A study of the inferior mesenteric and pelvic ganglia of guinea-pigs with intracellular electrodes.

1. Ganglion cells in the inferior mesenteric ganglion (IMG) and the pelvic plexus of the guinea-pig were studied using intracellular micro-electrodes.2. Ganglion cells had resting membrane potentials of 55-65 mV. Threshold for initiation of an action potential ranged from 10 to 20 mV depolarization. Action potentials often exceeded 100 mV in amplitude and were followed by an after-hyperpolarization of up to 20 mV.3. Synaptic responses were recorded from cells in the IMG in response to stimulation of the right and left hypogastric nerves, ascending mesenteric, inferior splanchnic and colonic ne

Transmission from pregnanglionic fibres in the hypogastric nerve to peripheral ganglia of male guinea-pigs.

1. Intracellular records were obtained from ganglion cells of the pelvic plexus of male guinea-pigs.2. The input resistance of cells which responded to intracellular stimulation varied from 40 to 150 MOmega. Slope resistance decreased when the membrane was hyperpolarized. Time constants varied from 5 to 200 msec. Resting membrane potentials ranged from 40 to 70 mV.3. Action potentials in response to direct stimulation were followed by a prolonged phase of after-hyperpolarization.4. A second type of cell was also impaled which did not respond to electrical stimulation. These cells had resting m

Neonatal isolation stress alters bidirectional long-term synaptic plasticity in amygdalo-hippocampal synapses in freely behaving adult rats.

The basolateral amygdala (BLA) is known to be involved in emotional and stress responses, while the dentate gyrus (DG), a subfield of the hippocampus, is implicated in learning and memory. Together, the BLA-DG neuronal pathway is thought to link memory with emotional and physiological stress responses. To assess whether neonatal isolation, a known early life stressor, has enduring effects on bidirectional neuroplasticity in adulthood, changes in long-term potentiation (LTP) and long-term depression (LTD) of BLA-DG synapses were recorded in neonatally isolated and non-handled freely behaving ad

Synaptic plasticity in local cortical network in vivo and its modulation by the level of neuronal activity.

Neocortical neurons maintain high firing rates across all behavioral states of vigilance but the discharge patterns vary during different types of brain oscillations, which are assumed to play an important role in information processing and memory consolidation. In the present study, we report that trains of stimuli applied to local neocortical networks of cats, at frequencies that mimic endogenous brain rhythms, produced depression or potentiation of postsynaptic potentials, which lasted for several minutes. This form of synaptic plasticity was not mediated through NMDA receptors since it per

Impaired spatial learning after saturation of long-term potentiation.

If information is stored as activity-driven increases in synaptic weights in the hippocampal formation, saturation of hippocampal long-term potentiation (LTP) should impair learning. Here, rats in which one hippocampus had been lesioned were implanted with a multielectrode stimulating array across and into the angular bundle afferent to the other hippocampus. Repeated cross-bundle tetanization caused cumulative potentiation. Residual synaptic plasticity was assessed by tetanizing a naïve test electrode in the center of the bundle. Spatial learning was disrupted in animals with no residual LTP

Lambert-Eaton sera reduce low-voltage and high-voltage activated Ca2+ currents in murine dorsal root ganglion neurons.

Voltage-gated Ca2+ channels are categorized as either high-voltage activated (HVA) or low-voltage activated (LVA), and a subtype (or subtypes) of HVA Ca2+ channels link the presynaptic depolarization to rapid neuro-transmitter release. Reductions in transmitter release are characteristic of the autoimmune disorder, Lambert-Eaton syndrome (LES). Because antibodies from LES patients reduce Ca2+ influx in a variety of cell types and disrupt the intramembrane organization of active zones at neuromuscular synapses, specificity of LES antibodies for the Ca2+ channels that control transmitter release

Synaptic-type acetylcholine receptors raise intracellular calcium levels in neurons by two mechanisms.

Nicotinic acetylcholine receptors (AChRs) located in the postsynaptic membrane on neurons are responsible for mediating fast, excitatory synaptic transmission. If synaptic AChRs are also highly permeable to calcium as reported recently for several kinds of neuronal AChRs, the synaptic receptors could regulate calcium-dependent events in the neurons in concert with normal transmission. Chick ciliary ganglion neurons have two classes of AChRs, one located predominantly in the synaptic membrane and responsible for synaptic signaling through the ganglion and the other located almost exclusively in

Androgenic modulation of the activity of lumbar neurons involved in the rat bulbocavernosus reflex.

The bulbocavernosus (BC) reflex, produced in the BC motor nerve in response to electrical stimulation of the contralateral pudendal sensory nerve, was investigated in intact, castrated, and testosterone-treated castrated male rats under urethane anesthesia. No significant group differences in the reflex latency, sensory or motor conduction velocity, or central delay were observed. A conditioning pulse to the pudendal sensory nerve caused suppression of the averaged antidromic field potential recorded in the contralateral spinal nucleus of the bulbocavernosus (SNB) after stimulation of the SNB

Receptive fields of rat lamina 1 projection cells move to incorporate a nearby region of injury.

