Antioxidants delay the onset of thyroiditis in obese strain chickens.

Dietary iodine has been shown to be important in the induction of thyroiditis in susceptible chicken strains although the underlying mechanism remains unknown. Iodine may exert its effects through the formation of reactive oxidative radicals which would cause thyroidal injury and initiate infiltration. We have tested this hypothesis by examining the ability of butylated hydroxyanisole (BHA), ethoxyquin, and other antioxidants to prevent thyroiditis in Obese strain (OS) chickens, a strain that develops severe disease by 4 weeks of age. BHA, when administered from hatching until death at 5 weeks…

Immunogenetic analysis of spontaneous autoimmune thyroiditis of obese strain chickens.

A variety of immunological, endocrinological, and virological abnormalities have been implicated in the etiopathogenesis of spontaneous autoimmune thyroiditis (SAT) of Obese strain (OS) chickens, e.g., a general T cell hyperreactivity, an increased uptake of iodine into the thyroid gland, a diminution of the glucocorticoid tonus, and an OS-specific endogenous virus. In crosses of the close-bred OS B15/B15 subline with the inbred normal CB B12/B12 strain we have studied the mode of inheritance of these aberrations and their putative association with SAT. The results indicate that none of these …

Lesions of right heart failure and ascites in broiler chickens.

A naturally occurring cardiomyopathy in broiler chickens from a single Ontario farm was studied in order to define the morphologic changes. Gross and histologic features of affected birds were compared with those in age-matched control penmates. Body weight and weight and volume of individual cardiac chambers were measured. Histologic sections of 18 different tissues were examined, and lesions observed were scored subjectively. Affected birds were stunted and had marked right ventricular dilation and hypertrophy, atrial hypertrophy, ascites, pulmonary congestion and edema, and hepatic capsular…

Infectious stunting and pancreatic fibrosis in broiler chickens in Saskatchewan.

Two broiler flocks contained 0.5 and 3% small chickens. The small chickens were approximately one-third the size of their penmates, were very active, and retained much of their chick down. They had distended abdomens with full intestinal tracts, often containing undigested feed. In many, the pancreases were thin, white, and firm because of loss of exocrine tissue and replacement by fibrous tissue. Many had skeletal changes suggestive of rickets. In a survey of 48 broiler flocks for small birds and pancreatic lesions, five flocks had a noticeable incidence of small chickens, varying from less t…

Hypothyroidism: an early sign of vitamin A deficiency in chickens.

The effect of a marginal vitamin A deficiency on thyroid function was investigated in chicks gaining weight. This study examined the hypothesis that hypothyroidism is one of the earliest manifestations of hypovitaminosis A in chicks. Day-old Leghorn cockerels were fed vitamin A-adequate and deficient diets. Blood vitamin A levels were reduced in the deficient chicks by 3 weeks of age. Body weight gains were less in the 5-, 6-, 7- and 8-week-old A-deficient chicks than in controls. Compared to controls, vitamin A-deficient chicks were characterized by thyroid follicle hypertrophy by 4 weeks, he

The course of Trypanosoma brucei ssp. infection in domestic chickens.

Three unrelated stocks of Trypanosoma (Trypanozoon) brucei were tested for their ability to infect domestic chickens. One of them, Lugala/55/EATRO/459, regularly produced a chronic infection. This was characterized by a low parasitemia (3-100 mouse infective organisms per ml of blood) that lasted for nine months in pullets and over one year in cockerels. Advances and remissions of the parasitemia were detected during the course of the infection. There was no obvious impairment of the health of the birds and their growth rate was indistinguishable from that of uninfected chickens. A second stoc…

Pathogenesis of spontaneous autoimmune thyroiditis in Obese strain (OS) chickens.

