The localization and specialized function of Ras-like proteins are largely determined by posttranslational processing events. In a highly regulated process, palmitoyl groups may be added to C-terminal cysteine residues, targeting these proteins to specific membranes. In the human fungal pathogen Cryptococcus neoformans, Ras1 protein palmitoylation is essential for growth at high temperature but is dispensable for sexual differentiation. Ras1 palmitoylation is also required for localization of this protein on the plasma membrane. Together, these results support a model in which specific Ras fun

Pseudohypoparathyroidism type Ia (PHP-Ia) results from the loss of one allele of G(salpha), causing resistance to parathyroid hormone and other hormones that transduce signals via G(s). Most G(salpha)mutations cause the complete loss of protein expression, but some cause loss of function only, and these have provided valuable insights into the normal function of G proteins. Here we have analyzed a mutant G(salpha) (alphas-AVDT) harboring AVDT amino acid repeats within its GDP/GTP binding site, which was identified in unique patients with PHP-Ia accompanied by neonatal diarrhea. Biochemical and

BACKGROUND: Breast cancer (BrCa) has become the most common malignant tumor among women worldwide. Inducing tumor cell pyroptosis has emerged as a potential anti-tumor strategy, although the biological effects of pyroptosis are context-dependent and may be dual-edged. The NLRP3 inflammasome is a major regulator of canonical pyroptosis. However, the role of ZDHHC7-mediated NLRP3 palmitoylation in BrCa remains unclear.
METHODS: ZDHHC7 expression was analyzed using UALCAN, Kaplan-Meier plotter, qPCR, and Western blotting. The growth and metastasis ability of cells were detected and evaluated, re

SARS-CoV-2 accessory protein ORF3a contributes to viral pathogenesis through membrane remodeling, immune evasion, and inflammation induction. However, the molecular mechanisms underlying ORF3a-mediated pathogenesis remain poorly characterized, and no therapeutic strategies targeting ORF3a currently exist. Here, we demonstrate that palmitoylation, a post-translational modification, governs ORF3a-mediated viral pathogenesis. Specifically, ORF3a undergoes ZDHHC18-mediated palmitoylation at evolutionarily conserved Cys130/Cys133 residues, which stabilizes the protein by masking an intrinsic protea

The homeostasis of protein palmitoylation and depalmitoylation is essential for proper physiological functions in various tissues, in particular the central nervous system (CNS). The dysfunction of PPT1 (PPT1-KI, infantile neuronal ceroid lipofuscinosis [INCL] mouse model), which catalyze the depalmitoylation process, results in serious neurodegeneration accompanied by severe astrogliosis in the brain. Endeavoring to determine critical factors that might account for the pathogenesis in CNS by palm-proteomics, glial fibrillary acidic protein (GFAP) was spotted, indicating that GFAP is probably

Alterations of cell adhesion are involved in cancer progression, but the mechanisms underlying the progression and cell adhesion have remained poorly understood. Focusing on the complex between EpCAM, claudins and tetraspanins, we described a sequence of events by which of the molecules associate each other in ovarian cancer. The interactions between molecules were evaluated by immunoprecipitations and then immunoblotting. To identify the effects of complex formation on the ovarian cancer progression, the different types of ovarian cancer cell lines were compared. In this study, we report the

HD (Huntington's disease) is caused by an expanded polyQ (polyglutamine) repeat in the htt (huntingtin protein). GABAergic medium spiny neurons in the striatum are mostly affected in HD. However, mhtt (mutant huntingtin)-induced molecular changes in these neurons remain largely unknown. The present study focuses on the effect of mhtt on the subcellular localization of GAD (glutamic acid decarboxylase), the enzyme responsible for synthesizing GABA (γ-aminobutyric acid). We report that the subcellular distribution of GAD is significantly altered in two neuronal cell lines that express either the

Ferroptosis represents a promising vulnerability to overcome therapeutic resistance in castration-resistant prostate cancer (CRPC). While S-palmitoylation of lipid peroxide-scavenging proteins such as GPX4 and SLC7A11 has been shown to suppress ferroptosis, whether palmitoylation modulates the lipid peroxidation generation remains unclear. Here, we identified the palmitoyltransferase ZDHHC2 as a critical driver of enzalutamide resistance through destabilizing ACSL4. ZDHHC2 is transcriptionally upregulated by a FOXA1/CXXC5/TET2 complex and promotes S-palmitoylation of the deubiquitinase USP19,

The precise regulation of synaptic integrity is critical for neuronal network connectivity and proper brain function. Essential aspects of the activity and localization of synaptic proteins are regulated by posttranslational modifications. S-palmitoylation is a reversible covalent modification of the cysteine with palmitate. It modulates affinity of the protein for cell membranes and membranous compartments. Intracellular palmitoylation dynamics are regulated by crosstalk with other posttranslational modifications, such as S-nitrosylation. S-nitrosylation is a covalent modification of cysteine

Soraphen A is a myxobacterial metabolite that blocks the acetyl-coenzyme A carboxylase of the host and was previously identified as a novel HIV inhibitor. Here, we report that soraphen A acts by reducing virus production and altering the gp120 virion content, impacting entry capacity and infectivity. These effects are partially reversed by addition of palmitic acid, suggesting that inhibition of HIV envelope palmitoylation is one of the mechanisms of antiviral action.

