Genome-Wide Identification of Novel miRNAs and Infection-Related Proteins in Leishmania major via Comparative Analysis of the Protozoa, Vectors, and Mammalian Hosts.

Leishmania major is a unicellular protozoan that causes cutaneous leishmaniasis in mammals and is mainly transmitted by the sand fly Phlebotomus papatasi. However, the contribution of microRNAs (miRNAs) and protein-coding genes to its pathogenic mechanisms remains largely unexplored. In this study, we systematically analyzed miRNAs and protein-coding genes in L. major, its insect vector, and mammalian hosts. Comparative genomic analysis revealed 2963 conserved proteins shared among the three groups, highlighting a core set of proteins across protozoa, vectors, and hosts. Among mammals, human p

Comparative proteome analysis reveals that blood and sugar meals induce differential protein expression in Aedes aegypti female heads.

Aedes aegypti females ingest sugar or blood to obtain the nutrients needed to maintain cellular homeostasis. During human blood ingestion, female mosquitoes may transmit different viruses such as dengue, yellow fever and, more recently, zika and chikungunya. Here, we report changes in protein expression in the heads of adult female Ae. aegypti mosquitoes in response to the ingestion of blood or sugar. Proteins extracted from the heads of Ae. aegypti fed exclusively on blood (BF) or sugar (SF) were trypsin hydrolyzed (off-gel) and analyzed by the reverse-phase nano-liquid chromatography coupled

Expression of the recombinant plasminogen activator (reteplase) by a non-lytic insect cell expression system.

Reteplase is a potent thrombolytic agent which is widely used in the management of acute myocardial infarction and stroke. It belongs to the third generation of the thrombolytic drugs and has been derived from native human tissue plasminogen activator by removing three domains of it and keeping the Kringle 2 and Serine protease domains. However, the high cost of this drug, has limited the application of this drug especially in the developing and third world countries. The most laborious steps in the bacterial production of this drug is its purification and refolding steps which keep the proces

Artificial Diets With Different Protein Levels for Rearing Spodoptera frugiperda (Lepidoptera: Noctuidae).

Spodoptera frugiperda (J.E. Smith, 1797) (Lepidoptera: Noctuidae) is a pest of great economic importance which can feed on more than 300 plant species. As it is polyphagous, its host plants may have variable physical and chemical constitutions. This may influence larval development, as protein and carbohydrate levels are important factors for adequate biological development. The aim of this study was to evaluate insect developmental parameters as well as to compare the food consumption of S. frugiperda larvae reared using diets with different protein levels under laboratory conditions. Three a

A horsefly saliva antigen 5-like protein containing RTS motif is an angiogenesis inhibitor.

The Ag5 proteins are the most abundant and immunogenic proteins in the venom secretory ducts of stinging insects. An antigen 5-like protein (named tabRTS) composed of 221 amino acid residues was purified and characterized from the salivary glands of the horsefly, Tabanus yao (Diptera, Tabanidae). Its cDNA was cloned from the cDNA library of the horsefly's salivary gland. TabRTS containing the SCP domain (Sc7 family of extracellular protein domain) was found in insect antigen 5 proteins. More interestingly, there is an Arg-Thr-Ser (RTS) disintegrin motif at the C-terminus of tabRTS. The RTS mot

Novel replication-competent reporter-expressing Rift Valley fever viruses for molecular studies.

UNLABELLED: Rift Valley fever virus (RVFV) is a mosquito-borne zoonotic disease that causes severe disease in both domestic and wild ungulates and humans, making it a significant threat to livestock and public health. The RVFV genome consists of three single-stranded, negative-sense RNA segments differing in size: small (S), medium (M), and large (L). Segment S encodes the virus nucleoprotein N and the virulence-associated factor non-structural (NSs) protein in opposite orientations, separated by an intergenic region (IGR). To overcome the current need to use secondary techniques to detect the

Lineage-specific innovations expand mitochondrial proteomes in kinetoplastids.

Trypanosoma brucei and Leishmania tarentolae are model kinetoplastids whose mitochondria contain a unique DNA network, kinetoplast DNA (kDNA), and exhibit specialized features, including developmental remodeling and RNA editing. By integrating mass spectrometry-based protein correlation profiling with published datasets, we assembled inventories of 1,666 mitochondrial proteins in T. brucei and 1,594 in L. tarentolae. These include 124 polypeptides not previously recognized as mitochondrial. Despite their phylogenetic distance, trypanosomes and leishmanias maintain highly conserved mitoproteome

Immunity to St. Louis encephalitis virus by sequential immunization with recombinant vaccinia and baculovirus derived PrM/E proteins.

