Five O-alkoxyamidine analogues of the prodrug 2,5-bis[4-methoxyamidinophenyl]furan were synthesized and evaluated against Trypanosoma brucei rhodesiense in the STIB900 mouse model by oral administration. The observed in vivo activity of these prodrugs demonstrates that compounds with an O-methoxyamidine or O-ethoxyamidine group effectively cured all trypanosome-infected mice, whereas prodrugs with larger side-chains did not completely cure the mice. Permeability across Caco-2 cell monolayers and microsomal metabolism were used to identify the underlying mechanisms of prodrug efficacy.

3-Nor-spongiolide A (1), belonging to the extremely rare 3-nor-spongian carbon skeleton, and spongiolides A (2) and B (3), having γ-butenolide instead of furan ring as usual for ring D, together with six related known metabolites were isolated from South China Sea sponge Spongia officinalis as its metabolic components. γ-Hydroxynitrile glucosides (prinsepicyanosides A-E) were isolated alongside 11 known compounds from seeds of Prinsepia utilis Royle. Their structures were determined by detailed analysis of NMR and MS spectroscopic data. The relative configuration of prinsepicyanoside C was established by Cu-Kα X-ray crystallography. Prinsepicyanoside A, osmaronin, and 4-(hydroxylmethyl)-5H-furan-2-one exhibited borderline antibacterial activity against Salmonella gallinarum, Vibrio parahaemolyticus, and Vibrio cholera with MIC values of 30.1, 20.7, and 22.8μg/mL, respectively.

The title ionic compound, (C(21)H(27)N(2))[AuCl(4)]·CH(2)Cl(2), was obtained from the reaction of 1,3-dimesitylimidazolidinium chloride with t-BuOK and a solution of AuCl(3) in tetra-hydro-furan. Second-generation ethanol is a promising renewable alternative to traditional transportation fuels. However, its large-scale production via fermentation faces economic and technical challenges, including microbial contamination. This study evaluates how ten common lignocellulosic inhibitors, formed during biomass pretreatment, affect the growth of the industrial bioethanol yeast strain Saccharomyces cerevisiae PE-2 and two bacterial contaminants Lactiplantibacillus plantarum I4a and Limosilactobacillus fermentum I3a. The bacteria demonstrated greater resilience to furanic compounds than yeast.

A novel trypanocide, 2,5-bis(4-amidinophenyl)furan (DB75), in its prodrug amidoxime-derivative form, 2,5-bis(4-amidinophenyl)furan-bis-O-methylamidoxime (DB289), is in trials as the first orally administered drug for human African trypanosomiasis.
