NADH dehydrogenase defects confer isoniazid resistance and conditional lethality in Mycobacterium smegmatis.

Isoniazid (INH) is a highly effective drug used in the treatment and prophylaxis of Mycobacterium tuberculosis infections. Resistance to INH in clinical isolates has been correlated with mutations in the inhA, katG, and ahpC genes. In this report, we describe a new mechanism for INH resistance in Mycobacterium smegmatis. Mutations that reduce NADH dehydrogenase activity (Ndh; type II) cause multiple phenotypes, including (i) coresistance to INH and a related drug, ethionamide; (ii) thermosensitive lethality; and (iii) auxotrophy. These phenotypes are corrected by expression of one of two enzym

Increased type II glucocorticoid-receptor numbers and glucocorticoid-sensitive enzyme activities in the brain of the obese Zucker rat.

The possibility that the glucocorticoid-dependence of obesity of the obese fa/fa rat reflects on overactivity of glucocorticoids on the brain has been investigated by studies of enzyme activities and glucocorticoid type II (GR) receptors. The activity of 2 glucocorticoid-sensitive enzymes, glycerol-3-phosphate dehydrogenase and glutamine synthetase, were increased in the hippocampus of obese rats. In contrast malate dehydrogenase and pyruvate kinase, glucocorticoid-insensitive enzymes, were normal. Adrenalectomy of obese rats reduced glycerol-3-phosphate dehydrogenase activity to the level of

Sample preparation for inositol measurement: Sep-Pak C18 use in detergent removal.

The enzymatic-fluorometric method of myo-inositol quantitation by L.C. MacGregor and F.M. Matschinsky (1984, Anal. Biochem. 141, 382-389) has proved to be very useful in accurately measuring small amounts of myo-inositol. Although this procedure is quite satisfactory, relatively high concentrations of detergents, salts, NADH, and malate interfere. To extend the usefulness of the MacGregor and Matschinsky method we report here an extraction procedure which removed the interfering substances, yet allowed the recovery of close to 100% of the inositol. The procedure involved first passing the samp

Hu'po Anshen Decoction Accelerated Fracture-Healing in a Rat Model of Traumatic Brain Injury Through Activation of PI3K/AKT Pathway.

Hu'po Anshen decoction (HPASD) is a traditional Chinese medicine formula comprising five herbal medicines for the treatment of concussion and fracture healing, but its pharmacological mechanism is still unclear. Ultra-performance liquid chromatography coupled with quadrupole time of flight mass spectrometry (UPLC/Q-TOF MS) was used to analyze the main active components of HPASD. Rats were randomly assigned to fracture group, fracture combined with traumatic brain injury (TBI) group (FBI) and FBI combined with HPASD treatment group (FBIH). Rats in the FBIH group were given oral doses of HPASD (

[Physiology and clinical aspects of the extracellular bicarbonate pool: plea for cognizant use of HCO3-].

OBJECTIVE: Derived from literature data the function of bicarbonate (HCO3-) is described together with its extracellular pool, the regulating organs, and the clinical variations. DATA SOURCES AND SELECTION CRITERIA: The medical German and English literature was reviewed. No special literature retrieval was performed. Results from measurements at the author's laboratory were used. RESULTS: HCO3- may be described as the most potential nonrespiratory buffer base. For a 65 kg patient, the extracellular HCO3- pool amounts to approximately 350 mmol with a maximum tolerance limit of +/- 200 mmol. A

Isolation and biochemical characterization of maleic-acid hydratase, an iron-requiring hydro-lyase.

A procedure for the isolation of maleic acid hydratase (D-malate hydro-lyase, EC 4.2.1.31) of about 95% purity from rabbit kidneys is described. The enzyme consists of a single polypeptide chain of 582 amino-acid residues with an approximate molecular mass of 68 kDa. The enzyme is very unstable and has an absolute requirement for chloride ions. Addition of sodium sulphide during the purification process was essential to maintain the enzyme in an activatable state. The pure preparation has low activity but responds to activation with Fe2+ ions, Na2S and a thiol. The sequence of adding the activ

Selective inhibition of enzyme synthesis under conditions of respiratory inhibition.

When Neurospora mycelium is transferred from a medium containing sucrose to one containing acetate as sole source of carbon, a preferential synthesis of many Krebs cycle, glyoxylate cycle, and associated enzymes occurs. Respiration was inhibited during preferential enzyme synthesis in the following ways. (i) The amount of aeration (shaking) was reduced, (ii) cyanide was added to the culture, (iii) the carbon source, acetate, was removed, (iv) a mutant strain was starved of its Krebs cycle intermediates, and (v) respiration was inhibited by mutation. The effect of this respiratory inhibition on

l-Malic Acid Facilitates Stem Cell-Driven Intestinal Epithelial Renewal through the Amplification of β-Catenin Signaling by Targeting Frizzled7 in Chicks.

l-Malic acid (l-MA) contributes to energy metabolism and nutrient digestion, which is an alternative to antibiotics for livestock; however, it is not clear whether l-MA can replace antibiotics to promote intestinal development in chicks. To investigate the effects of l-MA on intestinal stem cells (ISCs) driving epithelial renewal, we employed in vivo chick feeding experiments, chick intestinal organoid (IO) models, and in vitro chick intestinal epithelial cell models. The results showed that the feed conversion rate and diarrhea scores were decreased with improved jejunal morphology and barrie

Impact of low molecular weight organic acids (LMWOAs) on biochar micropores and sorption properties for sulfamethoxazole.

