Combined effect of hygienic and polygenic risk scores in children with allergic rhinitis.

BACKGROUND: Although the development of allergic rhinitis (AR) is associated with multiple genetic and hygienic environmental factors, previous studies have focused mostly on the effect of a single factor on the development of AR. OBJECTIVE: This study aimed to investigate the combined effect of multiple genetic and hygienic environmental risk factors on AR development in school children. METHODS: We conducted a cross-sectional study, comprising 1,797 children aged 9-12 years. Weighted environmental risk score (ERS) was calculated by using four hygienic environmental factors, including antib

Monogenic and Polygenic Risk for Kidney Cancer in Two Large Biobanks.

BACKGROUND: Pathogenic variants (PVs) in monogenic genes and polygenic risk scores (PRS) have been associated with kidney cancer risk. However, their joint contributions in the general population remain unclear. DESIGN, SETTING, AND PARTICIPANTS: Associations of kidney cancer with PVs in 15 core renal cell carcinoma (RCC) hereditary genes and 22 other cancer-susceptibility genes, and a published PRS (PGS004908) were evaluated in two large cohorts: the United Kingdom Biobank (UKB; N = 452 208) and the Genomic Health Initiative (GHI; N = 20 452). OUTCOME MEASUREMENTS AND STATISTICAL ANALYSIS:

Plant-based diet, metabolic signature, genetic susceptibility, and risk of musculoskeletal disorders: a large-scale population-based prospective cohort study.

AIMS: To explore the association between plant-based diets, diet-related metabolic signatures, and the risk of total and specific musculoskeletal disorders (MSDs), while accounting for genetic susceptibility. METHODS: We analyzed data from 107,983 participants in the UK Biobank. Plant-based diet indices [PDI, unhealthy PDI (uPDI), and healthy PDI (hPDI)] were calculated via 24-h dietary recalls. Elastic net regression was applied to generate PDIs-related metabolic signatures. Polygenic risk score (PRS) was constructed to quantify genetic susceptibility to MSDs. Cox proportional hazards regres

Genetic architecture of cytokine autoantibodies and associated risk of common diseases.

Anti-cytokine autoantibodies are increasingly recognized as modulators of immune function and determinants of disease risk, yet their genetic basis and population-level impact remain unclear. Here we perform genome-wide association studies of autoantibodies against IL-1α, IL-6, IL-10, IFN-α, IFN-β, IFN-γ and GM-CSF in 15,000 individuals from the Danish Blood Donor Study. We identify 52 genome-wide significant loci, implicating genes enriched in antigen-presentation pathways. Using these data, we derive cytokine-specific polygenic risk scores and evaluate their associations across 300,000 indiv

Pancreatitis polygenic risk score is associated with acute pancreatitis in multifactorial chylomicronemia syndrome.

BACKGROUND: Multifactorial chylomicronemia syndrome (MCS) is a severe form of hypertriglyceridemia associated with an increased risk of acute pancreatitis (AP). The risk of AP is heterogenous and is associated with increased level of triglycerides (TG) and presence of rare variants in TG metabolism-related genes. OBJECTIVE: To determine if the accumulation of common variants in pancreatitis susceptibility genes, measured with a weighted polygenic risk score (PRS), is associated with AP in MCS patients. METHODS: A total of 114 patients with MCS underwent genetic testing for eight single nucle

Causation and familial confounding as explanations for the associations of polygenic risk scores with breast cancer: Evidence from innovative ICE FALCON and ICE CRISTAL analyses.

A polygenic risk score (PRS) combines the associations of multiple genetic variants that could be due to direct causal effects, indirect genetic effects, or other sources of familial confounding. We have developed new approaches to assess evidence for and against causation by using family data for pairs of relatives (Inference about Causation from Examination of FAmiliaL CONfounding [ICE FALCON]) or measures of family history (Inference about Causation from Examining Changes in Regression coefficients and Innovative STatistical AnaLyses [ICE CRISTAL]). Inference is made from the changes in reg

Polygenic Risk and Cardiovascular Event Risk in Patients With Atrial Fibrillation With Low to Intermediate Stroke Risk.

BACKGROUND: The clinical utility of the polygenic risk score in predicting cardiovascular events in patients with atrial fibrillation (AF) has not yet been established. This study aimed to determine whether the polygenic risk score for AF might be useful in the risk stratification of AF-related cardiovascular events. METHODS AND RESULTS: This study included 9597 oral anticoagulation-naive patients with AF with a CHA2DS2-VA (congestive heart failure; hypertension; age ≥75 years; diabetes; prior stroke or transient ischemic attack or thromboembolism; vascular disease; and age 65-74 years) score

Gout in Older Adults: The Atherosclerosis Risk in Communities Study.

