Determination of N-acetylation phenotyping in a Greek population using caffeine as a metabolic probe.

Studies of isoniazid, the well known antituberculosis drug, have revealed that N-acetylation polymorphism, is of great clinical importance. In humans, N-acetylation is one of the most important pathways in the inactivation of isoniazid. Caffeine, which is also biotransformed by N-acetylation, has been widely used as an in vivo probe for the assessment of N-acetyltransferase polymorphism. The activity of N-acetyltransferase can be estimated from the urinary metabolic ratio of two caffeine metabolites, namely, 5-acetylamino-6-formylamino-3-methyluracil (AFMU), and 1-methylxanthine (1X) after the

In vitro characterization of Bacillus sp. LS100 for detoxifying deoxynivalenol.

The mycotoxin deoxynivalenol (DON) contamination in animal feed has been reported to reduce the growth performance of animals and compromise their immune system, especially pigs. Bacterial detoxification is considered a promising strategy to mitigate its harmful effects. We previously identified a bacterial isolate (Bacillus sp. LS100) from the chicken gut with the ability to detoxify DON. To explore its potential application in swine production, the current study was focused on in vitro characterization of Bacillus sp. LS100 with regard to its capacity to detoxify DON. The bacterium was stabl

Evaluation of Detoxification-Related Gene Expression, Oxidative Stress Biomarkers, and Blood Biochemical Parameters in Common Carp (Cyprinus carpio) Co-Exposed to Polyethylene Microplastics and Deltamethrin.

Microplastics (MPs) are emerging contaminants in aquatic ecosystems and are able to modify the bioavailability and toxicity of co-occurring chemicals. In the present study, it was investigated whether high-density polyethylene (HDPE)-MPs exacerbate the toxicological effects of the pyrethroid insecticide deltamethrin in juvenile common carp (Cyprinus carpio). Fish (n = 270; 25 ± 5 g) were randomly allocated to a 2 × 3 factorial design (three replicates per group; 15 fish per tank) and exposed for 30 days to sub-lethal deltamethrin (12.5 μg/L; 10% of 96-h LC₅₀) and/or HDPE-MPs (0, 350, or 700 μg

[Molecular dynamics simulation and experimental verification reveal processing detoxification mechanism of Pinelliae Rhizoma Praeparatum].

This study aims to investigate the effects of glycyrrhizic acid and calcium ions from adjuvant solutions on the toxic lectin in Pinelliae Rhizoma Praeparatum and clarify the processing detoxification mechanism. Molecular docking and molecular dynamics simulations were employed to construct a 3D model of lectin and evaluate the interactions of glycyrrhizic acid and/or calcium ions with lectin. Wild-type and mutant lectin proteins were produced via a prokaryotic expression system in Escherichia coli. The interactions of glycyrrhizic acid and/or calcium ions with the lectin proteins were verified

Hemozoin: a waste product after heme detoxification?

BACKGROUND: Hemozoin is considered a waste byproduct of heme detoxification following hemoglobin digestion; consequently, the biological functions of hemozoin in hemozoin-producing organisms have often been overlooked. However, recent findings indicate that Schistosoma hemozoin facilitates the transfer of iron from erythrocytes to eggs through its formation and degradation, thereby increasing interest in the role of malarial hemozoin. METHODS: Using transmission electron microscopy, we compared the formation of Schistosoma hemozoin and malaria hemozoin. Through transcriptome analysis of diffe

Prevalence of insecticide resistance and enhanced detoxifying enzymes in field populations of Tuta absoluta (Lepidoptera: Gelechiidae) in the major Tomato growing regions of South India.

South American pinworm Tuta absoluta (Meyrick) is an invasive and major tomato pest that attained resistance or reduced susceptibility to most insecticides used in their management due to repeated and intensive applications. This study aimed to find the levels of resistance in T. absoluta populations collected from South Indian states against novel insecticides such as chlorantraniliprole 18.5% SC, flubendiamide 39.35% SC, spinosad 45% SC, indoxacarb 14.5% SC, and emamectin benzoate 5% SG using leaf dip bioassay. Among the eight populations collected from major tomato growing belts spanning th

Organ-specific mercury stable isotopes, speciation and particle measurements reveal methylmercury detoxification processes in Atlantic Bluefin Tuna.

Identifying metabolism and detoxification mechanisms of Hg in biota has important implications for biomonitoring, ecotoxicology, and food safety. Compared to marine mammals and waterbirds, detoxification of MeHg in fish is understudied. Here, we investigated Hg detoxification in Atlantic bluefin tuna Thunnus thynnus using organ-specific Hg and Se speciation data, stable Hg isotope signatures, and Hg and Se particle measurements in multiple tissues. Our results provide evidence for in vivo demethylation and biomineralization of HgSe particles, particularly in spleen and kidney. We observed a ma

Biological removal of pharmaceuticals by Navicula sp. and biotransformation of bezafibrate.

