Melatonin, a hormone monitorable in vivo by voltammetry?

Melatonin, an indoleamine hormone synthesized in the pinealocytes, is electroactive at the surface of pre-treated carbon fibre microelectrodes (mCFE) in vitro when using differential-pulse voltammetry (DPV), at the specific oxidation potential of approximately +570 mV. In vivo DPV experiments have then been performed in melatonergic regions such as the pineal gland or the suprachiasmatic nucleus (SCH) of anaesthetized adult male rats. These experiments indicated the feasibility of simultaneous measurements of the indolaminergic peak 3, which occurred at approximately +280 mV, due mainly to the…

Light, melatonin and the sleep-wake cycle.

Blood levels of the pineal hormone melatonin are high at night and low during the day. Its secretion is regulated by a rhythm-generating system located in the suprachiasmatic nucleus of the hypothalamus, which is in turn regulated by light. Melatonin is regulated not only by that circadian oscillator but acts as a darkness signal, providing feedback to the oscillator. Melatonin has both a soporific effect and an ability to entrain the sleep-wake rhythm. It also has a major role in regulating the body temperature rhythm. Melatonin rhythms are altered in a variety of circadian rhythm disorders. …

The effects of melatonin on isolation distress in chickens.

Melatonin (0.1-1.0 mg/kg) reduced isolation-induced distress vocalizations (DVs) in young domestic chickens in a dose-dependent manner. This effect was unaffected by the administration of d-amphetamine (1.0 mg/kg) suggesting that melatonin's effects were not merely due to fatigue. The melatonin reduction in DVs was not naloxone reversible, indicating an action independent of the endogenous opioid system. However, chronic pretreatment with naltrexone facilitated the melatonin effect, suggesting a complex relationship between melatonin and the endogenous opioids in regulating distress vocalizati…

Melatonin, the pineal gland, and circadian rhythms.

Amniote circadian organization derives from the interactions of circadian oscillators and photoreceptors located in the hypothalamic suprachiasmatic nuclei (SCN), the pineal gland, and the eyes. In mammals, circadian organization is dominated by the SCN, which serve as "master pacemakers" in the control of a wide array of behavioral and physiological rhythms (including locomotion, sleep-wake, thermoregulation, cardiovascular function, and many endocrine processes). Among the rhythms under SCN control in mammals are the circadian synthesis and secretion of the pineal hormone melatonin, which re…

The melatonin rhythm: both a clock and a calendar.

The paper briefly reviews the data which shows that the circadian production and secretion of melatonin by the pineal gland can impart both daily, i.e., clock, and seasonal, i.e., calendar, information to the organism. The paper summarizes the 3 patterns of nocturnal melatonin production that have been described. Clearly, regardless of the pattern of nocturnal melatonin production a particular species normally displays, the duration of nightime elevated melatonin is proportional to the duration of the night length. Since daylength under natural conditions changes daily the melatonin rhythm, wh…

Melatonin and its receptors in the gastrointestinal tract.

Melatonin and its synthesizing enzymes have been demonstrated in the gastrointestinal tract, suggesting the in situ biosynthesis of melatonin by the gut tissues. There appears to be a diurnal rhythm of melatonin in the gastrointestinal tissues of birds and rodents, with high levels in the dark period. Release of gut melatonin into the general circulation, however, was recorded following tryptophan treatment. Melatonin has direct gastrointestinal functions: it decreased serotonin-induced gut concentration, alleviated serotonin-induced reduction in gastric glandular mucosal blood flow, diminishe…

Stimulation of murine splenocytes by melatonin and methoxytryptamine.

Male C57 mice kept under a 14:10 (L:D) photoperiod received vehicle (VEH), melatonin (MEL) and methoxytryptamine (MTA) in the drinking water for 2 weeks. Splenocytes from MEL-treated mice showed an augmented mitogenic response to concanavalin A and lipopolysaccharide (LPS) while splenocytes from MTA-treated mice demonstrated an enhanced mitogenic response to LPS when compared to the VEH-treated control. Splenocytes from MEL-treated and MTA-treated mice also produced higher levels of gamma interferon and interleukin-2. Lymphokines prepared from splenocytes of MEL-treated mice stimulated periton…

Regulatory mechanisms in melatonin biosynthesis in retina.

The vertebrate retina produces melatonin in a light-dependent rhythmic fashion, synchronized with, but independent from the rhythm of the hormone formation in the pineal gland. This review summarizes the current status of our knowledge on regulatory mechanisms involved in controlling the retinal melatonin biosynthesis. Special emphasis is given to the role and mode of action of dopamine and GABA, two established retinal neurotransmitters, as well as that of second messengers (cyclic AMP, calcium ions). Comparisons are made between lower vertebrates and mammals.

