Autophagy is a catabolic process involved in maintaining energy and organelle homeostasis. The relationship between obesity and the regulation of autophagy is cell type specific. Despite adverse consequences of obesity on cardiac structure and function, the contribution of altered cardiac autophagy in response to fatty acid overload is incompletely understood. Here, we report the suppression of autophagosome clearance and the activation of NADPH oxidase (Nox)2 in both high fat-fed murine hearts and palmitate-treated H9C2 cardiomyocytes (CMs). Defective autophagosome clearance is secondary to s

Retinyl palmitate and its metabolites retinol and retinoic acid control growth and epithelial differentiation. Systemic or local vitamin A deficiency induced by malnutrition, continuous chemical irritation or locally induced by inflammation causes squamous metaplastic changes in the epithelium of mucous membranes. We demonstrate that in an animal model topically applied retinyl palmitate can be taken up by the mucosal cells independently from the systemic supply. Under in vivo conditions metabolic changes in vaginal epithelium of rats were shown to be reversed by treatment with topical retinyl

To investigate aqueous metabolic profiles in Vogt-Koyanagi-Harada (VKH) and Behcet's disease (BD), we applied ultra-high-performance liquid chromatography equipped with quadrupole time-of flight mass spectrometry in aqueous humor samples collected from these patients and controls. Metabolite levels in these three groups were analyzed by univariate logistic regression. The differential metabolites were subjected to subsequent pathway analysis by MetaboAnalyst. The results showed that both partial-least squares discrimination analysis and hierarchical clustering analysis showed specific aqueous

The haruan (Channa striatus) is an indigenous, predatory freshwater fish of Malaysia. It is a common food fish among the local populace with traditionally identified pharmacological benefits in treating wound and pain and in boosting energy of the sick. Channa striatus is also a subject of renewed interest in Malaysian folk medicine in the search for a better cure for diseases and ailments. Amino acids and fatty acids, found in high concentrations in the fish, might have contributed to its pharmacological properties. Important amino acids of the fish include glycine, lysine and arginine, while

A few studies suggested high stereo-specifically numbered (sn)-2 palmitate in a formula might favor the gut Bifidobacteria of infants. The initial colonization and subsequent development of gut microbiota in early life might be associated with development and later life functions of the central nervous system via the microbiota-gut-brain axis, such as children with autism. This study aims to assess the hypothesized effect of increasing the amount of palmitic acid esterified in the sn-2 position in infant formula on neurodevelopment in healthy full-term infants and to explore the association of

Metabolic responses of epiphytic lichen Ramalina farinacea to cadmium (Cd) and/or nitric oxide (NO) scavenger (cPTIO) were studied. Accumulation of Cd and other metallic nutrients was not affected by cPTIO while total and absorbed amounts differed. Cd-induced NO formation was suppressed by cPTIO but ROS signal was synergistically enhanced, confirming that NO is essential to keep ROS under control. This excessive ROS generation could be a reason for depleted amount of all fatty acids, including SFAs, MUFAs and PUFAs. Total content of fatty acids reached 3.89 mg/g DW in control with linoleic (40

Liquid chromatography/mass spectrometry (LC/MS)-based methods were developed for the analysis of 10 resin acids and five fatty acids in process waters of paper industries. No fragmentation of target compounds was observed using atmospheric pressure chemical ionization (APCI) with negative ionization. The [M - H](-) ion permitted the individual quantification of fatty and aromatic resin acids, whereas the non-aromatic resin acids presented a single and common ion at m/z 301. Separation with two columns of different polarity permitted peak confirmation. The method that used a C(8) column with 2-

The lipid composition and content of breast milk are influenced by the lactation stage and maternal diet. While dairy products are recommended during pregnancy, the effects of fermented milk on breast milk lipids remain unclear. In this study, we investigated the impact of fermented milk intake on the glyceride and phospholipid composition of breast milk. Pregnant women consumed fermented milk containing glycerides and phospholipids from early pregnancy until the end of lactation. Fermented milk intake significantly increased the number and content of triacylglycerols, including glycerides wit

The major constituent free fatty acid in rat parotid and submandibular glands was palmitic acid (about 40% of the total), followed by saturated and unsaturated fatty acids with 18 carbon atoms and arachidonic acid. The composition was changed by chronic administration of isoproterenol, with an increase of linoleic acid and a decrease of arachidonic acid coinciding with changes in the acyl composition of phospholipids. With respect to the metabolism of unsaturated fatty acids, delta 6-desaturation activity converting linoleic acid to gamma-linolenic acid was detected using [14C]linoleoyl-CoA in

The effect of lipid peroxidation on fatty acid composition in membrane phospholipids of transparent and cataractous lens nuclei was investigated. The preliminary results demonstrate a trend toward an increase in the content of palmitic acid with a decrease in the content of oleic acid, linolenic acid and docosahexaenoic acid in the opaque nuclei as compared to the transparent ones of senile cataractous lenses.

