Effect of Nail Thickness on Visible Radiation Transmittance: Implications for New Photodynamic Therapy Technologies in Onychomycosis.

Photodynamic therapy is taking importance as a nonintrusive treatment for nail onychomycosis. Knowledge of true transmittance values across nails could lead to qualitative and quantitative improvements in light-based treatments. We have characterized the spectral transmittance of healthy and fungally infected human fingernails and toenails according to nail thickness, and we propose a surface transmittance model for the small-scale optimization of light-based treatments. Transmittance of fingernails and toenails was analyzed by means of spectroradiometric measurements under solar-simulated vis

Elucidation the binding interaction of hypericin-loaded P84 copolymeric micelles by using 1D and 2D NMR techniques.

Hypericin (HYP) is an active compound of Hypericum perforatum. Associated with photodynamic therapy (PDT), HYP has shown a broad therapeutic potential against microorganisms and cancer cells. Due to the low water solubility of HYP, its application in the biological medium becomes limited. To solve this limitation, our research group has been used copolymeric micelles to carrier HYP with high efficiency. However, there is no elucidated mechanism for HYP delivery mediated by copolymeric micelles. In this sense, we believed that the study of binding-sites of copolymeric micelles and HYP is the fi

Photodynamic therapy in dermatology: state-of-the-art.

Photodynamic therapy (PDT) has become an established treatment modality for dermatooncologic conditions like actinic keratosis, Bowen's disease, in situ squamous cell carcinoma and superficial basal cell carcinoma. There is also great promise of PDT for many non-neoplastic dermatological diseases like localized scleroderma, acne vulgaris, granuloma anulare and leishmaniasis. Aesthetic indications like photo-aged skin or sebaceous gland hyperplasia complete the range of applications. Major advantages of PDT are the low level of invasiveness and the excellent cosmetic results. Here, we review th

Recent advances in the prevention and treatment of skin cancer using photodynamic therapy.

Photodynamic therapy (PDT) is a noninvasive procedure that involves a photosensitizing drug and its subsequent activation by light to produce reactive oxygen species that specifically destroy target cells. Recently, PDT has been widely used in treating non-melanoma skin malignancies, the most common cancer in the USA, with superior cosmetic outcomes compared with conventional therapies. The topical 'photosensitizers' commonly used are 5-aminolevulinic acid (ALA) and its esterified derivative methyl 5-aminolevulinate, which are precursors of the endogenous photosensitizer protoporphyrin IX. Update on the current indications, practice and results of photodynamic therapy (PDT) in early central lung cancer (ECLC).

OBJECTIVES: The aim of this study was to investigate the practice and results of photodynamic therapy (PDT) in early central lung cancer (ECLC) based on the review of the literature and personal experience and to suggest the future role of PDT in such patients. MATERIAL AND METHODS: Literature search was made for articles on PDT in lung cancer. Only original articles with >10 patients with ECLC treated by PDT were selected. Data collection aimed at providing information on number of patient/lesions in the series, photosensitisers used, illumination method, mortality and morbidity, pathologica

Narrowband TL-01 phototherapy for patch-stage mycosis fungoides.

BACKGROUND: Although patch-stage mycosis fungoides (MF) has a generally good prognosis, and long-term survival rates with current therapies (UV-B, photochemotherapy, topical nitrogen mustards, electron-beam therapy) are similar, there is concern regarding their potential adverse effects. Narrowband or TL-01 UV-B phototherapy (311 nm), in use for more than 10 years, is more effective than broadband UV-B for the treatment of psoriasis, with an efficacy approaching that of psoralen UV-A. This open study assesses TL-01 as an alternative therapy for patch-stage MF. OBSERVATIONS: Eight white patien…

Uncaging Iridium(III) Complexes for Combined Photodynamic and Photoactivated Chemotherapy.

Inorganic photochemotherapy enables precise spatial and temporal control over anticancer drug activation, offering a promising strategy to reduce systemic toxicity. Two iridium(III) complexes of general formula [Ir(L)(ppy)(terpy)]PF6 with L = N-acetyl-L-cysteinato methyl ester (cys) or N-acetyl-L-selenocysteinato methyl ester (sec), were developed as light-activatable prodrugs with sulfur- and selenium-based "caging" groups. Both complexes are stable in the dark but undergo light induced bond cleavage upon visible light activation, following distinct photochemical pathways dictated by the coor…

Photodynamic therapy-induced inflammation and adverse effects: An updated review.

Photodynamic therapy (PDT) is a non-invasive medical treatment that uses a photosensitizing agent and light to treat various medical conditions and ophthalmology including chronic central serous chorioretinopathy, cancer, skin disorders, and infections. Local inflammation in tumor environment after PDT can be effective in eliciting an immune response to improve treatment efficiency but it causes some early and long-term side effects in local and surround tissues, resulting leads to incidental effects and unwilling consequences such as pain, erythema and infection. Moreover, the mechanism of in

D-A Structured High-Performance Photothermal/Photodynamic Thionin-Synthetic Melanin Nanoparticles for Rapid Bactericidal and Wound Healing Effects.

Synthesized melanin nanoparticles (SMNPs) are used as advanced photothermal materials. However, their internal structures are complex and disordered, and tuning the photothermal performance of nanoparticles is still a hot spot of concern. This article presents thionin (Th)-doped SMNPs, namely Th-SMNPs, which are the first SMNPs formed using the one-pot polymerization of Th with Levodopa. Th can undergo Michael addition and Schiff base reaction between indole dihydroxy/indolequinone and their oligomers to form donor-acceptor pairs in the structure to modulate the photothermal performance of SMN

Photodynamic therapy in dermatology--an update.

Topical photodynamic therapy (PDT) is a well-established treatment modality which has mainly shown to be effective for dermatooncologic conditions like actinic keratoses (AK), Bowen's disease, in situ squamous cell carcinoma and superficial basal cell carcinoma (BCC). However, a therapeutical benefit of PDT is also evident for inflammatory dermatoses like localized scleroderma, acne vulgaris and granuloma annulare. Recent work has been focused on the development and evaluation of topical photosensitizers like the heme precursor 5-aminolevulinic acid (5-ALA) or its methyl ester (methyl aminolev

Effects of porphyrins on skin.

Porphyrins and radiation induce activation of the complement system, release of mediators from mast cells, dermal accumulation of neutrophils, and activation of factor XII-dependent pathways. Recent studies have also shown that porphyrins and radiation have profound effects on the eicosanoid metabolism of mast cells, endothelial cells, macrophages and epidermal and dermal homogenates. In vitro, prostaglandin D2 (PGD2) was generated when mast cells were exposed to protoporphyrin and radiation. Exposure of cultured human umbilical vein endothelial cells or human omental microvascular endothelial

Concept of 2D van der Waals Nanohybrids for Key Biomedical Applications.

The past decade has witnessed a fundamental shift in 2D nanomaterials, from isolated single-component sheets to vdW nanohybrids, architected as stacked, stitched, or surface-engineered assemblies of chemically distinct layers. Enabled by weak interlayer forces, these hybrids permit modular integration of photonic, catalytic, electronic, and bioactive functions without lattice matching or harsh chemistries. In biomedicine, this modularity is transformative: one layer can absorb Near Infrared, NIR light for photothermal or photodynamic therapy, another can intercalate and release drugs or nuclei
