Plasmid growth hormone releasing hormone therapy in healthy and laminitis-afflicted horses-evaluation and pilot study.

BACKGROUND: In vivo electroporation dramatically improves the potency of plasmid-mediated therapies, including in large animal models. Laminitis and arthritis are common and debilitating diseases in the horse, as well as humans. METHODS: The effects of growth hormone releasing hormone (GHRH) on healthy horses and on horses with laminitis that were followed for 6 months after a single intramuscular injection and electroporation of 2.5 mg of an optimized myogenic GHRH-expressing plasmid were examined. RESULTS: In the first study on six healthy horses, we observed a significant increase in body

Equine laminitis: bites by Bothrops spp cause hoof lamellar pathology in the contralateral as well as in the bitten limb.

The envenoming caused by Bothrops snakebite includes local symptoms, such as pronounced edema, hemorrhage, intense pain, vesicles, blisters and myonecrosis. The principal systemic symptom consists in the alteration of blood clotting, due to fibrinogen consumption and platelet abnormalities. The horses involved in this study had this symptomatology and one of them exhibited symptoms consistent with laminitis in the bitten and in the contralateral limbs. Laminitis lesions were characterized by separation of the hoof lamellar basement membrane (BM) from basal cells of the epidermis. These results

Evidence for vascular and enzymatic events in the pathophysiology of acute laminitis: which pathway is responsible for initiation of this process in horses?

To date, there is a substantial amount of data to support the hypotheses that vascular and enzymatic changes are ongoing in experimental laminitis. Furthermore, there is substantial in vitro evidence that the enzymatic changes weaken the dermo-epidermal attachments leading to mechanical failure of the hoof-bone interface of the equine digit. However, investigators of both the vascular and enzymatic theories have, to date, been unable to substantiate the effects of these pathophysiological changes in vivo on laminar tissues of horses afflicted with experimentally induced or naturally acquired l

Expression of a mutant lamin A that causes Emery-Dreifuss muscular dystrophy inhibits in vitro differentiation of C2C12 myoblasts.

Autosomal dominantly inherited missense mutations in lamins A and C cause several tissue-specific diseases, including Emery-Dreifuss muscular dystrophy (EDMD) and Dunnigan-type familial partial lipodystrophy (FPLD). Here we analyze myoblast-to-myotube differentiation in C2C12 clones overexpressing lamin A mutated at arginine 453 (R453W), one of the most frequent mutations in EDMD. In contrast with clones expressing wild-type lamin A, these clones differentiate poorly or not at all, do not exit the cell cycle properly, and are extensively committed to apoptosis. These disorders are correlated w…

Properties of lamin A mutants found in Emery-Dreifuss muscular dystrophy, cardiomyopathy and Dunnigan-type partial lipodystrophy.

Autosomal dominant Emery-Dreifuss muscular dystrophy is caused by mutations in the LMNA gene, which encodes lamin A and lamin C. Mutations in this gene also give rise to limb girdle muscular dystrophy type 1B, dilated cardiomyopathy with atrioventricular conduction defect and Dunnigan-type partial lipodystrophy. The properties of the mutant lamins that cause muscular dystrophy, lipodystrophy and dilated cardiomyopathy are not known. We transfected C2C12 myoblasts with cDNA encoding wild-type lamin A and 15 mutant forms found in patients affected by these diseases. Immunofluorescence microscopy…

Cerebellar microfolia and other abnormalities of neuronal growth, migration, and lamination in the Pit1dw-J homozygote mutant mouse.

The Snell dwarf mouse (Pit1dw-J homozygote) has a mutation in the Pit1 gene that prevents the normal formation of the anterior pituitary. In neonates and adults there is almost complete absence of growth hormone (GH), prolactin (PRL), thyroxin (T4), and thyroid-stimulating hormone (TSH). Since these hormones have been suggested to play a role in normal development of the central nervous system (CNS), we have investigated the effects of the Pit1dw-J mutation on the cerebellum and hippocampal formation. In the cerebellum, there were abnormalities of both foliation and lamination. The major folia

Adult stem cell maintenance and tissue regeneration in the ageing context: the role for A-type lamins as intrinsic modulators of ageing in adult stem cells and their niches.

