Cholesterol biosynthesis defines oligodendrocyte precursor heterogeneity between brain and spinal cord.

Brain and spinal cord oligodendroglia have distinct functional characteristics, and cell-autonomous loss of individual genes can result in different regional phenotypes. However, a molecular basis for these distinctions is unknown. Using single-cell analysis of oligodendroglia during developmental myelination, we demonstrate that brain and spinal cord precursors are transcriptionally distinct, defined predominantly by cholesterol biosynthesis. We further identify the mechanistic target of rapamycin (mTOR) as a major regulator promoting cholesterol biosynthesis in oligodendroglia. Oligodendrogl

Inhibited oligodendrogenesis impairs attention and motivation in adult mice regardless of repeated mild traumatic brain injury.

INTRODUCTION: The brain's white matter is responsible for communication across the brain and because attention relies on coordinated communication among distributed brain networks, white matter dysfunction may contribute to attentional impairment. Attention problems are among the most common long-lasting cognitive symptoms of mild traumatic brain injury (mTBI) and as attention is fundamental to many aspects of cognition, the effects of attentional impairment can be broad. mTBI-induced effects on oligodendrocytes and myelin contribute to cognitive deficits following injury and myelin plasticity

Molecular and cellular dynamics of the developing human neocortex.

The development of the human neocortex is highly dynamic, involving complex cellular trajectories controlled by gene regulation1. Here we co … iate progenitor cells (Tri-IPCs)-that is responsible for the local production of GABAergic neurons, oligodendrocyte precursor cells and astrocytes. Notably, most glioblastoma cells resemble Tri-IPCs at the transcriptomic level, suggesting that cancer cells hijack developmental processes to enhance growth and heterogeneity. Furthermore, by integrating our atlas data with large-sc …

Environmental phthalate exposure impairs cognition in aged males by coupling myelin injury to synaptic dysfunction via Nogo-A/S1PR2 signaling.

Di-(2-ethylhexyl) phthalate (DEHP) is a widespread environmental contaminant linked to endocrine disruption, neurotoxicity, and cognitive aging. However, its effects on hippocampal function in aging populations remain unclear. In this study, 22-month-old male C57BL/6J mice were orally exposed to DEHP (0.2, 20, or 200  mg/kg/day) for 5 weeks to model environmentally relevant and toxicological exposure scenarios. Cognitive performance was evaluated using the Morris water maze and novel object recognition tests, while hippocampal alterations were assessed through histology, immunofluorescence, an

A Question of Origins: Non-neuronal Sources of Amyloid-β.

Amyloid-β (Aβ) plaques and neurofibrillary tau tangles are hallmarks of Alzheimer's disease (AD). While the intracellular localization of tau tangles within neurons nominates them as the primary producers of tau, the cellular origin of Aβ is less clear as plaques accumulate extracellularly. Neurons have been considered the sole source of Aβ, leading to the generation of many AD animal models expressing familial AD protein variants specifically in neurons. However, emerging evidence showed that non-neuronal cells abundantly express amyloid precursor protein (APP) and its processing machinery. A

Characterizing oligodendrocyte-lineage cells and myelination in the basolateral amygdala: Insights from a novel methodology in postmortem human brain.

The basolateral amygdala (BLA) plays a key role in the pathophysiology of depressive disorders and trauma, yet oligodendrocyte (OL)-lineage cells and myelin in this region remain understudied in humans. This may be due in part to the lack of a cost-effective, antibody-based method to isolate OL and OL precursor cells (OPC) from postmortem brain tissue. This study aimed to 1) create and validate a method for isolating OPC and OL nuclei from postmortem grey matter; 2) compare OPC and OL gene expression in the BLA between individuals with depression who died by suicide (with or without a history

Myelin endocytosis by brain endothelial cells causes endothelial iron overload and oligodendroglial iron hunger in hypoperfusion-induced white matter injury.

AIMS: Hypoperfusion induces significant white matter injury in cerebral vascular disorders, including arteriosclerotic cerebral small vessel disease (aCSVD), which is prevalent among the elderly. Iron transport by blood vessel endothelial cells (BVECs) from the periphery supports oligodendrocyte maturation and white matter repair. This study aims to elucidate the association between iron homeostasis changes and white matter injury severity, and explore the crosstalk between BVECs and oligodendroglial lineage cells. METHODS: In vivo: C57BL/6 mice were subjected to unilateral common carotid art

The Cell-Specific Effects of the Human Remyelination-Promoting rHIgM22 on Sphingolipid Metabolism in Cultured Glial Cells.

