Neurons have well-developed membrane microdomains called "rafts" that are recovered as a detergent-resistant membrane microdomain fraction (DRM). Neuronal tissue-enriched acidic protein of 22 kDa (NAP-22) is one of the major protein components of neuronal DRM. To determine the cellular function of NAP-22, interacting proteins were screened with an immunoprecipitation assay, and calcineurin (CaN) was detected. Further studies with NAP-22 prepared from DRM and CaN expressed in bacteria showed the binding of these proteins and a dose-dependent inhibitory effect of the NAP-22 fraction on the phosp

Cholesterol accumulation in late endosomes is a prevailing phenotype of Niemann-Pick type C1 (NPC1) mutant cells. Likewise, annexin A6 (AnxA6) overexpression induces a phenotype reminiscent of NPC1 mutant cells. Here, we demonstrate that this cellular cholesterol imbalance is due to AnxA6 promoting Rab7 inactivation via TBC1D15, a Rab7-GAP. In NPC1 mutant cells, AnxA6 depletion and eventual Rab7 activation was associated with peripheral distribution and increased mobility of late endosomes. This was accompanied by an enhanced lipid accumulation in lipid droplets in an acyl-CoA:cholesterol acyl

The effect of altering cell membrane lipids on ion transport across isolated corneas was studied. Corneas mounted in Ussing-type chambers showed a rapid increase in short-circuit current following treatment with a variety of unsaturated fatty acids of varying chain length and unsaturation. Measurements of membrane fluidity which utilize immunofluorescence labelling of membrane proteins showed corneal epithelial cell membranes to be significantly more fluid following linoleic acid treatment. Uptake studies indicate rapid incorporation of [14C]linoleic acid into corneal cell membranes. Highly un

OBJECTIVES: The usefulness of omega-3 lipid emulsions has been extensively studied. The objectives of the present study were to examine the effect of an omega-3 lipid emulsion in reducing oxidative stress in a rat model of intestinal ischemia-reperfusion injury and the underlying mechanism.
METHODS: A total of 66 rats were divided into three dietary groups (lipid-free, soybean oil, and fish oil groups). Each animal was administered total parenteral nutrition for 3 days, followed by induction of intestinal ischemia for 100 min. Animals subjected to sham surgery served as the controls. Intestin

We have investigated the influence of the neurotoxic Alzheimer's disease peptide amyloid-beta (25-35) on the dynamics of phospholipid membranes by means of quasi-elastic neutron scattering in the picosecond time-scale. Samples of pure phospholipids (DMPC/DMPS) and samples with amyloid-beta (25-35) peptide included have been compared. With two different orientations of the samples the directional dependence of the dynamics was probed. The sample temperature was varied between 290K and 320K to cover both the gel phase and the liquid-crystalline phase of the lipid membranes. The model for describ

We report that soluble serine proteases such as trypsin and chymotrypsin trigger the shedding and/or detachment of SARS-CoV-2 particles bound to lipid membranes. Shedding kinetics were evaluated upon fluorescence imaging of the labeled virus particles on native membrane mimics. The temporal change in the fluorescence signal, supported by kinetic data from SPR, confirms the fastest shedding of the Delta variant. B.1.1.8 (near Wuhan) and Omicron exhibit comparable shedding kinetics. Membrane-bound Delta variants become laterally mobile upon protease treatment. The other variants are shed from th

To elucidate abnormalities in lipid metabolism in patients with primary biliary cirrhosis (PBC), plasma lipoproteins and apoproteins were analyzed in 10 such patients. Lipoprotein X was present in sera from five of the patients. Another abnormal lipoprotein of slow alpha-mobility on agarose gel electrophoresis was observed in sera of eight of the patients. The slow alpha-lipoprotein was distributed in the range of densities between low density and high density lipoproteins and was rich in apoprotein E. This abnormal lipoprotein of PBC was observed in those in Stages II and III but not in those

The MarR family of transcription factors in Mycobacterium tuberculosis plays a critical role in bacterial adaptation to host stresses, yet the function of many members remains unknown. Here, we characterize the novel MarR regulator Rv0737 and its homolog Ms_1492 in M. smegmatis. Overexpression of Rv0737 severely impaired bacterial growth, cell division, and biofilm formation, while increasing susceptibility to oxidative stress and the cell wall-targeting drug isoniazid. Conversely, deletion of ms_1492 altered cell envelope permeability and lipid composition, enhanced ATP synthesis, and conferr