Chickens of the Obese strain (OS) develop a spontaneous, hereditary autoimmune thyroiditis during the first weeks of life which parallels human Hashimoto's thyroiditis in all clinical, histopathological and serological aspects. This review summarizes the results from investigations on this strain with special emphasis on the pathogenic effector mechanisms leading to the autoimmune destruction of the thyroid gland. The fact that this model disease arises in an avian species is particularly advantageous because of the clear-cut anatomical and functional division of the immune system. Data are di…

Susceptibility of broiler chickens to Clostridium botulinum type C toxin.

As broiler chickens aged during the first eight weeks of life, they decreased in susceptibility to the lethal effects of Clostridium botulinum type C toxin. Eight-week-old broilers were 2,000 times less susceptible to the lethal effects of the toxin than hatched chicks. Different broiler crosses showed similar susceptibility to the lethal effects of type C toxin at seven weeks of age, but 2.5 times more toxin was required to produce lethality by oral inoculation than was required by subcutaneous inoculation.

Spontaneous autoimmune thyroiditis in chickens. II. Evidence for autoresponsive thymocytes.

A major factor involved in the spontaneously occurring autoimmune thyroiditis of OS chickens is the presence of thyroid reactive cells in the thymus of young OS chicks. This was determined by transferring OS and normal strain thymocytes to T cell-depleted histocompatible normal chickens and examining the recipients for thyroiditis. Although both thymocyte populations reconstituted T cell function, only the OS thymocytes induced thyroiditis. Additional evidence for hyperactivity of the young OS thymus was provided by studying the in situ uptake of a radiolabeled thymidine analogue.

[Protective effect of alpha-linolenic acid in encephalomalacia in chickens].

Encephalomacia is a vitamin E deficiency syndrome which affects the cerebellum of young chicks. The lesion includes degenerative alterations of cellular and fibrillar elements, apparently as the result of the ischaemia caused by thrombotic events in the microvascular system. A supply of linoleic acid, as fatty acid methyl esters prepared from safflower oil (Carthamus tinctorius), caused a high incidence of encephalomalacia. On the other hand, linseed oil esters, rich in alpha-linolenic acid, did not induce any symptoms and protected the chicks to a large extend against the development of signs

Inhibition of the glycogen phosphorylase system during ochratoxicosis in chickens.

Graded doses of ochratoxin A incorporated into the diet (0, 0.5, 1.0, 2.0, 4.0, and 8.0 micrograms/g) of broiler chickens significantly (P < 0.05) inhibited activity of protein kinase, the initiator enzyme of the glycogen phosphorylase system, in the livers at all dose levels. Only the highest dose, 8.0 micrograms/g, significantly reduced the total activity of phosphorylase kinase, which is activated by protein kinase. The total activity of phosphorylase, which is activated by phosphorylase kinase, was unaltered by ochratoxin A at any level. Additon of ochratoxin A to liver extracts control bi…

Reduced symptoms of anaphylaxis in chickens by ACTH or heat.

Exogenous ACTH, when administered intramuscularly in four equal doses of 2 I.U./100 gm. of body weight at 24, 23, 22, and 21 hours prior to challenge with BSA, reduced the severity of anaphylaxis in BSA sensitized juvenile chickens. Additionally, a single heat exposure (45 degrees C.) of 30 minutes at 24 hours prior to challenge consistently reduced the severity of anaphylaxis. Single heating exposures at 24 hours prior to sensitization or at 48 hours prior to challenge did not influence the anaphylactic response. In one of two experiments single heat exposures at one hour prior to challenge r…

Hypergammaglobulinemia in chickens congenitally infected with an avian leukosis virus.

Significantly elevated (2- to 5-fold higher than controls) serum levels of IgG were found in chickens congenitally infected with F42 strain of avian leukosis (ALV-F42) a subgroup A avian leukosis virus (ALV). A further increase in IgG levels in congenitally infected birds was found to be induced by injection of influenza virus in complete Freund's adjuvant(CFA). Serum immunoglobulin M (IgM) levels were not significantly elevated in ALV congenitally infected chickens except in those animals that had been injected with influenza virus in CFA. Hypergammaglobulinemia in ALV infected birds resulted…
