INTRODUCTION: In recent years, educators and clinicians have advocated moving away from race-based medicine toward race-conscious medicine. Yet, few studies have evaluated the impact of teaching preclerkship medical students skills to critically evaluate various frameworks for understanding the role of race in clinical decision-making. This educational innovation was designed to review various frameworks, prompt clinical evaluation, and evaluate how student perspectives were shaped by their learning.
METHODS: We delivered a 90-minute interactive session focused on recognizing, appraising, and

Little is known about how the initial endothelial plexus is remodelled into a mature and functioning vascular network. Studying postnatal remodelling of the retina vasculature, we show that a critical step in vascular maturation, namely pericyte recruitment, proceeds by outmigration of cells positive for (alpha)-smooth muscle actin from arterioles and that coverage of primary and smaller branches lags many days behind formation of the endothelial plexus. The transient existence of a pericyte-free endothelial plexus coincides temporally and spatially with the process of hyperoxia-induced vascul

BACKGROUND: Systemic sclerosis (SSc) is an autoimmune disease characterized by vascular injury and progressive fibrosis. Although microvascular injury is an inciting event and pericytes are recognized as a major source of myofibroblasts, the precise phenotypic heterogeneity of pericytes in the SSc microenvironment and the genetic mechanisms driving their pathological transition remain elusive.
METHODS: This study systematically explored the cellular and genetic basis of pericyte dysfunction by integrating single-cell RNA sequencing (scRNA-seq) data from SSc patients with genome-wide associati

Platelet-derived growth factor subunit B (PDGF-B) signaling plays a critical role in pericyte activation and neurovascular repair following ischemic injury. Although enhancing PDGF-B signaling in pericytes represents a promising therapeutic strategy for ischemic stroke, practical approaches have not yet been developed. In this study, we evaluated the therapeutic efficacy and underlying mechanisms of PDGF-B-modified heat shock protein nanoparticles (PDGF-B HSPNPs) in a transient middle cerebral artery occlusion mouse model of either sex. Mice were treated with single intravenous injection of PD

The ETS transcription factor Fli-1, known for regulating the vitality of pericyte in mice, has not been thoroughly investigated in traumatic brain injury (TBI). In this study, we used a mouse TBI model to demonstrate that Fli-1 expression in pericyte within the ipsilateral hippocampus is significantly increased following TBI and is associated with pericyte loss. Interfering with Fli-1 expression in pericyte disrupted their interactions with microglia, which in turn inhibited the transformation of microglia to a pro-inflammatory phenotype. Administration of Fli-1 siRNA via lateral ventricle inj

Oncogenic osteomalacia (OOM), also known as tumor-induced osteomalacia (TIO), is a rare paraneoplastic syndrome caused by excessive production of fibroblast growth factor 23 (FGF23) by mesenchymal tumors. This leads to renal phosphate wasting, hypophosphatemia, decreased 1,25-dihydroxy vitamin D levels, and impaired bone mineralization. Patients often present with progressive muscle weakness, diffuse bone pain, and recurrent fractures, with symptoms frequently preceding diagnosis by several years due to the non-specific nature of hypophosphatemia and its exclusion from routine laboratory panel

BACKGROUND: Glioblastoma (GBM) is the most common and lethal primary brain tumor characterized by extensive vascularization. Anti-angiogenic therapy for this cancer offers the possibility of universal efficacy. However, preclinical and clinical studies suggest that anti-VEGF drugs, such as Bevacizumab, actively promote tumor invasion, which ultimately leads to a therapy-resistant and recurrent phenotype of GBMs. Whether Bevacizumab can improve survival over chemotherapy alone remains debated. Herein, we emphasize the importance of small extracellular vesicles (sEVs) internalization by glioma s

Congestive heart failure produces fluid volume overload, central and renal venous pressure elevation, and consequently renal congestion, which results in worsening renal function. Pericyte detachment and pericyte-myofibroblast transition (PMT) were linked to renal interstitial fibrosis. Dahl salt-sensitive hypertensive (DahlS) rats are a non-surgical renal congestion model. The relation, however, between renal interstitial damage, pericyte morphology, and PMT in the renal congestion of DahlS rats has not been reported. DahlS rats (8-week-old) were fed normal salt (NS, 0.4% NaCl) or high salt (

PURPOSE: Choroidal neovascularizations (CNV) are partially stabilized through a coverage of pericytes leading to a partial anti-VEGF resistence. Drugs licensed for neovascular AMD (nAMD) do not take this mechanical and growth factor-driven CNV stability into account. The purpose of this work was to see if inhibiting the mammalian target of rapamycin (mTOR) may successfully block angiogenic cellular pathways in primary human retinal pericytes in an in vitro model of nAMD.
METHODS: The mTOR inhibitor rapamycin was used to treat human retinal pericytes (HRP) at doses ranging from 0.005 to 15 g/m

BACKGROUND: Chronic cerebral hypoperfusion (CCH) is the leading cause of cerebral small vessel disease (CSVD). CCH is strongly associated with blood-brain barrier (BBB) dysfunction and white matter lesions (WMLs) in CSVD. However, the effects of CCH on BBB integrity and components and the cellular and molecular mechanisms underlying the effects of BBB dysfunction remain elusive. Whether maintaining BBB integrity can reverse CCH-induced brain damage has also not been explored.
METHODS: In this study, we established a rat model of CSVD via permanent bilateral common carotid artery occlusion (2V

