Visceral leishmaniasis (VL) is a neglected parasitic disease, and the first-line treatments for VL include drugs that exhibit serious toxicological issues. In this sense, new molecules are sought for VL treatment, such as Carvacrol (Car), a phenolic monoterpene that has shown strong activity against Leishmania spp. However, its low solubility prevents its free administration, requiring a new therapeutic strategy such as encapsulation in chitosan biopolymeric nanoparticles. This study aimed to develop chitosan biopolymeric nanoparticles (NPChi) encapsulating Car (NPCar) and evaluate their in vi

BACKGROUND: Streptococcus mutans is a well-known oral pathogen that plays a critical role in the development of dental caries. Many studies have been directed to discover the chemical compounds present in natural products to inhibit the growth and biofilm formation activity of S. mutans. Thymus essential oils exhibit good inhibition on the growth and pathogenesis of S. mutans. However, details about the active compounds in Thymus essential oil and the inhibition mechanism still remain unclear. The aim of this study was to investigate the antimicrobial activity of 6 Thymus species (Three sample

The antileishmanial activity of the essential oil (EO) from Chenopodium ambrosioides L. has been demonstrated in vitro and in animal models, attributed to the major components of the EO. This study focused on the effects of the three major EO compounds carvacrol, caryophyllene oxide (Caryo), and the antileishmanial endoperoxide ascaridole (Asc) on mitochondrial functions in Leishmania tarentolae promastigotes (LtP). EO and Caryo were able to partially inhibit the leishmanial electron transport chain, whereas other components failed to demonstrate a direct immediate effect. Caryo demonstrated i

Carvacrol has been described as an agonist/antagonist of different transient receptor potential (TRP) channels and voltage-dependent calcium channels (Cavs). The aim of this study was to evaluate the role of Cav and TRP channels following carvacrol stimulation. Initially, in mesenteric artery rings carvacrol relaxed phenylephrine-induced contractions. Furthermore, carvacrol inhibited contraction elicited by CaCl2 in depolarizing nominally without Ca2+ medium and antagonized the contractions induced by S(-)-Bay K 8644 and inhibited Ca2+ currents indicating the inhibition of Ca2+ influx through

BACKGROUND: Biofilms of fungi such as Candida albicans (C. albicans) can survive even at cold temperatures and are generally difficult to eradicate with well-known antimicrobials. The aim of this study was to develop a novel synergistic technique against C. albicans biofilms using low concentrations of propylene glycol (PG), as a penetration enhancer. It helps naturally-derived antimicrobials [caprylic acid (CA) and carvacrol (CAR)] permeate C. albicans biofilms and cell membranes within short times even in cold environments.
METHODS: C. albicans biofilms formed on stainless steel coupons wer

Aging is one of the risk factors involved in the development of erectile dysfunction (ED). Growing evidence suggests that oxidative stress is the critical mediator of changes in endothelial function and penile vascular tone in the aging process. Thus, reducing reactive oxygen species (ROS) levels may preserve the bioactivity of the penile vasculature. Antioxidant compounds, such as carvacrol, limit the damage caused by ROS and, therefore, benefit the treatment of ED. Thus, this study aims to evaluate the effects of carvacrol on ED using the D-( +)-galactose aging model. The animals were divide

Arsenic (As) is a highly toxic metalloid. Carvacrol (CAR) is the active ingredient of Lamiaceae plants and has various biological and pharmacological properties. The present study investigated the protective effects of carvacrol (CAR) against testicular toxicity induced by sodium arsenite (SA). Rats were given SA (10 mg/kg) and/or CAR (25 or 50 mg/kg) for 14 days. Semen analyzes showed that CAR increased sperm motility and decreased the percentage of abnormal and dead sperm. It was determined that the oxidative stress induced by SA decreased with the increase of Nrf-2 and HO-1 expressions, SOD

Chemical stability and metal speciation are key determinants of the biological behavior and translational potential of metal-based chemotherapeutic agents, for which ligand exchange and chemical transformation frequently occur under physiological conditions. In these systems, pharmacological activity is defined not by a single molecular entity but by the distribution and interconversion of metal-containing species in solution and biological media. Here, we investigate the time-dependent speciation of the highly water-soluble ruthenium complex [(η6-p-cymene)Ru(κ-N,O-Ph₃P=N-CO-2-NC₅H₄)]Cl (Ru-IM
