BACKGROUND: Primary biliary cirrhosis (PBC) is a complex disease with genetic and environmental influences. The disease is more prevalent in families with PBC and candidate gene case-control studies have linked PBC with DRB1(*)08 human leucocyte antigen class II alleles.
AIMS: The goal of this study was to characterize a MER115 intergenic region on chromosome 4 as a putative genetic variant associated with PBC.
METHODS/RESULTS: This region was incidentally identified during investigations to discover candidate microbial agents using representational difference analysis (RDA) with liver sampl

The genetic basis of hybrid vigor or heterosis has been debated for more than a century. A popular hypothesis to explain this phenomenon is that there are different slightly deleterious recessive homozygous alleles in the two parents and that these alleles are complemented in the hybrid so that the biomass and fertility exceed both parents. To address the complementation hypothesis in a direct manner, heterosis was examined in diploid inbreds and reciprocal hybrids and compared with matched triploid inbred derivatives and two types of triploid hybrids that differ in the number of genomes from

Adenoviral, retroviral/lentiviral, adeno-associated viral, and herpesviral vectors are the major viral vectors used in gene therapy. Compared with non-viral methods, viruses are highly-evolved, natural delivery agents for genetic materials. Despite their remarkable transduction efficiency, both clinical trials and laboratory experiments have suggested that viral vectors have inherent shortcomings for gene therapy, including limited loading capacity, immunogenicity, genotoxicity, and failure to support long-term adequate transgenic expression. One of the key issues in viral gene therapy is the

The translation of cell-derived extracellular vesicles (EVs) into biogenic gene delivery systems is limited by relatively inefficient loading strategies. In this work, the loading of various nucleic acids into small EVs via their spontaneous hybridization with preloaded non-lamellar liquid crystalline lipid nanoparticles (LCNPs), forming hybrid EVs (HEVs) is described. It is demonstrated that LCNPs undergo pH-dependent structural transitions from inverse hexagonal (HII) phases at pH 5 to more disordered non-lamellar phases, possibly inverse micellar (L2) or sponge (L3) phases, at pH 7.4, which

Hexaploid triticale is an important genetic resource for genetic improvement of common wheat, which can broaden the genetic basis of wheat. In order to lay a foundation for the subsequent research and utilization of triticale germplasm materials, the chromosomal genetic characteristics of cross and backcross offspring of hexaploid triticale×hexaploid wheat were investigated in the process of transferring rye chromatin from hexaploid triticale to hexaploid wheat. Hybrid and backcross combinations were prepared with hexaploid triticale 16yin171 as the maternal parent and hexaploid wheat Chuanmai

Introgression among parasite species has the potential to transfer traits of biomedical importance across species boundaries. The parasitic blood fluke Schistosoma haematobium causes urogenital schistosomiasis in humans across sub-Saharan Africa. Hybridization with other schistosome species is assumed to occur commonly, because genetic crosses between S. haematobium and livestock schistosomes, including S. bovis, can be staged in the laboratory, and sequencing of mtDNA and rDNA amplified from microscopic miracidia larvae frequently reveals markers from different species. However, the frequency

The grasshopper Trimerotropis thalassica (Bruner) has a diploid count of 2n=23 male (XO), 24 female (XX). The two largest autosomes pairs are regularly metacentric, a consequence of fixed pericentric inversions. The X-chromosome is also a fixed metacentric. The remaining nine pairs of autosmes are polymorphic for floating percentric inversions so that the complement consists of a mixture of telocentric and metacentric members. Trimerotropis occidentalis (Bruner) is polymorphic for comparable inversions in only two of its autosome pairs and has a telocentric X. It is however, unique among the s

Speciation typically involves a stage in which species can still exchange genetic material. Interspecific gene flow is facilitated by the hybrid zones that such species establish upon secondary contact. If one member of a hybridizing species pair displaces the other, their hybrid zone would move across the landscape. Although theory predicts that moving hybrid zones quickly stagnate, hybrid zones tracked over one or a few decades do not always follow such a limitation. This suggests that hybrid zones have the potential to traverse considerable distances over extended periods of time. When hybr

BACKGROUND: Honey bees are essential pollinators supporting agricultural production and wild plant diversity. In evolutionary lineage M, some populations are threatened by genetic erosion caused by the widespread introduction of commercially bred queens. To assess this risk, wing images from existing and new datasets were used to assign them to 4 evolutionary lineages (A, C, M, and O). The new dataset consisted of 29,043 wing images representing 1,342 colony samples from 10 countries.
RESULTS: Overall, 63.7% of colonies belonged to lineage M, whereas 27.5% were classified as A, 7.9% as C, and

Diploid potato breeding enables faster genetic improvement via selection against deleterious alleles in inbred lines, unlike breeding by intercrossing tetraploid varieties. Starch is the major source of calories in potato tubers, but the starch properties of diploid lines have rarely been reported. In this study, we provide a comprehensive characterisation of tuber and starch properties in two diploid lines that are early isolates from the Solynta breeding program, B26 and B100, and their F1 hybrids. B100 produced fewer, but larger tubers compared to B26, and both diploid lines produced tubers

