RELMβ in intestinal helminth infection: Mechanisms and research advances.

Intestinal helminth infections remain a major global public health challenge, affecting both human and animal health. The immune mechanisms by which hosts resist and expel helminths are multifaceted, with intestinal epithelial cells and their secretory products playing pivotal roles. Resistin-like molecule β (RELMβ), also known as Found in Inflammatory Zone 2 (FIZZ2), is a secreted protein predominantly produced by intestinal goblet cells. Recent studies have revealed that RELMβ exerts important functions in intestinal inflammation and immune regulation, particularly in type 2 immunity-driven

Evaluating evidence for co-geography in the Anopheles-Plasmodium host-parasite system.

The often tight association between parasites and their hosts means that under certain scenarios, the evolutionary histories of the two species can become closely coupled both through time and across space. Using spatial genetic inference, we identify a potential signal of common dispersal patterns in the Anopheles gambiae and Plasmodium falciparum host-parasite system as seen through a between-species correlation of the differences between geographic sampling location and geographic location predicted from the genome. This correlation may be due to coupled dispersal dynamics between host and

Structural definition of babesial RAP-1 proteins identifies a novel protein superfamily across Apicomplexa.

Apicomplexan protozoa are intracellular parasites of medical and economic importance. These parasites contain specialized apical complex organelles, including rhoptries, that participate in the process of host cell invasion. Conserved antigens expressed in the rhoptries are rational vaccine targets, but whether conservation of protein structure is a functional requirement for invasion remains unknown. Novel protein structural modeling enables identification of structurally conserved protein families that are not evident by sequence analysis alone. Here we show by AlphaFold2 structural modeling

No evidence for schistosome parasite fitness trade-offs in the intermediate and definitive host.

BACKGROUND: The trematode parasite Schistosoma mansoni uses an aquatic snail intermediate and a vertebrate definitive host to complete its life cycle. We previously showed that a key transmission trait-the number of cercariae larvae shed from infected Biomphalaria spp. snails-varies significantly within and between different parasite populations and is genetically controlled by five loci. We investigated the hypothesis that the success of parasite genotypes showing high propagative fitness in the intermediate snail host may be offset by lower reproductive fitness in the definitive vertebrate h

Stickleback mass occurrence driven by spatially uneven parasite pressure? Insights into infection dynamics, host mortality, and epizootic variability.

Since 2012, a massive invasion of the three-spined stickleback (Gasterosteus aculeatus) has taken place into the pelagic area of Lake Constance. This species, which had previously been restricted to the littoral zone, is now the dominant pelagic fish and the previously dominant whitefish (Coregonus wartmanni) has suffered severe reductions in growth and recruitment. In this study, in total, 2871 sticklebacks were collected via monthly sessions over a 4-year period in pelagic and benthic areas of Lake Constance and examined for signs of infection with Schistocephalus solidus, a parasite known t

Polar and neutral lipid composition of the copepod Lernaeocera lusci and its host Merluccius merluccius in relationship with the parasite intensity.

Parasitic copepod Lernaeocera lusci is a common mesoparasite of the hake Merluccius merluccius. Although widely distributed throughout the Mediterranean, little is known about this pathogen. The current study was designed to assess the impact of different L. lusci infection loads on lipid classes and their fatty acid (FA) composition in both parasite and the host organs (gills, liver, and muscle). Results showed a significant decrease in total lipid, neutral lipid (NL), and polar lipid (PL) contents in all analyzed host's organs in relationship with parasite intensity. Gills appeared to be the

Cold water reduces the severity of parasite-inflicted damage: support for wintertime recuperation in aquatic hosts.

The reduction in host fitness caused by parasite infections (virulence) depends on infection intensity and the degree of damage caused per parasite. Environmental conditions can shape both virulence components, but in contrast to infection intensity, environmental impacts on per-parasite damage are poorly understood. Here, we studied the effect of ambient temperature on per-parasite damage, which is jointly determined by the ability of parasites to induce harm (per-parasite pathogenicity) and the ability of hosts to limit damage (tolerance). We experimentally exposed two salmonid species, Atla

Gall-Inducing Parasites: Convergent and Conserved Strategies of Plant Manipulation by Insects and Nematodes.

