Lysosomal acid hydrolase alterations in gastric mucosa from an experimental peptic ulcer model.

Ligation of the common bile duct in pigs causes esophagogastric ulceration in 100 per cent of instances within seven days postoperatively. The results of previous studies have suggested a peptic cause for these ulcers. The possible role of lysosomal acid hydrolases in the primary pathogenesis was investigated herein. The total activities of two lysosomal acid hydrolases in mucosal specimens taken at biopsy from ulcerated pars esophagea and one in ulcerated gastric cardia were significantly lower in pigs undergoing ligation of the bile duct postoperatively, than in comparable pigs undergoing sh…

Differential metabolism of acrylonitrile to cyanide is responsible for the greater sensitivity of male vs female mice: role of CYP2E1 and epoxide hydrolases.

Acrylonitrile (AN) is a potent toxicant and a known rodent carcinogen. AN epoxidation to cyanoethylene oxide (CEO) via CYP2E1 and its subsequent metabolism via epoxide hydrolases (EH) to yield cyanide is thought to be responsible for the acute toxicity and mortality of AN. Recent reports showed that male mice are more sensitive than females to the acute toxicity/mortality of AN. The present work was undertaken to assess the metabolic and enzymatic basis for the greater sensitivity of male vs female mice to AN toxicity. Male and female wild-type and CYP2E1-null mice received AN at 0, 2.5, 10, 2…

Inhibition of soluble epoxide hydrolase ameliorates renal injury in IgA nephropathy by restoring epoxyeicosatrienoic acids.

Immunoglobulin A (IgA) nephropathy (IgAN) is a major cause of end-stage renal disease with limited therapies. Altered lipid metabolism is implicated in chronic kidney disease, but its role in IgAN is unclear. Plasma oxylipin profiling and renal single-cell RNA sequencing (scRNA-seq) revealed reduced epoxyeicosatrienoic acids (EETs) and increased soluble epoxide hydrolase (sEH) in IgAN. Two sEH inhibitors, t-AUCB and macamide, reduced proteinuria, improved renal function, attenuated IgA deposition, and restored EET levels. Mechanistically, sEH overexpression activated NF-κB (p65 phosphorylation

Structural and functional analysis of peptidyl-tRNA hydrolase 2 from Candidatus Lokiarchaeum sp. GC14_75.

Peptidyl-tRNA hydrolase (Pth2) hydrolyzes peptidyl-tRNA, an immature product of aborted translation, into peptide and tRNA, thereby maintaining cellular protein synthesis through peptide release and tRNA recycling. Here, we present the crystal structure of Pth2 from Candidatus Lokiarchaeum sp. GC14_75 (LokiPth2) at 2.12 Å resolution. This is the first structure of Pth2 from a lineage within Promethearchaeati, a kingdom of archaea closely related to eukaryotes. The structure reveals that LokiPth2 forms a homodimer and closely resembles Pth2 structures from other species. However, LokiPth2 exhib

Hydration at C5'-C6' double bond of adenosine substrates by S-adenosyl-L-homocysteine hydrolase.

It was previously demonstrated that (E)-5',6'-didehydro-6'-deoxy-6'-fluorohomoadenosine (EDDFHA) undergoes addition of water across the isolated C5'-C6' double bond by S-adenosyl-L-homocysteine (AdoHcy) hydrolase without prior oxidation at C3' (J. Biol. Chem.1993, 268, 17030). Addition of water at the 5' position results in the formation of 6'-deoxy-6'-fluoro-5'-hydroxyhomoadenosine (DFHHA). We now describe preparation of EDDFHA, its Z-isomer and their 6'-[2H]-labeled analogues as well independent syntheses of DFHHA and 6'-deuterio-DFHHA. These compounds allow the assignment of absolute config

Fatty Acid Amide Hydrolase Signaling and Ovarian Disorders: From Molecular Mechanism to Clinical Significance.

Fatty acid amide hydrolase (FAAH) is a central component of the endocannabinoid system (ECS), where it primarily regulates intracellular levels of anandamide (AEA) through enzymatic hydrolysis. Although FAAH has been extensively studied in neural and immune contexts, its involvement in female reproductive physiology is receiving increasing attention. Accumulating evidence indicates that FAAH participates in several important ovarian processes, including follicular development, steroid hormone synthesis, ovulation, and luteal function. In this review, we outline the biochemical properties of FA

In Silico Optimization of a Bifunctional Lipase-Polyethylene Terephthalate (PET) Hydrolase for Enhanced PET and Lipid Hydrolysis.

Polyethylene terephthalate (PET) and lipid-rich waste pose urgent environmental challenges that need multifunctional biocatalysts. LipMRD9, a lidless lipase with natural PETase activity, exemplifies such a scaffold. Here, we used an integrated approach combining automated in silico mutagenesis (AsiteDesign), Protein Energy Landscape Exploration (PELE), and molecular dynamics (MD) simulations, structure-guided rational engineering, and bioprospecting of homologous sequences to improve the dual lipase-PETase activity of LipMRD9. Computational analyses pinpointed mutational hotspots extending the

Acyloxyacyl Hydrolase Prevents Colitis and Colitis-Associated Colorectal Cancer by Inactivating Stimulatory LPS in the Intestine.

