Functional Characterization of Trypsin Genes in Spodoptera litura and Their RNA Silencing in Combination with Insecticides.

Trypsin is one of the major digestive proteinases secreted by the insect midgut. It plays important roles in digestion, molting, diapause, zymogen activation, innate immunity, reproduction and insecticide resistance. However, the functions of trypsin and its potential as a target for the control of Spodoptera litura, a polyphagous agricultural pest, remain poorly understood. In this study, seven trypsin genes related to S. litura development were characterized. Temporal-spatial expression results showed that the seven trypsin genes were predominantly expressed during the larval stage and mainl

Unveiling the hidden threat: flupyradifurone soluble liquid's toxic impact on silkworms.

Bombyx mori Linnaeus (Lepidoptera: Bombycidae) is an economically significant insect that consistently contributes to the enhancement of the national economy in all countries with predominant sericulture industries. Flupyradifurone, a butenolide insecticide, is recognized for its minimal toxic effects on bees when applied at standard dosages; however, its potential toxicity or threat to B. mori has not been fully elucidated. This study aims to investigate and evaluate the toxicity of flupyradifurone soluble liquid (SL) on B. mori by administering various concentrations to third instar silkworm

An ovarian insulin-like peptide specifically regulates energy allocation and oocyte development in nutrition-restricted cockroaches.

BACKGROUND: Insects exhibit remarkable resilience and maintain high levels of reproduction despite frequently encountering nutritional restriction. However, the mechanisms governing their adaptive reproductive strategies under nutrition-restricted conditions remain poorly understood. The German cockroach Blattella germanica, a widespread urban pest, exhibits remarkable reproductive capabilities even in domestic environments where food resources are frequently limited. RESULTS: In this study, we demonstrate that the ovary plays a crucial role in promoting vitellogenin biosynthesis and oocyte d

Ovary Activation Dynamics in the Bean Weevil Zabrotes subfasciatus (Bruchinae): The Essential Roles of Seeds and Males.

Phytophagous beetles, particularly those within the superfamilies Chrysomeloidea and Curculionoidea, constitute one of the most diverse and ecologically significant groups of insect herbivores. Within this group, the subfamily Bruchinae is especially notable for its close association with leguminous plant seeds. As most Bruchinae species do not feed during the adult stage, the timing and regulation of vitellogenesis remain unclear. Previous studies suggest that vitellogenesis may be triggered by volatile organic compounds emitted by host seeds, which promote juvenile hormone (JH) synthesis. Th

Ellagic Acid Alleviates Imidacloprid-Induced Thyroid Dysfunction via PI3K/Akt/mTOR-Mediated Autophagy.

Imidacloprid (IMI) is a widely used insecticide known for its high selectivity toward insects. Ellagic acid (EA) is a plant-derived polyphenolic compound recognized for its therapeutic potential and favorable safety profile in the treatment of various diseases. This study aimed to evaluate the therapeutic effects of EA, formulated as novasomes (NOV), against IMI-induced thyroid dysfunction and to investigate the underlying mechanisms. Rats were divided into four equal groups: control, EA-NOV, IMI, and IMI + EA-NOV. Thyroid function was assessed by measuring free triiodothyronine (T3), free thy

Farnesol dehydrogenase from Metisa plana (Lepidoptera: Psychidae): Biochemical characterization, in silico analysis and inhibitors discovery.

Metisa plana (Lepidoptera: Psychidae), a foliar-feeding bagworm, is a major pest in oil palm plantations, particularly in Peninsular Malaysia. Government of Malaysia has classified it as a dangerous pest under the Plant Quarantine Act 1976. Effective and sustainable solutions remain elusive despite various management strategies, including chemical, cultural and biological methods. This study explored an alternative approach by targeting the juvenile hormone biosynthetic pathway, crucial for insect growth, metamorphosis, and reproduction, as part of the Integrated Pest Management (IPM) strategy

Juvenile Hormone Involved in the Defensive Behaviors of Soldiers in Termite Reticulitermes aculabialis.