Red blood cells (RBCs) are the most abundant human cell type, and interactions with the RBC membrane are at the heart of many processes relevant for human health, such as immune system modulation, as well as binding by foreign pathogens and pharmacological drugs. To better study such membrane interface interactions, it would be useful to employ surface-attached model lipid membranes derived from RBCs to enable surface-sensitive biophysical and biochemical measurements. Here, we present approaches to prepare two such types of RBC-derived model lipid membranes─tethered RBC liposomes and supporte

The ability to generate, store, and mobilize energy is fundamental to life, and disruptions in these processes underlie metabolic disease. This review examines how these mechanisms evolved to rely profoundly on lipids, a large family of molecules that serve an unusually wide range of biological functions. From early evolutionary times, lipids have enabled chemiosmotic energy transduction, organized the endomembrane systems that synthesize and package neutral lipids, served as concentrated reservoirs of metabolic energy, and functioned as signaling molecules that coordinate cellular and systemi

A subsection of integral membrane proteins partition into chloroform during a chloroform/methanol/water extraction primarily designed to extract lipids. Traditionally, these proteins were called proteolipids due to their lipid-like properties; the c-subunit of the ATP synthase integral FO component is the best known due to its abundance. In this manuscript, we investigate purification of proteolipid proteins away from lipids for high-resolution mass spectrometry. Size-exclusion chromatography on silica beads using a chloroform/methanol/aqueous formic acid (4/4/1; v/v) mobile phase allowed the

UNLABELLED: Excessive production of reactive oxygen species (ROS) in cells results in oxidative stress, which can promote lipid oxidation in cellular membranes. This oxidation of membrane lipids accompanies various diseases and can even result in cell death through processes such as ferroptosis. The complex compositions and diverse morphologies of cellular membranes make understanding the mechanisms of lipid oxidation challenging, especially when attempting to investigate membrane composition and curvature simultaneously. Here, we utilize reconstituted lipid membranes and the fluorescent oxida

As an electric field-operated and relatively noninvasive technique, dielectrophoresis (DEP) has been frequently employed in the manipulation, assembly and sorting of bioparticles, including cells, organelles and liposomes. Of the latter category, however, prior investigations have been mostly generic, treating liposomes as just another type of dielectric colloids and leaving specific lipid properties, such as their type and phase, largely unprobed. In this work, we subject microsized liposomes in either liquid-ordered or liquid-disordered phase, or their combination, under DEP, and follow thei

Aging and obesity are closely connected, with obesity being a major risk factor for many age-related diseases caused by chronic inflammation and mobility problems. Lifestyle choices such as good nutrition, including medicinal herbs, and exercise are believed to influence body weight. Ashwagandha, an Ayurvedic adaptogen containing bioactive withanolides, is known to reduce stress, fight cancer, and improve energy metabolism. Its effects on lipid metabolism and obesity remain unclear. CARF (Collaborator of ARF), a protein involved in stress response and cancer regulation, was recently found to c

Lipids make up an important class of biomolecules with diverse structures and varied chemical functions. This diversity is a major challenge in chemical analysis and limits our understanding of biological functions and regulation. A major way lipid isomers differ is by double-bond (db) position, and analyzing db-isomers is especially challenging for mass spectrometry imaging (MSI). Ozonolysis can be used to determine the db-position and has been paired with MSI before. However, previous techniques require increased analysis time to allow for gas-phase reactions within an ion trap or ion mobili

The glycolipid glycosylphosphatidylinositol anchor (GPI-A) plays an important role in lipid raft formation, which is required for proper expression on the cell surface of two inhibitors of the complement cascade, CD55 and CD59. The absence of these markers from the surface of blood cells, including erythrocytes, makes the cells susceptible to complement lysis, as seen in patients suffering from paroxysmal nocturnal haemoglobinuria (PNH). However, the explanation for why PNH-affected hematopoietic stem/progenitor cells (HSPCs) expand over time in BM is still unclear. Here, we propose an explana
