Tumor Imaging Using Radiolabeled Matrix Metalloproteinase-Activated Anthrax Proteins.

Increased activity of matrix metalloproteinases (MMPs) is associated with worse prognosis in different cancer types. The wild-type protective antigen (PA-WT) of the binary anthrax lethal toxin was modified to form a pore in cell membranes only when cleaved by MMPs (to form PA-L1). Anthrax lethal factor (LF) is then able to translocate through these pores. Here, we used a 111In-radiolabeled form of LF with the PA/LF system for noninvasive in vivo imaging of MMP activity in tumor tissue by SPECT. Methods: MMP-mediated activation of PA-L1 was correlated to anthrax receptor expression and MMP acti

A comparative analysis of CXCR4 gene expression from published datasets and cell surface protein levels in breast and prostate cancer models.

The CXCL12/CXCR4 signaling axis is a critical mediator of cancer metastasis, particularly in the homing of breast and prostate cancer cells to the bone. Consequently, characterizing the CXCR4 status of preclinical cancer models is essential for designing meaningful experiments. However, prior reports show conflicting CXCR4 status in these models, with reported positivity rates varying from <5 % to >90 % across studies, highlighting the urgent need for systematic comparison. This study provides a systematic comparison of CXCR4 expression patterns by analyzing gene expression data from breast ca

Peptide surface structure flexibility controls gold nanoparticle uptake in triple-negative breast cancer cells.

Peptide-functionalized gold nanoparticles (AuNPs) provide a model for decoding how the surface-bound secondary structure regulates nano-bio interactions. AuNPs (30-40 nm) coated with small peptides, GSH, ECECG, GSSG and cyclic CEIEIEC exhibited a progressive increment in antiparallel β-sheet content (37-89%) upon surface binding with an increase in the number of amino acids. Cellular studies in MDA-MB-231 cells revealed efficient uptake for flexible, mixed-structure coronas (GSH, ECECG, GSSG), while the rigid, antiparallel β-sheet-dominant cyclic CEIEIEC corona suppressed internalization despi…

Early Radiation Therapy Response Assessment Using Multi-Scale Photoacoustic Imaging.

There is a critical unmet clinical need to identify biomarkers that predict and detect radiation therapy (RT) response in cancer. Using the unique capabilities of multi-scale photoacoustic imaging (PAI) to depict tumor oxygenation and vasculature in vivo, we identified surrogate biomarkers of RT response in two human breast cancer xenograft models (MCF7 and MDA-MB-231), comparing hypofractionated delivery with an ablative single-dose scheme. Mesoscopic and multispectral tomographic PAI were performed 24h pre-RT, 24h post-RT and at endpoint and were supported by ex vivo immunohistochemistry. MC

Lipidomic profiling of extracellular vesicles from breast and metastatic triple-negative breast cancer cell lines for identification of potential biomarkers.

Triple-negative breast cancer (TNBC) remains a highly aggressive disease with limited therapeutic options, and metastasis is the leading cause of patient mortality which highlights a major unmet medical need. Nevertheless, the mechanisms regulating metastatic progression remain poorly understood. Extracellular vesicles (EVs) have recently emerged as key mediators of the intercellular communication. Ubiquitously secreted, these nanoparticles provide a convenient and informative glimpse into cellular physiology given that their molecular composition shall reflect the state of their parent cell.

Actinolactomycin from Streptomyces sp. CB00271: Isolation, Identification, and Potent Anti-Triple-Negative Breast Cancer.

Triple-negative breast cancer (TNBC) is a highly aggressive subtype with limited targeted therapies, underscoring an urgent need for novel agents. The soil-derived Streptomyces sp. CB00271, isolated from a biodiversity hotspot, was investigated for its bioactive metabolites. Bioassay-guided isolation led to the identification of actinolactomycin (1), alongside daidzein (2) and genistein (3). Remarkably, actinolactomycin (1) exhibited potent cytotoxicity against TNBC models, with IC50 values of 0.72 ± 0.12 μM (MDA-MB-231) and 0.15 ± 0.02 μM (4T1), demonstrating approximately 9-fold and 31-fold

Combinatorial Treatment with Chlorogenic Acid and Cinnamaldehyde Disrupts Intracellular pH and Metabolic Transport in Breast Cancer Cells.

Breast cancer cells exhibit a reversed pH gradient and metabolic plasticity that promote proliferation, invasion, and resistance to therapy. Natural products such as chlorogenic acid (CGA) and cinnamaldehyde (CA) have shown emerging anticancer potential. However, their effects on intracellular pH and metabolic transport systems remain undefined. Therefore, the aim of this study was to characterize these parameters in breast cancer and non-tumorigenic breast cells. This study evaluated the physiochemical properties of CGA and CA using LC-MS, under pH conditions (pH 1.2, 7.4, and 9.0) mimicking

Identification of a gene (GPR30) with homology to the G-protein-coupled receptor superfamily associated with estrogen receptor expression in breast cancer.

Using the technique of differential cDNA library screening, a cDNA clone was isolated from an estrogen receptor (ER)-positive breast carcinoma cell line (MCF7) cDNA library based upon the overexpression of this gene compared to an ER-negative cell line (MDA-MB-231). Sequence analysis of this clone determined that it shared significant homology to G-protein-coupled receptors. This receptor, GPCR-Br, was abundantly expressed in the ER-positive breast carcinoma cell lines MCF7, T-47D, and MDA-MB-361. Expression was absent or minimal in the ER-negative breast carcinoma cell lines BT-20, MDA-MB-231

Exploiting novel placental homing peptides for targeted drug delivery in breast cancer.

More effective drug formulations are needed to increase the selectivity and efficacy of available chemotherapeutics. We have previously shown that nanoparticles decorated with the tumour homing peptide CGKRK can selectively deliver payloads to the placenta. In this study, we investigated whether two novel placental homing peptides NKGLRNK (NKG) and RSGVAKS (RSG) can be utilized to selectively deliver doxorubicin (DOX) to breast cancer cells. Fluorescence microscopy and flow cytometry showed that NKG and RSG bind to and accumulate in MDA-MB-231 and MCF-7 cells in a time-dependent manner, to a s

Recombinant pigment epithelium-derived factor reverses epithelial-to-mesenchymal transition (EMT) and impedes proliferation of triple-negative breast cancer cells through downregulation of Wnt/β-catenin signaling.

Triple-negative breast cancer (TNBC) remains the most aggressive breast cancer subtype due to aberrant activation of EMT, predominantly driven by the Wnt/β-catenin signaling. Pigment epithelium-derived factor, a multifunctional protein and negative regulator of the Wnt pathway, is markedly downregulated in TNBC, providing an opportunity to restore the inhibition of this signaling axis for therapeutic benefit. In this study, we present a systematic method for the production of bioactive recombinant pigment epithelium-derived factor protein expressed in Escherichia coli BL21(DE3) and characteriz

Long-chain fatty acid esters of 5-androstene-3 beta, 17 beta-diol: composition and turnover in human mammary cancer cells in culture.

Long-chain fatty acid esters of the adrenal-derived estrogen 5-androstene-3 beta, 17 beta-diol (ADIOL) were found to accumulate in four human mammary cancer cell lines (MCF-7, ZR-75-1, MDA-MB-231 and MDA-MB-330) when explosed to 10-30 nM ADIOL for variable time periods. At each time point examined, the monoester fraction, which represented the major component of the total lipoidal fraction, contained fatty acids linked to either the 3 beta- or 17 beta-positions. However, there was considerable variation in the ratio of 3 beta- to 17 beta-monoesters in the four cell lines. By means of reverse p
