Lentiviral-mediated GATA-3 RNAi decreases allergic airway inflammation and hyperresponsiveness.

GATA-3 is the key transcriptional factor for Th2 commitment in T cells and is strongly associated with asthma and allergic disease. We studied the silencing of the GATA-3 gene expression using RNA interference (RNAi) delivered by a lentiviral vector, to evaluate the therapeutic role of GATA-3 short hairpin RNAs (shRNAs) in a murine model of asthma. Mice were sensitized with OVA and instilled intratracheally (IT) with GATA-3 shRNAs lentiviral vector (Lenti-si-GATA-3) once, 48 hours before challenge. After three challenges with the OVA antigen, the mice were assessed for airway hyperresponsivene

Recruitment of dendritic cells and macrophages during T cell-mediated synovial inflammation.

Adoptive transfer of adjuvant-induced arthritis was used in this study to examine local macrophages and dendritic cells (DCs) during T cell-mediated synovial inflammation. We studied the influx of CD11b+CD11c+ putative myeloid DCs and other non-lymphoid CD45+ cells into synovium-rich tissues (SRTs) of the affected hind paws in response to a pulse of autoreactive thoracic duct cells. Cells were prepared from the SRTs using a collagenase perfusion-digestion technique, thus allowing enumeration and phenotypic analysis by flow cytometry. Numbers of CD45+ cells increased during the first 6 days, wi

RvE1 protects from local inflammation and osteoclast- mediated bone destruction in periodontitis.

Periodontitis is a well-appreciated example of leukocyte-mediated bone loss and inflammation that has pathogenic features similar to those observed in other inflammatory diseases such as arthritis. Resolvins are a new family of bioactive products of omega-3 fatty acid transformation circuits initiated by aspirin treatment that counter proinflammatory signals. Because it is now increasingly apparent that local inflammation plays a critical role in many diseases, including cardiovascular disease, atherosclerosis, and asthma, experiments were undertaken to evaluate the actions of the newly descri

Interleukin-7-mediated inflammation in unstable angina: possible role of chemokines and platelets.

BACKGROUND: Atherogenesis and plaque destabilization involve immune-mediated mechanisms, but the actual mediators have not been fully clarified. Interleukin (IL)-7 is a regulator of T-cell homeostasis but also may be involved in inflammation. We hypothesized that IL-7 could be involved in the inflammatory processes observed in atherosclerosis and acute coronary syndromes. METHODS AND RESULTS: To study the role of IL-7 in coronary artery disease, we analyzed IL-7 levels and the effect of this cytokine on inflammatory mediators in patients with stable and unstable angina and in healthy control

Hepcidin, a putative mediator of anemia of inflammation, is a type II acute-phase protein.

Hepcidin is a liver-made peptide proposed to be a central regulator of intestinal iron absorption and iron recycling by macrophages. In animal models, hepcidin is induced by inflammation and iron loading, but its regulation in humans has not been studied. We report that urinary excretion of hepcidin was greatly increased in patients with iron overload, infections, or inflammatory diseases. Hepcidin excretion correlated well with serum ferritin levels, which are regulated by similar pathologic stimuli. In vitro iron loading of primary human hepatocytes, however, unexpectedly down-regulated hepc

miR-369-3p Modulates LRRK2-Mediated Inflammation and Autophagy in RAW264.7 Macrophages.

Leucine-rich-repeat kinase 2 (LRRK2) is a multidomain protein highly expressed in immune cells and implicated in the regulation of immune functions including immune signaling, cytokine release and autophagy. LRRK2 is one of the therapeutic targets in Parkinson's Disease (PD). Aberrant activation of LRRK2 can also contribute to intestinal inflammation, mainly in inflammatory bowel disease (IBD). Hence the modulation of LRRK2 may influence gut inflammation providing an improvement in disease outcomes. Over the years, microRNAs (miRNAs) have acquired much attention thanks to their potential as th

Astrocytes in the paraventricular nucleus mediates inflammation-related depression in mice treated with lipopolysaccharide.

BACKGROUND: Inflammation has been implicated in the pathogenesis of depression, yet the role of astrocytes in modulating these processes remains poorly understood. This study aimed to explore the involvement of paraventricular nucleus (PVN) astrocytes in inflammation-related depression, focusing on their impact on depression-like behaviors, neuronal activity, and immune responses. METHODS: We used the lipopolysaccharide (LPS)-induced depression model to evaluate depression-like behaviors in mice through sucrose preference, open field, and forced swim tests. Calcium dynamics in PVN astrocytes

Peripheral inflammation mediates the relationship between early life stress and alcohol use.

RATIONALE: Early life stress (ELS) is associated with an increased risk for alcohol consumption and development of alcohol use disorder (AUD). Inflammation may be a sex-dependent link between ELS and alcohol use; however, there is limited evidence to support such an association in individuals with AUD. METHODS: This secondary analysis utilized cross-sectional mediation to investigate the role of peripheral inflammation in the relationship between ELS and alcohol consumption (i.e., drinks per drinking day (DPDD)) in treatment-seeking individuals with AUD (n = 99; 60 M/39 F). A cross-sectional

Immune Cell-Mediated Inflammation in Heart Failure: Subset Heterogeneity and Targeted Therapy.

