Cochlear Amyloid-β42 Accumulation Drives Progressive Auditory Neuropathy in 5XFAD Mice: A Potential Biomarker for Early Alzheimer's Disease.

BACKGROUND: Emerging evidence suggests auditory dysfunction may serve as an early biomarker of Alzheimer's disease (AD). This study investigates amyloid-beta 42 (Aβ42) accumulation in the cochlea and its relationship to auditory dysfunction in 5XFAD mice. METHODS: Immunofluorescence imaging assessed Aβ42 deposition in cochlear structures (spiral ganglion neurons [SGNs], vasculature) at 8 weeks. Auditory brainstem responses (ABR) were analyzed using multimetric methods (Wave I amplitude, signal-to-noise ratio [SNR], phase-locking precision, cross-correlation) at 8 and 16 weeks. RESULTS: Aβ42

Neural crest and periderm-specific requirements of Irf6 during neural tube and craniofacial development.

IRF6 is a key genetic determinant of cleft lip and palate. The ability to interrogate post-embryonic requirements of Irf6 has been hindered, as global Irf6 ablation in the mouse causes neonatal lethality. Prior work analyzing Irf6 in mice defined its role in the embryonic surface epithelium and periderm, where it regulates cell proliferation and differentiation. Several reports have also described Irf6 expression in other cell types, such as muscle, and neuroectoderm. However, analysis of a functional role in non-epithelial cells has been incomplete due to the severity and lethality of the Irf

Gating Attractor Dynamics of Frontal Cortex Under Acupuncture via Recurrent Neural Network.

Acupuncture can regulate the functions of human body and improve the cognition of brain. However, the mechanism of acupuncture manipulations remains unclear. Here, we hypothesis that the frontal cortex plays a gating role in information routing of brain network under acupuncture. To that end, the gating effect of frontal cortex under acupuncture is analyzed in combination with EEG data of acupuncture at Zusanli acupoints. In addition, recurrent neural network (RNN) is used to reproduce the dynamics of frontal cortex under normal state and acupuncture state. From low-dimensional view, it is sho

Variation in the human cervical neural canal.

BACKGROUND CONTEXT: The dimensions of the cervical spinal canal can impact the likelihood of an individual suffering longtime effects from a spinal neck injury as well as influence recovery time. Most studies have used radiographic studies to compare differences in the neural canal, but few have examined skeletal populations to determine variation in the neural canal dimensions without the presence of soft tissue. PURPOSE: To analyze variation seen in the cervical neural canal (anterior-posterior and transverse diameters) with respect to sex and ancestry and to define cervical canal narrowing

Long-latency reflexes in patients with Behçet's disease.

OBJECTIVE: Recent studies demonstrate that the subclinical involvement of motor pathways is frequently observed in patients with Behçet's disease (BD). Long-latency reflexes (LLR) provide information about the continuity of both ascending and descending neural pathways. Our aim was to evaluate the utility of LLR and somatosensory-evoked potentials (SEP) in demonstrating subclinical neural involvement in patients with BD. METHODS: Twenty-nine patients with BD were studied by means of SEP and LLR. Bilateral median nerve SEPs and LLRs evoked by electrical stimulation of both median nerves were r

New surgical paradigm for open neural tube defects.

INTRODUCTION: An open neural tube defect (ONTD) features an exposed, unclosed neural plate in the form of an expanded, flat, and frequently hefty neural placode. Traditional philosophy of ONTD repair aims at preserving function at any cost, which often means stuffing the entire thick and unwieldy but non-functional placode into a tight dural sac, increasing the likelihood of future tethering of the spinal cord. The same philosophy of attempting to save the whole perimetry of the placode also sometimes leads to inadvertent inclusion of parts of the squamous epithelial membrane surrounding the p

Unveiling Hearts: Deep Learning-Based Electrocardiogram Classification for Congenital Heart Disease Detection.

The electrical activity of the human heart, recorded via an electrocardiogram (ECG), is characterized by distinct waveforms such as the P wave, QRS complex, and T wave. By analyzing the duration, morphology, and intervals between these waveforms, various cardiac disorders can be identified. This study aims to develop a deep learning-based approach for the accurate classification of congenital heart disease (CHD) using ECG data. We employed convolutional neural networks (CNNs) and recurrent neural networks (RNNs) to analyze ECG signals, leveraging their ability to detect multiple features in ti

SCAN: Spatiotemporal Cloud Atlas for Neural cells.

The nervous system is one of the most complicated and enigmatic systems within the animal kingdom. Recently, the emergence and development of spatial transcriptomics (ST) and single-cell RNA sequencing (scRNA-seq) technologies have provided an unprecedented ability to systematically decipher the cellular heterogeneity and spatial locations of the nervous system from multiple unbiased aspects. However, efficiently integrating, presenting and analyzing massive multiomic data remains a huge challenge. Here, we manually collected and comprehensively analyzed high-quality scRNA-seq and ST data from

Ncx, a Hox11 related gene, is expressed in a variety of tissues derived from neural crest cells.

