Human T-cell function in experimental ascorbic acid deficiency and spontaneous scurvy.

Studies in animal models suggest that ascorbic deficiency impairs T-cell-mediated immunity. We studied five normal volunteers hospitalized on a metabolic unit and consuming a strictly controlled diet deficient in ascorbic acid I) after a 5-wk control period of ascorbic acid supplementation (75 mg/day) and 2) after a 9-wk period of no supplementation. Three of the subjects were restudied after a 5-wk period of ascorbic acid supplementation after the deficient period. At the end of both control periods ascorbic acid levels in plasma ranged from 0.9 to 1.3 mg/dl and in leukocytes from 19 to 30 mi

On the persistence of human serum resistance and isoenzyme patterns of Trypanozoon in experimentally infected pigs.

'Mini-pigs' were infected with salivarian Trypanozoon clones to examine the persistence and stability of the human serum resistance [Blood Incubation Infectivity Test (BIIT)] and isoenzyme characteristics during infection in a new host. A stock regarded as Trypanosoma brucei, derived from a domestic pig in the Ivory Coast, retained its BIIT negative (serum sensitive), alanine aminotransferase (ALAT) and peptidase 2 (PEP 2) characteristics throughout 343 days of infection in pigs. Similarly there was no change in the BIIT positive (serum resistant) and different ALAT and PEP characteristics of

[Xenotransplantation of cultures of islet cells of human fetal pancreas into experimentally diabetic rats].

Cultures of islet cells of fetal human pancreas were transplanted into the liver of alloxan diabetic rats. This xenotransplantation led to a long-term reduction of glycemia maintained during 8--18 weeks. Histological studies of the liver of recipients revealed the presence of the implanted islet cells.

[Oscillometry on various stape protheses. Experimental examination of human temporal bone preparations].

In human temporal bone specimens the vibrations of the labyrinthine fluid were measured by a piezoelectric system after application of various techniques of otosclerosis surgery. Compared with the amplitude in the case of a normal stapes, the surgical techniques according to Zangemeister, Shea and Schuknect resulted in considerable transmission losses in the high frequency range, while transmission of low frequencies to the inner ear was almost equivalent to normal transmission via the stapes. For frequencies of more than 3,000 Hertz the damping influence of the fibrous tissue implanted into t

An evaluation of oxygen therapy equipment. Experimental study of various devices on the human subject.

Six commercially available devices were tested to determine the alveolar oxygen concentration that is achieved and the volume of rebreathing that can occur in conditions simulating clinical use. All were tested at zero oxygen flow, since failure and interruption of the oxygen supply to such devices can occur in clinical use. In patients whose cardiorespiratory reserve is exhausted, any increase in dead space will be detrimental; for such patients devices should be selected which cannot cause rebreathing under any circumstances. Three of the devices tested could cause appreciable rebreathing, i

A common T-cell epitope between human thyroglobulin and human thyroid peroxidase is related to murine experimental autoimmune thyroiditis.

We have investigated functional common T-cell epitopes between human thyroglobulin (hTg) and human thyroid peroxidase (hTPO) in mice. Four hTg peptides, Tg-P1, Tg-P2, Tg-P3 and Tg-P4, in which 5 amino acid residues are identical to those of hTPO, and 1 hTPO peptide, TPO-P4 relevant to Tg-P4, were prepared. Among these peptides, only Tg-P4 (residues 2730-2743) and TPO-P4 (residues 118-131) were highly antigenic and both peptides shared the common T-cell epitope. In addition, when the spleen cells from mice immunized with mouse Tg (mTg) were restimulated in vitro by Tg-P4 or TPO-P4 as well as by

The effects of radiofrequency electromagnetic fields exposure on human self-reported symptoms: A protocol for a systematic review of human experimental studies.

BACKGROUND: The technological applications of radiofrequency electromagnetic fields (RF-EMF) have been steadily increasing since the 1950s across multiple sectors exposing large proportions of the population. This fact has raised concerns related to the potential consequences to people's health. The World Health Organization (WHO) is assessing the potential health effects of exposure to RF-EMF and has carried out an international survey amongst experts, who have identified six priority topics to be further addressed through systematic reviews, whereof the effects on symptoms is one of them. We

Association of mucosal neutrophil inflammation and cytokine responses with natural and experimental pneumococcal carriage in a randomised vaccine trial using experimental human pneumococcal carriage.

