A neuronal circuit driven by GLP-1 in the olfactory bulb regulates insulin secretion.

Glucagon-like peptide 1 (GLP-1) stimulates insulin secretion and holds significant pharmacological potential. Nevertheless, the regulation of energy homeostasis by centrally-produced GLP-1 remains partially understood. Preproglucagon cells, known to release GLP-1, are found in the olfactory bulb (OB). We show that activating GLP-1 receptors (GLP-1R) in the OB stimulates insulin secretion in response to oral glucose in lean and diet-induced obese male mice. This is associated with reduced noradrenaline content in the pancreas and blocked by an α2-adrenergic receptor agonist, implicating functio…

Effect of Xylazine-Tiletamine-Zolazepam on the Local Field Potential of the Rat Olfactory Bulb.

Neural oscillations of the mammalian olfactory system have been studied for decades. This research suggests they are linked to various processes involved in odor information analysis, depending on the vigilance state and presentation of stimuli. In addition, the effects of various anesthetics, including commonly used ones like chloral hydrate, pentobarbital, ketamine, and urethane, on the local field potential (LFP) in the olfactory bulb (OB) have been studied. In particular, the combination of xylazine and tiletamine-zolazepam has been shown to produce steady anesthesia for an extended period…

Not all sharks are "swimming noses": variation in olfactory bulb size in cartilaginous fishes.

Olfaction is a universal modality by which all animals sample chemical stimuli from their environment. In cartilaginous fishes, olfaction is … we have addressed these questions by directly examining interspecific variation in the size of the olfactory bulbs (OB), the region of the brain that receives the primary sensory projections from the olfactory nerve, in 58 species of cartilaginous fishes. Relative OB size was compared among species occupying different ecological niches. Our results show that th …

Decreased coherence between the two olfactory bulbs in Alzheimer's disease model mice.

Alzheimer's disease (AD) is a progressive, neurodegenerative disease characterized by cell and synapse losses and the ensued network disconnections. Olfactory impairment is common and occurs early in AD patients, suggesting its potential role in the early diagnosis of AD. However, the functional connectivity of the olfactory system in AD has received little attention. Using local field potentials symmetrically recorded from the two olfactory bulbs in awake mice, we found that in AD model mice with robust amyloid-β burden, the coherence between the two bulbs for spontaneous activities was signi…

Expanding the clinical and neuroradiological phenotype of 6q27 microdeletion: olfactory bulb aplasia and anosmia.

Subtelomeric deletions of chromosome 6q may result in a syndrome with brain malformations, comprising hydrocephalus and hypoplasia of the corpus callosum. Aplasia of the olfactory bulbs (OB) or anosmia has not been described in this syndrome. We describe a 3-year-old girl and a 25-year-old man with subtelomere 6q deletions. Both patients had aplastic OB and hydrocephalus. Subtelomeric 6q deletions might be underdiagnosed as anosmia can be the only symptom.

Optical imaging of postsynaptic odor representation in the glomerular layer of the mouse olfactory bulb.

Olfactory glomeruli are the loci where the first odor-representation map emerges. The glomerular layer comprises exquisite local synaptic circuits for the processing of olfactory coding patterns immediately after their emergence. To understand how an odor map is transferred from afferent terminals to postsynaptic dendrites, it is essential to directly monitor the odor-evoked glomerular postsynaptic activity patterns. Here we report the use of a transgenic mouse expressing a Ca(2+)-sensitive green fluorescence protein (GCaMP2) under a Kv3.1 potassium-channel promoter. Immunostaining revealed th

Long-term enhancement of synchronized oscillations by adrenergic receptor activation in the olfactory bulb.

The noradrenergic system is widely thought to be important for associative learning in the olfactory system through actions in the first processing structure, the main olfactory bulb (MOB). Here, we used extracellular local field potential (LFP) and patch-clamp recordings in rat MOB slices to examine norepinephrine (NE)-induced long-term changes in circuit properties that might underlie learning. During responses to patterned olfactory nerve stimulation mimicking the breathing cycle, NE induced a long-term increase in gamma frequency (30-70 Hz) synchronized oscillations. The enhancement persis…

A potential reservoir of immature dopaminergic replacement neurons in the adult mammalian olfactory bulb.

