Association between hyperuricemia and metabolic syndrome in patients suffering from bipolar disorder.

BACKGROUND: Clinical studies have shown that bipolar patients have increased serum uric acid levels. High serum uric acid levels could play a role contributing to high prevalence of metabolic syndrome. Metabolic syndrome is known to increase the risk of developing a number of life threatening diseases including coronary heart disease, hypertension, and type 2 diabetes. This study investigated the association between hyperuricemia and metabolic syndrome and its components in individuals suffering from bipolar disorders. METHODS: This study recruited 318 inpatients suffering from bipolar disord

Asymptomatic Hyperuricemia Without Comorbidities Predicts Cardiometabolic Diseases: Five-Year Japanese Cohort Study.

Whether asymptomatic hyperuricemia in the absence of comorbidities increases the risk for cardiometabolic disorders and chronic kidney disease remains controversial. This study was conducted to clarify the association between asymptomatic hyperuricemia and cardiometabolic conditions. Subjects consisting of Japanese adults between 30 and 85 years of age were enrolled in the study at Center for Preventive Medicine, St Luke's International Hospital, Tokyo, and were available at enrollment (2004) and at 5-year follow-up (2009). Subjects were excluded if they were overweight or obese, hypertensive,

Potassium oxonate induces acute hyperuricemia in the tree shrew (tupaia belangeri chinensis).

Potassium oxonate, a selectively competitive uricase inhibitor, produced hyperuricemia (HUA) in rodents in a previous study. In this study, we employed the tree shrew as an animal model to study potassium oxonate-induced HUA. The effect of allopurinol (ALLO), a uric acid reducer, was also examined in this model. Potassium oxonate at doses of 5, 20, 40, 60, 80, 100, and 1,000 mg/kg was given intraperitoneally to tree shrews. The results showed that potassium oxonate can effectively increase the levels of uric acid in tree shrews at doses ranging from 40 to 100 mg/kg. Semiquantitative RT-PCR sho

A two-phase digital tool to predict and prevent post-transplant hyperuricemia.

BACKGROUND: Post-transplant hyperuricemia (PTHU) is a frequent and clinically significant metabolic complication among kidney transplant recipients. Current models lack personalized pre-transplant prevention strategies, and few studies define the optimal tacrolimus level for PTHU prevention. To address this unmet need, we developed and evaluated an integrated digital health tool consisting of a pre-operative risk-prediction algorithm and a post-operative tacrolimus dosing management module. METHOD: We enrolled 195 kidney transplant recipients. Pre-transplantation laboratory assessments and se

Long-term exposure to ambient air pollution and an increased risk of hyperuricemia.

OBJECTIVES: Evidence suggests that ambient air pollution contributes to metabolic dysregulation, including hyperuricemia. However, previous studies were limited to specific groups or regions with relatively high air pollutant concentrations. This study examined the association between long-term exposure to ambient air pollutants and hyperuricemia in a general population exposed to relatively low pollution concentrations. METHOD: This cross-sectional study analyzed 20,117 adults who underwent health screenings in the Republic of Korea (2015-2019). Annual mean concentrations of PM2.5, PM10, NO2

Integrating network pharmacology and metabolomics to elucidate the mechanism of allopurinol in hyperuricemia treatment.

This study integrated network pharmacology and metabolomics to investigate the mechanism of allopurinol in treating hyperuricemia. Therapeutic targets were identified by screening drug and disease databases. A drug-target-disease network was constructed and validated by molecular docking using Cytoscape. A mouse model of hyperuricemia was established with yeast extract and potassium oxonate. Allopurinol efficacy was evaluated via body weight, renal histopathology, and serum biochemical indices. Enrichment analyses of Gene Ontology and Kyoto Encyclopedia of Genes and Genomes pathways were perfo

Effect of Probiotic Product Containing Heyndrickxia coagulans TBC169 on Hyperuricemia in Rats.

