Cardiovascular and renal interactions between cyclosporine and NSAIDs: Underlying mechanisms and clinical relevance.

Cyclosporine, the prototype calcineurin inhibitor, transformed immunosuppressant regimens and practices post-organ transplantation. Therapeutic uses of cyclosporine branched out to include management of different autoimmune disorders. However, multiple additional effects posed significant clinical challenges in face of the prolonged nature of cyclosporine use. Significantly, cyclosporine produced nephrotoxic, cardiotoxic and neurotoxic effects in addition to alteration of hemodynamic function. These adverse effects are shared with other drug groups further complicating the therapeutic situatio

Aquaporin 1-mediated changes in pulmonary arterial smooth muscle cell migration and proliferation involve β-catenin.

Exposure to hypoxia induces migration and proliferation of pulmonary arterial smooth muscle cells (PASMCs), leading to vascular remodeling and contributing to the development of hypoxic pulmonary hypertension. The mechanisms controlling PASMC growth and motility are incompletely understood, although aquaporin 1 (AQP1) plays an important role. In tumor, kidney, and stem cells, AQP1 has been shown to interact with β-catenin, a dual function protein that activates the transcription of crucial target genes (i.e., c-Myc and cyclin D1) related to cell migration and proliferation. Thus the goal of th

Inflammatory micro-environmental cues of human atherothrombotic arteries confer to vascular smooth muscle cells the capacity to trigger lymphoid neogenesis.

BACKGROUND: Experimental atherosclerosis is characterized by the formation of tertiary lymphoid structures (TLOs) within the adventitial layer, which involves the chemokine-expressing aortic smooth muscle cells (SMCs). TLOs have also been described around human atherothrombotic arteries but the mechanisms of their formation remain poorly investigated. Herein, we tested whether human vascular SMCs play the role of chemokine-expressing cells that would trigger the formation of TLOs in atherothrombotic arteries. RESULTS: We first characterized, by flow cytometry and immunofluorescence analysis,

Interleukin-17A promotes functional activation of systemic sclerosis patient-derived dermal vascular smooth muscle cells by extracellular-regulated protein kinases signalling pathway.

INTRODUCTION: Dermal vascular smooth muscle cells (DVSMCs) are important for vascular wall fibrosis in microangiopathy of systemic sclerosis (SSc). T helper 17 cell-associated cytokines, particularly interleukin-17A (IL-17A), have been demonstrated to play a role in the pathogenesis of SSc. However, the effect of IL-17A on the DVSMCs in microangiopathy of SSc has not been established. In the present study, we investigated the effect of IL-17A on the SSc patient-derived DVSMCs. METHODS: DVSMCs from patients with SSc and healthy subjects were incubated using IL-17A or serum derived from patient

Pathophysiology of voltage-gated K+ channels in vascular smooth muscle cells: modulation by protein kinases.

In this review, the pathological alteration and clinical relevance of voltage-gated K(+) (Kv) channels and their specific regulation by protein kinase-dependent signaling in vascular smooth muscle cells are described, particularly focusing on the pulmonary vasculature. The physiological relevance, channel characteristics, pharmacological modulation, and expression of Kv channels vary between different arterial beds and between subdivisions of arteries within those vascular beds. Although detailed signaling cascades regulating Kv channels are not clearly elucidated, it is known that the Kv chan

[Reactive transformation of human urinary smooth muscular tissue in some diseases (structural-metabolic aspects of organization)].

Smooth muscular tissue (SMT) of human urinary system was studied at different levels (renal pelvis, proximal and distal parts of the ureter) in health and some urological diseases (vesicoureteral reflux, ureteral obstruction). The method of target cell dissociation was used. The volume of myocytes, nuclei, nuclear-cytoplasmic correlation were calculated. Content of DNA and cytoplasmic protein was studied cytospectrophotometrically. Electron-microscopic study was also made. Three types of myocytes with different structural-metabolic parameters were detected. There were significant differences i

Immunolocalization of a Na-K-2Cl cotransporter in human tracheobronchial smooth muscle.