A significant fraction of the interneurons added in adulthood to the glomerular layer (GL) of the olfactory bulb (OB) are dopaminergic (DA). In the OB, DA neurons are restricted to the GL, but using transgenic mice expressing eGFP under the tyrosine hydroxylase (TH) promoter, we also detected the presence of TH-GFP+ cells in the mitral and external plexiform layers. We hypothesized that these could be adult-generated neurons committed to become DA but not yet entirely differentiated. Accordingly, TH-GFP+ cells outside the GL exhibit functional properties (appearance of pacemaker currents, syna…

Soluble guanylyl cyclase appears in a specific subset of periglomerular cells in the olfactory bulb.

In the brain, nitric oxide acts as an atypical messenger in cellular nonsynaptic transmission. In the olfactory bulb, this gas is produced at the level of the olfactory glomeruli by a subpopulation of periglomerular cells that participates in the first synaptic relay of the olfactory information between the olfactory nerve and the dendritic tufts of principal cells. It has been proposed that nitric oxide modulates intraglomerular synaptic integration of sensory inputs, but its specific role in the glomerular circuitry remains to be understood. In this article, we demonstrate that, in the glome…

Changes in glutamate receptor subunit 4 expression in the deafferented olfactory bulb of zebrafish.

The distribution of ionotropic glutamate receptor subunit 4 (iGluR4) was examined in both normal and deafferented olfactory bulbs of adult zebrafish, Danio rerio. With the exception of the olfactory nerve layer, there was extensive labeling with antibodies to iGluR4 in the olfactory bulbs, specifically in juxtaglomerular cell bodies and their processes. These results are consistent with previous work, which has suggested differential distribution of glutamate receptors in the vertebrate olfactory system. Analysis of bulbs following olfactory organ removal revealed a significant loss of iGluR4 …

Spatiotemporal distribution of the insulin-like growth factor receptor in the rat olfactory bulb.

Insulin-like growth factor I (IGF-I) and its receptor (IGF-IR) are involved in growth of neurons. In the rat olfactory epithelium, we previously showed IGF-IR immunostaining in subsets of olfactory receptor neurons. We now report that IGF-IR staining was heaviest in the olfactory nerve layer of the rat olfactory bulb at embryonic days 18, and 19 and postnatal day 1, with labeling of protoglomeruli. In the adult, only a few glomeruli were IGF-IR-positive, some of which were unusually small and strongly labeled. Some IGF-IR-positive fibers penetrated deeper into the external plexiform layer, eve…

GABA immunoreactivity in the olfactory bulbs of the adult sea lamprey Petromyzon marinus L.

The distribution of gamma-aminobutyric acid (GABA) immunoreactivity in the olfactory bulbs of the adult sea lamprey was studied using an antibody against this transmitter. Five types of GABA-immunoreactive (GABAir) cells were observed. Medium-sized GABAir cells (periglomerular cells) were located around the olfactory glomeruli and occasionally within them. In the inner cellular layer of the bulbs and around the olfactory ventricles, two types of GABAir perikarya were present: some medium-sized GABAir cells and numerous small GABAir cells (granules). In the walls of the olfactory ventricle, som…

Assessment and discrimination of odor stimuli in rat olfactory bulb by dynamic functional MRI.

Dynamic blood oxygenation level-dependent functional MRI was applied at 7 T in the rat olfactory bulb (OB) with pulsed delivery of iso-amyl acetate (IAA) and limonene. Acquisition times for single-slice and whole OB data were 8 and 32 s, respectively, with spatial resolution of 220 x 220 x 250 micrometer. On an intrasubject basis, short IAA exposures of 0.6 min separated by 3.5-min intervals induced reproducible spatial activity patterns (SAPs) in the olfactory nerve layer, glomerular layer, and external plexiform layer. During long exposures ( approximately 10 min), the initially dominant dor…

Regulation of neuropeptide Y mRNA and peptide concentrations by copper in rat olfactory bulb.

