Fabrication of egg white peptides/konjac glucomannan co-assembly stabilized high internal phase Pickering emulsions: Application for co-delivery of vitamin D3 and riboflavin.

With the increasing demand for the co-encapsulation and controlled delivery of bioactive compounds, the design of liquid-based carriers has garnered considerable attention. Vitamin D3 and riboflavin sodium phosphate (RP) suffer from poor stability and low bioavailability; nevertheless, high internal phase Pickering emulsions (HIPPEs) feature a tunable microstructure that allows for effective compartmentalization of multiple components. In this study, HIPPEs stabilized by egg white peptide (EWP)-konjac glucomannan (KGM) co-assemblies were constructed as a versatile vehicle for the co-delivery o

Tuning bigel performance via emulsifier type and concentration: Structural, physicochemical, and molecular perspectives.

To develop bigels with tunable physical properties and enhanced chemical stability, this study investigated the effect of emulsifier type and concentration on the physical properties, microstructure, and oxidative stability of bigels composed of ethyl cellulose oleogel and deacetylated konjac glucomannan hydrogel. The marked improvement in bigel uniformity and dispersion observed at low concentrations (≤1 %) of glycerol monooleate (GM) and phosphatidylcholine (PC) was primarily attributed to the significant reduction in oil-water interfacial tension (from 18.10 to 12.90 mN/m). At concentration

False-positive results of galactomannan assays in patients administered glucose-containing solutions.

Galactomannan (GM) is a polysaccharide cell wall component released by Aspergillus spp., and an immunoenzymatic GM assay is used for the diagnosis of invasive pulmonary aspergillosis. We evaluated the cause of strong positivity for GM in patients with no typical signs of aspergillosis. Repeat assays were performed using different instruments and reagent lots, but there were no differences in results among the assays. Patients with strongly positive GM results were investigated. Medication histories revealed that 14 of 23 patients had been administered total parenteral nutrition solution from o

Systematic review and meta-analysis of galactomannan antigen testing in serum and bronchoalveolar lavage for the diagnosis of chronic pulmonary aspergillosis: defining a cutoff.

BACKGROUND: A clear cutoff value of galactomannan (GM) has not been established for chronic pulmonary aspergillosis (CPA) and is frequently extrapolated from invasive pulmonary aspergillosis. We performed a systematic review and meta-analysis to evaluate the diagnostic performance of serum and bronchoalveolar lavage (BAL) GM, and to propose a cutoff. METHODS: We extracted from the studies the cutoff of serum or/and BAL GM associated with true positives, false positives, true negatives, and false negatives. We performed a multi-cutoff model and a non-parametric random effect model. We estimate

Formation of complexes between hematite nanoparticles and a non-conventional galactomannan gum. Toward a better understanding on interaction processes.

The physicochemical characteristics of hematite nanoparticles related to their size, surface area and reactivity make them useful for many applications, as well as suitable models to study aggregation kinetics. For several applications (such as remediation of contaminated groundwater) it is crucial to maintain the stability of hematite nanoparticle suspensions in order to assure their arrival to the target place. The use of biopolymers has been proposed as a suitable environmentally friendly option to avoid nanoparticle aggregation and assure their stability. The aim of the present work was to

Uniqueness of Candida auris cell wall in morphogenesis, virulence, resistance, and immune evasion.

Candida auris has drawn global attention due to its alarming multidrug resistance and the emergence of pan resistant strains. C. auris poses a significant risk in nosocomial candidemia especially among immunocompromised patients. C. auris showed unique virulence characteristics associated with cell wall including cell polymorphism, adaptation, endurance on inanimate surfaces, tolerance to external conditions, and immune evasion. Notably, it possesses a distinctive cell wall composition, with an outer mannan layer shielding the inner 1,3-β glucan from immune recognition, thereby enabling immune

Expression of AtMed15 of Arabidopsis in yeast causes flocculation and increases ethanol production in yeast culture.

Mediator, a multiprotein complex involved in transcription of class II genes, was first discovered in yeast and then characterized in many metazoans revealing a striking structural conservation of the complex. However, sequences of Mediator subunits are not well conserved raising a question on the functional conservation of these individual subunits. In this study, expression of Med15 of Arabidopsis (AtMed15) in gal11∆ yeast could not complement the function of ScGal11 in galactose metabolism and resistance against cycloheximide. Surprisingly, AtMed15 changed the morphology of the yeast cells.