Inhibition of the Na-K-2Cl (NKCC) cotransporter by loop diuretics is associated with airway relaxation, but there has been no direct evidence for the expression of this protein in airway smooth muscle. Thus we hypothesized that a NKCC cotransporter is present and functional in airway smooth muscle cells. Monoclonal and polyclonal antibodies were used first to demonstrate the presence of a NKCC cotransporter protein in isolated human fetal trachea and normal human bronchial smooth muscle cells (BSMC) by Western blotting. The cotransporter protein was then localized by immunohistochemical staini

Hsa_circ_0044097 Serves as a Promising Biomarker of Atherosclerosis and Its Effects on Vascular Smooth Cell Proliferation and Migration.

Atherosclerosis (AS) is closely related to the pathogenesis and abnormal proliferation and migration of vascular smooth muscle cells (VSMCs). To explore the value of hsa_circ_0044097 in the clinical management of AS and to elucidate its role and mechanism in the injury of VSMCs induced by in vitro oxidized low-density lipoprotein (ox-LDL). The mRNA expressions of hsa_circ_0044097 and miR-3918 were determined using RT-qPCR, and transfection verification was also conducted. The content of IL-6 and TNF-α was determined using an ELISA kit. CCK-8 and transwell assays were performed to determine the

GLS1 governs vascular smooth muscle cell phenotypic switching and aortic dissection via glutamate metabolism.

Aortic dissection (AD) is a catastrophic vascular emergency with high mortality, and current pharmacological interventions to prevent its progression are limited. Vascular smooth muscle cells (VSMCs) undergo a pathological phenotypic switch from a contractile to a synthetic state during AD, compromising aortic wall integrity; however, the underlying metabolic mechanisms remain poorly understood. In this study, we performed integrative transcriptomic analyses and identified glutaminase 1 (GLS1) as a key regulator of VSMC phenotypic switching in AD. GLS1 expression was significantly downregulate

Duloxetine inhibits voltage-dependent K+ channels in rabbit coronary arterial smooth muscle: Electrophysiological evidence and mechanistic insights.

Duloxetine, a serotonin-norepinephrine reuptake inhibitor, has been associated with increased blood pressure and other vascular side effects. However, the mechanisms underlying these effects remain poorly understood. Voltage-dependent K+ (Kv) channels are key regulators of membrane potential and vascular tone in arterial smooth muscle cells. Therefore, we examined the effects of duloxetine on Kv channels in rabbit coronary arterial smooth muscle cells using the whole-cell patch-clamp technique. Duloxetine inhibited arterial Kv currents in a concentration-dependent manner, with an IC50 of 4.95

Clearance of senescent vascular smooth muscle cells retards aging-related restenosis following bioresorbable scaffolds implantation.

In contrast to bioinert metal stents, the degradation of bioresorbable scaffolds (BRS) induces complex mechanical changes and accumulation of degradation products, potentially leading to adverse events following implantation into stenotic arteries. Atherosclerosis (AS) is a typical age-related disease, plaque formation and changes in vascular mechanical properties can significantly affect the process of restenosis and vascular repair after BRS implantation. The aging of vascular smooth muscle cells (VSMCs) is earlier than that of endothelial cells (ECs) and plays a decisive role in the mechani

Differential effects of traditional cigarette, electronic cigarette, and tobacco heating product on human vascular smooth muscle cell phenotypic switch: A comparative functional perspective.

BACKGROUND AND AIMS: Vascular smooth muscle cell (SMC) phenotypic switching, from a contractile to a synthetic state, impacts on atherosclerotic plaque development. While traditional cigarette (TC) smoke is a known driver of cardiovascular disease, the effects of alternative nicotine delivery systems, i.e. electronic cigarettes (E-cigs) and tobacco heating products (THPs), on SMC behavior remains poorly understood. We evaluated the effects of water-soluble components collected in aqueous extracts (AEs) from TC, E-cig, and THP smoke, on human aortic SMC (HASMC) phenotypic switch. METHODS: HASM

p53 Regulates Mitochondrial Dynamics in Vascular Smooth Muscle Cell Calcification.

