Factor XIIIa-containing cells and fibrosis in oral and maxillofacial lesions: an immunohistochemical study.

The distribution of subunit A of blood coagulation factor XIII (FXIIIa) was investigated by the avidin-biotin-peroxidase complex (ABC) method in various oral and maxillofacial tissues. These tissues were from normal tongue, gingiva, lip, and submandibular gland, and from Dilantin gingival hyperplasia (one case), pyogenic granuloma (three cases), peripheral fibroma (four cases), squamous cell carcinoma (seven cases), chronic sclerosing submandibular adenitis (two cases), and fibrous dysplasia of the mandibular bone (one case). The distribution of collagenous components was examined in the same …

The impact of intrauterine growth on coagulation and factor XII function in preterm infants.

This study aims to examine the impact of intrauterine growth restriction (IUGR) on coagulation in preterm newborns and assess the administration of fresh frozen plasma. The study involved 30 preterm infants with IUGR and 32 preterm infants without it. Blood samples were collected within 4 h after birth to analyze blood gases, complete blood counts, renal and liver function tests, and coagulation factors. Differences between the two groups were assessed using SPSS version 15. IUGR significantly impacts coagulation in infants born before the 32nd week of gestation. Preterm infants with IUGR, spe

Spectrum of hemostatic disorders in Indian females presenting with bleeding manifestations.

INTRODUCTION: Hemostatic disorders are often missed in women with bleeding particularly menorrhagia. Preexisting hemostatic disorders are now known as common risk factor for postpartum hemorrhage and prolonged bleeding in puerperium. Females with bleeding complaints constitute an important population referred to hematology clinic. Hence, we aim to evaluate the type and frequency of hemostatic disorders among females presenting with bleeding in a tertiary care hospital and a basic hemostatic laboratory. METHODS: Three-year data were retrospectively analyzed for 200 females with various bleedin

Evaluation of expression of genes associated with post-thrombotic syndrome.

Prediction of the development of post-thrombotic syndrome (PTS) among patients with deep venous thrombosis (DVT) is currently based on clinical characteristics alone; no reliable biomarkers are available. Coagulation Factor XIII A chain (F13A1) of the clotting cascade stabilizes the thrombus; myeloperoxidase (MPO) interacts with the endothelium; and Fms-related tyrosine kinase 4 (FLT4), also known as Vascular Endothelial Growth Factor Receptor-3, encodes a vascular endothelium-derived growth factor receptor that participates in angiogenesis. In this study, MPO, FLT4, and F13A1 gene expression …

Factor XIIIa as a nerve-associated transglutaminase.

Recent findings have led to changes in the traditional concept of nerve recovery, including the realization that injured nerves, like any other injured tissue, need the assistance of blood-derived cells and factors in order to heal. We show that factor XIIIa (FXIIIa, the potentially active a2subunit of factor XIII), an enzyme that participates in blood coagulation by stabilizing the fibrin clot, is also active in the nervous system where it may play a key role in the healing of injured tissue. We demonstrate that the plasma, macrophages and nerves of fish contain a 55 kDa form of transglutamin…

Inhibitors to clotting factors.

Clot formation is the final result of interaction among multiple plasma proteins; after activation, it results in the conversion of fibrinogen to fibrin and cross-linking of fibrin by activated factor XIII, which stabilizes the formed clot. Deficiency or functional abnormality of the factors involved in these reactions causes bleeding disorders. Natural inhibitors of clotting factors include antithrombin III, protein S, and protein C. When activated, these proteins inactivate specific clotting factors, providing a regulatory mechanism that serves to control the coagulation response and limit t…

Identification of a novel thrombin-like phospholipase A2 from Gloydius ussuriensis snake venom.

