Plasmid profile of ESBL producing Gram-negative bacteria and correlation with susceptibility to beta-lactam drugs.

BACKGROUND: Members of family Enterobacteriaceae can acquire resistance to extended spectrum beta lactams by a number of mechanisms; most important being the plasmid encoded extended spectrum beta lactamase (ESBL) and AmpC beta lactamase. This study has been designed to look for the presence of plasmids and their correlation with drug resistance. METHODS: ESBL production was studied in different gram-negative bacteria and susceptibility testing of ESBL positive isolates was done for various beta lactams, cephalosporins and other commonly used drugs against them. Plasmid DNA isolation of all t

Defining an extended-spectrum beta-lactamase.

The term 'extended-spectrum beta-lactamase' (ESBL), initially 'extended-broad-spectrum beta-lactamase', was first coined for derivatives of TEM and SHV enzymes able to hydrolyse oxyimino-cephalosporins. These all belonged to beta-lactamase functional group 2be. Subsequently, the term has been stretched to include: (i) enzymes with spectra similar to those of TEM and SHV mutants but derived from other sources, e.g., the CTX-M and VEB types; (ii) TEM and SHV mutants with borderline ESBL activity, e.g., TEM-12; and (iii) various beta-lactamases conferring wider resistance than their parent types

Extended-release divalproex in bipolar and other psychiatric disorders: A comprehensive review.

Bipolar disorder can be a devastating disease state for individuals with the disease and also for family members. Proper recognition and treatment is vital to the successful management of this disease state. Through increased community and practitioner awareness, along with efforts to increase awareness for proper assessment, the rate of diagnosed bipolar disorder is increasing. Recent years have brought about the introduction of several new medications with approved indications for the treatment of bipolar disorder. In addition to new agents, traditional mood stabilizing medications have also

More Motivated to Help Male Leaders? Explaining Fatherhood Bonuses via Follower Helping.

UNLABELLED: Family amplifies gender biases in organizations, presenting an ethical dilemma for modern organizations. Specifically, women tend to face additional barriers to leadership when they become mothers, while men often experience the opposite. But what about leaders' family explains these fatherhood bonuses? Focusing on work-family conflict (WFC), we theorize that this ostensibly negative state paradoxically motivates followers because it signals an opportunity to expend extra effort to help their leaders. We further theorize this helping process is stronger for male (vs. female) leader

A pathogenic variant of AMOT leads to isolated X-linked congenital hydrocephalus due to N-terminal truncation.

Congenital hydrocephalus is a life-threatening condition that might affect brain development by increasing the pressure on the brain parenchyma. Here, we describe 6 male patients from 1 family, all presenting with an isolated X-linked congenital hydrocephalus. Exome sequencing identified a likely pathogenic variant of angiomotin (AMOT) that segregated with the phenotype in the extended family. We show that the variant, affecting the first methionine, translated into a shorter AMOT protein lacking 91 amino acids from the N-terminus. Mechanistically, we unraveled that the absence of the N-termin…

Infants and toddlers in the United States with more close relationships have larger vocabularies.

Young children learn language from their caregivers, family members, and friends. However, with few exceptions, contemporary developmental scientists have studied language input and language learning through the lens of the primary caregiver and the nuclear family, rather than the infants' broader communities. In many communities-and increasingly in the United States-nonnuclear family structures are common, and extended kin, fictive kin, and intergenerational relationships are relied upon for child care. Understanding children's relationships within kinship networks can allow for more inclusiv

A syndrome of congenital hyperinsulinism and rhabdomyolysis is caused by KCNJ11 mutation.

BACKGROUND: Congenital hyperinsulinism is a genetically heterogeneous disorder, but mutations in the components of the ATP-sensitive potassium channel K(ATP) account for more than a third of all isolated congenital hyperinsulinism cases. The association between congenital hyperinsulinism and rhabdomyolysis has not been reported. OBJECTIVE: To describe significant skeletal muscle manifestations in a family with a novel mutation in KCNJ11 (encoding the Kir6.2 component of K(ATP)). METHODS: Cross-sectional analysis of members of a large multiplex consanguineous family with congenital hyperinsul

Mitral valve prolapse as a new finding in branchio-oto-renal syndrome.