Heart failure (HF) is the end-stage manifestation of various cardiovascular diseases, affecting over 64 million patients globally. Current standard therapies have substantially improved the prognosis of patients with HF with reduced ejection fraction (HFrEF), but show limited efficacy in HF with preserved ejection fraction (HFpEF), and the overall cure rate remains unsatisfactory. Persistent low-grade sterile inflammation mediated by immune cells is a core pathological driver of HF onset and progression. The high complexity of the cardiac immune microenvironment, along with the phenotypic and

Central neurons encode interleukin-1β signals and mediate stress-induced inflammation.

The brain encodes and stores information about peripheral inflammation and can directly recapitulate prior inflammatory responses. However, whether individual cytokines activate specific neural circuits to produce distinct physiological responses remains unknown. To address this fundamental question, we mapped brain-wide responses to IL-1β and found prominent engagement of the bed nucleus of the stria terminalis (BNST). Using targeted recombination in active populations, snRNA sequencing, and circuit tracing, we discovered that corticotropin-releasing hormone-expressing BNST neurons encode IL-

Adiposity and inflammation mediate altered metabolic profiles in individuals with opioid use disorder.

Previous studies have linked opioid use to altered metabolic profiles, but findings have been inconsistent and mechanisms remain unclear. One potential mechanism involves increased adiposity, leading to chronic low-grade inflammation that elevates metabolic risk. Here, we examined metabolic profiles in individuals with opioid use disorder (OUD) and matched non-OUD controls, focusing on the sequential mediating roles of BMI and inflammation. Data from individuals with OUD (n=281) and non-OUD (n=246) were drawn from a natural history screening protocol from the National Institute on Alcohol Abus

Curcumin analogue C66 attenuates hypertensive nephropathy by inhibiting JNK-mediated inflammation.

Hypertensive nephropathy (HTN) is one of the common, highly disabling, and lethal complication of hypertension and is characterized by increased fibrosis, oxidative stress, and chronic inflammation. Notably, chronic inflammation is throughout the pathogenesis of HTN, even renin-angiotensin-aldosterone system (RAAS) was blocked. Our previous study demonstrated that C66, an anti-inflammatory curcumin derivative compound, prevented various metabolic diseases via inhibiting c-Jun N-terminal kinase (JNK), a key inflammatory target. In the present study, we speculate that C66 could prevent HTN by su

TSPO Regulates TLR4-Mediated Inflammation Through Calcium Homeostasis and Immunometabolic Adaptation.

Bacterial infection triggered excessive inflammatory responses, yet the mechanisms linking inflammatory activation to immunometabolic adaptation remained incompletely understood. The mitochondrial translocator protein (TSPO) has been implicated in inflammatory activation and cellular metabolism. This study aimed to investigate the role of TSPO in inflammation mediated by Toll-like receptor 4 (TLR4). Herein, we integrated transcriptomic data from the human peripheral blood dataset GSE72829, and single-cell transcriptomic profiles from the CELLxGENE platform with cellular mechanistic experiments

Regulation of keratinocyte proliferation and epidermal inflammation by meprin α-mediated cleavage of dermokine.

Dysregulations within the epidermal proteolytic network can cause hyperproliferative and inflammatory disorders. Although the metalloprotease meprin α is localized in the stratum basale in healthy skin, increased levels are found in the upper epidermal layers in wound healing and psoriatic lesions. To investigate a link between meprin α expression and keratinocyte proliferation, we developed a mouse model for inducible expression of pathological meprin α levels (ie, K5Mα mice). K5Mα mice developed a skin phenotype characterized by hyperkeratosis, acanthosis, parakeratosis, and barrier defect.

Emerging Roles of Regulated Cell Death-mediated Inflammation in Pathophysiology of Ocular Diseases.

Regulated cell death pathways are vital for proper developmental and homeostatic processes. Dysregulation of these pathways contributes to the pathogenesis of many diseases, including ocular inflammatory and neurodegenerative diseases such as glaucoma, diabetic retinopathy, age-related macular degeneration, retinitis pigmentosa, and ocular surface diseases. Our knowledge of the regulated cell death pathways, including apoptosis, necroptosis, pyroptosis, and ferroptosis, has been extensively expanded in the recent years. The targeting of these pathways as a potential therapy for various ocular

Targeted Modulation of Lipid Mediator-Biosynthetic Enzymes as Strategy for Inflammation Resolution Pharmacology.

Lipid mediators (LMs) comprise a large variety of signaling molecules that are enzymatically produced from polyunsaturated fatty acids (PUFA) in a wide biosynthetic network. These pivotal mediators regulate all stages of inflammation, from initiation, progression, and maintenance to resolution and regain of tissue homeostasis. While leukotrienes and prostaglandins, produced from arachidonic acid (AA) by the 5-lipoxygenase (LOX) and cyclooxygenase (COX) pathways, respectively, mediate/promote inflammatory reactions and related symptoms, the specialized pro-resolving mediators (SPMs) terminate a