This study investigates the high-temperature interactions between lead palmitate (Pb(C16)2) and linseed oil, revealing complex phase transformations and structural changes critical for understanding metal soap formation in oil paintings. Through complementary analytical techniques including DSC, XRPD, FTIR, and solid-state NMR, we identified two significant phenomena: the formation of mixed lead palmitate-stearate compounds and the emergence of a high-temperature lead palmitate polymorph. When heated to 112 °C, lead palmitate in linseed oil undergoes partial conversion to mixed lead palmitate-

Lipotoxicity caused by excess free fatty acids, particularly saturated fatty acids (SFAs) such as palmitic acid (PA), is one of the most important pathogenesis of nonalcoholic fatty liver disease (NAFLD). However, unsaturated fatty acids (UFAs), such as oleic acid (OA), are nontoxic and can combat SFA-induced toxicity through alleviation of cell apoptosis, endoplasmic reticulum stress (ER stress) and lipids metabolism disorder. However, whether OA is able to regulate autophagy is largely unknown. So, this study aims to investigate the mechanism underlying OA mediated modulation of autophagy in

Acute pancreatitis (AP) is an inflammatory disease characterized by localized pancreatic injury and a systemic inflammatory response. Fatty acids (FAs), produced during the breakdown of triglycerides (TGs) in blood and peripancreatic fat, escalate local pancreatic inflammation to a systemic level by damaging pancreatic acinar cells (PACs) and triggering M1 macrophage polarization. This paper provides a comprehensive analysis of lipases' roles in the onset and progression of AP, as well as the effects of long-chain fatty acids (LCFAs) on the function of pancreatic acinar cells (PACs). Abnormali

OBJECTIVES: Metabolic dysfunction-associated steatotic liver disease, the most common chronic liver disease worldwide, is a major cause of end-stage liver failure requiring transplant. Its hallmark, free fatty acid accumulation in hepatocytes, leads to lipotoxicity. We examined lipid droplet and mitochondrial changes in HepG2 cells under lipotoxic conditions.
MATERIALS AND METHODS: Lipotoxic stress was induced in HepG2 cells (ATCC HB8065) by treating cells with 1 mM palmitic acid. We studied cells at 0, 12, and 24 hours. Cells were fixed in 2.5% glutaraldehyde (pH 7.4), postfixed with 1% osmi

The key aroma components and (furan) fatty acids in fresh red goji berries were determined and their variations after hot-air drying were investigated to understand the impacts of drying. 22 of the 46 detected aroma components were found to be the key aroma components in fresh red goji berries, including (E)-β-damascenone, 1-octen-3-one, and trans-4,5-epoxy-(E)-2-decenal, which elicited the cooked apple-like, mushroom-like, and metallic odor impressions. Linoleic acid, oleic acid, and palmitic acid were the predominant fatty acids. Furan fatty acid (FuFA) in fresh red goji berries was measured

In this study, the combined effect of different packaging materials (transparent PET, transparent glass, glass-PET bottle and tin), some aromatic herbs (thyme, rosemary, sage and olive leaf) and also their essential oils (thyme, rosemary and sage) on fatty acid composition of virgin olive oil was investigated during storage period. The initial amounts of the main fatty acids as oleic, palmitic and linoleic acids were determined as 72.89%, 11.89% and 8.96%, respectively. The addition of aromatic plants and essential oils did not effect the fatty acid profile. Also, packaging materials had a min

Meconium, an ether extract of meconium, and the major free fatty acids of meconium (palmitic, stearic, and oleic acids) were all found to increase the surface tension minimum of dog lung extract in a Wilhelmy balance. Each of these fractions was instilled into the lungs of dogs (15 experimental, eight saline solution controls), and cardiac output, venous and arterial blood gases, pulmonary, atrial, and systemic pressures, airway pressure, and static lung compliance were serially monitored for 2 hours. Mean airway pressure increased and static lung compliance decreased significantly in all of t

In this research, we utilized an efficient approach to synthesize superparamagnetic graphene oxide (SPGO) rapidly in a one-pot method using microwave irradiation of graphene oxide (GO), urea, and Fe(III) ion. Tannic acid (TA) was introduced to the surface of SPGO through a straightforward and eco-friendly process. Methods were devised to furnish GO nanosheets and modify their surfaces with TA in an environmentally friendly manner. Two series of nanosheets, namely, SPGO/TA-COOH and SPGO/TA-IM, were engineered on the surface and used for immobilizing lipase enzyme. Through various analytical too

PURPOSE: Sodium-glucose cotransporter 2 (SGLT2) inhibitors prevent heart failure and decrease cardiovascular mortality in patients with type 2 diabetes. Heart failure is associated with detrimental changes in energy metabolism, and the preservation of cardiac mitochondrial function is crucial for the failing heart. However, to date, there are no data to support the hypothesis that treatment with a SGLT2 inhibitor might alter mitochondrial bioenergetics in diabetic failing hearts. Thus, the aim of this study was to investigate the protective effects of empagliflozin on mitochondrial fatty acid
