Spectrum and functions of ion channels and transporters in osteoclasts.

Osteoclasts are essential for bone resorption and interact with osteoblasts during bone remodeling. Ion channels and transporters located in the ruffled border or intracellular vesicles coordinate the transport of various ions and substrates, which is fundamental to the primary functions of osteoclasts. Numerous channels and transporters are implicated in bone metabolic disorders and genetic diseases. Among these, the voltage-gated chloride channel 7 (ClC-7) and vacuolar proton ATPases (V-ATPase) represent the most well-characterized examples in osteoclasts. Using the classification system of

Ion Clustering Regulated by Extreme Nanoconfinement Enables Mechanosensitive Nanochannels.

Mechanosensitive ion nanochannels regulate transport by undergoing conformational changes within the nanopores. However, achieving precise control over these conformational states remains a major challenge for both artificial soft and solid pores. Here, we propose an alternative mechanism that modulates the charge carrier density inside nanopores, inspired by transistors in solid-state electronics. This strategy leverages a novel phenomenon of confinement-regulated ion clustering in two-dimensional extremely confined nanochannels, revealed by extensive μs-scale enhanced-sampling molecular simu

Voltage-gated sodium channel toxins: poisons, probes, and future promise.

Neurotoxins have served as invaluable agents for identification, purification, and functional characterization of voltage-gated ion channels. Multiple classes of these toxins, which target voltage- gated Na+ channels via high-affinity binding to distinct but allosterically coupled sites, have been identified. The toxins are chemically diverse, including guanidinium heterocycles, a variety of structurally unrelated alkaloids, and multiple families of nonhomologous polypeptides having either related or distinct functions. This review describes the biochemistry and pharmacology of these agents, a

Some assembly required: SOCE and Orai1 channels couple to NFAT transcriptional activity via calmodulin and calcineurin.

Advances in our ability to monitor the temporal and spatial dynamics of intracellular second messengers such as Ca(2+) and cyclic nucleotides at millisecond and sub-micron levels of resolution have greatly increased our understanding of cellular signal transduction mechanisms. Thus, it is now well appreciated that second messengers can rise and fall within discrete regions of the intracellular compartment, as opposed to global changes, and on a time scale determined by the local collection of signaling molecules responsible for the synthesis and degradation/re-uptake of the second messenger. Voltage-gated Na+ channels: potential for beta subunits as therapeutic targets.

BACKGROUND: Voltage gated Na(+) channels (VGSCs) contain a pore-forming alpha subunit and one or more beta subunits. VGSCs are involved in a wide variety of pathophysiologies, including epilepsy, cardiac arrhythmia, multiple sclerosis, periodic paralysis, migraine, neuropathic and inflammatory pain, Huntington's disease and cancer. Increasing evidence implicates the beta subunits as key players in these disorders. OBJECTIVE: To review the recent literature describing the multifunctional roles of VGSC beta subunits in the context of their role(s) in disease. METHODS: An extensive review of th

Supramolecular Ion Channels to Engineer Zn2+ Ion Transport Mediated Chemical-to-Optical Signal Transduction.

Artificial ion channels have significant potential for various applications, including molecular communication, bio-sensing, and constructing artificial cells. In this report, we demonstrated the development of a molecular communication system that transports ions across the lipid bilayers via the formation of supramolecular ion channels and harnesses chemical reactions to overcome the challenges of executing signal-processing functions at the molecular level. The potent ionophore self-assembles into nanochannels within the lipid bilayers and selectively transports Zn2+ ions. The movement of Z

Modulation of Ion Channels by Cysteine-Rich Peptides: From Sequence to Structure.

Venom peptides are natural ligands of ion channels and have been used extensively in pharmacological characterization of various ion channels and receptors. In this chapter, we survey all known venom peptide ion-channel modulators. Our survey reveals that the majority of venom peptides characterized to date target voltage-gated sodium or potassium channels. We further find that the majority of these peptides are found in scorpion and spider venoms. We discuss the influence of the pharmacological tools available in biasing discovery and the classical "toxin-to-sequence" approach to venom peptid

A novel epileptic encephalopathy mutation in KCNB1 disrupts Kv2.1 ion selectivity, expression, and localization.

The epileptic encephalopathies are a group of highly heterogeneous genetic disorders. The majority of disease-causing mutations alter genes encoding voltage-gated ion channels, neurotransmitter receptors, or synaptic proteins. We have identified a novel de novo pathogenic K+ channel variant in an idiopathic epileptic encephalopathy family. Here, we report the effects of this mutation on channel function and heterologous expression in cell lines. We present a case report of infantile epileptic encephalopathy in a young girl, and trio-exome sequencing to determine the genetic etiology of her dis

Muscle acetylcholine receptor conversion into chloride conductance at positive potentials by a single mutation.

Charge selectivity forms the basis of cellular excitation or inhibition by Cys-loop ligand-gated ion channels (LGICs), and is essential for physiological receptor function. There are no reports of naturally occurring mutations in LGICs associated with the conversion of charge selectivity. Here, we report on a CHRNA1 mutation (α1Leu251Arg) in a patient with congenital myasthenic syndrome associated with transformation of the muscle acetylcholine receptor (AChR) into an inhibitory channel. Performing patch-clamp experiments, the AChR was found to be converted into chloride conductance at positiv

Zinc Oxide Nanoparticles and Voltage-Gated Human Kv 11.1 Potassium Channels Interact through a Novel Mechanism.