Eusocial insects have evolved specific defensive strategies to protect their colonies. In termite colonies, soldiers perform a colony-level defense by displaying mechanical biting, head-banging and mandible opening-closing behaviors. However, few studies have been reported on the factors modulating defensive behaviors in termites. Owing to JH (juvenile hormone) being involved in soldier differentiation, JH was speculated to affect defensive behaviors in termite soldiers. To determine the effect of JH on the defensive behaviors of termite soldiers, we performed a JHA-feeding and RaSsp1-silencin

A Drosophila model to study retinitis pigmentosa pathology associated with mutations in the core splicing factor Prp8.

Retinitis pigmentosa (RP) represents genetically heterogeneous and clinically variable disease characterized by progressive degeneration of photoreceptors resulting in a gradual loss of vision. The autosomal dominant RP type 13 (RP13) has been linked to the malfunction of PRPF8, an essential component of the spliceosome. Over 20 different RP-associated PRPF8 mutations have been identified in human patients. However, the cellular and molecular consequences of their expression in vivo in specific tissue contexts remain largely unknown. Here, we establish a Drosophila melanogaster model for RP13

[Amphibian skin as a source of therapeutic peptides].

The search for new bioactive molecules that could be used in therapeutics is a major public health issue, particularly in the treatment of certain diseases such as cancer. In this context the exploration of the venom of animals (snakes, amphibians, cones, scorpions, insects...) that produce molecules of various structures and biological activities, is a very promising direction. Research in this area led to the discovery of neuropeptides, hormones, toxins, antimicrobial peptides and other extremely potent mediators. These are now used in many areas both in fundamental research and in translati

Transcriptome profile analysis of cell proliferation molecular processes during multicellular trichome formation induced by tomato Wov gene in tobacco.

BACKGROUND: Trichomes, developing from the epidermis of nearly all terrestrial plants, provide good structural resistance against insect herbivores and an excellent model for studying the molecular mechanisms underlying cell fate determination. Regulation of trichomes in Rosids has been well characterized. However, little is known about the cell proliferation molecular processes during multicellular trichome formation in Asterids. RESULTS: In this study, we identified two point mutations in a novel allele (Wov) at Wo locus. Ectopic expression of Wov in tobacco and potato induces much more tri

Physiology of invertebrate oxytocin and vasopressin neuropeptides.

Neuropeptides and regulatory peptide hormones control many developmental, physiological and behavioural processes in animals, including humans. The nonapeptides oxytocin and arginine vasopressin are produced and released by the pituitary gland and have actions on many organs and tissues. Receptive cells possess particular receptors to which the peptides bind as ligands, leading to activation of G-protein-coupled receptors, hence cellular responses. In humans and other mammalian species, oxytocin and vasopressin mediate a range of peripheral and central physiological functions that are importan

Full sequence and characterization of two insect defensins: immune peptides from the mosquito Aedes aegypti.

We report the complete amino acid sequence and biological activity of two immune peptides, from the yellow fever mosquito Aedes aegypti, that are induced in response to infection. Both peptides display biological activity against the Gram positive microbe Micrococcus luteus and substantial sequence homology to insect defensins, small heat-stable, antibiotic peptides previously described from several non-vector insects. These mosquito peptides, designated Ae. aegypti defensins A and B, are isoforms. Defensin B is the most abundant antibacterial peptide in this species whereas defensin A is much

Dosage compensation in Drosophila: the X-chromosomal binding of MSL-1 and MLE is dependent on Sxl activity.

In Drosophila, dosage compensation, i.e. the equalization of levels of X-linked gene products in the two sexes, is achieved by the hypertranscription of most X-linked genes in males relative to females. The products of at least four genes, collectively termed male-specific lethal (msl) genes, are required for this process and, at least in the case of three of them, mediate this function through an association with the X chromosome in males. We have studied some of the parameters that affect the association of the msl-1 gene product and found that its presence is dependent on the wild-type func

A corazonin G protein-coupled receptor gene in the tick Ixodes scapularis yields two splice variants, each coding for a specific corazonin receptor.

