Synthesis of acid-stabilized iron oxide nanoparticles and comparison for targeting atherosclerotic plaques: evaluation by MRI, quantitative MPS, and TEM alternative to ambiguous Prussian blue iron staining.

UNLABELLED: To further optimize citrate-stabilized VSOPs (very small iron oxide particles, developed for MR angiography) for identification of atherosclerotic plaques, we modified their surface during synthesis using eight other acids for electrostatic stabilization. This approach preserves effective production for clinical application. Five particles were suitable to be investigated in targeting plaques of apoE(-/-) mice. Accumulation was evaluated by ex vivo MRI, TEM, and quantitatively by magnetic particle spectroscopy (MPS). Citric- (VSOP), etidronic-, tartaric-, and malic-acid-coated part

Thallium poisoning from maliciously contaminated food.

Four young adults presented two days after one of them had received marzipan balls packaged in a box from an expensive candy manufacturer. Two ate one candy ball, while two others shared a third. The next day, variable gastrointestinal symptoms developed. On the third day, two patients developed painful paresthesiae of the hands and feet, an early but nonspecific clinical marker of thallium poisoning. A tentative diagnosis of thallium poisoning was made based on symptoms, and treatment was initiated. The remaining candies were radiographed. Metallic densities in the candies supported the diagn

Zinc phthalocyanine and sulfasalazine coloaded nanoaggregates with Prussian blue functionalization for sensitizing tumor photothermal/photodynamic therapy.

Photothermal therapy/photodynamic therapy (PTT/PDT), as a noninvasive therapeutic modality, has been extensively applied in superficial tumor treatment. However, their curative effects were largely weakened due to hypoxia and an elevated glutathione (GSH) microenvironment. Herein, zinc phthalocyanine (ZnPc) and sulfasalazine (SAS) coloaded nanoaggregates (Z-S@B NAs) with Prussian blue (PB) functionalization (PB/Z-S@B NAs) were fabricated via self-assembly and using an in situ oxidative polymerization method for tumor PTT/PDT sensitization. The designed PB/Z-S@B NAs were capable of triggering l

Near-Infrared Radiation-Assisted Drug Delivery Nanoplatform to Realize Blood-Brain Barrier Crossing and Protection for Parkinsonian Therapy.

Mitochondrial dysfunction, which is directly involved in Parkinson's disease (PD), is characterized by the production of reactive oxygen species (ROS) and aberrant energy metabolism. Thus, regulating mitochondrial function might be an effective strategy to treat PD. However, the blood-brain barrier (BBB) presents a significant challenge for the intracerebral delivery of drugs. Here, we synthesized a zeolitic imidazolate framework 8-coated Prussian blue nanocomposite (ZIF-8@PB), which was encapsulated with quercetin (QCT), a natural antioxidant, to treat PD. ZIF-8@PB-QCT exhibited superior near

Modulation of β secretase and neuroinflammation by biomimetic nanodelivery system for Alzheimer's disease therapy.

Alzheimer's disease (AD) is an irreversible and progressive neurodegenerative disorder. The vicious circle between amyloid-β peptide (Aβ) overgeneration and microglial dysfunction is an important pathological event that promotes AD progression. However, therapeutic strategies toward only Aβ or microglial modulation still have many problems. Herein, inspired by the Aβ transportation, an Aβ-derived peptide (CKLVFFAED) engineered biomimetic nanodelivery system (MK@PC-R NPs) is reported for realizing BBB penetration and reprogram neuron and microglia in AD lesion sites. This hollow mesoporous Prus

Biofilm-modified Prussian blue improves memory function in late-stage Alzheimer's disease mice with triple therapy.

