The capsule of Cryptococcus neoformans is its dominant virulence factor and plays a key role in the biology of this fungus. In this essay, we focus on the capsule as a cellular structure and note the limitations inherent in the current methodologies available for its study. Given that no single method can provide the structure of the capsule, our notions of what is the cryptococcal capsule must be arrived at by synthesizing information gathered from very different methodological approaches including microscopy, polysaccharide chemistry and physical chemistry of macromolecules. The emerging pic

Activation of autophagosomes in cell death has been described since the late 1950s as a form of cell death characterized by consumption of the bulk of the cytoplasm by lysosomal derivatives. However, it is not yet established that autophagy is a primary, causative mechanism of death rather than a response to initial problems. Methods to assess autophagic cell death are similar to those used to detect and measure autophagy, with further evidence that the affected cells are indeed dying. These methods include structural analysis using electron microscopy, examination of the activity of lysosomal

While it has been well demonstrated that quantum dots (QDs) play an important role in biological labeling both in vitro and in vivo, there is no report describing the cellular nanostructure basis of receptor-mediated endocytosis. Here, nanostructure evolution responses to the endocytosis of transferrin (Tf)-conjugated QDs were characterized by atomic force microscopy (AFM). AFM-based nanostructure analysis demonstrated that the Tf-conjugated QDs were specifically and tightly bound to the cell receptors and the nanostructure evolution is highly correlated with the cell membrane receptor-mediate

Annexin I is an abundant protein in U937 cells differentiated towards a macrophagic phenotype. These cells become able to kill Escherichia coli, however, the intracellular pathogen Brucella suis, known to interfere with phagosome maturation, multiply in these differentiated cells. We have analysed by confocal and electron microscopy the cellular localization of annexin I during phagocytosis of yeast, non-pathogenic E. coli and the intracellular pathogen B. suis. Using immunocytochemical detections annexin I was found mainly as patches in the cytoplasm of uninfected cells. Upon phagocytosis of

Acute myeloid leukemia (AML) has a poor prognosis and is associated with aberrant NF-κB activation, providing a tumor-selective therapeutic window. Therefore, a combination therapy involving FDA-approved ferumoxytol (Feraheme, FeNPs) and NF-κB-responsive GBA interference was developed in this study. GBA is a key enzyme that maintains lysosomal lipid metabolism and membrane composition homeostasis. Its inhibition causes lysosomal storage disorders and functional impairment, leading to the release of labile iron that contributes mainly to ferroptosis in AML. Using a miR30-based shRNA system, we

Salmonella Enteritidis, a major foodborne bacterial pathogen commonly linked to poultry products, necessitates effective control measures. This study investigated the antibacterial activity and underlying mechanism of lauroyl arginate ethyl (LAE) against S. Enteritidis by integrating physiological, proteomic, and functional analyses. LAE exhibited potent concentration- and time-dependent antibacterial effects in vitro. In chicken meat, LAE at 0.2 and 0.4 mg/mL completely inactivated S. Enteritidis within 30 min. Scanning electron microscopy and selective recovery assays demonstrated that LAE c

To date, numerous studies have examined the etiological relationship between vacuoles and DNA fragmentation in human sperm, with vacuoles typically assessed using differential interference contrast microscopy. To explore the optimal dye for visualizing vacuoles, we collected 12 commercially available reactive dyes and compared their staining performance. Labeling protamines with 9.0 μmol/L Reactive Black-5 (RB5) stained the nuclear body bluish and visualized vacuoles as unstained spots. Next, we synthesized 30 molecularly designed RB5 derivatives to identify an optimal molecular structure for

Like other invertebrates, Litopenaeus vannamei lacks adaptive immunity and relies mainly on innate immunity for defense against foreign pathogens. In this study, three distinct Reeler domain-containing molecules were discovered in L. vannamei, designated as LvReeler1, LvReeler2, and LvReeler3. Analysis of tissue-specific expression patterns indicated that LvReeler1 showed predominant expression in the stomach, whereas LvReeler2 and LvReeler3 demonstrated peak transcriptional activity within gill tissues. The expression of these molecules was induced by Vibrio parahaemolyticus. In vivo interfer

Superparamagnetic iron oxide nanoparticles (SPIONs) with tailored surface modifications were employed to fabricate ordered thin films through a drop-casting technique. By systematically varying the ligand chain length using stearic acid, decanoic acid, and hexanoic acid, we precisely controlled the interparticle distances within the films. Comprehensive investigations utilizing superconducting quantum interference device (SQUID) magnetometry elucidated the films' superparamagnetic behaviour at room temperature, as well as notable exchange interactions at lower temperatures. Notably, these exch

A new variety of nitrogen-doped carbon dots (NCDs) was produced using a hydrothermal synthesis method, based on propanedioic acid and barbituric acid as the sources of carbon and nitrogen. The NCDs were analyzed by X-ray photoelectron spectroscopy (XPS), transmission electron microscopy (TEM), Zeta Potential,X-ray Diffraction(XRD),Thermogravimetry-Derivative Thermogravimetry(TG-DTG),Fourier transform infrared spectroscopy (FTIR) and Fluorescence Lifetime. The characterization results indicate that NCDs possess an average diameter of approximately 2.35 nm, and blue fluorescence is visualized un

