Disentangled diffusion model for 3D molecular generation with protein-ligand interaction priors.

MOTIVATION: Structure-based drug design (SBDD) aims to generate ligand molecules that tightly bind to specific protein targets, a critical step in drug discovery. Diffusion models have shown promise for this task, yet existing methods struggle to effectively incorporate protein-ligand interaction priors during generation. Most approaches rely on protein-specific structural priors that remain fixed throughout generation, limiting molecular diversity and failing to capture the dynamic interplay between protein pockets and ligand atoms, which is essential for achieving high binding affinity. The Effects of Early Postnatal Alcohol Exposure on Bone Molecular Composition in a Mouse Model.

BACKGROUND: Perinatal exposure to alcohol is a critical risk factor for long-term skeletal health in humans and has been demonstrated in rodent models. Early postnatal alcohol gavage exposure in mice is used to study third trimester toxicity in humans. However, further research is required to fully characterize the effects of alcohol on bone molecular composition and distinguish them from the alterations associated with gavage-related stress in mouse models. METHODS: Female mouse pups were assigned to untreated control (C), gavage control (GC), and alcohol-treated (A) groups. Between postnata

Synthesis, Molecular Modeling and Assessment of Anticancer Activity of New Potential CYP17A1 Inhibitors.

Castration-resistant prostate cancer (CRPC) remains a significant clinical challenge due to the ability of tumor cells to undergo intratumoral androgen synthesis, a process catalyzed by the CYP17A1 enzyme. The only CYP17A1 inhibitor available in therapy, abiraterone acetate, faces significant limitations due to its steroidal structure, which causes off-target effects and generates agonistic metabolites that paradoxically stimulate the androgen receptor (AR). This study presents the development of the D2AAK1M series, a novel class of non-steroidal potential CYP17A1 inhibitors based on a pyridin

Curcumin-loaded beta-glucan nanoparticles: preparation, physicochemical properties, and molecular modeling.

OBJECTIVES: This study introduces the application of wet milling, a well-established top-down technique, as a novel approach for preparing beta-glucan nanoparticles (BG-NPs) as carriers for poorly soluble drugs, using curcumin (CC) as a model compound. This presents one of the first studies illustrating wet milling as a feasible way to produce BG-NPs with enhanced drug delivery capabilities, potentially improving the bioavailability of CC in particular and other poorly soluble drugs in general. METHODS: A two-step process was employed: first, optimizing beta-glucan (BG) nanocarrier preparatio

Zebrafish Model Reveals Early Electrocardiographic and Molecular Signatures of Doxorubicin-Induced Cardiotoxicity.

Doxorubicin (DOX) is a highly effective anthracycline widely used in cancer therapy but limited by its dose-dependent cardiotoxicity, which may result in arrhythmia, dilated cardiomyopathy, and heart failure. Conventional surveillance tools, including echocardiography and serum biomarkers, often identify injury only after substantial cardiac dysfunction has occurred. This underscores the need for early markers with mechanistic relevance. In this study, we developed an integrated zebrafish platform combining pathophysiological evaluation, electrocardiography (ECG), and transcriptomic profiling

Modeling Liver Cancer Molecular and Histological Features by CRISPR Editing of Porcine Hepatocytes.

BACKGROUND: /Aims: Hepatocellular carcinoma (HCC) is an aggressive disease with limited response to available therapies. Progress in therapeutic development has been hampered by the lack of preclinical models that recapitulate human HCC and support evaluation of multimodal therapies. Here, we employ CRISPR editing to identify alterations that drive hepatocyte transformation and to establish genetically defined HCC models. METHODS: Hepatocytes were isolated from pigs (n=7) and subjected to combinations of 2-4 gene alterations, including CRISPR-mediated knockout of tumor suppressor genes (TP53,

Accelerated Chlorination at the Air-Organic Interface Revealed by Molecular Simulations and Kinetic Modeling.

An interface is a distinct chemical environment where reactivity can proceed differently than the bulk. Chemical reactions can speed up at air-water interfaces due to the rapidly varying electrostatic environment between highly polar liquid and nonpolar gas phases. Here, we show that reaction acceleration can also occur at nonpolar air-organic interfaces, which are prototypical chemical environments important for understanding atmospheric aerosols, industrial membranes, and biological cells. We examine the uptake dynamics of chlorine gas at the air/squalene interface to better interpret the an

Molecular Pathophysiology of Parathyroid Tumorigenesis-The Lesson from a Rare Disease: The "MEN1 Model".

Primary hyperparathyroidism represents the third most prevalent endocrine disease in the general population, consisting of an excessive secretion of parathyroid hormone from one or, more frequently, more of the parathyroid glands, leading to a dysregulation of calcium homeostasis. Schematically, its development occurs primarily by pathophysiological events with genetic mutation, at the germline and/or somatic level, that favor the neoplastic transformation of parathyroid cells and promote their aberrant proliferation, and mutations determining the shift in the PTH "set-point", thus interfering

PED: a novel predictor-encoder-decoder model for Alzheimer drug molecular generation.

Alzheimer's disease (AD) is a gradually advancing neurodegenerative disorder characterized by a concealed onset. Acetylcholinesterase (AChE) is an efficient hydrolase that catalyzes the hydrolysis of acetylcholine (ACh), which regulates the concentration of ACh at synapses and then terminates ACh-mediated neurotransmission. There are inhibitors to inhibit the activity of AChE currently, but its side effects are inevitable. In various application fields where Al have gained prominence, neural network-based models for molecular design have recently emerged and demonstrate encouraging outcomes. H