Alzheimer's disease (AD) is a neurodegenerative disease that is significantly characterized by cognitive and memory impairments, which worsen significantly with age. In the late stages of AD, metal ion disorders and an imbalance of reactive oxygen species (ROS) levels occur in the brain microenvironment, which causes abnormal aggregation of β-amyloid (Aβ), leading to a significant worsening of the AD symptoms. Therefore, we designed a composite nanomaterial of macrophage membranes-encapsulated Prussian blue nanoparticles (PB NPs/MM). Prussian blue nanoparticles (PB NPs) are capable of chelatin

Pd, S co-modified Prussian blue analogues nanocomposites for MRI guided combined mitochondria-targeting cancer therapy with tumor microenvironment remodeling, multienzyme-like catalysis and mild photothermal therapeutic effect.

Developing appropriate nanoplatforms with enhanced properties to increase the efficiency of the nano-based strategies is an urgent challenge for cancer therapy. In this work, a S, Pd modified hollow CoNiMnFe Prussian blue analogue nanocomposite (Pd-S-CNMF) was prepared as multifunctional nanoplatform for magnetic resonance imaging (MRI) guided for combined cascade catalyzed therapy and low-temperature photothermal therapy. We found that the co-modification of S and Pd on CNMF could enhance the responsive feature the as-prepared nanocomposite to acidic environment, which endowed the Pd-S-CNMF w

Biomimetic Nanoplatform Based on the Neutrophil Membrane for Targeted Therapy of Spinal Cord Injury.

INTRODUCTION: Spinal cord injury (SCI) triggers a cascade of secondary damage, including oxidative stress and neuroinflammation, for which effective treatments remain limited. To address this, we developed a biomimetic nanoplatform, NEU@MPBNPs-HED, consisting of mesoporous Prussian blue nanoparticles (MPBNPs) loaded with hederagenin (HED) and cloaked with neutrophil membranes (NEUm) to enhance lesion targeting and immune evasion. METHODS: The physicochemical properties, drug release characteristics, and cellular uptake of NEU@MPBNPs-HED were characterized. Therapeutic efficacy was evaluated i

Investigation of neural progenitor cell induced angiogenesis after embolic stroke in rat using MRI.

Using MRI, we investigated dynamic changes of brain angiogenesis after neural progenitor cell transplantation in the living adult rat subjected to embolic stroke. Neural progenitor cells isolated from the subventricular zone (SVZ) of the adult rat were labeled by superparamagnetic particles and intracisternally transplanted into the adult rat 48 h after stroke (n = 8). Before and after the transplantation, an array of MRI parameters were measured, including high resolution 3D MRI and quantitative T1, T1sat (T1 in the presence of an off-resonance irradiation of the macromolecules of brain), T2,

Systematic expression of immediate early genes and intensive astrocyte activation induced by intrastriatal ferrous iron injection.

The potential role(s) of transitional metals such as iron have been implicated in neurodegeneration through biochemical processes, particularly oxidative stress. We injected ferrous chloride (FeCl2) and ferric chloride (FeCl3) into the striatonigral system of Sprague-Dawley rats to investigate the biological and toxic effects of ferrous iron in the central nervous system. When FeCl2 was injected into the ventral midbrain, rats showed a characteristic behavior which indicated ipsilateral dopaminergic hyperactivity. FeCl2 injection into the striatum induced a dose-dependent damage, the activatio

Tracking the healing effect of human Wharton's jelly stem cells labeled with superparamagnetic iron oxide nanoparticles seeded onto polyvinyl alcohol/chitosan/carbon nanotubes in burn wounds by MRI and Prussian blue staining.

Regenerative medicine through the application of tissue engineering and cell transplantation has provided a new door for wound healing. In this study, the healing effect of human Wharton's jelly stem cells (WJSCs) labeled with superparamagnetic iron oxide nanoparticles (SPIONs) seeded onto polyvinyl alcohol/chitosan/carbon nanotubes (PVA/CS/CNTs) in burn wounds was investigated by performing magnetic resonance imaging (MRI) and Prussian blue staining. Human WJSCs were prepared from umbilical cord and characterized. PVA/CS/CNTs were fabricated via electrospinning. Forty-eight rats were divided

Neuroprotective effects and magnetic resonance imaging of mesenchymal stem cells labeled with SPION in a rat model of Huntington's disease.

