Human Papillomavirus Vaccination in India: Paving the Way to Cervical Cancer Prevention.

Cervical cancer remains a major public health concern in India, contributing significantly to global morbidity and mortality. Persistent infection with high-risk human papillomavirus (HPV) is the primary etiological factor, making vaccination a critical preventive strategy. Recent developments, including the introduction of the indigenous quadrivalent HPV vaccine Cervavac and its proposed inclusion in the Universal Immunization Programme (UIP) for girls aged 9-14 years, represent a significant milestone. However, challenges such as vaccine hesitancy, logistical barriers, and limited awareness

A Personalised Vaccination Program Based on Immune Reconstitution in Paediatric Cancer Survivors.

AIMS: Paediatric cancer survivors often experience treatment-induced immunosuppression, requiring post-treatment revaccination. However, immune recovery timelines vary, and current revaccination guidelines, largely based on data of varied quality derived from studies on acute-lymphoblastic-leukaemia (ALL), may not be applicable across all paediatric malignancies. This study evaluated a personalised revaccination approach based on immune recovery. METHODS: Immune recovery was evaluated using antibody titers to common childhood vaccines, immunoglobulin levels, B- and T-cell subpopulations, and

Clinical research progress and challenges of vaccine for human papillomavirus-associated cancers.

As a major cause of cancer-related morbidity in women, cervical cancer is strongly related to chronic infection by high-risk human papillomavirus (HPV). This viral persistence also underpins the pathogenesis of a broad spectrum of cancers. In this review, we summarize the research progress of prophylactic and therapeutic HPV vaccines in clinical trials. Furthermore, we present a critical analysis of future challenges, specifically discussing the type replacement phenomenon and its potential implications for long-term vaccine efficacy. We also expand the discourse to a pan-gender and pan-cancer

Optimizing Cancer Vaccinations Using a Physiologically Based Pharmacokinetic (PBPK) Model.

UNLABELLED: Antigen-based tumor vaccines rely on adjuvants to stimulate local inflammation, recruit antigen-presenting cells (APCs), and enhance immune activation. However, the complex interplay between antigen transport, lymphatic drainage, and immune cell dynamics across organs remains poorly understood, limiting the rational design of vaccination strategies. Here, we present a multiscale compartmental Physiologically Based Pharmacokinetic (PBPK) model of antigen vaccination that integrates systemic circulation, lymphatic connectivity, and immune cell activation at the whole-body level. The

Immunotherapy of Oncovirus-Induced Cancers: A Review on the Development and Efficacy of Targeted Vaccines.

BACKGROUND: A number of viruses are oncogenic. These include the human papilloma virus (HPV), Epstein-Barr virus (EBV), Kaposi sarcoma human herpes virus 2/human herpes virus 8 (KSHHV/HHV8), hepatitis B virus, (HBV), hepatitis C virus (HCV), Merkel cell polyoma virus (McPyV), and the human T-cell leukemia virus type 1 (HTLV-1). These viruses cause malignancies ranging from carcinomas, sarcomas, lymphomas, to leukemias. This review aims to study the effects and efficacy of vaccines against these viruses and the cancers they cause in their prevention and treatment. METHODS: The literature in th

Engineered mitochondria exert potent antitumor immunity as a cancer vaccine platform.

The preferable antigen delivery profile accompanied by sufficient adjuvants favors vaccine efficiency. Mitochondria, which feature prokaryot … and TRP2-containing mitochondria (OVA-MITO and TRP2-MITO) were extracted and evaluated as potential cancer vaccines. Impressively, the engineered mitochondria vaccine demonstrated efficient antitumor effects when used as both prophylactic and therapeutic vaccines in murine tumor models. Mechanistically, OVA-MITO and TRP2-MITO potently recruited and activated dend …

Side effects of COVID-19 vaccinations in patients treated for breast cancer.

Lymph node swelling is a side effect of the mRNA COVID-19 vaccines, a distressing side effect for women treated for breast cancer. The purpose of this study is to present side effects reported by a cohort of patients treated for breast cancer. A survey link was sent to 4945 women who received breast cancer treatment and were prospectively screened for breast cancer-related lymphedema. In total, 621 patients who received an mRNA vaccine and responded to the survey were included in analysis. We assessed the frequency and predictors of side effects. The most frequent side effects reported were in

COVID-19 in cancer patients: The impact of vaccination on outcomes early in the pandemic.

BackgroundWith the rapid evolution of the severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) pandemic, the development of effective and safe vaccines was of utmost importance to protect vulnerable individuals, including cancer patients. Studies comparing the clinical outcomes of cancer patients with or without vaccination against coronavirus disease 2019 (COVID-19) have not demonstrated clear benefit. We aimed to determine the protective effects of COVID-19 vaccination by comparing vaccinated and unvaccinated cancer patients after the initial phase of vaccine roll-out and to identify

Human Papillomavirus Infection in Penile Cancer: Multidimensional Mechanisms and Vaccine Strategies.