Distinct usage of three C-type lectins by Japanese encephalitis virus: DC-SIGN, DC-SIGNR, and LSECtin.

Infection with West Nile virus and dengue virus, two mosquito-borne flaviviruses, is enhanced by two calcium-dependent lectins: dendritic cell-specific intercellular adhesion molecule-3-grabbing nonintegrin (DC-SIGN), and its related molecule (DC-SIGNR). The present study examined the relationship between Japanese encephalitis virus (JEV) infection and three lectins: DC-SIGN, DC-SIGNR, and liver sinusoidal endothelial cell lectin (LSECtin). Expression of DC-SIGNR resulted in robust JEV proliferation in a lymphoid cell line, Daudi cells, which was otherwise non-permissive to infection. DC-SIGN

Differential gene expression of 36-kDa microfibril-associated glycoprotein (MAGP-36/MFAP4) in rat organs.

By using quantitative Western blot analysis and the real time polymerase chain reaction technique, we investigated the differential gene expression of microfibril-associated glycoprotein (MAGP-36) in rat organs. The gene was expressed highly in sites rich in elastic fibers, such as aorta, skin, and esophagus. However, MAGP-36 was also expressed highly in some other sites containing no elastic fibers. In lung and trachea, the expression levels of MAGP-36 mRNA were about seven times higher than those in other elastic tissues, although the protein abundances were almost at the same levels as othe

Recombinant human serum albumin.

Human serum albumin (HSA) is responsible for 80% of the colloid osmotic pressure of plasma (25-33 mmHg). Its main clinical use is in maintaining colloid oncotic pressure and increasing circulating plasma volume with the typical dosage in excess of 10 g per dose. HSA is isolated by fractionating human plasma, which entails possible contamination by viruses or prions. Recombinant HSA (rHSA) has been successfully produced using a methylotrophic yeast, Pichia pastoris. Due to the fact that the clinical usage of HSA infusion often exceeds 10 g, rHSA preparation requires a high level of purity. rHSA

Role of the complement-lectin pathway in anaphylactoid reaction induced with lipopolysaccharide in mice.

We show that Proteus vulgaris O25 (PO25) lipopolysaccharide (LPS) induced an anaphylactoid reaction not only in wild-type and in lipid A non-responding mice but also in recombinase-activating gene-2-deficient (RAG-2(-/-)) and in mast cell-deficient (W/Wv) animals. Western blot analysis indicated that PO25 LPS bound to Ra-reactive factor (RaRF), the complex of mannan-binding lectins (MBL) and MBL-associated serine proteases. Binding of RaRF to PO25 LPS led to the activation of C4 component without participation of either C1 or Ig, via the lectin pathway. Relative concentration of RaRF and hemol

[Non-culture based laboratory diagnosis of sepsis].

Blood culture is clearly the most important diagnostic procedure for sepsis. In the majority of cases, however, it yields negative results when bacterial or fungal sepsis is strongly suspected in view of an excellent response to antibacterial or antifungal medication. This is likely because infecting microbes have already been too seriously damaged in the blood stream to grow in culture media. Therefore, various tests have been devised to detect cellular components of pathogens including endotoxin for gram-negative bacteria, (1-->3)-beta-D-glucan for fungi, a non-mannan heat-labile candida ant

Serodiagnosis of candidiasis, aspergillosis, and cryptococcosis.

During the past decade, substantial progress has been made in the development of new approaches and methods for the serological diagnosis of the mycoses. Clinically relevant antigens have been adapted for use in immunoassays for the detection of specific antibodies, and methods that detect fungal antigens in body fluids have been progressively refined. Nevertheless, few of the novel serological techniques have as yet found widespread clinical use. Reasons for this apparent discrepancy include the paucity of clinical data that substantiate improved predictive value for disease of antibody tests

Galactomannans from Neltuma spp. as Emerging Functional Dietary Fibre for Human Nutrition: Extraction Technologies and Health Potential.

Galactomannans are reserve polysaccharides widely distributed in legume seeds and recognized for their technological and physiological properties. Their structure, consisting of a β-(1→4)-linked mannose backbone with α-(1→6)-galactose side chains, determines their solubility, viscosity, and functional behavior. In recent years, galactomannans have gained increasing attention as functional dietary fibre due to their potential effects on glycemic regulation, cholesterol reduction, gut microbiota modulation, and short-chain fatty acid production. This review critically analyzes the current knowle…
