Time course of muscle atrophy and recovery following a phenol-induced nerve block.

Clinically, phenol is used often as a neurolytic agent to treat pain and spasticity. The purpose of this study was to examine the time course of denervation and recovery in several hindlimb muscles following application of a 5% aqueous solution of phenol to the sciatic nerve. Phenol was applied to the sciatic nerve of adult female rats either by intraneural or perineural injection. Axonal degeneration was evident within the sciatic nerve 2 days following phenol application, although variable amounts of damage were observed. By 2 weeks, the soleus and tibialis anterior had atrophied to 63% and

Muscle Expression of SOD1G93A Triggers the Dismantlement of Neuromuscular Junction via PKC-Theta.

AIM: Neuromuscular junction (NMJ) represents the morphofunctional interface between muscle and nerve. Several chronic pathologies such as aging and neurodegenerative diseases, including muscular dystrophy and amyotrophic lateral sclerosis, display altered NMJ and functional denervation. However, the triggers and the molecular mechanisms underlying the dismantlement of NMJ remain unclear. RESULTS: Here we provide evidence that perturbation in redox signaling cascades, induced by muscle-specific accumulation of mutant SOD1G93A in transgenic MLC/SOD1G93A mice, is causally linked to morphological

Electrical stimulation of embryonic neurons for 1 hour improves axon regeneration and the number of reinnervated muscles that function.

Motoneuron death after spinal cord injury or disease results in muscle denervation, atrophy, and paralysis. We have previously transplanted embryonic ventral spinal cord cells into the peripheral nerve to reinnervate denervated muscles and to reduce muscle atrophy, but reinnervation was incomplete. Here, our aim was to determine whether brief electrical stimulation of embryonic neurons in the peripheralnerve changes motoneuron survival, axon regeneration, and muscle reinnervation and function because neural depolarization is crucial for embryonic neuron survival and may promote activity-depend…

Surgical Treatment Strategies for Injuries of the "Unforgiving" Superficial Branch of the Radial Nerve.

BACKGROUND: Neuropathic pain caused by adhesions or neuroma formation of the superficial branch of the radial nerve (SBRN) is difficult to treat. The authors evaluated the effectiveness of different routinely used surgical techniques for SBRN neuralgia to provide a basis for future studies on SBRN neuralgia and explored the prognostic value of a preoperative diagnostic nerve block. METHODS: The authors performed a retrospective cohort study by reviewing surgical records for procedures to treat SBRN neuralgia. Patient satisfaction was scored as satisfied or unsatisfied, and pain intensity was

Motor nerve transplantation.

The motor nerve transplantation (MNT) technique is used to transfer an intact nerve into a denervated muscle by harvesting a neurovascular pedicle of muscle containing motor endplates from the motor endplate zone of a donor muscle and implanting it into a denervated muscle. Thirty-six adult New Zealand White rabbits underwent reinnervation of the left long peroneal (LP) muscle (fast twitch) with a motor nerve graft from the soleus muscle (slow twitch). The right LP muscle served as a control. Reinnervation was assessed using microstimulatory single-fiber electromyography (SFEMG), alterations i

Activity-driven synaptic and axonal degeneration in canine motor neuron disease.

The role of neuronal activity in the pathogenesis of neurodegenerative disease is largely unknown. In this study, we examined the effects of increasing motor neuron activity on the pathogenesis of a canine version of inherited motor neuron disease (hereditary canine spinal muscular atrophy). Activity of motor neurons innervating the ankle extensor muscle medial gastrocnemius (MG) was increased by denervating close synergist muscles. In affected animals, 4 wk of synergist denervation accelerated loss of motor-unit function relative to control muscles and decreased motor axon conduction velociti

Determining C5, C6 and C7 myotomes through comparative analyses of clinical, MRI and EMG findings in cervical radiculopathy.

OBJECTIVE: There are many myotome charts in the literature, but few studies have presented actual data to support their identification. We aimed to determine C5/C6/C7 myotomes based on clinical and EMG data of patients with cervical spondylotic radiculopathy (CSR) having a single-root lesion confirmed by MRI. METHODS: Medical Research Council (MRC) scores and EMG findings were retrospectively reviewed for patients enrolled from our EMG database. RESULTS: Enrolled were 25 patients (10 C5, 6 C6, and 9 C7 CSR). In C5 CSR, weakness or denervation potentials in EMG, or both, were observed in the

Circulatory responses to acute asphyxia in intact and chemodenervated fetal sheep near term.

The study concerned the role of the carotid sinus nerves in the effects of acute asphyxia on the fetal circulation. Fetal sheep (n = 12) were instrumented at approximately 130 days' gestation for placement of fetal vascular catheters and an occluder around the maternal descending aorta below the renal arteries. In 7 fetuses the carotid sinus nerves were cut. On the third post-operative day, asphyxia was produced by occluding the maternal aorta for 2 min and fetal blood flows were determined by the radio-labelled microsphere technique. In control, there were no differences between intact and de

MR patterns of denervation around the shoulder.

