Comparative transcriptome analysis provides insights into dwarfism in cherry tomato (Solanum lycopersicum var. cerasiforme).

Tomato, which can be eaten as a vegetable or fruit, is one of the most popular and nutritionally important crops around the world. Although most plants of the cherry tomato cultivar 'Minichal' have a normal phenotype, some plants have a stunted phenotype with reduced plant height, leaf size, and fruit size, as well as altered leaf and fruit shape. To investigate the molecular mechanisms underlying these differences, we generated RNA-seq libraries from pooled leaf samples of 10 normal (N) and 10 stunted (S) plants. Using the Illumina sequencing platform, we obtained a total of 115.45 million hi

Glycine max DREB1D transcription factor regulates plant height by reducing gibberellin levels and response.

Overexpression of GmDREB1D reduces plant height of soybean. Overexpression of GmDREB1D reduced active GA content and GmDREB1D can regulate GmGA2ox8. GmDREB1D may inhibit growth by reducing the levels of active GAs and response. Dehydration responsive element binding (DREB) proteins play an important role in growth and abiotic stress. Although the DREB1D transcription factor contributes to drought tolerance, the molecular mechanisms underlying growth inhibition are unclear. In this study, we reveal the function of GmDREB1D as a transcriptional regulator in growth. Overexpression of GmDREB1D red

Mechanism and Application of Developmental Factors in Plant Genetic Transformation.

Genetic transformation serves as a critical tool for gene function research and crop improvement. However, its efficiency is often low and highly dependent on species, genotypes, and explant types, significantly restricting its broader application. Many developmental factors have been proven pivotal not only for plant growth and development but also for the regulation of callus formation and shoot regeneration, which are key steps in the process of genetic transformation. Thus, this review focuses on the application of developmental factors in enhancing transformation efficiency across species

Genome-Wide Characterization and Heat Responsive Expression Profiling of the HSP10 Co-Chaperonin Gene Family in Brassica juncea.

Heat stress disrupts protein homeostasis, yet the thermoregulatory roles of HSP10/CPN10 co-chaperonins in polyploid crops remain unexplored. This study conducted the first genome-wide characterization of the HSP10 gene family in allotetraploid Brassica juncea to identify heat tolerance candidates. Genome-wide identification of BjuHSP10 genes was performed across A and B subgenomes, followed by phylogenetic, gene structure, duplication, synteny, promoter, and structural analyses. Transcriptional responses were evaluated under moderate (28/24 °C) and severe (36/32 °C) heat stress conditions. Ten

Effects of loss of CO2-Responsive CCT Protein interaction with 14-3-3 proteins and its ectopic expression on starch synthesis in rice.

CO2-Responsive CCT Protein (CRCT) is a transcription factor that positively regulates the expression of starch synthesis-related genes in the vegetative organs of rice. CRCT interacts with 14-3-3 proteins. Yeast two-hybrid analysis showed that the CRCT with the S217A mutation was unable to interact with CRCT. This mutated CRCT was introduced into the CRCT knockout rice. The transcript of CRCT-S217A was detected significantly by qRT-PCR, but not by immunoblotting. Nevertheless, starch accumulation and the expression of starch synthesis-related genes in the leaf sheath of the transformants were

The major plant sphingolipid long chain base phytosphingosine inhibits growth of bacterial and fungal plant pathogens.

Sphingolipid long chain bases (LCBs) are building blocks of sphingolipids and can serve as signalling molecules, but also have antimicrobial activity and were effective in reducing growth of a range of human pathogens. In plants, LCBs are linked to cell death processes and the regulation of defence reactions against pathogens, but their role in directly influencing growth of plant-interacting microorganisms has received little attention. Therefore, we tested the major plant LCB phytosphingosine in in vitro tests with the plant pathogenic fungi Verticillium longisporum, Fusarium graminearum and

Identification of MTP gene family in tea plant (Camellia sinensis L.) and characterization of CsMTP8.2 in manganese toxicity.