We have identified a corazonin G protein-coupled receptor (GPCR) gene in the tick Ixodes scapularis, which likely plays a central role in the physiology and behavior of this ectoparasite. This receptor gene is unusually large (1.133 Mb) and yields two corazonin (CRZ) receptor splice variants, where nearly half of the coding regions are exchanged: CRZ-Ra (containing exon 2, exon 3, and exon 4 of the gene) and CRZ-Rb (containing exon 1, exon 3, and exon 4 of the gene). CRZ-Ra codes for a GPCR with a canonical DRF sequence at the border of the third transmembrane helix and the second intracellula

Unveiling tomato defense mechanisms against Bradysia impatiens (Diptera: Sciaridae) feeding: insights from transcriptomic and metabolomic analyses.

BACKGROUND: Tomato (Solanum lycopersicum) is an important agricultural crop frequently threatened by pests such as Bradysia impatiens (B. impatiens), a globally distributed sciarid fly whose larvae inflict severe damage on roots, stems, and leaves, leading to substantial yield losses. Despite its economic importance, the molecular and metabolic mechanisms underlying tomato defense against B. impatiens remain poorly understood. RESULTS: In this study, integrated transcriptomic and metabolomic approaches were employed to investigate the responses of tomato leaves to B. impatiens larval infestat

Spatial and temporal coordination of Duox/TrpA1/Dh31 and IMD pathways is required for the efficient elimination of pathogenic bacteria in the intestine of Drosophila larvae.

Multiple gut antimicrobial mechanisms are coordinated in space and time to efficiently fight foodborne pathogens. In Drosophila melanogaster, production of reactive oxygen species (ROS) and antimicrobial peptides (AMPs) together with intestinal cell renewal play a key role in eliminating gut microbes. A complementary mechanism would be to isolate and treat pathogenic bacteria while allowing colonization by commensals. Using real-time imaging to follow the fate of ingested bacteria, we demonstrate that while commensal Lactiplantibacillus plantarum freely circulate within the intestinal lumen, p

Identification of juvenile hormone-induced posttranslational modifications of methoprene tolerant and Krüppel homolog 1 in the yellow fever mosquito, Aedes aegypti.

Recent studies reported that JH-regulated phosphorylation status of the JH-receptor complex contributes to its transcription activity in Aedes aegypti. However, phosphorylation sites of these proteins have not yet been identified. In this study, we found that the fusion of an EGFP tag to Ae. aegypti Kr-h1 (AaKr-h1) and Met (AaMet) improved their stability in mosquito Aag-2 cells, which allowed their purification. The liquid chromatography and tandem mass spectrometry analysis of the purified AaKr-h1 showed that the phosphoserine residue at position 694, located in the evolutionarily conserved

A case of anaphylaxis: horse-fly or hymenoptera sting?

In literature it has been described a high risk of systemic reaction after blood-sucking Dyptera bites, like mosquitoes and horsefly, in people sensitive to hymenoptera. A 51 year old man, allergic to hymenoptera venom and with a history of i.v. reaction after Mueller, who has been treated with Vespula sp. ITS for the last 3 years, was stung by a yellow, black and green insect on the neck. Five minutes after the bite, he suffered generalized hitching and urticaria, oral cavity and lower limbs paresthesia, followed by lost of consciousness. At the Emergency Room he was successfully treated with

Hormonal activation of phosphorylase in cockroach fat body trophocytes: A correlation with trans-membrane calcium flux.

This study is an investigation of the temporal relationship between transmembrane Ca(2+) fluxes, and glycogen phosphorylase activation in dispersed trophocytes from the fat body of the cockroach, Periplaneta americana. Phosphorylase is maximally activated within 5 min after treating the trophocytes with either of the hypertrehalosemic hormones, Pea-HTH-I and Pea-HTH-II. Activation caused by Pea-HTH-II is sustained for a longer period than that produced by Pea-HTH-I. Chelation of extracellular Ca(2+) with EGTA blocks the activation of phosphorylase by HTH. Similarly, chelation of intracellular