Chickens are believed to have inhabited the Hawaiian island of Kauai since the first human migrations around 1200AD, but numbers have peaked since the tropical storms Iniki and Iwa in the 1980s and 1990s that destroyed almost all the chicken coops on the island and released large numbers of domestic chickens into the wild. Previous studies have shown these now feral chickens are an admixed population between Red Junglefowl (RJF) and domestic chickens. Here, using genetic haplotypic data, we estimate the time of the admixture event between the feral population on the island and the RJF to 1981

BACKGROUND: Minke whales are separated into two genetically distinct species: the Antarctic minke whale found in the southern hemisphere, and the common minke whale which is cosmopolitan. The common minke whale is further divided into three allopatric sub-species found in the North Pacific, southern hemisphere, and the North Atlantic. Here, we aimed to identify the genetic ancestry of a pregnant female minke whale captured in the North Atlantic in 2010, and her fetus, using data from the mtDNA control region, 11 microsatellite loci and a sex determining marker.
RESULTS: All statistical parame

Advances in CRISPR-Cas9-based genome editing have simplified the generation of isogenic induced pluripotent stem cell (iPSC) pairs differing by a single genetic perturbation, minimizing interindividual variability in disease modeling. However, imprecise editing can cause unintended mutations that distort genotype-phenotype interpretation. Here, we investigated the molecular consequences of CRISPR-Cas9 editing in human iPSCs carrying a tandem duplication of the STAG2 locus. Using hybridization capture-based targeted next-generation sequencing of genomic regions surrounding Cas9-induced double-s

Identification of the genomic regions that contribute to reproductive isolation and how they interact is a major goal of evolutionary genetics. Much effort has focused on locating candidate genes and potential barrier loci by scanning genomes for regions of excess differentiation (FST). An alternative, and perhaps more robust approach, is to scan for genomic regions exhibiting steep clines in allele frequency across a hybrid zone. We develop a computationally efficient method for approximating cline parameters for large number of loci, and apply it to genomic data from across a hybrid zone bet

BACKGROUND: Primary trisomy is a powerful genetic tool in plants. However, trisomy has not been detected in Populus as a model system for tree and woody perennial plant biology.
RESULTS: In the present study, a backcross between Populus alba × Populus glandulosa 'YXY 7#' (2n = 2x = 38) and the triploid hybrid 'Beilinxiongzhu 1#' (2n = 3x = 57) based on the observation of microsporogenesis and an evaluation of the variations in pollen was conducted to create primary trisomy. Many abnormalities, such as premature migration of chromosomes, lagging of chromosomes, chromosome bridges, asymmetric s

OBJECTIVE: Comparative genomic hybridization (CGH) has been established as an informative technique in genetic analysis. However, differences in the ratio of hybridization intensities were reported for particular chromosomes, which may affect CGH results. The aim of this study was to define these differences in more detail. For this purpose, CGH results of 70 samples of bone marrow cells (BMC) with normal karyotype in conventional cytogenetics (CC) were evaluated using seven different reference DNAs and two different DNA labeling systems.
METHODS AND RESULTS: CGH using fluorochrome-conjugated

The genomes of two new human papillomavirus (HPV) types, named HPV54 and HPV55, were cloned from penile lesions of 2 patients. HPV54 was isolated from a verrucous carcinoma (Buschke-Löwenstein tumour) together with full-length HPV6 genomes and HPV6 DNA molecules with a deletion of about 0.3 kb located in the non-coding region. HPV55 was isolated from a condyloma acuminatum. No cross-hybridization was observed between HPV54 DNA and the DNAs of the known cutaneous and genital HPVs by blot hybridization experiments performed under stringent conditions. In contrast, significant cross-hybridization

Disomic alien addition lines (DAALs, 2n = 42) were obtained from an intersubgeneric cross between Glycine max [L.] Merr. cv. Dwight (2n = 40, G1G1) and Glycine tomentella Hayata (PI 441001, 2n = 78, D3D3CC). These DAALs had two homologous G. tomentella chromosomes. In some progenies of the DAALs the extra G. tomentella chromosomes were eliminated. One objective of this research was to identify differences in gene expression between a DAAL (LG13-7552) and one of its 2n = 40 progenies (LG12-7063). RNA-sequencing documented that many genes critical to fundamental plant growth and related to stres

We report on a genome-wide scan for introgression between the house mouse (Mus musculus domesticus) and the Algerian mouse (Mus spretus), using samples from the ranges of sympatry and allopatry in Africa and Europe. Our analysis reveals wide variability in introgression signatures along the genomes, as well as across the samples. We find that fewer than half of the autosomes in each genome harbor all detectable introgression, whereas the X chromosome has none. Further, European mice carry more M. spretus alleles than the sympatric African ones. Using the length distribution and sharing pattern

A foundation population derived from nine different rat strains was subjected to genetic selection for high and low rearing activity. A rapid response to selection was demonstrated, the two selected lines showing virtually no overlap in scores after 10 generations. Heritability estimates were assessed from parent-offspring regressions in the unselected control line and from comparisons of the achieved response with the selection pressure applied in the selected lines. Overall, the heritability of rearing activity in the rat was assessed to be about 0.25. In addition, a major gene effect was re