Cation diffusion facilitators (CDFs) play central roles in metal homeostasis and tolerance in plants, but the specific functions of Camellia sinensis CDF-encoding genes and the underlying mechanisms remain unknown. Previously, transcriptome sequencing results in our lab indicated that the expression of CsMTP8.2 in tea plant shoots was down-regulated exposed to excessive amount of Mn2+ conditions. To elucidate the possible mechanisms involved, we systematically identified 13 C. sinensis CsMTP genes from three subfamilies and characterized their phylogeny, structures, and the features of the enc

Genome-Wide Analysis of the PYL Gene Family and Its Expression Dynamics in Response to Abscisic Acid in Tomato.

The plant hormone abscisic acid (ABA) plays a crucial role throughout the plant life cycle and in adaptive responses to environmental stresses. The pyrabactin resistance 1-like (PYR/PYL/RCAR) proteins act as key regulators in the ABA signal transduction pathway by functioning as direct receptors for ABA. Although PYL genes have been identified in a variety of plant species, their evolutionary and structural characteristics in tomatoes (Solanum lycopersicum) remain elusive. To address this gap, we identified nine SlPYL genes, which were classified into three subfamilies: I (two genes), II (thre

Comparative Analyses of Anatomical Structure, Phytohormone Levels, and Gene Expression Profiles Reveal Potential Dwarfing Mechanisms in Shengyin Bamboo (Phyllostachys edulis f. tubaeformis).

Moso bamboo (Phyllostachys edulis) is one of the most important bamboo species in China and the third most important plant species for timber production. However, the dwarf variant of moso bamboo, P. edulis f. tubaeformis (shengyin bamboo), which has shortened internodes, is not well studied. We used anatomical, hormonal, and transcriptomic approaches to study internode shortening and shoot growth in dwarf shengyin and wild moso bamboo. Phenotypic and anatomical observations showed that dwarfing in shengyin bamboo is due to reduced internode length, and the culm fibers in shengyin bamboo are s

Hydrogen sulfide alleviates the cold stress through MPK4 in Arabidopsis thaliana.

Hydrogen sulfide (H2S) is a gaseous signaling molecule that mediates physiological processes in animals and plants. In this study, we investigated the relationship of H2S and mitogen activated protein kinase (MAPK) under cold stress in Arabidopsis. H2S up-regulated MAPK expression levels and was involved in the cold stress-related upregulation of MAPK genes expression. We then chose MPK4 whose expression level was influenced the most by H2S as a target and found that H2S's ability to alleviate cold stress required MPK4. Both H2S and MPK4 regulated the expression levels of the cold response gen

Gene expression profiling in wounded and systemic leaves of Fagus sylvatica reveals up-regulation of ethylene and jasmonic acid signalling.

Wounding is a crucial threat to plants because of the physical damage caused and the possible entry of pathogens. Little is known about the wound reaction in forest trees. Therefore, leaves of young beech trees were wounded and the transcriptional response of wounded leaves and leaves directly above and below was analysed. A total of 123 genes exhibited significant regulation. The magnitude of regulation was slightly weaker in the downward leaves but the regulation pattern resembles that of the local and upward reactions. Thus, the signal was transduced in both vertical directions. Genes exhib

Ectopic and transient expression of VvDIR4 gene in Arabidopsis and grapes enhances resistance to anthracnose via affecting hormone signaling pathways and lignin production.

BACKGROUND: DIR (Dirigent) proteins play important roles in the biosynthesis of lignin and lignans and are involved in various processes such as plant growth, development, and stress responses. However, there is less information about VvDIR proteins in grapevine (Vitis vinifera L). RESULTS: In this study, we used bioinformatics methods to identify members of the DIR gene family in grapevine and identified 18 VvDIR genes in grapevine. These genes were classified into 5 subfamilies based on phylogenetic analysis. In promoter analysis, various plant hormones, stress, and light-responsive cis-ele

Overview of molecular mechanisms underlying stress responses; signaling pathways and secondary metabolite synthesis in plants.