BACKGROUND: Mucosal inflammation is associated with increased nasal pneumococcal colonisation, but the specific mechanisms are not fully understood. We aimed to find innate immune factors associated with pneumococcal carriage using a controlled human infection model. METHODS: Healthy Malawian adults participating in a randomised trial of pneumococcal conjugate vaccine (PCV13) were inoculated with one of three doses of Streptococcus pneumoniae 6B. We categorised the participants into 4 pneumococcal carriage outcome groups - no carriage; natural carriage; experimental carriage; and dual carriag

Safety, tolerability and pharmacokinetics/pharmacodynamics of the adrenomedullin antibody adrecizumab in a first-in-human study and during experimental human endotoxaemia in healthy subjects.

AIMS: Adrenomedullin (ADM) is an important regulator of endothelial barrier function and vascular tone, and may represent a novel treatment target in sepsis. The non-neutralizing ADM antibody adrecizumab has shown promising results in preclinical sepsis models. In the present study, we investigated the safety, tolerability and pharmacokinetics (PK)/pharmacodynamics of adrecizumab in a first-in-man study and in a second study during experimental human endotoxaemia. METHODS: Forty-eight healthy male volunteers were enrolled in two randomized, double-blind, placebo-controlled phase I studies. In

Human-induced expanded distribution of Anopheles plumbeus, experimental vector of West Nile virus and a potential vector of human malaria in Belgium.

For the majority of native species, human-created habitats provide a hostile environment that prevents their colonization. However, if the conditions encountered in this novel environment are part of the fundamental niche of a particular species, these low competitive environments may allow strong population expansion of even rare and stenotopic species. If these species are potentially harmful to humans, such anthropogenic habitat alterations may impose strong risks for human health. Here, we report on a recent and severe outbreak of the viciously biting and day-active mosquito Anopheles plum

Interaction of a biologically active Zn2+ complex with human serum albumin: experimental and theoretical insights.

We report the synthesis and characterization of a novel Zn2+ complex derived from a coumarin-based azo dye ligand. The complex was confirmed by 1H NMR and ESI–MS analyses and further examined for its biological activity. Antibacterial assays revealed significant inhibition against Escherichia coli (DH5α), while cytotoxicity tests on the human breast cancer cell line MDA-MB-468 indicated moderate dose-dependent effects. Protein-binding studies using UV–Vis, fluorescence, circular dichroism, and time-resolved fluorescence spectroscopy demonstrated that the complex interacts with human serum albu

Primary culture and characterization of human upper limb muscle satellite cells: an experimental study.

BACKGROUND: Human muscle satellite (stem) cells (MuSCs) are essential for investigating muscle physiology, regeneration, and disease mechanisms. Primary cultures derived directly from human tissues offer a more physiologically relevant model than immortalized cell lines. However, the isolation and characterization of MuSCs from human upper limb tissues are limited. Therefore, this study aimed to establish and characterize a primary culture system for MuSCs obtained from human upper limb muscle tissue. METHODS: Human muscle tissues were obtained from upper limb surgical specimens. Muscle sampl

Plasma Metabolomics Reveals a Shared Metabolomic Profile in Experimental and Human Chronic Kidney Disease.

Chronic kidney disease (CKD) affects nearly 10% of the global population, yet diagnosis and disease monitoring still rely primarily on plasma creatinine. Because creatinine levels are strongly influenced by non-renal factors, such as age, sex, muscle mass, and diet, its accuracy as a kidney function marker is limited. To identify plasma biomarkers that reflect kidney injury, we applied untargeted and targeted metabolomics in the adenine-induced CKD mouse model, a well-known tubular damage model, and validated the findings in plasma from patients with advanced CKD and healthy controls. We ident

Candida albicans: Insights into the Biology and Experimental Innovations of a Commonly Isolated Human Fungal Pathogen.

Candida albicans is an opportunistic pathogen associated with healthy humans. It is the major causative agent for superficial and invasive candidiasis in immunocompromised individuals globally. Lack of awareness toward fungal infections, poor disease management, and increasing drug resistance have contributed to the burden of C. albicans-associated diseases. C. albicans displays polymorphism, assisting in host interaction, virulence, and sustenance of the pathogen. Well-known polymorphic forms of C. albicans include yeast, hyphal, pseudohyphal, white, opaque, gray, GUT, and chlamydospores. Whi
