Casein Kinase 2 Inhibitor, CX-4945, Induces Apoptosis and Restores Blood-Brain Barrier Homeostasis in In Vitro and In Vivo Models of Glioblastoma.

Background: In oncology, casein kinase 2 (CK2), a serine/threonine kinase, has a dual action, regulating cellular processes and acting as an oncogenic promoter. Methods: This study examined the effect of CX-4945, a selective CK2 inhibitor, in a human U-87 glioblastoma (GBM) cell line, treated with CX-4945 (5, 10, and 15 μM) for 24 h. Similarly, the hCMEC/D3 cell line was used to mimic the blood-brain barrier (BBB), examining the ability of CX-4945 to restore BBB homeostasis, after stimulation with lipopolysaccharide (LPS) and then treated with CX-4945 (5, 10, and 15 μM). Results: We reported t

Maternal supplementation with a casein hydrolysate and yeast beta-glucan from late gestation through lactation improves gastrointestinal health of piglets at weaning.

Improving maternal nutrition during pregnancy/lactation is a promising strategy to maximise the intestinal health of piglets undergoing abrupt weaning under commercial production conditions. This experiment investigated the effects of maternal supplementation of a casein hydrolysate and yeast β-glucan (CH-YBG) from day 83 of gestation until weaning (day 28) on sow faecal microbial populations and measures of piglet gastrointestinal health parameters at weaning. Sows (n = 10 sows/group) were assigned to: (1) control diet, and (2) control diet + CH-YBG. Maternal supplementation increased the abu

Antibodies to bovine serum albumin in human sera: problems and solutions with casein-based ELISA in the detection of natural Japanese encephalitis virus infections.

An ELISA system for measuring antibodies to nonstructural protein 1 (NS1) of Japanese encephalitis virus has already been established. This system uses an ELISA diluent containing casein, instead of bovine serum albumin (BSA). During a survey, we found that 21 (21%) of 102 children aged 1-5 years, who had no history of Japanese encephalitis vaccination and were without detectable neutralizing antibodies, showed positive results with this ELISA system. Western blotting analysis showed that sera from 19 (91%) of these 21 subjects had antibodies to BSA, but not NS1. These sera reacted with BSA an

Purified chickpea or lentil proteins impair VLDL metabolism and lipoprotein lipase activity in epididymal fat, but not in muscle, compared to casein, in growing rats.

BACKGROUND: It is well known that the legume proteins have a lowering effect on plasma cholesterol and triacylglycerols (TG) concentrations compared to animal proteins. The protein itself, as well as non-protein constituents, naturally present in legumes may be implicated. AIM OF THE STUDY: The effects of various dietary purified legumes proteins compared to casein, were determined on plasma TG level, VLDL concentration and composition. Moreover, lipoprotein lipase (LPL) activity in epididymal fat, gastrocnemius and heart was investigated to evaluate in these tissues their capacity to release

Adjunct effect of immunostimulating hexapeptide analogous to human beta-casein fragment (54-59) to sodium stibogluconate against experimental visceral leishmaniasis.

Visceral leishmaniasis (VL) is a major public health problem in many tropical countries of the world. The available chemotherapeutics require parenteral administration and have other limitations like cost, toxicity, variable efficacy or restricted supplies. There is no effective treatment for immunosuppressed patients with leishmaniasis- HIV co-infection. Hence, new therapies, that are effective when treatment with the currently available drugs fails, must be developed. One of the major strategies for effective and safe treatment of leishmaniasis and other infectious diseases, in the last deca

α- and β-Casein Differentially Protect Anthocyanins from Three Red Grape (Vitis vinifera L.) Cultivars During Simulated Gastrointestinal Digestion.

Grapes are valued for their high anthocyanin content, but the biological efficacy of these compounds is limited by their low digestive recovery. This study evaluated whether milk caseins improve the digestive recovery of anthocyanins from three red grape (Vitis vinifera L.) cultivars. Anthocyanins were profiled by HPLC-DAD-ESI-MS, total phenolic and flavonoid contents and antioxidant capacity were determined spectrophotometrically, the effect on cell viability was assessed on B16-F10 murine melanoma cells, and the digestive recovery of anthocyanins was measured after standardized static in vit…

β-Casein Polymorphism as a Potential Evolutionary Trade-Off: The Rise of A1 Under Intensive Selection and Its Implications for Gastrointestinal Tolerance and Agroecological Resilience.