Adult stem cells have been identified in most mammalian tissues of the adult body and are known to support the continuous repair and regener … tations in the LMNA gene. The concept is beginning to emerge that nuclear filament proteins, A-type lamins, may act as signalling receptors in the nucleus required for receiving and/or transducing upstream cytosolic signals in a number of pathways central to adult stem cell maintenance as well as adaptive responses to stress. We propose that during agein …

A Bimaterial Beam Strategy for Suppressing Thermal Deformation of Arc-Shaped CFRP Ribs via Asymmetric Laminate Design.

Deployable reflector antennas demand high geometric precision; the Ruze equation directly links surface error to RF gain. Arc-shaped CFRP ribs are vulnerable to thermal deformation, as their curvature converts in-plane expansion into out-of-plane displacement, which symmetric laminates cannot suppress. Classical laminate theory (CLT) underestimates the coefficient of thermal expansion (CTE) of cross-ply laminates by factors of 1.75-2.38 for the laminate configurations investigated in this study, causing up to 79.4% of displacement prediction errors in symmetric designs. Here, we present an asy

Spatiotemporal coordination of Slit-Robo repulsion and neurturin-Gfrα attraction guides multipolar migration during retinal lamination.

Multipolar migration is a conserved neuronal migration mode in the developing brain, enabling emerging neurons to navigate in crowded environments and reach precise laminar positions. Yet, how these cells interpret external cues to guide their migration is not fully understood. We investigate this question using retinal horizontal cells as a model. Combining transcriptomics, targeted CRISPR screening, and live imaging, we reveal the spatiotemporal guidance system underlying horizontal cell lamination: repulsive Slit1b/2-Robo2 signaling in the amacrine cell layer initiates apical horizontal cel

Sensor-Based Dynamic Response and Stress Estimation in Monolithic and Laminated Glass Beams Under Soft Impact Loading.

Monolithic and laminated glass elements are increasingly used in structural applications, where their response to impact loading must be assessed to ensure safety. For soft impacts, simplified simulations based only on the initial impact velocity may be inaccurate because the transient coupling between the impactor and the glass depends on the impactor stiffness and mass. This study investigates the dynamic response of monolithic and laminated glass beams under soft impact loading using accelerometers, an instrumented impact hammer, strain gauges, operational modal analysis, and simplified fin

Constructing Intrinsically Safe Lithium-Ion Battery Energy Storage via Gradient-Laminated Ceramifiable Silicone Foams.

Achieving high safety in energy storage systems is paramount but hindered by the catastrophic risks of thermal runaway propagation (TRP). This study develops a gradient-laminated ceramifiable silicone foam composite to resolve the inherent trade-off between thermal insulation and dynamic impact toughness. By integrating a polydimethylsiloxane foam matrix with a load-bearing glass fiber fabric skeleton, the material utilizes silane coupling agents for robust interfacial adhesion, while multiscale fillers promote synergistic ceramicization. Characterization reveals robust mechanical durability,

Multiscale Transfer of Cohesive-Zone Parameters for Opening-Dominated Interlaminar Fracture in Carbon-Fiber-Reinforced Aluminum Laminates.

Specimen-scale fitting can reproduce interlaminar fracture in carbon-fiber-reinforced aluminum laminates (CARALL) but obscures the physical origin of cohesive-zone parameters. We present a multiscale framework integrating molecular dynamics (MD), a representative volume element (RVE), finite element (FE) modeling, and opening-dominated asymmetric double-cantilever-beam (DCB) tests. At modeled high rates, an ideal nonbonded Al/epoxy interface exhibited normal and tangential strengths of 470.09 and 352.93 MPa, respectively. Across 0.001-0.005 Å/fs, normal and tangential peak tractions increased

Performance of Multi-Gradient Biomimetic POSS-Toughened CFRP Laminates with a Pearl-Inspired Hard-Soft Interfacial Architecture.

