Comparative Investigation of Coincident Single Nucleotide Polymorphisms Underlying Avian Influenza Viruses in Chickens and Ducks.

Avian influenza is a severe viral infection that has the potential to cause human pandemics. In particular, chickens are susceptible to many highly pathogenic strains of the virus, resulting in significant losses. In contrast, ducks have been reported to exhibit rapid and effective innate immune responses to most avian influenza virus (AIV) infections. To explore the distinct genetic programs that potentially distinguish the susceptibility/resistance of both species to AIV, the investigation of coincident SNPs (coSNPs) and their differing causal effects on gene functions in both species is imp…

Hedyotis diffusa alleviate aflatoxin B1-induced liver injury in ducks by mediating Nrf2 signaling pathway.

Aflatoxin B1 (AFB1), the most harmful aflatoxins, is a frequent contamination in feed and food items, raising global concerns in animal production and human public health. Also, AFB1 induces oxidative stress, cytotoxicity, mutations, and DNA lesions through its metabolic transformation into aflatoxin B1-8,9-epoxide (AFBO) by cytochrome P450 (CYP450). Hedyotis diffusa (HD) is a traditional Chinese herbal medicine known for its multiple pharmacological activities, including antioxidant, anti-inflammatory, and immunomodulatory. Yet, the influence of HD on AFB1-induced liver injury in ducks is sti…

Alleviation of Catching and Crating Stress by Dietary Supplementation of Bacillus subtilis in Pekin Ducks.

Catching and crating may elicit stress and fear reactions in poultry because the procedures involve human contact and exposure to a novel environment. This study determined the effects of dietary probiotic supplementation on physiological stress, underlying fear, and growth performance of Pekin ducks subjected to catching and 4 h of crating. The study used a 2 × 2 factorial arrangement; the main factors were diet (basal or basal + probiotic) and crating durations (0 or 4 h). From 1 to 21 days of age (doa), birds were fed a basal or basal + probiotic (CLOSTAT® (Bacillus subtilis) (Kemin Industr…

The Microbiota Contributes to the Control of Highly Pathogenic H5N9 Influenza Virus Replication in Ducks.

Ducks usually show little or no clinical signs following highly pathogenic avian influenza virus infection. In order to analyze whether the microbiota could contribute to the control of influenza virus replication in ducks, we used a broad-spectrum oral antibiotic treatment to deplete the microbiota before infection with a highly pathogenic H5N9 avian influenza virus. Antibiotic-treated ducks and nontreated control ducks did not show any clinical signs following H5N9 virus infection. We did not detect any significant difference in virus titers neither in the respiratory tract nor in the brain …

Dietary supplementation of 25-hydroxycholecalciferol increases tibial mass by suppression bone resorption in meat ducks.

Leg problems often result from the rapid weight gain and poor bone quality in modern ducks, leading to a high risk of fractures and continuous pain. We hypothesized that improving bone quality in combination with delaying weight gain via a low nutrient density (LND) diet probably reverses these skeletal abnormalities. Studies indicated that 25-hydroxycholecalciferol (25-OH-D3), a vitamin D3 metabolite, is effective in treating bone-related disorders. Therefore, Exp. 1 evaluated the effects of 25-OH-D3 on tibial mass of meat ducks. Male meat ducklings were fed a standard nutrient density diet (…

Development of a colloidal gold immunochromatographic strip for rapid detection of Riemerella anatipestifer in ducks.

Riemerella anatipestifer is one of the major bacterial pathogens of ducks and causes significant economic losses in poultry agriculture. Usually, methods for detecting R. anatipestifer infection need specialized equipment and highly skilled personnel. In this study, a novel colloidal gold immunochromatographic strip was developed for rapid detection of R. anatipestifer in ducks. The monoclonal antibodies 2D5 and 2A6 against R. anatipestifer were used as colloidal gold-labeled protein and capture protein, respectively, to recognize the bacteria in tryptic soy broth medium culture and in hearts …

Host specificity and age-dependent resistance to Cryptosporidium avium infection in chickens, ducks and pheasants.

Host- and age-specificity of Cryptosporidium avium were studied in 1-, 21- and 365-day-old chickens (Gallus gallus), domestic ducks (Anas platyrhynchos) and ring-necked pheasants (Phasianus colchicus) under experimental conditions. Cryptosporidium avium was not infectious for ring-necked pheasants, but it was infectious for ducks and chickens at all age categories. The course of infection in ducks did not differ among age categories, but 365-day-old chickens had less severe infections than 1- and 21-day-old chickens. The patent period in chickens and ducks was >30 DPI, but ducks started to she…

Isolation and genetic characterization of a novel adeno-associated virus from Muscovy ducks in China.