St. Louis encephalitis (SLE) is an important mosquito-borne disease of great public health concern in parts of the United States. South America and Canada. Protective immunogens of flaviviruses produced in different expression systems have been shown to be effective against virulent virus infection in laboratory animal models. Here we show that the pre-membrane and envelope (PrM/E) of SLE virus expressed in insect and mammalian cell systems using baculovirus and vaccinia virus, respectively, are processed correctly and showed similar antigenic characteristics as the authentic proteins. Neurotoxins active on insects: amino acid sequences, chemical modifications, and secondary structure estimation by circular dichroism of toxins from the scorpion Androctonus australis Hector.

Two scorpion neurotoxins active only on insects, the insect toxins AaH IT1 and AaH IT2, were purified from the venom of scorpions Androctonus australis Hector collected in Tozeur (Tunisia) and characterized. AaH IT2 was sequenced and found to differ in four amino acid positions from AaH IT, the single previously sequenced insect toxin [Darbon, H., Zlotkin, E., Kopeyan, C., Van Rietschoten, J., & Rochat, H. (1982) Int. J. Pept. Protein Res. 20, 320-330] which possessed an equal potential for paralyzing fly larvae. The basic amino acid residues of AaH IT1, which differs from AaH IT by one amino

Sublethal doses of fipronil intensify synapsin immunostaining in Atta sexdens rubropilosa (Hymenoptera: Formicidae) brains.

BACKGROUND: Although ants are common insects in agricultural ecosystems, few studies have considered how xenobiotics might induce physiological and morphological alterations in these insects. This study aimed to verify the neurotoxic action of sublethal doses of fipronil on the mushroom bodies of brains from the leaf-cutting ant Atta sexdens rubropilosa through immunocytochemistry analysis for the protein synapsin. RESULTS: The LD50 value was established as 1.42 ng ant(-1), and the sublethal doses used were LD50/10 and LD50/100. Synapsin labelling was more evident in the brains extracted from

[Antimicrobial peptides/proteins--application to the therapy of sepsis].

Many antimicrobial peptides and proteins were discovered recently in various animals. Cecropins are insect-derived antimicrobial peptides which contain 35-39 amino acid residues. Magainins are amphibian-derived antimicrobial peptides with 21-27 amino acid residues. In mammals, defensins, 29-35 amino acid peptides, were identified in the granules of neutrophils and various epithelial cells. In addition, the granules of neutrophils in the mammal have been shown to have several antimicrobial proteins. Among them, bactericidal/permeability increasing protein (BPI) and cationic antimicrobial peptid

Cloning and characterization of a ribosomal P protein from Taenia solium, the aetiological agent of human cysticercosis.

A cDNA encoding the complete open reading frame of the Taenia solium acidic ribosomal phosphoprotein P2 has been cloned. The 417 bp cDNA (arpP2) was isolated from a T. solium cDNA expression library by PCR using P protein specific pimers. The ORF encodes a protein of 121 amino acids exhibiting 40-55% identity to mammalian, fungal, insect, and parasite P2 proteins. The inferred molecular mass of the protein is 12,592 Daltons, similar to that reported for other ribosomal P2 proteins. After subcloning and expression, the recombinant protein was purified by affinity chromatography under non-denatu

The transcription factor RUNT-like regulates pupal cuticle development via promoting a pupal cuticle protein transcription.

Holometabolous insects undergo morphological remodeling from larvae to pupae and to adults with typical changes in the cuticle; however, the mechanism is unclear. Using the lepidopteran agricultural insect Helicoverpa armigera, cotton bollworm, as a model, we revealed that the transcription factor RUNT-like (encoded by Runt-like) regulates the development of the pupal cuticle via promoting a pupal cuticle protein gene (HaPcp) expression. The HaPcp was highly expressed in the epidermis and wing during metamorphosis and was found being involved in pupal cuticle development by RNA interference (R

Insecticidal effects of Buthus occitanus tunetanus BotIT6 toxin expressed in Escherichia coli and baculovirus/insect cells.