Melatonin: a chronobiotic with anti-aging properties?

Recently, it has been reported that melatonin administration extends the lifespan of mice, a finding which supports previous research on the effects of pinealectomy and pineal extract administration. The prolongation of lifespan by melatonin has been interpreted in favour of an upregulation of the immune system as well as due to anti-stress properties of melatonin acting via the brain opioid system. In this paper we offer an alternative explanation of melatonin's anti-aging effect: the circadian pacemaker system has a diminished amplitude with age as indexed by a decrease in circulating melato…

Pineal melatonin in schizophrenia: a review and hypothesis.

It has long been suggested that abnormal functions of the pineal gland may be implicated in the pathophysiology of schizophrenia. We present evidence proposing that diminished melatonin secretion may be associated with the pathophysiology of a subgroup of schizophrenic patients characterized by cerebral atrophy and ventricular enlargement, negative symptoms, impaired cognitive and psychosexual development, onset at pubescence, poor response to neuroleptic medication, and possible increased risk of extrapyramidal symptoms. This view holds that a subnormal plasma melatonin level may be a marker …

Effects of melatonin on vertebrate circadian systems.

In many species of vertebrates the pineal gland and its indoleamine hormone melatonin play central roles in the control of circadian rhythms, whereas in some species, the pineal gland appears to hold little importance. However, recent research indicates that the circadian rhythms of many species of reptiles, birds and mammals, including humans, are synchronized by the administration of exogenous melatonin. These studies have led to questions concerning the role of this hormone in circadian organization in general. Studies of the sites and mechanisms of melatonin action further indicate that me…

Regulatory sites in the melatonin system of mammals.

The hormone melatonin was first identified about 30 years ago as a secretory product of the pineal gland. In mammals, the daily rhythm of pineal melatonin synthesis is controlled by neural inputs. The CNS is thought to be a primary target organ involved in mediating the influence of melatonin on a variety of physiological and behavioral processes, including biological rhythms, neuroendocrine function, activity levels and sleep. It now appears that melatonin is also produced in the retina and affects various aspects of retinal physiology. The purpose of this article is to provide a brief overvi…

The circadian rhythm of melatonin in hypothyroidism and hyperthyroidism.

UNLABELLED: In order to determine the influence of thyroid function on the pineal gland in humans, the circadian rhythm of the serum melatonin concentration was estimated in 16 women with thyroid disorders: 8 with hypothyroidism and 8 with hyperthyroidism, as well as in 5 healthy controls. A significant melatonin circadian rhythm was observed in all the three groups studied. The melatonin rhythm parameters derived from cosinor analysis: mesor (controls: 0.163 +/- 0.03 nmol/l (+/- SEM), hypothyroid patients: 0.176 +/- 0.22 nmol/l, and hyperthyroid patients: 0.167 +/- 0.04 nmol), amplitude (0.15…

Changes in melatonin secretion with age and pubescence.

At the turn of the century, reports on a coincidence of pineal tumors and precocious puberty in man initiated a host of animal experiments. Results of these experiments ultimately revealed a modulating effect on sexual maturation and on gonadal activity in several species, brought about by the pineal hormone melatonin (aMT) and particularly its circadian secretion pattern. In addition, it renewed interest in a possible interaction of the pineal and human sexual function. The introduction of specific and precise aMT assays provided exact evaluation of the circadian aMT secretion pattern in adul…

Serum Daytime Melatonin Levels Reflect Cerebrospinal Fluid Melatonin Levels in Alzheimer's Disease but Are Not Correlated with Cognitive Decline.

BACKGROUND: Nocturnal cerebrospinal fluid (CSF) and blood melatonin levels are altered in Alzheimer's disease (AD). However, literature remains inconclusive on daytime blood melatonin levels. A positive correlation between melatonin levels and Mini-Mental State Examination (MMSE) scores in AD subjects has been evidenced following cross-sectional analyses. Whereas a correlation between serum and spinal CSF melatonin has been shown in healthy volunteers, an equal investigation in AD patients still has to be undertaken. OBJECTIVE: 1) To evaluate whether serum melatonin levels correlate with spin…

Effect of melatonin on motility pattern of small intestine in rats and its inhibition by melatonin receptor antagonist S 22153.

Melatonin is synthesized during the night by the pineal gland. Recently, melatonin binding sites have been identified in the gut. Despite few studies, the physiological role of melatonin in gut function remains unclear. The objective of the present study was to investigate the effects of melatonin in the regulation of intestinal motility by using the melatonin receptor antagonist S 22153 in rats. Twenty-four male Wistar rats (400 +/- 25 g) were equipped with intraparietal electrodes along the small intestine. Rats were subjected to a 12:12 hr light:dark schedule. During the dark phase, intesti…