Hyperuricemia (HUA) is a metabolic disease characterized by elevated serum uric acid, which is closely related to the gut microbiota. Probiotics have great potential in improving HUA. The purpose of this study was to evaluate the effect and mechanism of probiotic product (SQK) containing Heyndrickxia coagulans TBC169 on HUA rats. Forty SD rats (6 weeks old, 200 ± 20 g) were randomly divided into four groups (Ctrl group, HUA group, SQK1 group, and SQK2 group) of 10 rats each. Rats were given potassium oxonate (100 mg potassium oxonate/100 g BW/day) for 12 weeks to establish HUA model and simult

Integration of Metabolomics and Proteomics Reveals the Molecular Characterization of High-Altitude Hyperuricemia.

Hypobaric hypoxia-induced hyperuricemia (HUA) is a major health challenge for high-altitude populations, yet its molecular mechanisms remain poorly understood. This study employed metabolomic and proteomic profiling of a high-altitude cohort (10 HUA patients and 10 matched controls) to address this gap. HUA patients exhibited significantly elevated uric acid (UA), creatine kinase, and lactate dehydrogenase levels alongside reduced platelet counts. Metabolomic analysis identified 79 significantly altered metabolites, which were associated with amino acids, lipids, and other pathways, including

Diagnostic Significance of SLC2A9 Gene Polymorphisms and Serum Biomarkers in Gout and Hyperuricemia.

BACKGROUND: Uric acid, the final product of purine metabolism, is primarily excreted through the kidneys and intestines. Dysregulation in uric acid production or excretion can result in hyperuricemia and gout. Several proteins play important roles in uric acid metabolism. Among them, GLUT9, a key protein for uric acid excretion, has garnered significant attention, particularly for two SNPs (rs3733591 and rs1014290) on its encoding gene SLC2A9. However, their relationship with gout and hyperuricemia in the Han Chinese population has not been researched. METHODS: This study investigated 498 ind

[Miliary palmoplantar gout tophi in a patient with hyperuricemia and chronic renal insufficiency].

We report on a woman with palmoplantar whitish-yellowish aggregated, partly ulcerated papules on a slightly erythematous base. Histological examination showed pale amorphous nodular deposits in the dermis. On radiological examination, calcifications could be ruled out. Laboratory tests showed marked hyperuricemia and chronic renal insufficiency. We diagnosed miliary gout (MG) and initiated uric acid-reducing therapy with allopurinol 100 mg/day. First described in 2007, MG presenting with multiple whitish-yellowish papules is a rare intradermal variant of classic gout that is characterized by s

Associations of Bisphenols Exposure and Hyperuricemia Based on Human Investigation and Animal Experiments.

Hyperuricemia is characterized by elevated blood uric acid (UA) levels, which can lead to certain diseases. Epidemiological studies have explored the association between environmental contaminant exposure and hyperuricemia. However, few studies have investigated the role of chemical exposure in the development of hyperuricemia. Here, we sought to investigate the effects of bisphenol exposure on the occurrence of hyperuricemia. Fifteen bisphenol chemicals (BPs) were detected in human serum and urine samples collected from an area with a high incidence of hyperuricemia in China. Serum UA levels

Mechanism of anti-hyperuricemia of isobavachin based on network pharmacology and molecular docking.

BACKGROUND: Hyperuricemia is a more popular metabolic disease caused by a disorder of purine metabolism. Our previous study firstly screened out a natural product Isobavachin as anti-hyperuricemia targeted hURAT1 from a Chinese medicine Haitongpi (Cortex Erythrinae). In view of Isobavachin's diverse pharmacological activities, similar to the Tranilast (as another hURAT1 inhibitor), our study focused on its potential targets and molecular mechanisms of Isobavachin anti-hyperuricemia based on network pharmacology and molecular docking. METHODS: First of all, the putative target genes of compoun

Influence of intestinal microecology in the development of gout or hyperuricemia and the potential therapeutic targets.

Gout and hyperuricemia are common metabolic diseases. Patients with purine metabolism disorder and/or decreased uric acid excretion showed increased uric acid levels in the blood. The increase of uric acid in the blood leads to the deposition of urate crystals in tissues, joints, and kidneys, and causes gout. Recent studies have revealed that imbalance of the intestinal microecology is closely related to the occurrence and development of hyperuricemia and gout. Disorder of the intestinal flora often occurs in patients with gout, and high purine and high fructose may induce the disorder of inte