Membrane-nanoparticle interactions are important in determining the effects of manufactured nanomaterials on cell physiology and pathology. Here, silica, titanium, zinc, and magnesium oxide nanoparticles are screened against human hERG (Kv 11.1) voltage-gated potassium channels under a whole-cell voltage clamp. 10 µg mL-1 ZnO uniquely increases the amplitude of the steady-state current, decreases the rate of hERG current inactivation during steady-state depolarization, accelerates channel deactivation during resurgent tail currents, and shows no significant alteration of current activation rat

A Light-Driven Molecular Machine Controls K+ Channel Transport and Induces Cancer Cell Apoptosis.

The design of artificial ion channels with high activity, selectivity and gating function is challenging. Herein, we designed the light-driven motor molecule MC2, which provides new design criteria to overcome these challenges. MC2 forms a selective K+ channel through a single molecular transmembrane mechanism, and the light-driven rotary motion significantly accelerates ion transport, which endows the irradiated motor molecule with excellent cytotoxicity and cancer cell selectivity. Mechanistic studies reveal that the rotary motion of MC2 promotes K+ efflux, generates reactive oxygen species

Nanotechnology in Auditory Research: Membrane Electromechanics in Hearing.

The soft, thin membranes that envelop all living cells are 2D, nanoscale, fluid assemblies of phospholipids, sterols, proteins, and other molecules. Mechanical interactions between these components facilitate membrane function, a key example of which is ion flow mediated by the mechanical opening and closing of channels. Hearing and balance are initiated by the modulation of ion flow through mechanoreceptor channels in stereocilia membranes. Cochlear amplification by the outer hair cell involves modulation of ion movement by the membrane protein prestin. Voltage-gated ion channels shape the re

TMC1 and TMC2 function as the mechano-electrical transduction ion channel in hearing.

Mechanotransduction within the specialized cochlea hair cells is fundamental to perceiving sound waves. This intricate mechanism converts mechanical vibrations into electrical signals that the brain can interpret as sound. The molecular identity of the mechanoelectrical transducer continues to be a subject of intense debate. Transmembrane channel-like protein 1 (TMC1) was initially recognized as a deafness gene in humans, with subsequent studies revealing the hearing loss phenotype in Tmc1 mutant mice. Mechanotransduction currents were lost in the hair cells of Tmc1;Tmc2 double knockout mice,

Bio-Inspired Gating-Controlled Electronic Skin for Ultrafast Liquid Recognition and Precise Droplet Sliding Sensing in Robotic Intelligence.

Endowing intelligent robots with the ability to perceive and analyze complex liquid environments is essential for autonomous decision-making. However, conventional liquid-sensing technologies remain constrained by a fundamental trade-off between transient response and recognition accuracy, a limitation stemming from the sluggish kinetics of interfacial charge transfer and complex analytical procedures. Herein, we present a bionic liquid-sensing electronic skin (BLSE) inspired by the gating-controlled signal transduction mechanism of ion channels on the surface of biological sensory cells. By e

Shaping the Action Potential in Dorsal Root and Trigeminal Ganglia Neurons: Relevance to Pain Mechanisms.

Sensory neurons of the dorsal root ganglia (DRG) and trigeminal ganglia (TG) are critical for transmitting somatosensory information, including pain, to the central nervous system. In these neurons, as in other neurons, the shape of the action potential (AP) is a key determinant of excitability and signal transmission and is governed by the coordinated activity of voltage-gated sodium (NaV), potassium (KV), calcium (CaV), and calcium-activated potassium (BK) channels. This review synthesizes current knowledge about how these ion channels shape AP waveform and influence neuronal function across

CACNA1B mutation is linked to unique myoclonus-dystonia syndrome.

Using exome sequencing and linkage analysis in a three-generation family with a unique dominant myoclonus-dystonia-like syndrome with cardiac arrhythmias, we identified a mutation in the CACNA1B gene, coding for neuronal voltage-gated calcium channels CaV2.2. This mutation (c.4166G>A;p.Arg1389His) is a disruptive missense mutation in the outer region of the ion pore. The functional consequences of the identified mutation were studied using whole-cell and single-channel patch recordings. High-resolution analyses at the single-channel level showed that, when open, R1389H CaV2.2 channels carried

Roles of Microglial Ion Channel in Neurodegenerative Diseases.

As the average age and life expectancy increases, the incidence of both acute and chronic central nervous system (CNS) pathologies will increase. Understanding mechanisms underlying neuroinflammation as the common feature of any neurodegenerative pathology, we can exploit the pharmacology of cell specific ion channels to improve the outcome of many CNS diseases. As the main cellular player of neuroinflammation, microglia play a central role in this process. Although microglia are considered non-excitable cells, they express a variety of ion channels under both physiological and pathological co

Is lipid bilayer binding a common property of inhibitor cysteine knot ion-channel blockers?

Recent studies of several ICK ion-channel blockers suggest that lipid bilayer interactions play a prominent role in their actions. Structural similarities led to the hypothesis that bilayer interactions are important for the entire ICK family. We have tested this hypothesis by performing direct measurements of the free energy of bilayer partitioning (DeltaG) of several peptide blockers using our novel quenching-enhanced fluorescence titration protocol. We show that various ICK peptides demonstrate markedly different modes of interaction with large unilamellar lipid vesicles. The mechanosensiti