The interaction between biochar (BC) and antibiotics with the presence of low molecular weight organic acids (LMWOAs) is largely unknown, although it is crucial for understanding the role of BC in reducing the bioavailability of antibiotics in rhizosphere. The impacts of two typical LMWOAs (citric and malic acids) on sorption of sulfamethoxazole (SMX) by crop-straw BCs produced at 300 °C (BCs300) and 600 °C (BCs600), respectively, were examined. The sorption of SMX on BCs increased more than 5 times with the concentration of LMWOAs increasing from 0 to 100 mmol/L, which was mainly attributed t

Challenges associated with current and future TB treatment.

Current tuberculosis (TB) treatment is based on a combination of drugs that were developed mostly in the central decades of the last century. Cure rates are high for drug sensitive strains of Mycobacterium tuberculosis (M. tb) when the recommended complex and lengthy treatment protocols are adhered to. However the difficulty in correctly prescribing and adhering to these protocols, the emergence of M tb strains resistant to multiple drugs, and drug-drug interactions that interfere with optimal treatment of HIV and TB coinfected patients have generated a pressing need for improved TB therapies.

Human tear enzyme changes as indicators of the corneal response to anterior hypoxia.

A non-invasive biochemical technique for quantifying the effects of anterior corneal hypoxia on the in vivo corneal epithelium of the human eye is described. Following short-term exposure of the cornea to low atmospheric oxygen pressures, lactate dehydrogenase (LDH) and malate dehydrogenase (MDH) activities in tears are altered so that the tear LDH/MDH ratio is elevated. The degree of elevation of the ratio and its timing are related to the severity of hypoxia. Possible explanations for the elevation of the tear LDH/MDH ratio include unbinding of intracellular LDH and increased cell membrane p

Cardiac malic enzyme in Friedreich's disease.

We measured the activity of cytosolic and of mitochondrial malic enzyme in the hearts from 4 patients with Friedreich's disease and from two non-ataxic control subjects. There was a wide variability in the results and the slight overall decreases in both enzyme activities were not considered to be statistically significant. From these and other results, we conclude that deficient mitochondrial malic enzyme activity is not a constant or primary feature of Friedreich's disease.

Physiological basis for meso-tartrate sensitivity in some strains of Salmonella typhimurium.

meso-Tartrate inhibited the growth of non-meso-tartrate-utilizing strains of Salmonella typhimurium in peptone water media and mineral salts media with some, but not all, carbon sources. C-R intermediates of the tricarboxylic acid cycle or compounds readily converted to them and substrates metabolized independently of the C-6 part of the cycle spared bacteria from the inhibitory effects of meso-tartrate when added to cultures along with meso-tartrate. Experiments with cell-free extracts of non-meso-tartrate-utilizing strains from batch and continuous cultures showed that meso-tartrate was a co

[Cyanide-resistant respiration of Pseudomonas aeruginosa bacteria].

The transition of the bacterial culture into the stationary growth phase is accompanied by an appearance of cyanide-resistant respiration. Chloramphenicol inhibits the development of cyanide-resistant respiration. The cyanide-resistant oxidase is localized in the bacterial membrane. Its appearance is not due to the quantitative and qualitative changes of flavins, non-heme iron, ubiquinone and cytochromes of the b and c types, but is accompanied by an increase in the copper content of the membrane preparations. Neither cyanide-sensitive, nor cyanide-resistant chains of the bacterial electron tr

l-Malic Acid Potentiates Bifidobacterium breve Bb18 Tolerance and Synergistically Ameliorates Colitis via Gut Microbiota-Metabolite Modulation.

This study investigated l-malic acid (l-MA) as a protectant enhancing Bifidobacterium breve Bb18 (Bb18) bile tolerance and its synergistic therapeutic efficacy against DSS-induced colitis in mice. In vitro, 0.4% l-MA optimally enhanced Bb18 tolerance to glycodeoxycholic acid (GDCA) by 52.3%, preserving membrane integrity, boosting energy metabolism, and upregulating bile salt efflux genes, thus promoting a multidimensional cellular adaptive response. In vivo, the combination of Bb18 and l-MA synergistically alleviated the colon length shortening by 20.1%, reduced inflammation, and restored gut

Antisense reduction in NADP-ME in the C4 species Flaveria bidentis alters stomatal sensitivity to intercellular [CO2].

Stomata have the crucial role of balancing the uptake of CO2 for photosynthesis and water loss via transpiration by adjusting their aperture. In C3 plants, coordination between photosynthesis and stomatal conductance can be disrupted by mutations in Calvin-Benson-Bassham (CBB) cycle expression, but it is not clear whether the disruption of the C4 carbon concentrating mechanism would have similar effects. In this study, it was hypothesized that perturbing the C4 cycle, resulting in reduced carbon flux into the bundle sheath cells and overall net CO2 assimilation rates, may alter stomatal regula