Arterial calcification is an important characteristic of cardiovascular disease. It has key parallels with skeletal mineralization; however, the underlying cellular mechanisms responsible are not fully understood. Mitochondrial dynamics regulate both bone and vascular function. In this study, we therefore examined mitochondrial function in vascular smooth muscle cell (VSMC) calcification. Phosphate (Pi)-induced VSMC calcification was associated with elongated mitochondria (1.6-fold increase, p < 0.001), increased mitochondrial reactive oxygen species (ROS) production (1.83-fold increase, p < 0

Drebrin attenuates atherosclerosis by limiting smooth muscle cell transdifferentiation.

AIMS: The F-actin-binding protein Drebrin inhibits smooth muscle cell (SMC) migration, proliferation, and pro-inflammatory signalling. Therefore, we tested the hypothesis that Drebrin constrains atherosclerosis. METHODS AND RESULTS: SM22-Cre+/Dbnflox/flox/Ldlr-/- (SMC-Dbn-/-/Ldlr-/-) and control mice (SM22-Cre+/Ldlr-/-, Dbnflox/flox/Ldlr-/-, and Ldlr-/-) were fed a western diet for 14-20 weeks. Brachiocephalic arteries of SMC-Dbn -/-/Ldlr-/- mice exhibited 1.5- or 1.8-fold greater cross-sectional lesion area than control mice at 14 or 20 weeks, respectively. Aortic atherosclerotic lesion surf

Vascular Smooth Muscle Cell Subpopulations and Neointimal Formation in Mouse Models of Elastin Insufficiency.

OBJECTIVE: Using a mouse model of Eln (elastin) insufficiency that spontaneously develops neointima in the ascending aorta, we sought to understand the origin and phenotypic heterogeneity of smooth muscle cells (SMCs) contributing to intimal hyperplasia. We were also interested in exploring how vascular cells adapt to the absence of Eln. Approach and Results: We used single-cell sequencing together with lineage-specific cell labeling to identify neointimal cell populations in a noninjury, genetic model of neointimal formation. Inactivating Eln production in vascular SMCs results in rapid intim

Induced osteogenic differentiation of human smooth muscle cells as a model of vascular calcification.

Vascular calcification is a severe pathological event in the manifestation of atherosclerosis. Pathogenic triggers mediating osteogenic differentiation of arterial smooth muscle cells (SMC) in humans remain insufficiently understood and are to a large extent investigated in animal models or cells derived thereof. Here, we describe an in vitro model based on SMC derived from healthy and diseased humans that allows to comprehensively investigate vascular calcification mechanisms. Comparing the impact of the commonly used SMC culture media VascuLife, DMEM, and M199, cells were characterised by im

AIM2 regulates vascular smooth muscle cell migration in atherosclerosis.

BACKGROUND: Atherosclerosis (AS) is a common pathological basis of various cardiovascular and cerebrovascular diseases. Plaque formation is initiated and triggered by vascular smooth musclecells (VSMCs) migration in vascular wall, which gradually aggravates atherosclerosis progression. Absent in melanoma 2 (AIM2), a member of HIN-200 family, plays an important role in activating inflammasome. However, the role of AIM2 in atherosclerotic plaque progression outside of the inflammasome has not yet been reported. METHODS: The potential effect and the underlying mechanism of AIM2 were investigated

Myostatin, a profibrotic factor and the main inhibitor of striated muscle mass, is present in the penile and vascular smooth muscle.

Myostatin is present in striated myofibers but, except for myometrial cells, has not been reported within smooth muscle cells (SMC). We investigated in the rat whether myostatin is present in SMC within the penis and the vascular wall and, if so, whether it is transcriptionally expressed and associated with the loss of corporal SMC occurring in certain forms of erectile dysfunction (ED). Myostatin protein was detected by immunohistochemistry/fluorescence and western blots in the perineal striated muscles, and also in the SMC of the penile corpora, arteries and veins, and aorta. Myostatin was f