Demyelinating diseases are heterogeneous in their etiology, clinical course, and manifestations. In the long run, however, they lead to irreversible dysfunction of the nervous system. Although myelin regeneration occurs in response to myelin damage in both animal models of demyelination and human patients, the outcome is usually less favorable in humans. This explains the interest in treatments that could improve the effectiveness of myelin regeneration. Among these, treatment with the monoclonal antibody rHIgM22 has been shown to effectively enhance myelin regeneration in both immune and non-

Detection of JC virus DNA in human tonsil tissue: evidence for site of initial viral infection.

Progressive multifocal leukoencephalopathy is a demyelinating disease of the human central nervous system that results from lytic infection of oligodendrocytes by the polyomavirus JC (JCV). Originally, JCV was thought to replicate exclusively in human glial cells, specifically oligodendrocytes. However, we have recently shown that JCV can replicate in cells of lymphoid origin such as hematopoietic precursor cells, B lymphocytes, and tonsillar stromal cells. To determine whether tonsils harbor JCV, we tested a total of 54 tonsils, 38 from children and 16 from adult donors. Nested PCRs with prim

miR-381 Regulates Neural Stem Cell Proliferation and Differentiation via Regulating Hes1 Expression.

Источник пометил публикацию как отозванную

Neural stem cells are self-renewing, multipotent and undifferentiated precursors that retain the capacity for differentiation into both glial (astrocytes and oligodendrocytes) and neuronal lineages. Neural stem cells offer cell-based therapies for neurological disorders such as Alzheimer's disease, Parkinson's disease, Huntington's disease and spinal cord injuries. However, their cellular behavior is poorly understood. MicroRNAs (miRNAs) are a class of small noncoding RNAs involved in cell development, proliferation and differentiation through regulating gene expression at post-transcriptional

Angiotensin II impairs the remyelination mediated by brain endothelial cell-derived exosomes.

White matter hyperintensities (WMH), characterized by myelin loss and axonal injury, are largely driven by hypertension, their most significant modifiable risk factor. The role of endothelial cell-derived exosomes (EC-Exos) in oligodendrocyte precursor cell (OPCs)-mediated remyelination within WMH remains critical but poorly defined. This study investigated the function of EC-Exos in remyelination by comparing exosomes from angiotensin II-treated brain microvascular endothelial cells (AngII-Exos; modeling hypertensive conditions) with those from untreated cells (BMEC-Exos; modeling normotensiv

IFN-γ-Responsive Microglia-Derived Extracellular Vesicles Impair White Matter Repair After Stroke Through a miR-9-5p/NEDD4-Dependent Mechanism.

Promoting white matter repair is crucial for functional recovery after ischemic stroke. Our previous study has identified an IFN-γ-responsive microglia phenotype in the brain after stroke. However, its role in white matter repair remains poorly understood. Herein, we find that extracellular vesicles derived from IFN-γ-responsive microglia (IFN-γ EVs) are internalized by oligodendrocyte precursor cells (OPCs), leading to impaired OPCs proliferation, survival, and differentiation and reduced white matter repair in a murine transient middle cerebral artery occlusion (tMCAO) model. Mechanistically…

PD-L1/PD-1 checkpoint pathway regulates astrocyte morphogenesis and myelination during brain development.

Programmed cell death protein 1 (PD-1) and its primary ligand PD-L1 are integral components of a significant immune checkpoint pathway, wide … t coverage of blood vessels. Additionally, aberrant secretion of CSRP1 by astrocytes interacts with oligodendrocyte precursor cells (OPCs) membrane proteins annexin A1 (ANXA1) and annexin A2 (ANXA2), leading to the exclusion of migrating OPCs from blood vessels. This disruption in OPC migration and differentiation results in abnormal myelination and is associated with cognitive …

The dimensions of white matter injury in preterm neonates.

White matter injury (WMI) represents a frequent form of parenchymal brain injury in preterm neonates. Several dimensions of WMI are recognized, with distinct neuropathologic features involving a combination of destructive and maturational anomalies. Hypoxia-ischemia is the main mechanism leading to WMI and adverse white matter development, which result from injury to the oligodendrocyte precursor cells. Inflammation might act as a potentiator for WMI. A combination of hypoxia-ischemia and inflammation is frequent in several neonatal comorbidities such as postnatal infections, NEC and bronchopu…

Developmental defects and aberrant accumulation of endogenous psychosine in oligodendrocytes in a murine model of Krabbe disease.

Krabbe disease (KD), or globoid cell leukodystrophy, is an inherited lysosomal storage disease with leukodystrophy caused by a mutation in the galactosylceramidase (GALC) gene. The majority of patients show the early onset form of KD dominated by cerebral demyelination with apoptotic oligodendrocyte (OL) death. However, the initial pathophysiological changes in developing OLs remain poorly understood. Here, we show that OLs of twitcher mice, an authentic mouse model of KD, exhibited developmental defects and impaired myelin formation in vivo and in vitro. In twitcher mouse brain, abnormal myel
