Isolation of Salmonella and Shigella from fish harvested from the Winam Gulf of Lake Victoria, Kenya.

BACKGROUND: Human infections caused by pathogens transmitted from fish are quite common. The aim of this study was to isolate enteric pathogenic bacteria from fish that might be transmitted to humans after the handling or consumption of such fish. METHODOLOGY: One hundred and twenty Nile tilapia fish harvested using various fishing methods were collected from fishermen in five fish landing beaches within Winam Gulf and disinfected externally using 70% ethyl alcohol for 2 minutes then washed three times with autoclaved distilled water. Isolation of Salmonella and Shigella species from fish sam

Social opportunity rapidly regulates expression of CRF and CRF receptors in the brain during social ascent of a teleost fish, Astatotilapia burtoni.

In social animals, hierarchical rank governs food availability, territorial rights and breeding access. Rank order can change rapidly and typically depends on dynamic aggressive interactions. Since the neuromodulator corticotrophin releasing factor (CRF) integrates internal and external cues to regulate the hypothalamic-pituitary adrenal (HPA) axis, we analyzed the CRF system during social encounters related to status. We used a particularly suitable animal model, African cichlid fish, Astatotilapia burtoni, whose social status regulates reproduction. When presented with an opportunity to rise

The use of mitochondrial and nuclear sequences in prospecting for cryptic species in Tabascotrema verai (Digenea: Cryptogonimidae), a parasite of Petenia splendida (Cichlidae) in Middle America.

In this study, we used sequences of mitochondrial and nuclear markers to test the hypothesis that Tabascotrema verai Lamothe-Argumedo and Pineda-López, 1990, a parasite of the cichlid Petenia splendida Günther, 1862 in Middle America, represents a single species, following a molecular prospecting approach. One-hundred and five individuals of T. verai were collected from the intestine of 43 specimens of P. splendida in eight localities of Mexico, Belize and Guatemala. A portion (n=53) of the collected specimens was used for molecular studies, whereas the remaining individuals were processed for

Fine mapping and functional annotation of a QTL for resistance to tilapia lake virus in Nile tilapia (Oreochromis niloticus).

Disease resistance is one of the main targets of animal breeding programs. In recent years, incorporating genomic information to accelerate genetic progress has become one of the priorities of the industry. Here, we combined population-scale whole-genome sequencing with differential gene expression and functional annotation analyses to study resistance to tilapia lake virus (TiLV) in a breeding Nile tilapia (Oreochromis niloticus) GIFT population. Fish with survival data from a natural TiLV outbreak were sampled and genotyped for 6.7 M SNPs using whole-genome resequencing and imputation. Our r

Time-dependent biphasic dysregulation of autophagy and innate immunity underlies polystyrene microplastics-induced hepatotoxicity in Nile tilapia.

The immunotoxicity of microplastic pollution in aquatic species is an issue of growing concern. However, the time course and mechanistic crosstalk underlying microplastic-induced hepatic injury remain poorly defined. This study examined the temporal hepatic response of Nile tilapia (Oreochromis niloticus) to 100 nm polystyrene microplastics (PS) over a 21-day exposure period, covering acute (7 days) and sub-chronic (14 and 21 days) phases. Histopathological evaluation showed no obvious liver lesions in exposed fish relative to controls at 7 days post-exposure (dpe). In contrast, progressive ti

Effects of dietary phytic acid on growth, antioxidant status, hepatic metabolism, and immune-related gene expression in Nile tilapia (Oreochromis niloticus).

Phytic acid (PA) is a common anti-nutritional factor in plant-based aquafeeds that can affect nutrient utilization and physiological homeostasis in fish. However, its dose-dependent physiological effects on Nile tilapia (Oreochromis niloticus) remain poorly characterized. This study investigated the effects of graded dietary levels of PA (0, 10, 15, 20, and 25 g/kg) on growth performance, antioxidant status, hepatic metabolism, and immune regulation in Nile tilapia. Results showed that dietary PA significantly reduced body weight gain and increased liver somatic index in a dose-dependent manne

Synergistic impact of temperature rises and ferric oxide nanoparticles on biochemical and oxidative stress biomarkers in Oreochromis niloticus: relevant environmental risk assessment under predicted global warming.

Global warming and contamination of freshwater environments with nanoparticles (NPs) pose a global threat to biodiversity. Numerous studies demonstrated the effects of increasing temperatures and NPs separately, but their combined impact on aquatic life remains poorly understood or unstudied, particularly under predicted rising temperatures resulting from global warming (+ 2 and + 4 °C). So, the present study aims to determine how the temperature rises affect the toxicological characteristics of ferric oxide nanoparticles (Fe2O3 NPs) on the prevalent freshwater fish, Nile tilapia (Oreochromis

Immune Response and Protective Efficacy of β-Glucan and Alkoxyglycerol as Adjuvant in Streptococcus agalactiae Formalin-Inactivated Vaccine in Nile Tilapia (Oreochromis niloticus).

Nile tilapia (Oreochromis niloticus) farms face streptococcus agalactiae infections causing mass mortality following economic losses. To date, no commercial fish vaccine is available in India to protect against streptococcosis due to a lack of proper adjuvants and vaccine targets. Multiple studies justify that immunomodulators like β-glucan and Alkoxy glycerol (AKG) have improved immunity in fish. β-glucan and AKG are natural homopolymers and lipids, respectively, which have proven immunostimulant capabilities in fish and can be effectively used as a prominent adjuvant to improve vaccine effic

dmrt1 Is Responsible for Androgen-Induced Masculinization in Nile Tilapia.

17α-Methyltestosterone (MT) is a widely used androgen for all-male fish production in aquaculture. However, the molecular mechanism underlying MT-induced masculinization remains unclear. In this study, we aim to identify the key gene responsible for MT-induced masculinization using the Nile tilapia (Oreochromis niloticus) amhy, dmrt1, and gsdf mutants, which exhibit male-to-female sex reversal. Nile tilapia fry from these three mutant lines were treated with 50 μg/g MT from 5 to 30 days after hatching (dah). The results showed that amhy and gsdf mutants, but not dmrt1 mutants, were masculinize

Designing and evaluating a novel blood-brain barrier in vitro model of teleost for reproducing alterations in brain infecting.

A new blood-brain barrier (BBB) in vitro model of tilapia (Oreochromis niloticus) was successfully established by co-culture with brain microvascular endothelial cell line TVEC-01 and brain astrocyte cell line TA-02 of tilapia. Experiments with the expression levels of BBB-related genes, TEER value detection and 4-h water-leaking test have shown that the BBB in vitro model has an excellent barrier effect. In bacterial penetration experiments, the pathogenic strain of Streptococcus agalactiae was able to pass through the BBB in vitro model and initiate a series of immune responses, among which