We report a patient and his family, who have branchio-oto-renal (BOR) syndrome and coexisting mitral valve prolapse. A literature review of BOR syndrome failed to identify any similar families and we report this as a new observation. During the preoperative assessment of a patient with BOR syndrome, tachycardia was noted and a cardiologic examination including echocardiography revealed mitral valve prolapse. Members of his extended family were investigated, including carrying out cardiology and otolaryngology examinations to determine whether they had signs of either BOR syndrome or cardiac pr…

Phenotypic variation of tuberous sclerosis in a single extended kindred.

Tuberous sclerosis complex (TSC) is an autosomal dominant disorder with a great degree of phenotypic variability. Given the presence of two gene loci underlying this disorder, locus heterogeneity may account for some of the variability. However, significant within family variation suggests that different genes do not explain the majority of this variation. The purpose of this research is to identify physical and behavioural variation in expression of TSC in a single large extended kindred. TSC in this kindred is cosegregating with markers localised to chromosome 16p13.3. The expression of TSC

Characterization of a novel large deletion and single point mutations in the BRCA1 gene in a Greek cohort of families with suspected hereditary breast cancer.

BACKGROUND: Germline mutations in BRCA1 and BRCA2 predispose to breast and ovarian cancer. A multitude of mutations have been described and are found to be scattered throughout these two large genes. We describe analysis of BRCA1 in 25 individuals from 18 families from a Greek cohort. METHODS: The approach used is based on dHPLC mutation screening of the BRCA1 gene, followed by sequencing of fragments suspected to carry a mutation including intron--exon boundaries. In patients with a strong family history but for whom no mutations were detected, analysis was extended to exons 10 and 11 of the

Late diagnosis of Fabry disease caused by a de novo mutation in a patient with end stage renal disease.

BACKGROUND: We present the case of a white 35-year-old male with a diagnosis of Fabry disease and negative family history. CASE PRESENTATION: At the age of 31, he underwent a renal biopsy with a diagnosis of hypertension-induced nephroangiosclerosis. At the age of 35, he was referred to our hospital and started dialysis: the unusual finding of left ventricular hypertrophy with a normal ejection fraction and of myocardial fibrosis at the cardiac magnetic resonance suggested a diagnosis of Fabry disease, although there was no apparent family history-so extensive tests were subsequently undertak

Donor, dad, or…? Young adults with lesbian parents' experiences with known donors.

In this exploratory qualitative study of 11 young adults, ages 19-29 years, we examine how young people who were raised by lesbian parents m … perceived their donors in one of three ways: as strictly donors and not members of their family; as extended family members but not as parents; and as fathers. The more limited role of donors in participants' construction of family relationships sheds light on how children raised in lesbian, gay, and bisexual families are contributing to the redefinition and reco …

Identification of small-molecule elastase inhibitors as antagonists of IL-36 cytokine activation.

IL-1 family cytokines act as apical initiators of inflammation in many settings and can promote the production of a battery of inflammatory cytokines, chemokines and other inflammatory mediators in diverse cell types. IL-36α, IL-36β and IL-36γ, which belong to the extended IL-1 family, have been implicated as key initiators of skin inflammation in psoriasis. IL-36γ is highly upregulated in lesional skin from psoriatic individuals, and heritable mutations in the natural IL-36 receptor antagonist result in a severe form of psoriasis. IL-36 family cytokines are initially expressed as inactive pre

Experiences and beliefs related to exclusive breastfeeding and early supplementation in low income urban slums of Karachi, Pakistan- a qualitative study.

In Pakistan, exclusive breastfeeding (EBF) is often difficult to establish and sustain in low-income urban settings due to limited access to antenatal and postnatal care, maternal undernutrition, socioeconomic constraints, sociocultural myths, and extended family influence. These factors frequently lead to early supplementation when breastfeeding is perceived as insufficient. We conducted a qualitative exploratory study using focus group discussions in four low-income urban slums of Karachi, Pakistan in January 2020. Our study aimed to explore the community beliefs and barriers relating to EBF…

The genome evolution and domestication of tropical fruit mango.

BACKGROUND: Mango is one of the world's most important tropical fruits. It belongs to the family Anacardiaceae, which includes several other economically important species, notably cashew, sumac and pistachio from other genera. Many species in this family produce family-specific urushiols and related phenols, which can induce contact dermatitis. RESULTS: We generate a chromosome-scale genome assembly of mango, providing a reference genome for the Anacardiaceae family. Our results indicate the occurrence of a recent whole-genome duplication (WGD) event in mango. Duplicated genes preferentially