We have isolated the murine homeobox gene (Ncx) that belong to a Hox11 gene family. Expression of the Ncx gene was analyzed in total RNAs from embryos by reverse transcribed polymerase chain reaction (RT-PCR). The mRNA was detected in embryos after 9.5 days of embryogenesis (E9.5) and was maximal at E12.5. The RT-PCR also detected the message in total RNAs from adrenal glands and intestine in adult mice. The expression was further examined in various tissues from embryos by in situ hybridization. It was detected in dorsal root ganglia, cranial nerve ganglia (V, IX, X), enteric nerve ganglia an

Ultrastructural analysis of basal neuroepithelial cells in dysraphic mice.

Ultrastructural aspects of the cellular pathology in the basal neuroepithelium of the hindbrain and spinal cord were analyzed in dysraphic loop-tail mice at nine days of gestation. Whereas the basal cytoplasm of the neuroepithelium in normal littermates showed a consistent electron density, the neuroepithelium in abnormal embryos was characterized by "light" and "dark" cells scattered randomly along the basal aspect of the hindbrain and spinal cord. In the abnormals, gaps occurred in the neural basal lamina, and the neuroepithelial cells often were in direct contact with cytoplasmic processes

The mechanism of sciatic pain relief by chemonucleolysis.

Chemonucleolysis must relieve sciatic pain by some means other than the removal of the disc protrusion from the neural canal, because the protrusion characteristically persists despite the resolution of sciatic pain. This paper analyzes the mechanism of symptom production in sciatica and identifies how chemonucleolysis relieves these symptoms.Pressure can be exerted on a nerve root by a disc protrusion without compression against the posterior wall of the neural canal. This pressure is the result of tension produced in the nerve root when it is deformed by the disc protrusion. A previously unr

Fetal Autopsy: Insights Into the Spectrum of Dysraphisms With Associated Anomalies.

Introduction and aim Malformations of the central nervous system (CNS) are one of the most devastating, yet most common congenital anomalies. Congenital CNS anomalies are structural or functional abnormalities of the brain and spinal cord that occur during the intrauterine developmental period and have a high intrauterine mortality. This study aimed to determine the frequency and distribution of neural tube defects (NTDs) in a tertiary care center for a period of 4.5 years. The neural tube defect was analyzed along with the other associated anomalies in the fetus. Its association with maternal

Effects of early childhood otitis media-related conductive hearing loss on speech perception, neural processing, and working memory.

Otitis media with effusion (OME) is a common childhood ear disease that can cause temporary conductive hearing loss during a critical period of auditory development. This condition is hypothesized to be related to deficits in neural, perceptual, and cognitive abilities. This study examined the relation between conductive hearing loss due to OME in early childhood and its effects on neural and perceptual speech processing, as well as working memory (WM) function in early school-aged children. A total of 21 children aged 6-8 years with normal hearing who had a history of OME in early childhood w

Detecting central fixation by means of artificial neural networks in a pediatric vision screener using retinal birefringence scanning.

BACKGROUND: Reliable detection of central fixation and eye alignment is essential in the diagnosis of amblyopia ("lazy eye"), which can lead to blindness. Our lab has developed and reported earlier a pediatric vision screener that performs scanning of the retina around the fovea and analyzes changes in the polarization state of light as the scan progresses. Depending on the direction of gaze and the instrument design, the screener produces several signal frequencies that can be utilized in the detection of central fixation. The objective of this study was to compare artificial neural networks

How the Human Brain Represents Perceived Dangerousness or "Predacity" of Animals.

UNLABELLED: Common or folk knowledge about animals is dominated by three dimensions: (1) level of cognitive complexity or "animacy;" (2) dangerousness or "predacity;" and (3) size. We investigated the neural basis of the perceived dangerousness or aggressiveness of animals, which we refer to more generally as "perception of threat." Using functional magnetic resonance imaging (fMRI), we analyzed neural activity evoked by viewing images of animal categories that spanned the dissociable semantic dimensions of threat and taxonomic class. The results reveal a distributed network for perception of

Hybrid artificial intelligence approaches and bioimpedance spectroscopy for classifying pancreatic disease.

This research develops bioimpedance spectroscopy methods aimed at improving the differential diagnosis of pancreatic diseases. A novel approach for forming descriptors from bioimpedance data is introduced, which involves analyzing four amplitude-phase-frequency characteristics obtained from quasi-orthogonal leads. This method establishes informative feature spaces utilized by a hybrid classifier specifically designed to differentiate between pancreatitis and pancreatic cancer. The hybrid classifier comprises five macro layers, integrating probabilistic neural networks and fuzzy logical inferen

Application of automatic image analysis using a Deep Learning Neural Network for assessing the growth of green algae containing carotenoids - importance for environment, health and aquaculture.

Using deep learning and neural networks enables us to greatly speed-up quantitative studies and provide a useful tool for analyzing microscopic images. Studies conducted on selected algae Haematococcus and Coelastrum sp. confirm the feasibility of using the deep learning neural network. The confusion matrix demonstrated the numbers of errors generated by the YOLO v8 network in relation to the validation dataset. It indicated a higher number of errors in the detection of Haematococcus than Coleastrum. The F1 score, as the harmonic mean of precision and recall, is significantly higher for the cl