Penile cancer (PC) is a rare male malignant tumor, with early lymph node metastasis and poor prognosis. Human papillomavirus (HPV) plays a key role in the carcinogenesis of PC. This review aims to summarize the association between HPV infection and PC in terms of virus-host genome integration patterns (the disrupted regions in the HPV and PC genome), genetic alterations, and epigenetic regulation (methylation and microRNA modification) occurring in HPV and PC DNA, as well as tumor immune microenvironment reprogramming. In addition, the potential of HPV vaccination strategies for PC prevention

Cancer testis antigen subfamilies: Attractive targets for therapeutic vaccine (Review).

BACKGROUND: Conflict affected refugees and internally displaced persons (IDPs) are at increased vulnerability to gender-based violence (GBV). Health, psychosocial, and protection services have been implemented in humanitarian settings, but GBV remains under-reported and available services under-utilized. To improve access to existing GBV services and facilitate reporting, the ASIST-GBV screening tool was developed and tested for use in humanitarian settings. This process was completed in four phases: 1) systematic literature review, 2) qualitative research that included individual interviews a

Vaccination Strategies for the Control and Treatment of HPV Infection and HPV-Associated Cancer.

Human papillomavirus (HPV) is the most common sexually transmitted infection, currently affecting close to 80 million Americans. Importantly, HPV infection is recognized as the etiologic factor for numerous cancers, including cervical, vulval, vaginal, penile, anal, and a subset of oropharyngeal cancers. The prevalence of HPV infection and its associated diseases are a significant problem, affecting millions of individuals worldwide. Likewise, the incidence of HPV infection poses a significant burden on individuals and the broader healthcare system. Between 2011 and 2015, there were an estimat

The impact of HPV vaccination beyond cancer prevention: effect on pregnancy outcomes.

While the benefits of human papillomavirus (HPV) vaccination relating to cervical cancer prevention have been widely documented, recent published evidence is suggestive of an impact on adverse pregnancy outcomes (APOs) in vaccinated mothers and their infants, including a reduction in rates of preterm births and small for gestational age infants. In this review, we examine this evidence and the possible mechanisms by which HPV vaccination may prevent these APOs. Large-scale studies linking HPV vaccination status with birth registries are needed to confirm these results. Potential confounding fa

Seroconversion rates following COVID-19 vaccination among patients with cancer.

As COVID-19 adversely affects patients with cancer, prophylactic strategies are critically needed. Using a validated antibody assay against SARS-CoV-2 spike protein, we determined a high seroconversion rate (94%) in 200 patients with cancer in New York City that had received full dosing with one of the FDA-approved COVID-19 vaccines. On comparison with solid tumors (98%), a significantly lower rate of seroconversion was observed in patients with hematologic malignancies (85%), particularly recipients following highly immunosuppressive therapies such as anti-CD20 therapies (70%) and stem cell t

Safety Evaluation of Autologous Tissue Vaccine Cancer Immunotherapy in a Canine Model.

BACKGROUND/AIM: Previous work in rodent models showed that an autologous tissue vaccine is both a safe and effective approach for treating cancer; however, as a translational step, safety must first be evaluated in a more clinically-relevant model. MATERIALS AND METHODS: An autologous immunotherapy produced from resected tumors, was evaluated in a clinically-relevant canine model to assess safety. Ninety-three dogs with spontaneously occurring tumors received vaccination with inactivated autologous tumor tissue combined with an adjuvant of particulate porcine small intestinal submucosa extrac

Oncolytic virotherapy enhances the efficacy of a cancer vaccine by modulating the tumor microenvironment.

The efficacy of cancer vaccines has been limited by the immunosuppressive tumor microenvironment, which can be alleviated by immune checkpoint inhibitor (ICI) therapy. Here, we tested if oncolytic viruses (OVs), similar to ICI, can also synergize with cancer vaccines by modulating the tumor microenvironment. VSV-GP, a chimeric vesicular stomatitis virus (VSV) pseudotyped with the glycoprotein (GP) of the lymphocytic choriomeningitis virus, is a promising new OV candidate. Here, we show that in mouse B16-OVA melanoma, combination treatment of VSV-GP with an ovalbumin (OVA) peptide-loaded dendri…

Personalized Cancer Vaccine Platform for Clinically Relevant Oncolytic Enveloped Viruses.

The approval of the first oncolytic virus for the treatment of metastatic melanoma and the compiling evidence that the use of oncolytic viruses can enhance cancer immunotherapies targeted against various immune checkpoint proteins has attracted great interest in the field of cancer virotherapy. We have developed a novel platform for clinically relevant enveloped viruses that can direct the virus-induced immune response against tumor antigens. By physically attaching tumor-specific peptides onto the viral envelope of vaccinia virus and herpes simplex virus 1 (HSV-1), we were able to induce a st

Multi-target chimaeric VLP as a therapeutic vaccine in a model of colorectal cancer.

BACKGROUND: Colorectal cancer is responsible for almost 700,000 deaths annually worldwide. Therapeutic vaccination is a promising alternative to conventional treatment for colorectal cancer, using vaccines to induce targeted immune responses against tumour-associated antigens. In this study, we have developed chimaeric virus-like particles (VLP), a form of non-infectious non-replicative subunit vaccine consisting of rabbit haemorrhagic disease virus (RHDV) VP60 capsid proteins containing recombinantly inserted epitopes from murine topoisomerase IIα and survivin. These vaccines were developed i