Plants are susceptible to several biotic and abiotic stressors, including pathogens, extreme salinity, temperature, and drought. Continuous exposure to these conditions activates complex molecular mechanisms in plants. These include stress-signaling pathways mediated by phytohormones such as cytokins, salicylic acid, auxins, abscisic acid, jasmonic acid, and ethylene. These signaling molecules further interact with cellular pathways regulating stress-responsive gene expression through transcription factors such as DREBs, MYBs, bZIPs, and NACs. Key cellular pathways include mitogen-activated pr

Comparative analysis of ultraviolet-B radiation and abscisic acid revealing distinct mechanistic approaches to drought tolerance in highland barley.

Drought tolerance in highland barley (qingke) is modulated by distinct responses to ultraviolet-B radiation (UVB) and abscisic acid (ABA). Plants were exposed to two drought levels (moderate: 50 % soil water content and severe: 30 % soil water content), alongside two UVB intensities (low: 5-6 kJ m-2 d-1 and high: 14-15 kJ m-2 d-1), and two ABA concentrations (low: 70 μM and high: 150 μM). Physiological analyses revealed that UVB exposure significantly reduced the photosynthetic rate by 46.5 % under severe drought conditions. In contrast, ABA treatment helped to maintain the rate and improved w

Anti-auxin treatment and co-expression networks reveal auxin involvement in the transition to ripening in sweet cherry fruits.

The transition from development to ripening is governed by several hormones that interplay with abscisic acid (ABA) in non-climacterics, including auxin. In sweet cherry (Prunus avium), auxin has been shown to promote fruit coloration and ABA accumulation, in contrast to other non-climacteric models. Thus, this work aims to explore the role of auxin during the transition to ripening in sweet cherry by assessing the global gene expression response to an auxin homeostasis disturbance. For this, we used 5d, a fluorescently labeled compound with previously reported anti-auxin activity, which was a

Fungal endophytes relieve the growth-defence trade-off of their plant hosts through the production of bioactive alkaloids.

A central paradigm in plant biology is that there is a trade-off between growth and defence. We propose that Epichloë fungal endophytes relieve this trade-off in plants by the fungal-derived production of antiherbivore alkaloids. Lolium perenne plants without (nil) and with Epichloë LpTG-3 sp. strain AR37 able (wild type (wt), ΔidtA) and unable (ΔidtM) to produce bioactive indole diterpene alkaloids were subjected to an exogenous application of gibberellin (GA) hormone followed by a challenge with Rhopalosiphum padi aphids. The GA-derived plant growth promotion increased the susceptibility to

Nitric oxide interacts with brassinosteroid signalling to regulate plant growth and development in an organ-specific manner.

Nitric oxide (NO) is a plant gasotransmitter that regulates plant growth and development by interacting with different regulatory pathways. Among these, brassinosteroids (BRs) have become candidate phytohormones that interact antagonistically and closely with NO to regulate plant growth. Seedling growth in NO-deficient mutants is enhanced and hyper-responds to BR treatments, while NO overaccumulator mutants show some developmental defects. Interestingly, this hyper-response was also observed in mutants that had both constitutively activated BR signalling and reduced NO levels. BR signalling mu

Phytohormone-dependent plant defense signaling orchestrated by oral bacteria of the herbivore Spodoptera litura.

A vast array of herbivorous arthropods live with symbiotic microorganisms. However, little is known about the nature and functional mechanism of bacterial effects on plant defense responses towards herbivores. We explored the role of microbes present in extracts of oral secretion (OS) isolated from larvae of Spodoptera litura, a generalist herbivore, in phytohormone signaling-dependent defense responses in Arabidopsis thaliana (Arabidopsis). In response to mechanical damage (MD) with application of bacteria-free OS (OS- ) prepared by sterilization or filtration of OS, Arabidopsis leaves exhibi