Pharmaceutically active compounds are of great concern due to their detection frequency in the environment and the unexpected risks. In this study, the simultaneous removal of mixed pharmaceuticals by microalgae was explored using a typical freshwater diatom Navicula sp. Results showed that Navicula sp. could efficiently remove atenolol, carbamazepine, ibuprofen and naproxen with the efficiencies of >90% after 21 d of exposure. As compared to the removal efficiencies of each pharmaceutical in the individual pharmaceutical treatments, the degradation of sulfamethoxazole, bezafibrate, and naprox

Bacteria-mediated modification of insecticide toxicity in the yellow fever mosquito, Aedes aegypti.

The incidence of mosquito-borne disease poses a significant threat to human and animal health throughout the world, with effective chemical control interventions limited by widespread insecticide resistance. Recent evidence suggests that gut bacteria of mosquitoes, known to be essential in nutritional homeostasis and pathogen defense, may also play a significant role in facilitating insecticide resistance. This study investigated the extent to which bacteria contribute to the general esterase and cytochrome P450 monooxygenase (P450)-mediated detoxification of the insecticides propoxur and nale

Acetylator phenotyping in patients with malignant lymphomas, using caffeine as the metabolic probe.

OBJECTIVE: The study was designed to answer the question whether a particular (slow/fast) acetylator phenotype is a risk for malignant lymphoma patients. Differences in acetylator phenotypes were previously described in urinary bladder and colorectal carcinoma patients compared to controls. Addressing this issue may help understanding the possible role of xenobiotics' metabolism in the pathogenesis of malignant lymphomas. Caffeine is currently preferred as a metabolic probe due to its noninvasiveness. DESIGN: Case-control study. SETTING: Hematology/oncology inpatient unit and outpatient clin

Approaches to cancer prevention based on an understanding of N-nitrosamine carcinogenesis.

Understanding carcinogenesis is critical for development of rational approaches to cancer prevention. This paper uses N-nitrosamine carcinogenesis as an example. N-Nitrosamines are a large group of potent carcinogens. Approximately 300 different N-nitrosamines are carcinogenic. At least 30 animal species are responsive to their effects. There is little doubt that humans exposed to sufficient amounts of N-nitrosamines would also be susceptible to their carcinogenic effects. Human exposure to preformed N-nitrosamines occurs through the diet, in certain occupational settings, and through the use

[Acute toxicity of trichloroethylene. Description of a case series at the Autonomous Service of Toxicology of Florence during the 1977-1988 period].

The authors report the number of acute trichloroethylene intoxications admitted to the Toxicological Unit of Florence University from January 1977 to December 1988. The identification of the solvent metabolic pathway allowed to clarify the pathogenesis of hepatorenal dysfunction observed during acute intoxications. Together with gastrointestinal decontamination and cardiac arrhythmia control we have studied the effect of drugs supposed to act as blockers of trichloroethylene metabolism or as inactivators of the hepatotoxic free radical metabolite 2-2(1)-3 trichloroxirane. The prognostic modifi

The Antibiotic Fosfomycin Mimics the Effects of the Intermediate Metabolites Phosphoenolpyruvate and Glyceraldehyde-3-Phosphate on the Stenotrophomonas maltophilia Transcriptome.

Stenotrophomonas maltophilia is an opportunistic pathogen with an environmental origin, which presents a characteristically low susceptibility to antibiotics and is capable of acquiring increased levels of resistance to antimicrobials. Among these, fosfomycin resistance seems particularly intriguing; resistance to this antibiotic is generally due to the activity of fosfomycin-inactivating enzymes, or to defects in the expression or the activity of fosfomycin transporters. In contrast, we previously described that the cause of fosfomycin resistance in S. maltophilia was the inactivation of enzy

Is maternal opioid use hazardous to breast-fed infants?

Over the last few decades, the rate of breastfeeding has increased steadily in the developed countries of the world. During this time, opioid use in the general population has steadily increased as well. Despite this, clinicians remain unclear whether opioid use is safe during breastfeeding. While the vast majority of medications used during breastfeeding occur without incident, case reports and studies have reported possible opioid toxicity in breast-fed infants. Multiple enzymes are involved in the metabolism of opioids. CYP2D6 catabolizes O-demethylation of codeine, tramadol, oxycodone, and

Proteomic analysis of chicken bone marrow-derived dendritic cells in response to an inactivated IBV + NDV poultry vaccine.

Inactivated poultry vaccines are subject to routine potency testing for batch release, requiring large numbers of animals. The replacement of in vivo tests for cell-based alternatives can be facilitated by the identification of biomarkers for vaccine-induced immune responses. In this study, chicken bone marrow-derived dendritic cells were stimulated with an inactivated vaccine for infectious bronchitis virus and Newcastle disease virus, as well as inactivated infectious bronchitis virus only, and lipopolysaccharides as positive control, or left unstimulated for comparison with the stimulated s
