaid by an NS. Surgical excision revealed a lesion consistent with a GCT b

Membrane proteomics and transcriptomic profiling analysis of hepatic stellate cells co-incubated with Schistosoma japonicum eggs.

Schistosomiasis has been recognized as the second most prevalent parasitic disease worldwide, following malaria. Schistosome eggs can persist for extended periods within host hepatic tissues, leading to hepatic fibrosis primarily through the activation of hepatic stellate cells (HSCs). However, the mechanisms by which egg-secreted products modulate the activation of HSCs remain incompletely elucidated. In this study, purified Schistosoma japonicum (S. japonicum) eggs (Egg group) or corresponding unused medium (Control group) were placed in the upper chamber of a Transwell system, with HSCs cul

Regulatory T Cells Kinetics in Immune Reconstitution Inflammatory Syndrome in HIV-Tuberculosis Co-Infected Individuals.

BACKGROUND: Combination antiretroviral therapy (cART) can suppress human immunodeficiency virus (HIV-1) replication, but some patients develop worsening of co-infections, termed immune reconstitution inflammatory syndrome. Regulatory T cells (Tregs) are a population of CD4+ T cells that modulate immune responses. We hypothesized that immune reconstitution inflammatory syndrome (IRIS) is associated with Tregs dysfunction. METHODS: We prospectively enrolled antiretroviral naive HIV patients with co-infection with Mycobacterium tuberculosis (MTB; N = 26) or controls with no prior opportunistic i