BACKGROUND: It is unclear whether traditional and genetic risk factors in middle age predict the onset of gout in older age. METHODS: We studied the incidence of gout in older adults using the Atherosclerosis Risk in Communities study, a prospective U.S. population-based cohort of middle-aged adults enrolled between 1987 and 1989 with ongoing follow-up. A genetic urate score was formed from common urate-associated single nucleotide polymorphisms for eight genes. The adjusted hazard ratio and 95% confidence interval of incident gout by traditional and genetic risk factors in middle age were es

Advising women at high risk of breast cancer.

UNLABELLED: Insulin secretion varies widely in preclinical type 1 diabetes. To understand the pathogenesis of this metabolic heterogeneity, we asked whether genetic predisposition to type 2 diabetes, quantified by a type 2 diabetes genetic risk score (T2D-GRS), modulates β-cell function and disease progression in individuals at risk of type 1 diabetes. We analyzed 4,324 islet autoantibody–positive TrialNet Pathway to Prevention participants with genome-wide genotyping and oral glucose tolerance testing. Both T2D-GRS and the type 1 diabetes genetic risk score 2 (T1D-GRS2) differed significantly…

Genetic factor common to schizophrenia and HIV infection is associated with risky sexual behavior: antagonistic vs. synergistic pleiotropic SNPs enriched for distinctly different biological functions.

Schizophrenia (SZ) and HIV infection are serious disorders with a complex phenotypic relationship. Observational studies have described their comorbidity; their genetic correlation is not well studied. We performed extensive analysis in search of common genetic factors for SZ and HIV, and their relationship with risky sexual behavior (RSB). Summary statistics from genome-wide association studies of HIV infection and schizophrenia were obtained and 2379 European Americans were genotyped and assessed for RSB score. Genetic relationships between traits were analyzed in three ways: linkage disequi

HMG-Coenzyme A Reductase as a Drug Target for the Prevention of Ankylosing Spondylitis.

Statins are an inhibitor of 3-hydroxy-3-methylglutaryl coenzyme A reductase (HMGCR). Growing evidence indicates that statins may have an anti-inflammatory effect. Whether genetically proxied HMGCR inhibition can reduce the risk of ankylosing spondylitis is unknown. We constructed an HMGCR genetic score comprising nearly randomly inherited variants significantly associated with LDL cholesterol levels within ± 100 kb from HMGCR to proxy for inhibition of HMGCR. We also constructed PCSK9 and NPC1L1 scores as well as the LDL polygenetic score to proxy for the inhibition of these drug targets as we

Transethnic Genome-Wide Association Study Provides Insights in the Genetic Architecture and Heritability of Long QT Syndrome.

BACKGROUND: Long QT syndrome (LQTS) is a rare genetic disorder and a major preventable cause of sudden cardiac death in the young. A causal rare genetic variant with large effect size is identified in up to 80% of probands (genotype positive) and cascade family screening shows incomplete penetrance of genetic variants. Furthermore, a proportion of cases meeting diagnostic criteria for LQTS remain genetically elusive despite genetic testing of established genes (genotype negative). These observations raise the possibility that common genetic variants with small effect size contribute to the cli

Patient Perspectives on Psychiatric Polygenic Risk Scores in Reproductive Decision-Making and Polygenic Embryo Screening.

Polygenic risk scores (PRS) estimate individuals' genetic risk for developing multifactorial conditions. Recent genome-wide association studies have enabled development of psychiatric PRS, which hold potential to streamline diagnosis and treatment of psychiatric conditions. As individuals with mental illness often identify concern about passing on psychiatric conditions as a key factor in family planning, some may also seek to use psychiatric PRS in reproductive decisions like childbearing or embryo selection for in vitro fertilization. Despite calls for increased regulation, there remains no

Genomic Structural Equation Modeling Provides an Initial View of the Genetic Architecture Related to Type 1 Gaucher Disease.

The genetic architecture underlying traits associated with Type 1 Gaucher disease (GD1) remains insufficiently explored. We integrated genomic structural equation modeling and multiple post-genome-wide association study (GWAS) methodologies to prioritize candidate SNPs associated with GD1-related variation, identifying 15 loci with strong statistical support. Subsequently, diverse transcriptome-wide association approaches were employed to pinpoint susceptibility gene signals strongly correlated with GD1. For selected candidate genes, we explored the potential structural consequences of missens