Bone marrow mesenchymal stem cells (MSC) have been tested and proven effective in some neurodegenerative diseases, but their tracking after transplantation may be challenging. Our group has previously demonstrated the feasibility and biosafety of rat MSC labeling with iron oxide superparamagnetic nanoparticles (SPION). In this study, we investigated the therapeutic potential of SPION-labeled MSC in a rat model of Huntington's disease, a genetic degenerative disease with characteristic deletion of striatal GABAergic neurons. MSC labeled with SPION were injected into the striatum 1h after quinol

The response of cerebral cortex to haemorrhagic damage: experimental evidence from a penetrating injury model.

Understanding the response of the brain to haemorrhagic damage is important in haemorrhagic stroke and increasingly in the understanding the cerebral degeneration and dementia that follow head trauma and head-impact sports. In addition, there is growing evidence that haemorrhage from small cerebral vessels is important in the pathogenesis of age-related dementia (Alzheimer's disease). In a penetration injury model of rat cerebral cortex, we have examined the neuropathology induced by a needlestick injury, with emphasis on features prominent in the ageing and dementing human brain, particularly

[Thallium poisoning. Experience with 50 patients].

We have studied fifty cases of thallium intoxication during the past nine years. Twenty-eight occurred in women and twenty-two in men. One of the patients was a new born whose mother had this type of intoxication during her third trimester of pregnancy. The ages varied from one day to 84 years and in all cases the source of thallium was ingestion of rat poison, except for the baby who received it across the placenta and an other patient whose source was transdermal. Twenty-three of the cases of intoxication were accidental, twenty-one were suicidal attempts and five were homicidal. One case di

Photothermal therapy improves the efficacy of a MEK inhibitor in neurofibromatosis type 1-associated malignant peripheral nerve sheath tumors.

Malignant peripheral nerve sheath tumors (MPNSTs) are aggressive tumors with low survival rates and the leading cause of death in neurofibromatosis type 1 (NF1) patients under 40 years old. Surgical resection is the standard of care for MPNSTs, but is often incomplete and can generate loss of function, necessitating the development of novel treatment methods for this patient population. Here, we describe a novel combination therapy comprising MEK inhibition and nanoparticle-based photothermal therapy (PTT) for MPNSTs. MEK inhibitors block activity driven by Ras, an oncogene constitutively acti

Blood-spinal cord barrier disruption contributes to early motor-neuron degeneration in ALS-model mice.

Humans with ALS and transgenic rodents expressing ALS-associated superoxide dismutase (SOD1) mutations develop spontaneous blood-spinal cord barrier (BSCB) breakdown, causing microvascular spinal-cord lesions. The role of BSCB breakdown in ALS disease pathogenesis in humans and mice remains, however, unclear, although chronic blood-brain barrier opening has been shown to facilitate accumulation of toxic blood-derived products in the central nervous system, resulting in secondary neurodegenerative changes. By repairing the BSCB and/or removing the BSCB-derived injurious stimuli, we now identify

MRI of iron oxide nanoparticle-labeled ADSCs in a model of hindlimb ischemia.

Adipose-derived stem cells (ADSCs) exhibit tremendous potential for repair of ischemic diseases. However, studies on the fate, migration, differentiation, and body distribution of the labeled ADSCs are rarely reported. In this study, magnetic iron oxide nanoparticles were designed, synthesized, and coated with meso-2,3-dimercaptosuccinic acid (DMSA) to produce DMSA nanoparticles (DMSA-NPs). The properties, size distribution, and characterization of DMSA-NPs were evaluated. Green fluorescent protein expressing ADSCs (GFP-ADSCs) were obtained and labeled with DMSA-NPs. The viability, cytotoxicit