OBJECTIVE: The diagnosis of denervation injury as a cause of shoulder pain is conventionally based on clinical findings and electrophysiologic studies. MRI has an important role in identifying direct and indirect signs of neuropathy and can confirm the presence of nerve compression, depict space-occupying lesions, and exclude other intrinsic lesions of the shoulder. In this article, the relevant anatomy, causes, clinical features, and MR appearances of nerve injury and muscle denervation of the shoulder girdle are presented. CONCLUSION: MRI is commonly performed in patients with shoulder pain…

Nerve injury affects the capillary supply in rat slow and fast muscles differently.

The goal of this study was to determine the acute effects of permanent denervation on the length density of the capillary network in rat slow soleus (SOL) and fast extensor digitorum longus (EDL) muscles and the effect of short-lasting reinnervation in slow muscle only. Denervation was performed by cutting the sciatic nerve. Both muscles were excised 2 weeks later. Reinnervation was studied 4 weeks after nerve crush in SOL muscle only. Capillaries and muscle fibres were visualised by triple immunofluorescent staining with antibodies against CD31 and laminin and with fluorescein-labelled Griffo

Electromyographic recording and muscle biopsy in lepromatous leprosy.

The aim of this study was to detect early muscular changes in lepromatous leprosy using simultaneously electromyography and muscle biopsy. In 13 subjects a single clinically normal muscle innervated by the popliteal nerve was studied. Three were found to be normal. All the others were electromyographically denervated. Histopathologic findings included only 3 cases of fascicular atrophy. In 8 cases inflammatory nodules were observed in the connective tissue of the muscle and acid-fast bacilli were present in Virchow cells in 5. In only one patient were intact acid-fast bacilli found in muscle c

Configuration of the glottis in laryngeal paralysis. II: Animal experiments.

It is widely believed that in isolated recurrent laryngeal nerve paralysis, the paralyzed vocal fold assumes a median or paramedian position, due to the action of the cricothyroid muscle. A review of the literature reveals that support for this theory is not conclusive and, in particular, experiments indicate that the cricothyroid muscle does not appreciably affect vocal fold position in acute paralysis. The research in this study compares the configuration of the glottis in chronic unilateral recurrent laryngeal nerve paralysis in cats, with and without concomitant denervation of the cricothy

Surgical management of V-pattern strabismus and oblique dysfunction in craniofacial dysostosis.

INTRODUCTION: Strabismus affects as many as 60% to 70% of patients with craniofacial dysostosis. V-pattern strabismus with severe oblique muscle dysfunction is the most common ocular motility problem seen and can be difficult to manage. Few studies have reported on the results of strabismus surgery in this condition. METHODS: We retrospectively reviewed the surgical management and outcomes of 14 patients with craniofacial dysostosis who underwent 16 operations to determine the optimal surgical procedure and to report on extraocular muscle anomalies noted at the time of surgery. Operations per

Partial denervation of ankle extensors prior to spinalization in cats impacts the expression of locomotion and the phasic modulation of reflexes.

Following peripheral nerve sections some locomotor deficits appear which are gradually compensated for by spinal and supraspinal mechanisms. The present work is aimed at identifying contributions of both types of mechanisms. We performed a denervation of the left lateral gastrocnemius-soleus (LGS) muscles in three cats which was followed by a spinalization at the 13th thoracic segment. Three other cats were not denervated prior to spinalization (i.e. intact) and served as controls. Over the years, in our laboratory, there have been no instances in which cats did not express spinal locomotion w

From the Padua Muscle Days, the Basic and Applied Myology and the European Journal of Translational Myology to the A&CM Carraro Foundation for Translational Myology.

As a young researched I had the option to work on skeletal muscle at the University of Padova, Italy. Introduced to the study of muscle denervation/reinnervation, I started a project on long term denervated muscle that still is my primary interest and took me from rodents' models of chronic muscle denervation to human spinal cord injury-related muscle denervation and its managements. On the way, I organized a series of conferences in Euganei Hills, Padua, Italy and an international journal, the Basic and Applied Myology. From 2010 this journal changed name to European Journal of Translational …

Altered expression of ARPP protein in skeletal muscles of patients with muscular dystrophy, congenital myopathy and spinal muscular atrophy.

OBJECTIVES: Ankyrin-repeated protein with PEST and a proline-rich region (ARPP) is a recently identified protein with 4 ankyrin-repeated motifs in its central portion. Type 1 myofibers of skeletal muscle express high levels of ARPP. Recently, we have found that ARPP expression was induced in mouse denervated skeletal muscle. This led us to hypothesize that ARPP expression might be induced in skeletal muscle under some pathological conditions. METHODS: In this study, we performed immunohistochemical analysis of ARPP expression in biopsy specimens of muscle tissue from 15 patients with muscular

The other half of the sprinting coin: the neuromuscular profile of a 91-yr-old female world record sprinter.

A 91-yr-old female set the W90+ world record in the 200 m sprint in 2024. We characterized her neuromuscular function, fatigability, denervation markers, and single-fiber contractile properties, and, where possible, compared these outcomes with published reference data. Knee extensor muscle architecture, force, power, fatigability, and motor unit (MU) behavior were assessed in vivo, and a vastus lateralis biopsy was analyzed for single-fiber contractile properties and markers of denervation. Quadriceps cross-sectional area was 36.5 cm2, with fascicle length, pennation angle, and muscle thickne