Gall-inducing insects and nematodes engage in sophisticated interactions with their host plants. These parasites can induce major morphological and physiological changes in host roots, leaves, and other tissues. Sedentary endoparasitic nematodes, root-knot and cyst nematodes in particular, as well as gall-inducing and leaf-mining insects, manipulate plant development to form unique organs that provide them with food from feeding cells. Sometimes, infected tissues may undergo a developmental switch resulting in the formation of aberrant and spectacular structures (clubs or galls). We describe h

New scope on the relationship between rotifers and Biomphalaria alexandrina snails.

OBJECTIVE: To investigate the effect of rotifer internalization into snail tissue on the development of schistosomes. METHODS: Susceptible laboratory-bred Biomphalaria alexandrina (B. alexandrina) snails were exposed to lab-maintained rotifers; Philodina spp., two weeks before and after being infected with Schistosoma mansoni (S. mansoni) miracidia. The consequent histopathological impact on snail tissues and cercarial biology were investigated before and after emergence from snails. RESULTS: Contamination of B. alexandrina snails with philodina, two weeks before miracidial exposure, was fou

An unexpected advantage of whiteness in horses: the most horsefly-proof horse has a depolarizing white coat.

White horses frequently suffer from malign skin cancer and visual deficiencies owing to their high sensitivity to the ultraviolet solar radiation. Furthermore, in the wild, white horses suffer a larger predation risk than dark individuals because they can more easily be detected. In spite of their greater vulnerability, white horses have been highly appreciated for centuries owing to their natural rarity. Here, we show that blood-sucking tabanid flies, known to transmit disease agents to mammals, are less attracted to white than dark horses. We also demonstrate that tabanids use reflected pola

Intra-ChIP: studying gene regulation in an intracellular pathogen.

Intracellular bacteria that reside within a host cell use a variety of strategies to exploit this unique niche. While these organisms are technically challenging to study in the context of an infected host cell, recent advances have led to an improved understanding of how the intracellular environment impacts bacterial gene expression. We recently demonstrated that chromatin immunoprecipitation (ChIP) can be used to quantify transcription factor binding in the obligate intracellular pathogen Chlamydia trachomatis within infected cells. Furthermore, we showed it was possible to experimentally m

Insights into the pathogenesis of disease in human lymphatic filariasis.

Although two thirds of the 120 million people infected with lymph-dwelling filarial parasites have subclinical infections, ∼40 million have lymphedema and/or other pathologic manifestations including hydroceles (and other forms of urogenital disease), episodic adenolymphangitis, lymphedema, and (in its most severe form) elephantiasis. Adult filarial worms reside in the lymphatics and lymph nodes and induce lymphatic dilatation. Progressive lymphatic damage and pathology results primarily from the host inflammatory response to the parasites but also perhaps from the host inflammatory response t

Mitochondria of a human skeletal muscle cell line associate with the parasitophorous vacuoles of three archetypal lineages of Toxoplasma gondii and influence fatty acid import.

Toxoplasma gondii, an obligate intracellular parasite, acquires host nutrients to sustain its intracellular replication. A key interaction involves host mitochondrial association (HMA) with the parasitophorous vacuole membrane, previously thought to be strain- and cell-type-dependent, and notably absent in type 2 strains in fibroblasts. Here, we report that in human skeletal muscle KD3 myotubes, all three archetypal T. gondii strains-including type 2-demonstrate significant HMA. This association was confirmed by mitotracker staining and transmission electron microscopy. Notably, HMA appears to

Environmental temperature variation influences fitness trade-offs and tolerance in a fish-tapeworm association.

