Collagen organization and structure in FLBN5-/- mice using label-free microscopy: implications for pelvic organ prolapse.

Pelvic organ prolapse (POP) is a gynecological disorder described by the descent of superior pelvic organs into or out of the vagina as a co … nalysis of images acquired using second harmonic generation microscopy. Furthermore, coherent Raman hyperspectral imaging revealed elevated disorder in the secondary structure of collagen in prolapsed tissues. Our results underscore the need for in situ multimodal monitoring of collagen organization to improve POP predictive capabilities.

Feasibility of the optical imaging of thrombus formation in a rotary blood pump by near-infrared light.

Blood coagulation is one of the primary concerns when using mechanical circulatory support devices such as blood pumps. Noninvasive detection and imaging of thrombus formation is useful not only for the development of more hemocompatible devices but also for the management of blood coagulation to avoid risk of infarction. The objective of this study is to investigate the use of near-infrared light for imaging of thrombus formation in a rotary blood pump. The optical properties of a thrombus at wavelengths ranging from 600 to 750 nm were analyzed using a hyperspectral imaging (HSI) system. A sp…

Targeted Imaging of VCAM-1 mRNA in a Mouse Model of Laser-Induced Choroidal Neovascularization Using Antisense Hairpin-DNA-Functionalized Gold-Nanoparticles.

Mouse laser-induced choroidal neovascularization (mouse LCNV) recapitulates the "wet" form of human age-related macular degeneration (AMD). … nd that they are stable at reduced pH. LCNV mice received tail-vein injections of AS-VCAM-1 hAuNPs. Hyperspectral imaging revealed the delivery of AS-VCAM-1 hAuNPs to excised choroidal tissues. Fluorescent images of CNV lesions were obtained, presumably in response to the hybridization of AS-hAuNPs to LCNV-induced VCAM-1 mRNA. This is the first demonstration of systemi …

Spatial and spectral mapping of traffic-related nanoparticles in hippocampal subregions of an Alzheimer disease model.

Chronic exposure to traffic-related air pollution (TRAP) is linked to increased risk of neurodegenerative diseases, including Alzheimer disease (AD). Ultrafine particulate matter (UFPM) is a suspected driver of TRAP neurotoxicity, but its spatial interactions with AD pathology remain poorly defined. We investigated the distribution, composition, and pathological context of TRAP-derived UFPM in the hippocampus of TgF344-AD rats chronically exposed to TRAP or filtered air (FA) for 14 months. Using a multimodal imaging workflow that combines enhanced darkfield hyperspectral imaging (EDF-HSI) with…

Raman imaging for the identification of Teflon microplastics and nanoplastics released from non-stick cookware.

The characterisation of microplastics is still difficult, and the challenge is even greater for nanoplastics. A possible source of these particles is the scratched surface of a non-stick cooking pot that is mainly coated with Teflon. Herein we employ Raman imaging to scan the surfaces of different non-stick pots and collect spectra as spectrum matrices, akin to a hyperspectral imaging process. We adjust and optimise different algorithms and create a new hybrid algorithm to extract the extremely weak signal of Teflon microplastics and particularly nanoplastics. We use multiple characteristic pe…

A new definition about the relationship of intercellular fluid in the brain with the mandibular and parotid lymph nodes.

This study was carried out to reveal the relationship of the brain with both the mandibular lymph node (MLN) and parotid lymph node (PLN) by the hyperspectral fluorescence imaging techniques of Qdot 800 (QD) nanoparticles using in vivo. This relationship of the brain with both lymph nodes offers the preliminary morphological definition of lymphatic drainage. QD was injected into the left parietal brain lobe of each rat at a depth of 2.50 mm. In 65% of the rats that were imaged in vivo, signals were received first from the right MLN and PLN, and then from the left MLN and PLN. In contrast, in t

AI-powered skin spectral imaging enables instant sepsis diagnosis and outcome prediction in critically ill patients.

With sepsis remaining a leading cause of mortality, early identification of patients with sepsis and those at high risk of death is a challenge of high socioeconomic importance. Given the potential of hyperspectral imaging (HSI) to monitor microcirculatory alterations, we propose a deep learning approach to automated sepsis diagnosis and mortality prediction using a single HSI cube acquired within seconds. In a prospective observational study, we collected HSI data from the palms and fingers of more than 480 intensive care unit patients. Neural networks applied to HSI measurements predicted se…

Deep neural network optimization for pigment classification: a case study on representative medieval illumination mock-ups.

Neural networks are increasingly used in conservation science to exploit hyperspectral imaging (HSI) datasets. The literature includes a wide range of models with different architectures and hyperparameters available to the community. Yet, defining the optimal configurations of such parameters for new datasets is not straightforward. This work focuses on state-of-the-art optimization strategies to ensure that the best performance of the models is achieved, offering new opportunities for improving the predictive capacity of the model on unseen data. In particular, a synthetic mixture dataset is…

Phasor Analysis of Multiband Deuterium Mid-Infrared Imaging Reveals Spatial Metabolic Organization in Tissues.

Heavy water (D2O) labeling provides a safe and universal approach for probing biosynthesis in mid-infrared (MIR) metabolic imaging. However, current analysis typically simplifies the C-D spectral region into two peaks associated with lipid and protein synthesis, overlooking the broader spectral complexity arising from multiple deuteration sites and chemical environments. Here, we show that in vivo D2O labeling produces multiband C-D spectral features spanning 2000-2300 cm-1, reflecting contributions from distinct d1, d2, and d3 configurations across lipids, proteins, and other biomolecules. Discrimination of lipid composition and cellular localization in human liver tissues by stimulated Raman scattering microscopy.

SIGNIFICANCE: The molecular mechanisms driving the progression from nonalcoholic fatty liver (NAFL) to fibrosing steatohepatitis (NASH) are insufficiently understood. Techniques enabling the characterization of different lipid species with both chemical and spatial information can provide valuable insights into their contributions to the disease progression. AIM: We extend the utility of stimulated Raman scattering (SRS) microscopy to characterize and quantify lipid species in liver tissue sections from patients with NAFL and NASH. APPROACH: We applied a dual-band hyperspectral SRS microscop

The effect of root-applied glycinebetaine and nutrient cutoff on the sweetness, bitterness, and nutrient content of hydroponically grown lettuce during storage.

Root-applied glycinebetaine (GB) is a novel method to enhance the edible quality of hydroponically grown lettuce. However, the effect of root-applied GB on lettuce postharvest quality is unknown. Here we compare the effect of root-applied GB and nutrient cutoff treatment on postharvest quality of lettuce. In nutrient cutoff treatments, the fertigation solution was replaced with tap water with or without GB. Hyperspectral imaging technique was used to investigate the visual quality. The contents of sugars, sesquiterpene lactones, nitrate, minerals, vitamin C, and GB were analyzed. Plants treate…

Isotope Encoded Spatial Biology Identifies Amyloid Plaque-Age-Dependent Structural Maturation, Synaptic Loss, and Increased Toxicity.

Understanding how amyloid beta (Aβ) plaques form and progress to neurotoxicity in Alzheimer's disease remains a significant challenge. This study aims to elucidate the processes involved in Aβ plaque formation and maturation using a knock-in Aβ mouse model (App NL-F/NL-F ). By employing mass spectrometry imaging and stable isotope labeling, we timestamped Aβ plaques from their initial deposition, enabling the spatial tracking of plaque aging. Correlating single-plaque spatial transcriptomics with time since seeding, allowed us to track gene-expression changes specifically associated with plaqu