Protein S-palmitoylation is a reversible post-translational modification mediated by palmitoyl acyltransferase enzymes, a group of Zn2+-finger DHHC-domain-containing proteins (ZDHHC). Here, for the first time, we show that Zdhhc13 plays a key role in anxiety-related behaviors and motor function, as well as brain bioenergetics, in a mouse model (luc) carrying a spontaneous Zdhhc13 recessive mutation. At 3 m of age, mutant mice displayed increased sensorimotor gating, anxiety, hypoactivity, and decreased motor coordination, compared to littermate controls. Loss of Zdhhc13 in cortex and cerebellu

The amino acid sequences and signaling pathways of D2-like dopamine receptors (D2R, D3R, and D4R) are highly conserved. D4R has been suggested to be associated with novelty seeking, and binds with high affinity to an atypical antipsychotic drug with fewer motor function-related side effects than typical neuroleptics. A study comparing D2R and D3R reported that palmitoylation is important for the proper functioning of D3R. Although D4R is a member of the D2-like receptor family, its palmitoylation status and the functional significance of any such posttranslational modification are unknown. In

Progressive cardiomyocyte loss constitutes the pathological hallmark of dilated cardiomyopathy (DCM), a process driven by an intricate network of programmed cell death (PCD) pathways. This review systematically examines the molecular underpinnings and reciprocal crosstalk among six principal PCD modalities implicated in DCM: apoptosis, necroptosis, pyroptosis, autophagy, ferroptosis, and cuproptosis. Apoptosis is triggered by genetic defects-most notably titin (TTN) truncating variants-epigenetic dysregulation, and endoplasmic reticulum stress, converging on the activation of both intrinsic an

BACKGROUND: Cuproptosis represents a promising therapeutic strategy for colorectal cancer (CRC), yet the epigenetic mechanisms governing cuproptosis sensitivity remain largely unexplored. Enhancer of zeste homolog 2 (EZH2), a key epigenetic regulator frequently overexpressed in CRC, may play a critical role in modulating copper-induced cell death. This study aimed to investigate the role of EZH2 and its downstream effectors in regulating cuproptosis sensitivity in CRC cells.
METHODS: Using elesclomol-copper complex treatment, a cuproptosis model in HCT116 and RKO CRC cell lines was establishe

Cuproptosis is a form of programmed cell death reported in 2022, characterized by copper ions directly binding to mitochondrial lipoylated proteins, inducing aberrant protein aggregation and destabilizing iron-sulfur cluster proteins, ultimately leading to cell death. Distinct from apoptosis, ferroptosis, and other known death modalities, cuproptosis does not rely on classical caspase pathways but exerts its cytotoxic effects by interfering with mitochondrial metabolism. Copper, an essential trace element for humans, maintains its homeostasis through precise regulation by transporters such as

Methamphetamine (METH) abuse can cause persistent cognitive impairment and neuronal injury, but exposure-dependent alterations in cell-death-associated markers during repeated exposure remain incompletely characterized. We combined behavioral testing, exposure-dependent molecular profiling, transmission electron microscopy, and terminal hippocampal RNA sequencing to characterize METH-induced hippocampal injury. Repeated METH exposure impaired spatial learning and memory in mice. qPCR and Western blot analyses from animals sampled across five days of repeated METH exposure revealed that changes

BACKGROUND: Osteoporosis is a prevalent metabolic bone disorder driven by an imbalance between osteoblast-mediated bone formation and osteoclast-mediated bone resorption, and current pharmacological options remain limited by adverse effects and incomplete mechanistic targeting. Copper, an essential trace element, has long been linked to bone mineral density through population-level dietary surveys, but whether and how copper mechanistically shapes osteoblast and osteoclast function at the subcellular level has not been systematically reviewed.
METHODS: We synthesised genetic, clinical, and ce

Primary biliary cirrhosis (PBC) is an autoimmune liver disease characterized by destruction of intrahepatic bile ducts. Although the pathogenesis of this disease is still unknown, high titers of antimitochondrial autoantibodies (AMA) have long been recognized in patient sera. However, little is known about the presence of AMA in bile. In this study, we investigated bile and sera from patients with PBC and healthy controls for the presence of AMA and mitochondrial autoantigens. AMA were detected in the bile of 17 of 19 patients (89.4%) with PBC; they were specifically directed against the pyruv

Jellyfish envenomation poses a significant clinical challenge, with Nemopilema nomurai (N. nomurai) venom capable of inducing life-threatening hepatotoxicity through mechanisms that remain poorly understood. Here, we demonstrate that its tentacle extract (TE) simultaneously triggers a reactive oxygen species (ROS) burst, activates the canonical NLRP3/Caspase-1/Gasdermin D (GSDMD) pyroptosis pathway, and induces ZDHHC9-dependent GSDMD palmitoylation, collectively contributing to hepatic injury. To counteract this dual-pathway activation, we engineered ultrasmall iridium nanoantidotes (Ir NAs) w

BACKGROUND: Breast cancer (BC) ranks as the third most fatal malignant tumor worldwide, with a strong reliance on fatty acid metabolism. CLDN6, a candidate BC suppressor gene, was previously identified as a regulator of fatty acid biosynthesis; however, the underlying mechanism remains elusive. In this research, we aim to clarify the specific mechanism through which CLDN6 modulates fatty acid anabolism and its impact on BC growth and metastasis.
METHODS: Cell function assays, tumor xenograft mouse models, and lung metastasis mouse models were conducted to evaluate BC growth and metastasis. Hu