A universal method for creating selective hydrogen (H2) gas sensors through the integration of microporous zeolitic imidazolate framework (ZIF) filter layers on metal oxide sensing layers is presented. The sensor design consists of an indium-modified tin oxide (In-SnO2) layer as the gas-sensitive component, topped by a size-selective ZIF filter layer. The ZIF layer is generated by first depositing zinc oxide (ZnO) of variable thickness (20-48 nm) onto the In-SnO2 layer, followed by in situ conversion to either ZIF-8 or ZIF-71 through solvothermal methods. The resulting bilayer structures are c

The combined use of immunolabeling and confocal microscopy offers a robust tool for the comprehensive examination of red blood cells, allowing clear visualization of specific proteins and their roles in the cell. While immunolabeling is a widely employed technique, it presents specific challenges in the study of red blood cells, including difficulties with membrane permeabilization, the appearance of background fluorescence, detection of very faint signals, interference from other blood components, and, notably, poor preservation resulting from an incorrect choice of fixation components. Human syncytin-1 and suppressyn are cellular proteins of retroviral origin involved in cell-cell fusion events to establish the maternal-fetal interface in the placenta. In cell culture, they restrict infections from members of the largest interference group of vertebrate retroviruses, and are regarded as host immunity factors expressed during development. At the core of the syncytin-1 and suppressyn functions are poorly understood mechanisms to recognize a common cellular receptor, the membrane transporter ASCT2. Here, we present cryo-electron microscopy structures of human ASCT2 in complexes

Microplastics and nanoplastics have become noteworthy contaminants, affecting not only outdoor ecosystems but also making a notable impact within indoor environments. The release of microplastics and nanoplastics from commonly used plastic items remains a concern, and the characterisation of these contaminants is still challenging. This study focused on evaluating the microplastics and nanoplastics produced from plastic building bricks. Using Raman spectroscopy and correlation analysis, the plastic material used to manufacture building blocks was determined to be either acrylonitrile butadiene

One of the key tenets of sustainable agriculture and food safety is the removal of toxic pesticides from the environment. However, developing reliable, affordable, and efficient methods for detecting and degrading pesticides into non-toxic degradable products remains an immediate matter of concern. Herein, we attempt to develop a strategy for the detection as well as degradation of highly toxic phosphorodithioate pesticide, Azinphos methyl (AZM), using hybrid zinc oxide nanoparticles (ZnO NPs). Considering the non-selectivity of bare ZnO and receptor R1, we have fabricated the heterocalixarene

T and B cells continually recirculate between blood and secondary lymphoid organs. To promote their trans-endothelial migration (TEM), chemokine receptors control the activity of RHO family small GTPases in part via GTPase-activating proteins (GAPs). T and B cells express several RHO-GAPs, the function of most of which remains unknown. The ARHGAP45 GAP is predominantly expressed in hematopoietic cells. To define its in vivo function, we describe two mouse models where ARHGAP45 is ablated systemically or selectively in T cells. We combine their analysis with affinity purification coupled to mas

Uniform-sized iron oxide nanoparticles obtained from the solution phase thermal decomposition of the iron-oleate complex were encapsulated inside the silica shell by the reverse microemulsion technique, and then thermal treatment under NH3 to transfer the iron oxide to iron nitride. The transmission electron microscopy images distinctly demonstrated that the as-prepared iron nitride at silica core/shell nanostructures were highly uniform in particle-size distribution. By using iron oxide nanoparticles of 6.1, 10.3, 16.2, and 21.8 nm as starting materials, iron nitride nanoparticles with averag

A graphene modified glassy carbon (GR/GCE) sensor has been developed for the determination of aspirin metabolites 2,3- and 2,5-dihydroxybenzoic acids (2,3- and 2,5-DHB). The modified sensor was characterized by Field Emission Scanning Electron Microscopy and Electrochemical Impedance Spectroscopy. The electrochemical behavior of 2,3- and 2,5-DHB was investigated by cyclic and square wave voltammetry. The modified sensor exhibited excellent electrocatalytic activity for the oxidation of 2,3- and 2,5-DHB, leading to a remarkable enhancement in the peak current as compared to the bare sensor. The

In this contribution, a simple and sensitive method for papain detection was established based on the increment of the resonance light-scattering (RLS) intensity of mercaptoacetic acid-capped CdSe quantum dots (MAA-QDs) in aqueous solution. Meanwhile, the RLS characteristics and the optimal conditions were investigated in detail. Under the optimized conditions, the linear range of MAA-QDs RLS intensity versus the concentration of papain was 1.0×10(-8) M to 6.0×10(-7) M, with a correlation coefficient (R(2)) of 0.9977 and a limit of detection (3σ black) of 5.1×10(-9) M. The relative standard de

This paper described the investigation of surface-modified quantum dots (QDs) as a fluorescence probe for the detection of cardiolipin. A single-step method for preparation of non-toxic and photo-stable cadmium telluride (CdTe) QDs capped by L-cysteine in aqueous solution was developed. The prepared QDs were characterized by high-resolution transmission electron microscopy, X-ray diffraction spectrometry, Fourier transform infrared spectrometry and spectrofluorometry. These functional QDs were used as a fluorescence probe for cardiolipin determination based on the fluorescence quenching. The o