BotIT6 is a neurotoxin polypeptide derived from the venom of the scorpion Buthus occitanus tunetanus (Bot). Its mature form is composed of 62 amino acids. BotIT6 has been reported to be the most potent toxin from Bot venom that has a strict selectivity for insects. Such toxin may have potential as a potent animal-harmless tool against insects. Using RT-PCR, we isolated and sequenced a cDNA encoding 62 amino acid residues corresponding to the known amino acid sequence of BotIT6. We have expressed a recombinant active form of BotIT6 in significantly high amounts in Escherichia coli. We have also

Blood meal drives de novo lipogenesis in the fat body of Rhodnius prolixus.

In insects, lipids are stored in the fat body mainly as triacylglycerol. Lipids can be directly provided by digestion and incorporated from the hemolymph, or synthesized de novo from other substrates such as carbohydrates and amino acids. The first step in de novo lipid synthesis is catalyzed by acetyl-CoA carboxylase (ACC), which carboxylates acetyl-CoA to form malonyl-CoA. Rhodnius prolixus is a hematophagous insect vector of Chagas disease and feeds exclusively on large and infrequent blood meals. Adult females slowly digest the blood and concomitantly accumulate lipids in the fat body. In

A streamlined cloning and toxicity evaluation strategy of toxin-antitoxin systems in insect cells.

Toxin-antitoxin (TA) systems, which are prevalent in bacteria and archaea, have significant potential applications in eukaryotic cells due to their characteristic ability to inhibit cell growth. In particular, type II TA systems, where both the toxin and antitoxin are proteins, can serve as lethal and rescue genes in genetic pest management. However, traditional methods of cloning these systems often encounter difficulties due to cryptic promoter driven expression of toxins during the cloning process. To address this issue, we have developed a novel cloning method that incorporates an addition

Three hydrophobic surface binding a proteins link surface hydrophobicity to sporulation, biofilm formation, and host interaction in Mucor lusitanicus.

INTRODUCTION: Mucor lusitanicus is a model organism for studying fungal development and physiology, as well as pathogenicity of Mucorales fungi. Hydrophobic surface-binding proteins (HsbA family) have been described in filamentous fungi as interface-associated factors involved in adhesion, enzymatic recruitment, and surface interactions; however, their functional diversification in Mucorales remains poorly understood. METHODS: Here, we present a comprehensive characterization of three HsbA proteins in M. lusitanicus. RESULTS: All examined HsbA proteins share a conserved α-helical fold with a

AtCYP6K1 regulates ovarian development and reproduction by catalyzing JH biosynthesis in the small hive beetle, Aethina tumida.

Insect cytochrome P450 monooxygenases (CYPs) exert essential functions in xenobiotic detoxification and growth and development. Nevertheless, the physiological roles of the majority of CYP genes remain poorly characterized in the small hive beetle (SHB), Aethina tumida. Screening of CYP genes from the ovarian transcriptome of SHBs identified AtCYP6K1 as an ovary-enriched gene. RT-qPCR analysis revealed that ovarian AtCYP6K1 expression increased with female adult age, reaching the highest level at day 7 post-eclosion. RNA interference (RNAi)-mediated silencing of AtCYP6K1 caused defective ovari

Jasmonate-Responsive MYB55 Regulates Herbivore Resistance Through the Phenylpropanoid Metabolism in Rice.

The jasmonate (JA) phytohormone signaling pathway is a key regulator of plant resistance to insect herbivores by mediating the biosynthesis of anti-herbivore metabolites and proteins. However, the JA-mediated regulatory network and its downstream defensive compounds in the monocot crop rice remain largely unknown. Here, we investigate the role of the MYB transcription factor MYB55 in JA-mediated rice defense against insect herbivores. Knockout of MYB55 enhances feeding by both brown planthopper (BPH) and leaf folder (LF), while having no effect on BPH oviposition. MYB55 directly binds to AC mo

Roles of spermary-specific carboxylesterases in Nilaparvata lugens reproduction: Opposite between insecticide-induced upregulation and resistance-associated overexpression.

Seminal fluid proteins (SFPs) in insect males are critical for reproduction, being transported into female ovary. In Nilaparvata lugens, a significant rice pest, seven spermary-specific carboxylesterases (CarEs) were found abundantly in SFPs, with over-expression in males of an imidacloprid-resistant (RES) strain compared to a susceptible (SUS) strain. This study aimed to evaluate roles of spermary-specific CarEs in N. lugens reproduction, especially in the reproduction disadvantage in RES. Sublethal doses of three insecticides significantly impacted N. lugens reproduction, concomitant with up