The effects of punctate burns to the skin on the response properties of a group of lamina 1 projection cells in the dorsal horn of the spinal cord has been studied in decerebrate rats. The burns were made 5-15 mm outside the boundaries of the receptive fields (RF) of the cells and they produced a slow increase in the excitability of the cells as seen by an enlargement of RF, towards the lesion, a decreased mechanical threshold and an increased responsiveness to transcutaneous electrical stimulation applied with needle electrodes both inside and outside the RF. These changes were first seen 10-

Electrical signs of an oligosynaptic visual projection to the rat hippocampus.

In rats with pons transection photic or optic nerve stimulation elicits a response in dorsal hippocampus with approximately with same latency, amplitude and time course as the response in striate cortex. After optic nerve stimulation a positive polyphasic deflection with the last peak at 10 ms is followed by a larger biphasic major deflection with peaks at 15 and 25 ms and later, slower deflections at intervals of 125 ms. The polyphasic deflection is maximal in the inferior part of the hippocampus but does not reverse polarity; the other deflections reverse above and below an isoelectric point

Pharmacological evidence that GABA-induced relaxation of rat proximal duodenum longitudinal muscle depends on NKCC cotransporter activity and Ca2+ influx.

γ-Aminobutyric acid (GABA) is the main inhibitory neurotransmitter in adult central nervous system (CNS) synapses, but it excites immature CNS neurons as well as neurons in the myenteric plexus. The present work aimed to determine whether GABA-induced nonadrenergic, noncholinergic (NANC) neuronal-mediated relaxation of the rat duodenum is dependent on the activity of Na+ K+ Cl- cotransporters (NKCC) and requires calcium influx. In the presence of guanethidine (3 µmol/L), atropine (3 µmol/L), and indomethacin (1 µmol/L), relaxations induced by GABA (100 µmol/L), KCl (5-10 mmol/L) and electrical

Presynaptic inhibition and homosynaptic depression: a comparison between lower and upper limbs in normal human subjects and patients with hemiplegia.

Presynaptic inhibition of Ia terminals and postactivation depression at the Ia fibre-motor neuron (MN) synapses were compared in the upper and lower limbs of both sides in subjects from different populations: 49 spastic patients with hemiplegia [mainly with a lesion in the middle cerebral artery (MCA) area], two tetraplegics and 35 healthy subjects. Presynaptic inhibition was assessed using D1 inhibition of the soleus and the flexor carpi radialis (FCR) H reflexes elicited by electrical stimuli applied to the nerve supplying antagonistic muscles, and postactivation depression was explored by v

RIM and RIM-Binding Protein Localize Synaptic CaV2 Channels to Differentially Regulate Transmission in Neuronal Circuits.

At chemical synapses, voltage-gated Ca2+ channels (VGCCs) translate electrical signals into a trigger for synaptic vesicle (SV) fusion. VGCCs and the Ca2+ microdomains they elicit must be located precisely to primed SVs to evoke rapid transmitter release. Localization is mediated by Rab3-interacting molecule (RIM) and RIM-binding proteins, which interact and bind to the C terminus of the CaV2 VGCC α-subunit. We studied this machinery at the mixed cholinergic/GABAergic neuromuscular junction of Caenorhabditis elegans hermaphrodites. rimb-1 mutants had mild synaptic defects, through loosening th

Calcium currents in striatal fast-spiking interneurons: dopaminergic modulation of CaV1 channels.

BACKGROUND: Striatal fast-spiking interneurons (FSI) are a subset of GABAergic cells that express calcium-binding protein parvalbumin (PV). They provide feed-forward inhibition to striatal projection neurons (SPNs), receive cortical, thalamic and dopaminergic inputs and are coupled together by electrical and chemical synapses, being important components of the striatal circuitry. It is known that dopamine (DA) depolarizes FSI via D1-class DA receptors, but no studies about the ionic mechanism of this action have been reported. Here we ask about the ion channels that are the effectors of DA act

Expression of the GABA(A) receptor gamma 4-subunit gene: anatomical distribution of the corresponding mRNA in the domestic chick forebrain and the effect of imprinting training.

The learning process of imprinting involves morphological, electrophysiological and biochemical changes in a region of the chick (Gallus gallus domesticus) forebrain known as the intermediate and medial part of the hyperstriatum ventrale (IMHV). The alterations include increases in the mean length of postsynaptic density profiles of axospinous synapses and the number of N-methyl-D-aspartate (NMDA) receptor binding sites, and changes in spontaneous and evoked electrical activity. Recent immunocytochemical and behavioural studies have suggested that inhibitory GABAergic neurotransmission plays a