BACKGROUND: Increasing temperatures are predicted to strongly impact host-parasite interactions, but empirical tests are rare. Host species that are naturally exposed to a broad temperature spectrum offer the possibility to investigate the effects of elevated temperatures on hosts and parasites. Using three-spined sticklebacks, Gasterosteus aculeatus L., and tapeworms, Schistocephalus solidus (Müller, 1776), originating from a cold and a warm water site of a volcanic lake, we subjected sympatric and allopatric host-parasite combinations to cold and warm conditions in a fully crossed design. Parasites and allergies: a complex bidirectional relationship from evolutionary origins to modern therapeutics.

Parasites and allergic diseases are linked by a complex, bidirectional relationship shaped by long-term host-parasite coevolution. This review discusses how different parasites may either promote or attenuate allergic responses through immunological, epithelial, and microbiome-mediated mechanisms. IgE-mediated immunity, mast cell activation, eosinophilia, and pruritus may have evolved as protective responses against helminths and blood-feeding ectoparasites. In contrast, modern allergies may partly reflect misdirected responses to harmless environmental antigens. The effects of parasites on al

Parasite infection disrupts escape behaviours in fish shoals.

Many prey species have evolved collective responses to avoid predation. They rapidly transfer information about potential predators to trigger and coordinate escape waves. Predation avoidance behaviour is often manipulated by trophically transmitted parasites, to facilitate their transmission to the next host. We hypothesized that the presence of infected, behaviourally altered individuals might disturb the spread of escape waves. We used the tapeworm Schistocephalus solidus, which increases risk-taking behaviour and decreases social responsiveness of its host, the three-spined stickleback, to

Phenotypic plasticity of pre-adult egg maturation in a parasitoid: Effects of host-starvation and brood size.

Larvae of parasitoid wasps develop on a single arthropod host, and often face resource limitation that induces a tradeoff between egg maturation and somatic growth. Part of the variation in the growth-reproduction allocation was shown to be heritable, but how the larval developmental environment affects this allocation is not well-known. Detection of life history tradeoffs is often facilitated under stress conditions. We therefore exposed developing female larvae of the polyembryonic parasitoid Copidosoma koehleri (Hymenoptera: Encyrtidae) to laboratory manipulations aimed to restrict host res

Detangling the Crosstalk Between Ascaris, Trichuris and Gut Microbiota: What´s Next?

Helminth infections remain a global public health issue, particularly in low- and middle-income countries, where roundworms from theTrichuris and Ascaris genera are most prevalent. These geohelminths not only impact human health but most importantly also affect animal well-being, in particular the swine industry. Host-helminth parasite interactions are complex and at the same time essential to understand the biology, dynamics and pathophysiology of these infections. Within these interactions, the immunomodulatory capacity of these helminths in the host has been extensively studied. Moreover, i

Spatial, Temporal, and Host-Specificity Patterns of Three Genera of Blood Coccidian Parasites in Two Syntopic Mediterranean Lizards.

We investigated the phenological variation of three genera of blood parasites of lizards (Schellackia, Karyolysus, and Lankesterella) using samples of 275 adult Psammodromus algirus and 138 Acanthodactylus erythrurus collected from April to September in 2021 and 2022. Both years experienced heatwaves, and 2022 had the warmest summer on historical record for the region. We captured lizards in two close areas that differed in vegetation quality due to differential human pressure. We found that Schellackia and Karyolysus more frequently infected P. algirus, whereas Lankesterella infected A. eryth

The parasitic dinoflagellate Hematodinium infects multiple crustaceans in the polyculture systems of Shandong Province, China.

The parasitic dinoflagellates of the Hematodinium genus have impacted wild and cultured stocks of commercial crustaceans worldwide. In the past decade, outbreaks of Hematodinium epizootics resulted in substantial mortalities in cultured Chinese swimming crabs Portunus trituberculatus in the polyculture ponds located in Shandong Peninsula, whereas the source and transmission of the parasite in the polyculture pond system remains to be determined. During April to December of 2018, 2034 crabs and 108 shrimps were collected from the polyculture pond systems in the highly endemic area of Hematodini