Neuropeptide Y is highly abundant in both the peripheral and central nervous systems and is known to have diverse functions including regulation of feeding behavior, blood pressure, circadian rhythms, reproductive behavior and the response to stress. Northern analysis showed that copper deficiency increased brain NPY mRNA abundance particularly in the olfactory bulb (OB). These increases were not accompanied by alterations in food intake or blood pressure. After 4 weeks of a copper-restricted diet, OB copper concentrations decreased to 44% of control and NPY mRNA increased 1.5-fold. Addition o…

Cellular and subcellular localization of gamma-aminobutyric acidB receptors in the rat olfactory bulb.

Olfactory nerve axons terminate in rounded regions of the olfactory bulb, termed glomeruli, where they make excitatory synapses with the dendrites of second-order neurons. Neurotransmission from the olfactory nerve to the postsynaptic targets is negatively regulated by gamma-aminobutyric acid (GABA), and there is evidence that inhibition of sensory input is mediated, at least in part, by GABA(B) receptors. Using an antiserum that recognizes two GABA(B) receptor splice variants (GBR1a and GBR1b), we show here that GABA(B) receptors are located on the axon terminals of the olfactory nerve, where…

The accessory olfactory bulb of the mink, Mustela vison: a morphological and lectin histochemical study.

The distribution of binding sites for the lectins Ulex europaeus agglutinin I, Soybean agglutinin, Bandeiraea simplicifolia agglutinin I-isolectin B4, and Vicia villosa agglutinin in the mink olfactory bulb was investigated. All lectins except Ulex europaeus agglutinin I bound exclusively and systematically to a single area of the olfactory bulb. This area corresponded to that in which the vomeronasal nerves terminate, indicating that it is the accessory olfactory bulb, as confirmed by microdissection and by the study of transverse and parasagittal series of the olfactory bulb. The results, mo…

Combinatorial and chemotopic odorant coding in the zebrafish olfactory bulb visualized by optical imaging.

Odors are thought to be represented by a distributed code across the glomerular modules in the olfactory bulb (OB). Here, we optically imaged presynaptic activity in glomerular modules of the zebrafish OB induced by a class of natural odorants (amino acids [AAs]) after labeling of primary afferents with a calcium-sensitive dye. AAs induce complex combinatorial patterns of active glomerular modules that are unique for different stimuli and concentrations. Quantitative analysis shows that defined molecular features of stimuli are correlated with activity in spatially confined groups of glomerula…

Development of 5'-nucleotidase staining in the olfactory bulbs of normal and naris-occluded rats.

The distribution of the adenosine-producing ecto-enzyme 5'-nucleotidase was investigated histochemically in the developing rat olfactory bulb. Rat pups underwent either unilateral surgical occlusion of the right external naris or sham surgery on postnatal day 1. At 10, 20, or 30 days postpartum, horizontal sections of the olfactory bulb were reacted histochemically to reveal the locus and intensity of 5'-nucleotidase activity. Relative staining levels were determined by optical densitometry in standardized bulb regions. A marked, age-related increase in staining density was observed. Reaction …

Influences of the paraventricular and suprachiasmatic nuclei and olfactory bulbs on melatonin responses in the golden hamster.

Removal of the pineal, or denervation of this gland by superior cervical ganglionectomy, blocks testicular regression in golden hamsters exposed to short photoperiods. Aspiration of the olfactory bulbs or lesions of the suprachiasmatic or paraventricular nuclei of the hypothalamus (SCNx or PVNx) have similar effects. We have examined the effects of these operations on pineal melatonin content and gonadal responses to various patterns of exogenous melatonin in order to examine the roles played by the olfactory bulbs, the SCN, and the PVN in hamster photoperiodism. SCNx and PVNx significantly re…
