The possible role of endocrine disrupting chemicals in the aetiology of cryptorchidism and hypospadias: a population-based case-control study in rural Sicily.

Abstract This was an open case-control study of the possible association between parental occupational and domestic exposures to potential endocrine disrupting chemicals (EDC) assessed by questionnaire and cryptorchidism and hypospadias in their offspring in the agricultural area of Ragusa. Cases of infants born between 1998 and 2002 with either of these two malformations (n=90), and controls (n=203), were recruited through the paediatric services (for cases) and a random sample of healthy infants attending the same services born in the same period of time (for controls). Data on occupational

Infant leukaemia after in utero exposure to radiation from Chernobyl.

There has been no documented increase in childhood leukaemia following the Chernobyl accident. However, different forms of childhood leukaemia may not be equally susceptible to radiation carcinogenesis. Infant leukaemia is a distinct form associated with a specific genetic abnormality. Outside the former Soviet Union, contamination resulting from the Chernobyl accident has been highest in Greece and Austria and high also in the Scandinavian countries. All childhood leukaemia cases diagnosed throughout Greece since 1 January 1980 have been recorded. Here we report that infants exposed in utero

Multiple generation distinct toxicant exposures and epigenetic transgenerational inheritance of sex‑specific outcross impacts on sperm epigenetics and obesity pathology.

Environmental factors can influence the epigenetic inheritance of pathology across generations. Prior studies found various toxicants increased pathology in their descendants through epigenetic transgenerational inheritance. Previously, three generations of rats were exposed separately to vinclozolin (F0 generation), a jet fuel hydrocarbon mixture (F1 generation), and dichlorodiphenyltrichloroethane (DDT) (F2 generation), producing a transgenerational F5 generation. The current study extends this study by breeding this transgenerational F5 generation with wild-type rats to create maternal and

Nutritional, pharmacological, and environmental programming of NAFLD in early life.

Nonalcoholic fatty liver disease (NAFLD) is the main liver disease worldwide, and its prevalence in children and adolescents has been increasing in the past years. It has been demonstrated that parental exposure to different conditions, both preconceptionally and during pregnancy, can lead to fetal programming of several metabolic diseases, including NAFLD. In this article, we review some of the maternal and paternal conditions that may be involved in early-life programing of adult NAFLD. First, we describe the maternal nutritional factors that have been suggested to increase the risk of NAFLD

The epidemiology of electric and magnetic field exposures in the power frequency range and reproductive outcomes.

Some laboratory studies have reported effects of extremely low frequency electric and magnetic fields on calcium homeostasis and pineal gland function which potentially could have consequences for reproductive health. The purpose of this review is to report results of research to date in humans on maternal, paternal and fetal exposure to the 50-60 Hz fields associated with electricity, and to propose populations, endpoints and strategies for future research.

Spontaneous adverse drug reaction reports for neonates and infants in the UK 2001-2010: content and utility analysis.

AIMS: The UK Medicines and Healthcare products Regulatory Agency (MHRA) runs a national spontaneous reporting system (Yellow Card [YC] Scheme) to collect 'suspected' adverse drug reaction (ADR) data. We aim to describe the content and utility of YC reports received for patients aged <2 years. METHODS: Data on all ADRs reported using YC in infants aged <2 years from the years 2001-10 were supplied by the MHRA. RESULTS: For infants age <2 years, 3496 suspected ADRs were reported using YC (paternal medication pre-conception n = 3, transplacental n = 246, transmammary n = 30, neonates n = 97, in

Developmental outcome of school-age children born to mothers with heroin dependency: importance of environmental factors.

Development of children aged 5 to 12 years born to mothers with heroin dependency raised at home or adopted was studied in comparison with: (1) children with environmental deprivation alone (i.e. low parental socioeconomic status [SES] and evidence of neglect), (2) children born to fathers with heroin dependency fathers, and (3) control individuals of average SES. One hundred and sixty children (84 males and 76 females; average age at examination 8 years) were evaluated between 1998 and 1999. All were attending mainstream schools. All participants were examined by a paediatrician and a psychol

Transgenerational toxicity of tralomethrin in zebrafish: Parental exposure induces developmental defects in offspring.

Tralomethrin (TRA), a widely used type II pyrethroid insecticide, belongs to a class of pesticides frequently detected in aquatic environments, raising concerns about its potential chronic and transgenerational toxicity. This study aimed to evaluate the long-term effects of environmentally relevant concentrations of TRA (0.05, 0.5, and 5 μg/L) on adult zebrafish and their offspring. Adult zebrafish were exposed to TRA for 150 days, during which phenotypic observations, histological assessments, biochemical assays, transcriptomic profiling, and developmental evaluations of F1 progeny were condu

Birth defects among offspring of Norwegian farmers, 1967-1991.

We investigated birth defects (N = 4,565) reported to the Medical Birth Registry of Norway among 192,417 births between 1967 and 1991 to parents identified as farmers in five agricultural and horticultural censuses between 1969 and 1989. The prevalences at birth of all and specific birth defects deviated little from those among 61,351 births to non-farmers in agricultural municipalities. We classified exposure indicators on the basis of information provided at the agricultural censuses. The main hypotheses were that parental exposure to pesticides was associated with defects of the central ner

Predator Communities With Invasive Pike Are Associated With Changes in Parental Care and Brain Gene Expression in Wild Alaskan Stickleback.

Invasive predators can influence prey through direct predation but also via nonconsumptive effects that alter behavior and physiology. Parental care may be particularly sensitive to these effects, as parents must balance offspring investment with survival under predation risk. Here, we examine how predator exposure and predator community composition influence paternal behavior and brain gene expression in wild populations of threespined stickleback (Gasterosteus aculeatus) in southcentral Alaska. We conducted field behavioral assays in lakes with either native predators only or both native pre

Prescribed analgesics in pregnancy and risk of childhood asthma.

Many epidemiological studies have reported a positive association between prenatal exposure to paracetamol and childhood wheezing and asthma. We investigated whether the link between prenatal analgesic exposure and asthma/wheeze is specific to paracetamol, and whether it is causal or confounded.Using linked Swedish health register data we investigated the relation between various prescribed analgesics in pregnancy and the risk of childhood asthma/wheeze in a population of 492 999, and used negative paternal control and sibling comparison approaches to explore unmeasured confounding. After contr

Risk Factors for Birth Defects.

IMPORTANCE: Major congenital abnormalities, or birth defects, carry significant medical, surgical, cosmetic, or lifestyle consequences. Such abnormalities may be syndromic, involving multiple organ systems, or can be isolated. Overall, 2% to 4% of live births involve congenital abnormalities. Risk factors for birth defects are categorized as modifiable and nonmodifiable. Modifiable risk factors require thorough patient education/counseling. The strongest risk factors, such as age, family history, and a previously affected child, are usually nonmodifiable. OBJECTIVE: This review focuses on ris
