Forty-five patients with myeloproliferative or myelodysplastic syndromes, treated with recombinant interferon-alpha (rIFN-alpha) for a minimum of 1 up to 4 years, were examined for the occurrence of thyroid autoimmunity. During treatment, the rate of thyroid autoimmunity rose to more than 20%. The decrease in severity and frequency of thyroid autoimmunity after withdrawal of IFN shows that this is a potentially reversible side effect. Interferon alpha-2b (αIF) was administered to 13 previously untreated patients with essential thrombocythaemia (ET). Recent evidence suggests that tumour necrosis factor alpha (TNF) is an autocrine growth factor for the chronic B-cell malignancies hairy cell leukaemia (HCL) and some cases of B-chronic lymphocytic leukaemia (B-CLL). The interferon system plays an important role in the control of viral infections and cell proliferation. These effects are mediated through the interaction of interferons with specific cell surface receptors. We report here the development of monoclonal antibodies against one of the subunits of the interferon alpha receptor. These antibodies detect a 110-kDa protein in surface-labeled cells and in Western blots, and 130- and 210-kDa bands after crosslinking to iodinated interferon alpha 2. No other subunits are disulfide-linked to the 130-kDa subunit or are coprecipitated by these antibodies.

Fifty-six patients with metastatic renal cell carcinoma (RCC) were treated with recombinant DNA-derived interferon alpha (rIFN alpha A). The first 30 patients were randomized between doses of 2 X 10(6) U/m2 and 20 X 10(6) U/m2 intramuscularly daily. PURPOSE: To evaluate the efficacy of topical interferon alpha-2b (tIFN a2b) and subcutaneous pegylated interferon alpha-2a (peg-IFN a2a) in the treatment of refractory pseudophakic (PME) and uveitic (UME) macular edema.
METHODS: Retrospective case series of patients with PME or UME that was non-responsive to conventional therapies. Topical IFN a2b drops (1 MIU/ml) were commenced four times a day. Non-responders were offered treatment with subcutaneous peg-IFN a2a starting at 180 mcg weekly.