This narrative review summarizes evidence on the bovine β-casein (CSN2) A1/A2 polymorphism as a case study of how intensive dairy selection and global gene flow can reshape allele frequencies in ways that matter for consumers, processing and agroecological resilience. We draw together evidence from (i) population-genetic surveys of CSN2 in contrasting cattle populations, including a descriptive summary of published genotype-frequency studies; (ii) controlled human studies that separate A1-containing from A2-only dairy exposure; and (iii) dairy technology and the authenticity literature relevan

Co-delivery of vitamin D and probiotics and their synergistic improvement of intestinal functions in simulated microgravity rats via zein/sodium caseinate-based microcapsules.

Long-term microgravity disrupts astronauts' intestinal homeostasis, causing gut dysbiosis, barrier injury and immune imbalance among other issues. Vitamin D (VD) and probiotics may provide synergistic protection, but their synchronous and stable gastrointestinal delivery remains a key challenge. In this study, zein and sodium caseinate (NaCas) were used as wall materials to fabricate Vitamin D3 (VD3)-loaded nanoparticles (ZND) by anti-solvent precipitation. ZND and Lactobacillus rhamnosus GG (LGG) were then co-encapsulated into microcapsules (ZND-loaded LGG microcapsules, ZND-L) via complex co

Casein hydrolysate promotes intestinal repair in adult mice with Salmonella enteritis through a lactate-GPR81-Wnt3A axis in intestinal stem cell niche.

BACKGROUND: Salmonella enterica serovar Typhimurium (S. Typhimurium) is a major enteric pathogen that causes severe intestinal damage, characterized by epithelial disruption, inflammation, and impaired regeneration. Nutritional intervention has emerged as a promising strategy to mitigate such injury and promote mucosal repair. Casein enzymatic hydrolysate (CEH) possesses well-documented anti-inflammatory and mucosa-protective properties. However, whether CEH exerts its protective effects against S. Typhimurium-induced intestinal injury by modulating the gut microbiota, and the underlying mecha

Casein kinase 1α mediates phosphorylation of the Merkel cell polyomavirus large T antigen for β-TrCP destruction complex interaction and subsequent degradation.

UNLABELLED: Merkel cell polyomavirus (MCPyV) is a double-stranded tumor virus that is the main causative agent of Merkel cell carcinoma (MCC). The MCPyV large T antigen (LT), an essential viral DNA replication protein, maintains viral persistence by interacting with host Skp1-Cullin 1-F-box (SCF) E3 ubiquitin ligase complexes, which subsequently induces LT's proteasomal degradation, restricting MCPyV DNA replication. SCF E3 ubiquitin ligases require their substrates to be phosphorylated to bind them, utilizing phosphorylated serine residues as docking sites. The MCPyV LT unique region (MUR) is

The chloroplast-localized casein kinase II α subunit, CPCK2, negatively regulates plant innate immunity through promoting S-nitrosylation of SABP3.

The casein kinase II (CK2) complex consists of catalytic (α) and regulatory (β) subunits and is highly conserved throughout eukaryotes. Plant CK2 plays critical roles in multiple physiological processes; however, its function in plant immunity remains obscure. In this study, we demonstrated that the unique chloroplast-localized CK2 α subunit (CPCK2) is a negative regulator of Arabidopsis thaliana innate immunity. cpck2 mutants displayed enhanced resistance against the fungal pathogen powdery mildew, Golovinomyces cichoracearum and the virulent bacterial pathogen, Pseudomonas syringae pv. tomat

Efficient in vitro digestion of lipids and proteins in bovine milk fat globule membrane ingredient (MFGMi) and whey-casein infant formula with added MFGMi.

The relative immaturity of the infant digestive system has the potential to affect the bioavailability of dietary lipids, proteins, and their digested products. We performed a lipidomic analysis of a commercial bovine milk fat globule membrane ingredient (MFGMi) and determined the profile of lipids and proteins in the bioaccessible fraction after in vitro digestion of both the ingredient and whey-casein-based infant formula without and with MFGMi. Test materials were digested using a static 2-phase in vitro model, with conditions simulating those in the infant gut. The extent of digestion and

Gouda Cheese with Modified Content of β-Casein as a Source of Peptides with ACE- and DPP-IV-Inhibiting Bioactivity: A Study Based on In Silico and In Vitro Protocol.