As a clinical manifestations of diabetic retinopathy (DR), pericytes (PCs) loss from the capillary walls is thought to be an initial pathological change responsible for the breakdown of the blood-retinal barrier (BRB). This study was performed to investigate the effects of ursodeoxycholic acid (UDCA) in PC depletion mice by injection of an antibody against platelet-derived growth factor reception-β (PDGFR-β clone APB5). To assess the integrity of the retinal vessels, their density, diameters, vessel branching points, and number of acellular capillaries were evaluated. While all types of retina

BACKGROUND: As a consequence of SARS-CoV-2 infection various neurocognitive and neuropsychiatric symptoms can appear, which may persist for several months post infection. However, cell type-specific routes of brain infection and underlying mechanisms resulting in neuroglial dysfunction are not well understood.
METHODS: Here, we investigated the susceptibility of cells constituting the blood-brain barrier (BBB) and the blood-cerebrospinal fluid barrier (BCSFB) of the choroid plexus (ChP) to SARS-CoV-2 infection using human induced pluripotent stem cell (hiPSC)-derived cellular models and a ChP

The specialized tightly controlled microcirculation of craniofacial neurosensory organs is an essential evolutionary adaptation and yet a dilemma where angiogenic remodeling occurs. Despite extreme plasticity of neurosensory structures, the capacity to reconcile barrier phenotype of the microcirculation with an angiogenic cascade is not known. Here we provide primary evidence for such a response in an elemental neurosensory structure, human dental pulp, following chronic carious insult. In response to hypoxic challenge neurosensory odontoblasts express hypoxia-inducible factor-1α and notch-1.

Arsenite is an environmental toxicant that is associated with vascular disease; however, the underlying mechanism of its toxicity has yet to be elucidated. Vascular stability appears to be tightly regulated by several vasoactive proteins produced by two adjacent vascular cells, endothelial cells (EC) and pericytes. The disruption of vascular stability may be involved in arsenite toxicity. The roles of angipoietins (Ang) and vascular endothelial growth factor (VEGF) in this process have been evaluated, but these studies have mostly been limited to EC. In this study, we used human brain microvas

Pericytes (PCs) are essential components of the retinal microvasculature and exhibit fibrotic activity, which is crucial for tissue repair and regeneration. However, chronic fibrotic activity can lead to harmful fibrosis that impairs retinal function. Treatment with faricimab, which targets angiopoietin-2 and vascular endothelial growth factor α (VEGFα) to prevent neovascularization in diabetic macular edema (DME), reduces epiretinal membrane (ERM) formation in DME, suggesting that faricimab has antifibrotic properties. Because PCs play an important role in fibrosis, we investigated the effect

Cerebral ischemic small vessel disease (SVD) is a leading cause of vascular dementia and stroke. Cerebral autosomal-dominant arteriopathy with subcortical infarcts and leukoencephalopathy (CADASIL), the most common monogenic SVD, is caused by dominant missense mutations in Notch3 expressed in smooth muscle cells (SMCs) and pericytes. However, cell-type-specific contributions driving CADASIL remain unknown. Here, we generate two conditional knockin mouse models carrying the CADASIL-causing Notch3R170C mutation in SMCs or pericytes. Both Notch3R170C models show perivascular accumulation of the N

BACKGROUND: In Alzheimer's disease (AD), neurovascular unit and blood-brain barrier (BBB) abnormalities are assessed using biologically distinct readouts, often across separate cohorts, complicating interpretation across clinical stages and biomarker domains.
METHODS: We searched major databases (inception to February 7, 2026) for human observational studies quantifying cerebrospinal fluid (CSF) soluble platelet-derived growth factor receptor-β (sPDGFRβ), dynamic contrast-enhanced magnetic resonance imaging (DCE-MRI) K trans, or the CSF/serum albumin quotient (QAlb) in cognitively normal (CN)

Lead (Pb), a persistent heavy metal, has caused widespread environmental contamination due to its extensive industrial use. Among its compounds, lead acetate (PbAc) is particularly hazardous in aqueous form, exhibiting neurotoxic potential. However, its toxic effects on the integrity of the blood-brain barrier (BBB) and the underlying mechanisms remain poorly understood. This study demonstrates that zebrafish embryos exposed to varying concentrations of PbAc exhibit dose-dependent morphological abnormalities, including growth retardation, delayed hatching, and cardiovascular dysfunction, along

Tissue engineering techniques for the regeneration of large bone defects require sufficient vascularisation of the applied constructs to ensure a sufficient supply of oxygen and nutrients. In our previous work, prevascularised 3D scaffolds have been successfully established by coculture of bone marrow derived stem cells (MSCs) and endothelial progenitor cells (EPCs). We identified stabilising pericytes (PCs) as part of newly formed capillary-like structures. In the present study, we report preliminary data on the interactions between MSCs and EPCs, leading to the differentiation of pericyte-li