Adeno-associated virus (AAV; genus Dependoparvovirus, family Parvoviridae) was first discovered in 1965 as a contaminant in adenovirus preparations. The AAVs are generally considered non-pathogenic, and they have the ability to attenuate the replication of other more pathogenic viruses, which makes them attractive as potential therapeutics or preventative measures. This study characterized a novel AAV isolated from Muscovy ducks in China. The novel virus (MHH-05-2015) was isolated after propagating a field isolate of the DAdV-3 virus (a type 3 duck adenovirus) in duck embryo fibroblasts. The f…

Cloacotaenia megalops (Nitzsch in Creplin, 1829) (Cestoda, Hymenolepididae) in wild ducks in Western Pomerania, Poland.

Cloacotaenia megalops (Nitzsch in Creplin, 1892) is a polyxenic and cosmopolitan tapeworm from the family Hymenolepididae. Its generic name derives from their typical location (cloaca), and the typical final hosts which are birds typically associated with water and marsh environments: Anseriformes, Galliformes and Gruiformes. In Poland, the presence of C. megalops has been observed so far in 16 species of ducks from the Baltic coast, the Mazurian Lake District, Wielkopolsko-Kujawska Lowland, Mazovian Lowland, and Podlasie Lowland. In Western Pomerania, quantitative structure analyses were only…

Differences in in vitro cerebellar neuronal responses to hypoxia in eider ducks, chicken and rats.

Ducks are well-known to be more tolerant to asphyxia than non-diving birds, but it is not known if their defences include enhanced neuronal hypoxia tolerance. To test this, we compared extracellular recordings of spontaneous activity in the Purkinje cell layer of 400 mum thick isolated cerebellar slices from eider ducks, chickens and rats, before, during and after 60 min hypoxia (95%N(2)-5%CO(2)) or chemical anoxia (hypoxia + 2 mM NaCN). Most slices rapidly lost activity in hypoxia, with or without recovery after rinse and return to normoxia (95%O(2)-5%CO(2)), but some maintained spontaneous a…

Mallard ducks - a waterfowl species with high risk of distributing Escherichia coli pathogenic for humans.

Mallard ducks may pose a so far underestimated risk to human and animal health by transmitting pathogenic Escherichia coli via their faecal deposits to various environmental sources. We processed Mallard duck faecal samples for E. coli and unique clones, as defined by pulsed-field gel electrophoresis (PFGE), were subsequently investigated for their virulence genotype and phylogenetic background. Multilocus sequence typing and in vivo experiments were performed for selected clones. Of 60 clones identified among 142 E. coli isolated from 175 samples, 15 (25%) were recovered from multiple individ…

Antibodies to flaviviruses in wild ducks captured in Hokkaido, Japan: risk assessment of invasive flaviviruses.

Recently, the distribution of Japanese encephalitis virus (JEV) and West Nile virus (WNV) has expanded into new territories. The invasion of WNV into Japan is of great concern. The migration of birds is suggested to be involved in the expanded distribution of these flaviviruses. In this study, 92 wild ducks--20 Anas poecilorhyncha (migratory breeders), 50 Anas platyrhynchos (undetermined), 16 Anas acuta (winter visitors), and 6 Anas penelope (winter visitors)--were captured in autumn of 2005 and 2006, in the central part of Hokkaido, a low JEV activity area. A seroepidemiologic analysis of fla…

Comparison of Sertoli cells in duck testes pre- and post-sexual maturation.

To investigate the role of Sertoli cells in duck testicular development and maturation, we conducted single-cell RNA sequencing on testes from immature (IMT) and mature (MT) ducks. Sertoli cells accounted for 27.75 % and 33.42 % of testicular cells in IMT and MT, respectively. We identified 2,630 differentially expressed genes (DEGs) in mature Sertoli cells, with 1,540 upregulated and 1,090 downregulated. Subtype analysis revealed 2,498 DEGs in Sertoli cell type 1 (SC1) (898 up, 1,600 down) and 2,515 DEGs in SC2 (1,291 up, 1,224 down), including 1,090 SC1-specific, 1,107 SC2-specific, and 1,40…

Brefeldin A inhibits duck plague virus replication by impairing virion envelopment.

Duck plague virus (DPV) is the causative agent of duck plague (DP), an acute and highly fatal disease that inflicts substantial economic losses in the waterfowl industry. The virus acquires its envelope from the endoplasmic reticulum (ER) and Golgi apparatus during virion assembly. Currently, there are no specific antiviral drugs available, making the development of novel anti-DPV drugs critically important. Brefeldin A (BFA) is an antibiotic known to disrupt endoplasmic reticulum-Golgi transport and inhibit the replication of various enveloped viruses. However, its inhibitory effect and mecha