In silico and in vitro methods were used to analyze ACE- and DPP-IV-inhibiting potential of Gouda cheese with a modified content of β-casein. Firstly, the BIOPEP-UWM database was used to predict the presence of ACE and DPP-IV inhibitors in casein sequences. Then, the following Gouda cheeses were produced: with decreased, increased, and normative content of β-casein after 1 and 60 days of ripening each (six variants in total). Finally, determination of the ACE/DPP-IV-inhibitory activity and the identification of peptides in respective Gouda-derived water-soluble extracts were carried out. The i

Post-weaning A1/A2 β-casein milk intake modulates depressive-like behavior, brain μ-opioid receptors, and the metabolome of rats.

The postnatal period is critical for brain and behavioral development and is sensitive to environmental stimuli, such as nutrition. Prevention of weaning from maternal milk was previously shown to cause depressive-like behavior in rats. Additionally, loss of dietary casein was found to act as a developmental trigger for a population of brain opioid receptors. Here, we explore the effect of exposure to milk containing A1 and A2 β-casein beyond weaning. A1 but not A2 β-casein milk significantly increased stress-induced immobility in rats, concomitant with an increased abundance of Clostridium hi

The regulation of protein kinase casein kinase II by apigenin is involved in the inhibition of ultraviolet B-induced macrophage migration inhibitory factor-mediated hyperpigmentation.

Ultraviolet (UV) radiation elicits melanogenesis and pigmentation in the skin. Apigenin (4',5,7-trihydroxyflavone [AGN]) is a plant flavone contained in various herbs, fruits, and vegetables. We herein investigated antimelanogenic properties of AGN and the molecular mechanisms of the action of AGN. In UVB-treated mice, AGN inhibited cutaneous hyperpigmentation and macrophage migration inhibitory factor (MIF) expression as a melanogenesis-related key factor. In mouse keratinocytes, AGN inhibited the expression of MIF and also the related factors (e.g., stem cell factor and proteinase-activated

Endoplasmic Reticulum Stress Signalling Induces Casein Kinase 1-Dependent Formation of Cytosolic TDP-43 Inclusions in Motor Neuron-Like Cells.

Motor neuron disease (MND) is a progressive neurodegenerative disease with no effective treatment. One of the principal pathological hallmarks is the deposition of TAR DNA binding protein 43 (TDP-43) in cytoplasmic inclusions. TDP-43 aggregation occurs in both familial and sporadic MND; however, the mechanism of endogenous TDP-43 aggregation in disease is incompletely understood. This study focused on the induction of cytoplasmic accumulation of endogenous TDP-43 in the motor neuronal cell line NSC-34. The endoplasmic reticulum (ER) stressor tunicamycin induced casein kinase 1 (CK1)-dependent

An Inhibitor of Casein Kinase 1ε/δ (PF670462) Prevents the Deterioration of Dextran Sodium Sulfate-induced Ulcerative Colitis Caused by UVB Eye Irradiation.

Although we previously reported the exacerbation of dextran sodium sulfate (DSS)-induced ulcerative colitis by ultraviolet (UV) B eye irradiation, we do not yet understand the mechanism behind this phenomenon. In this study, we examined the relationship between the deterioration of DSS-induced ulcerative colitis and clock genes. We induced a mouse model of ulcerative colitis by administering DSS for 5 days, and administered UVB eye irradiation on each day of the DSS treatment period. The DSS-induced ulcerative colitis was deteriorated by the UVB eye irradiation. The levels of Clock, brain and

Whey protein lowers systolic blood pressure and Ca-caseinate reduces serum TAG after a high-fat meal in mildly hypertensive adults.

Epidemiological studies show an inverse association between dairy consumption and blood pressure (BP) but there are few data on the postprandial effects of milk proteins. This study examined their effects, compared to maltodextrin, on postprandial BP and other CVD risk markers in volunteers with mild and pre-hypertension over an 8 h period. In this double-blinded, randomised, cross-over, controlled study 27 adults ingested a high-fat, isoenergetic breakfast and lunch with 28 g whey protein, 28 g Ca-caseinate or 27 g maltodextrin. Whey protein reduced systolic BP compared with Ca-caseinate (-15