Carbon-fiber-reinforced epoxy (CFRP) composites are widely used in aerospace structures because of their high specific strength and modulus; however, matrix cracking and interlaminar delamination induced by low-velocity impact compromise their structural safety. To overcome the difficulty of simultaneously improving interfacial strength and impact resistance through uniform nanomodification, a pearl-inspired gradient interfacial architecture incorporating a through-thickness distribution of polyhedral oligomeric silsesquioxane (POSS) was developed and compared with unmodified and uniformly mod

Tau Oligomerization Drives Neurodegeneration via Nuclear Membrane Invagination and Lamin B Receptor Binding in Alzheimer's disease.

The microtubule-associated protein tau aggregates into oligomeric complexes that highly correlate with Alzheimer's disease (AD) progression. Increasing evidence suggests that nuclear membrane disruption occurs in AD and related tauopathies, but whether this is a cause or consequence of neurodegeneration remains unclear. Using the optogenetically inducible 4R1N Tau::mCherry::Cry2Olig (optoTau) system in iPSC-derived neurons, we demonstrate that tau oligomerization triggers nuclear rupture and nuclear membrane invagination. Pathological tau accumulates at sites of invagination, inducing structur

Improved Selectivity of CeMnOx/Pt@SnO2 Laminated MOS Sensor for Hydrogen Cyanide Under Temperature Dynamic Modulation.

Poor selectivity is one of the main bottlenecks restricting the development of metal oxide semiconductor (MOS) sensors. In this paper, using hydrogen cyanide (HCN) as the target gas, CeMnOx as the catalytic layer material and Pt@SnO2 as the gas-sensitive layer material, we have proposed a scheme to improve the selectivity of a catalytic layer/gas-sensitive layer-laminated MOS sensor under dynamic temperature modulation. We tested HCN and 12 kinds of battlefield environment simulation gases, and the results showed that the CeMnOx/Pt@SnO2 sensor, under the condition of temperature dynamic modula

Carbon fiber-laminated epoxy resin causing chronic occupational dermatosis: presentation with erythroderma and dyschromia-a case report.

Epoxy resins are a common cause of occupational allergic contact dermatitis. Carbon fiber-laminated epoxy resin (CFLER) is a newer material with enhanced mechanical properties, but its health effects remain underreported. A 46-year-old male worker developed erythroderma involving >90% of his body surface and dyschromia after chronic exposure to CFLER in a confined workspace. Skin biopsy demonstrated features of chronic dermatitis, and immunohistochemistry revealed uneven melanocyte distribution. Laboratory studies showed elevated immunoglobulin E levels without internal organ dysfunction. Ches

Bonding performance and surface characterization of cold-bonded acetylated beech (Fagus sylvatica L.) laminated veneer lumber.

Acetylation of wood with acetic anhydride reduces the wood-moisture interaction, improves the dimensional stability and resistance against biodegradation. However, the adhesive bonding is affected by the modification, which is crucial to manufacture engineered wood products, such as laminated veneer lumber (LVL). In this study we report the bonding of 8-layered acetylated beech (Fagus sylvatica L.) LVL boards to 2-layered LVL beams. The beams were glued together at room temperature adding three common load-bearing construction adhesives: melamine-urea-formaldehyde (MUF), phenol-resorcinol-form

Co-cultures of cerebellar slices from mice with different reelin genetic backgrounds as a model to study cortical lamination.

Background: Reelin has fundamental functions in the developing and mature brain. Its absence gives rise to the Reeler phenotype in mice, the first described cerebellar mutation. In homozygous mutants missing the Reelin gene ( reln -/-), neurons are incapable of correctly positioning themselves in layered brain areas such as the cerebral and cerebellar cortices. We here demonstrate that by employing ex vivo cultured cerebellar slices one can reduce the number of animals and use a non-recovery procedure to analyze the effects of Reelin on the migration of Purkinje neurons (PNs). Methods: We gene