The coagulant effects of phospholipase A2 with Gln at 49 sites (Gln49-PLA2), purified from Gloydius ussurensis snake venom, were investigated on human citrated plasma and fibrinogen. Gln49-PLA2 clotted human plasma dose-dependently from 180.67 +/- 1.86 s to 19.00 +/- 0.58 s, and reduced the re-calcification time from 7.46 +/- 1.17 to 0.75 +/- 0.33 min and the prothrombin time from 12.4 +/- 0.29 s to 6.95 +/- 0.20 s, but it could not activate factor XIII, and the procoagulant effects were inhibited by heparin. The specific clotting activities of Gln49-PLA2 were equivalent to 1100 NIH thrombin U…

Acquired inhibitors.

Factor VIII auto-antibody inhibitors, though rare, may present significant and often life-threatening haemorrhage. These auto-antibodies, arising predominantly in older individuals, occur in association with autoimmune disorders, lymphoproliferative disorders, solid tumours, medications and the postpartum state. Almost half of the patients develop auto-antibodies spontaneously without an underlying medical condition. Factor VIII auto-antibody inhibitors are characterized as polyclonal IgG immunoglobulins directed against the FVIII procoagulant activity. Laboratory diagnosis is made by performi

A Case Report of Coagulation Factor XII Deficiency.

BACKGROUND: Coagulation factor XII (FXII) deficiency is a rare coagulation disorder, often incidentally detected during routine physical examinations, invasive procedures, or preoperative screenings, with no specific clinical manifestations. Here, we report a case of FXII deficiency. METHODS: A 46-year-old female patient was found to have unexplained prolonged activated partial thromboplastin time (APTT) during routine preoperative testing, without clinical bleeding symptoms. We performed an APTT mixing study, anti-cardiolipin antibody (ACA) testing, anti-β2-glycoprotein 1 (anti-β2-GP1) antib

Pharmacogenetics of antithrombotic drugs.

Antithrombotic therapy has improved the prognosis of patients with venous or arterial thrombosis. However, there is substantial interindividual variability in the response to antithrombotic drugs. This variability is due, in part, to genetics, which may affect the efficacy and safety of drugs used in the treatment and prevention of thrombosis. Pharmacogenetics studies the genetic factors related to interindividual variability in the response to drugs. Various polymorphisms in genes of the hemostatic system that have been reported to be markers of susceptibility to thromboembolic disease also s

The role of transglutaminase 2 in stabilizing fibrin clots and supporting hemostasis.

Transglutaminases are enzymes with pleiotropic functions in the human body, among them are the A-subunit of coagulation factor XIII (FXIII; FXIII-A) and tissue transglutaminase (TG2). FXIII has a crucial function in clot formation and stabilization. FXIII also contributes to extracellular matrix formation, wound healing and tissue regeneration, a function it shares with TG2. Whether TG2 may also have a role in hemostasis has never been studied in detail. The aim of our project was to study the role of TG2 in clot formation, stabilization and hemostasis. Here, with the use of a microfluidic end…

Cross-linking of alpha 2-plasmin inhibitor to fibrin by fibrin-stabilizing factor.

The concentration of alpha 2-plasmin inhibitor in blood plasma is higher than that in serum obtained from the blood clotted in the presence of calcium ions, but is the same as that in serum obtained in the absence of calcium ions. Radiolabeled alpha2-plasmin inhibitor was covalently bound to fibrin only when calcium ions were present at the time of clotting of plasma or fibrinogen. Whereas, when batroxobin, a snake venom enzyme that lacks the ability to activate fibrin-stabilizing factor, was used for clotting fibrinogen, the binding was not observed. When fibrin-stablizing, factor-deficient p

Rare inherited coagulation disorders and thrombosis.

The traditional clinical paradigm of rare inherited coagulation disorders (RICDs) implies that reduced levels of coagulation factors increase the bleeding tendency, although the relationship between factor activity and clinical phenotype is often variable and disease-related. However, emerging evidence suggests a more complex coagulation balance in RICD, so that a few inherited factor deficiencies predispose patients to arterial thrombosis and venous thromboembolism (VTE). By synthesizing data from international registries, clinical trials and biochemical studies, this review explores the mole