Ulcerative colitis (UC) is believed to be triggered by a dysregulated inflammatory response to the intestinal microbiota. Acyloxyacyl hydrolase (AOAH) is a unique host lipase that inactivates Gram-negative bacterial lipopolysaccharides (LPS). After finding that AOAH produced in the intestine decreases stimulatory LPS levels in colon contents, we used the dextran sodium sulfate (DSS) model to test the enzyme's ability to prevent colitis in mice. We found that AOAH played a protective role by decreasing colonic inflammation, tissue injury, and barrier permeability. Increasing or decreasing intes

Inhibition of soluble epoxide hydrolase confers neuroprotection and restores microglial homeostasis in a tauopathy mouse model.

BACKGROUND: The epoxyeicosatrienoic acids (EETs) are derivatives of the arachidonic acid metabolism with anti-inflammatory activities. However, their efficacy is limited due to the rapid hydrolysis by soluble epoxide hydrolase (sEH). Inhibition of sEH has been shown to stabilize the EETs and reduce neuroinflammation in Aβ mouse models of Alzheimer's disease (AD). However, the role of the sEH-EET signaling pathway in other CNS cell types and neurodegenerative conditions are less understood. METHODS: Here we investigated the mechanisms and functional role of the sEH-EET axis in tauopathy by tre

Hydroxylation near the epoxyketone of carfilzomib confers protection from microsomal epoxide hydrolase-mediated metabolism.

Carfilzomib (CFZ) is a tetrapeptide epoxyketone-based proteasome inhibitor (PI) drug approved for multiple myeloma therapy. Despite its improved efficacy and safety profiles over bortezomib (the first-in-class PI drug), CFZ has a short half-life in vivo (< 1 hour), possibly contributing to the lack of efficacy against solid cancers. Previous studies indicated that microsomal epoxide hydrolase (mEH) plays a predominant role in the metabolic degradation of CFZ. With that in mind, we synthesized a novel hydroxylated analog of CFZ (dubbed "CFZ-OH"), which was predicted to have a lower affinity to

The phosphatase activity of soluble epoxide hydrolase regulates vascular calcification through the metabolism of pyrophosphate anions.

While the hydrolase activity of soluble epoxide hydrolase (sEH) reduces vascular calcification, it is not known whether the phosphatase activity of sEH (sEH-P) is also involved. Pharmacological and genetic inhibition of sEH-P reduced the increased calcium deposition in rat aortic rings cultured under high-phosphate conditions. This was associated with decreased mRNA expression of the osteochondrogenic markers Msx2 and Sox9. Deendothelialization of the aortic rings abolished this anticalcifying effect, while the calcification of human aortic smooth muscle cells was unaffected by sEH-P inhibitio

Peptidyl-tRNA hydrolase 2 is a negative regulator of peripartum cardiomyopathy with heart failure in female mice.

The peptidyl-tRNA hydrolase 2 (PTRH2, Bit-1, BIT1) gene plays a pro-survival role during development with loss of function gene mutations causing congenital infantile multisystem disease (IMNEPD). In wild-type female mice hearts, Ptrh2 protein levels significantly increase during pregnancy and decrease postpartum, demonstrating a protective role in response to pregnancy-initiated cardiac stresses. Peripartum cardiomyopathy (PPCM) is due to dysregulated protective signaling in the pregnant heart. The genetic and molecular mechanisms underlying PPCM remain poorly defined with no specific therapi

Characterization of structure of peptidyl-tRNA hydrolase from Enterococcus faecium and its inhibition by a pyrrolinone compound.

In bacteria, peptidyl-tRNA hydrolase (Pth, E.C. 3.1.1.29) is a ubiquitous and essential enzyme for preventing the accumulation of peptidyl-tRNA and sequestration of tRNA. Pth is an esterase that cleaves the ester bond between peptide and tRNA. Here, we present the crystal structure of Pth from Enterococcus faecium (EfPth) at a resolution of 1.92 Å. The two molecules in the asymmetric unit differ in the orientation of sidechain of N66, a conserved residue of the catalytic site. Enzymatic hydrolysis of substrate α-N-BODIPY-lysyl-tRNALys (BLT) by EfPth was characterized by Michaelis-Menten parame

Inhibition of soluble epoxide hydrolase reverses bone loss in periodontitis by upregulating EMCN and inhibiting osteoclasts.

BACKGROUND: Improving the microenvironment to augment endogenous regenerative potential has emerged as a fundamental concept for stimulating and expediting periodontal tissue repair and regeneration. Previous studies have demonstrated that TPPU, a soluble epoxide hydrolase inhibitor (sEHi), mediates the suppression of inflammatory bone loss in periodontitis models. However, the underlying mechanisms remain largely elusive. METHODS: In this study, we constructed a human umbilical vein endothelial cell (HUVEC) and periodontal ligament stem cell (PDLSC) coculture system in vitro and tested the a

Hypomorphic variants of lactase-phlorizin hydrolase in congenital lactase deficiency are trafficking incompetent and functionally inactive.

Patients with the rare autosomal recessive disorder congenital lactase deficiency (CLD) present with severe, potentially life-threatening symptoms shortly after birth. Several variants have been characterized within the gene for lactase-phlorizin hydrolase (LCT) that are associated with CLD. Here, we analyze at the biochemical and cellular levels LCT mutants harboring the genetic variants p.Y1390*, p.E1612*, p.S1150Pfs*19, p.S1121L, p.R1587H, and p.S688P. Our data unequivocally demonstrate that these mutants are absolutely transport incompetent, some of which are readily degraded, and are enzy
